Stem cell is an ideal target cell with the ability of self renewing and multi differentiation, and can proliferate and keep multi differentiation potential when it is suitably cultivated in vitro. Marrow stem cell can differentiate into cardiomyocytes and vascular structures through transplanting into damaged heart of experimental animal, and ameliorate the function of the ischemia heart.
2.Significance of serum fibrotic indexes for the judgement of infarction area in acute myocardial infarction

Objective To investigate the relationship between the infarction area and the changes of serum fibrotic indexes in acute myocardial infarction (AMI) patients.Medthods Serum procollagen type Ⅲ (PCⅢ),collagen IV(IV?C) and laminin (LN) were measured in 42 AMI patients with successful recanalization,22 AMI patients with unsuccessful recanalization and 37 healthy controls.Michelle's method was used to calculate infarction area and correlative analysis was made between indexes and infarction area.Results Serum PCⅢ,IV.C and LN were significantly increased in AMI patients with unsuccessful recanalization as compared with the other two groups,which were positively correlated with the infarction area.However,there was no statistical significance between the indexes in the other two groups and infarction area.Conclusion Serum PCⅢ,IV.C and LN can be serum fibrotic indexes in AMI patients,which can reflect the infarction area indirectly.
3.The study of the role of blockade of CD40 ligand-CD40 costimulatory pathway on T lymphocytes typing and cytokines

Objective:To investigate the role of blockade of CD40 ligand-CD40 costimulatory pathway by anti-CD40L mAb on T lymphocytes typing and cytokines, and provide the experimental clues of inducing donor-specific T cell anergy.Methods:To monitor primary MLR, C57BL/6 H-2b spleen T cells were isolated as responder cells, and BALB/C H-2d spleen cell as stimulator cells. Anti-CD40L mAb was added to primary MLR cultures(termed anti-CD40L mAb group), and in control group(no anti-CD40L mAb). Incubated for 7 days, the cells responsiveness rates were detected by 3 H-TdR methods at the indicated time points, and supernatants were assayed for IFN-?,IL-2,IL-4,IL-10 cytokines levels by commercial ELISA kits. Day 5 MLR-cultured cells were assessed for the expressions of CD4, CD8, CD25, CD69, CD40L and CD45RA with flow cytometry(FCM).Cells proliferation for 5 days and cytokines in secondary MLR were assayed according above-mentioned method.Results:Blockade of the CD40-CD40L pathway by anti-CD40L mAb can induce the hyporesponsiveness to alloantigen in primary and secondary MLR culture. In primary MLR culture, the expressions of CD4 +T and CD8 +T cells and CD4 +CD25 +T, CD4 +CD69 +T, CD4 +CD40L +T, CD8 +CD25 +T and CD8 +CD69 +T cells in anti-CD40L mAb group were lower than that in control group ( P 0.05).The expression of CD4 +CD45RA + T in anti-CD40L mAb-cultured group was higher than that in control group( P
4.Long-term follow-up study of percutaneous transluminal septal myocardial ablation for hypertrophic obstructive cardiomyopath

36 mm?Hg. No permanent cardiac pacemaker was implanted. Transit trifascicular was seen in 3 cases, of whom, 2 disappeared with 1 hour after the procedure and, 1 trifascicular disappeared with 3 days. Permanent branch blocks were demonstrated in 36% patients. Conclusion PTSMA was feasible for HOCM with controllable complications. Precise position was the key point to prevent the alcohol leakage which will induce large area myocardial infarction and cardiac shock. Further study was needed to appreciate the more long-term follow-up.

6.Study of lesion characteristics and stenting for diseased grafts in older patients

Objective To determine the lesion characteristics and effect of intervention on diseased grafts in older patients. Methods Thirty-eight patients with angina after coronary artery bypass grafts (CABG) were enrolled into Nanjing First Hospital. Interventions were performed after multiple-view angiogram and formal consent from relatives received. Angiographic follow-up was conducted within 6 months. Cardiac events were recorded during the 6-month clinical follow-up. Results Of the 38 patients (36 males and 2 females), the average age was 63.2 years. Angina occurred 11.5 months after CABG. Angipgrams were performed 4.8 years after CABG. Diabetes was complicated in 10 cases and old myocardial infarction in 8, respectively. 7.Predicting cardiac events with myocardial fractional flow resever (FFRmyo) after coronary stenting

Objective To investigate the ability of myocardial fractional flow resever (FFRmyo) in predicting the incidence of major cardiac events (death, myocardial infarction, revascularization and recurrance of angina) 3 months after stent inplantation and also try to find out the cutoff point. Methods Coronary pressure was measured in 79 patients with stable angina immediately after stent implantation. The values of poststenting FFRmyo was calculated and found to be related to the presence of major adverse cardiac events during first 3 months. Results In 14 patients (18.4%), at least one adverse event occurred. Two out of 12 patients experienced recurrance of angina and underwent revascularization. By Logistic Regress analysis, FFRmyo immediately after stenting was the most significant independent variable for predicting major cardiac events. By receiver-operating characteristic (ROC) analysis, the cutoff point of FFRmyo was 0.92. Conclusion FFRmyo immediately after stenting is an independent predictor of clinical events at first 3 months.
8.Glycoprotein Ⅱb/Ⅲa receptor inhibitors (tirofiban) in percutaneous coronary intervention for elderly high-risk patients with acute coronary syndrome

Objective To investigate the efficacy and safety of glycoprotein Ⅱb/Ⅲa inhibitor (tirofiban) for elderly high-risk patients with acute coronary syndrome (ACS) undergoing percutaneous coronary interventional therapy (PCI). Methods Ninety four high-risk ACS patients admitted between Dec 2004 and Jul 2006 were randomly divided into the tirofiban group (n=46, mean age 68.5?11.4 years old) whom received tirofiban and half-dose of heparin, and the conventional interventional group (n=48, mean age 66.8?9.9 years old) whom were treated by full dose heparin before PCI. The coronary reperfusion flow (TIMI grades) of culprit vessel (CV) after PCI, cardiac enzyme changed 6 hrs and 12 hrs afterwards, major adverse cardiac events (MACE) rates within 30 days, bleeding and transfusion rates were analyzed and compared between the two groups. 9.Significance of coronary collaterals in patients with acute coronary syndrome

AIM: To study the relation between development of coronary collaterals and clinical outcome in patients with acute coronary syndrome(ACS).METHODS:251 patients with clinical diagnosed ACS were consecutively enrolled into the present study and divided into two groups according to the presence of coronary collaterals by quantitative coronary angiography(QCA).Serial venous blood samples were collected at admission for determining of high-sensitive C-reactive protein(hs-CRP),brain natriuretic peptide(BNP),and cardiac troponin I(cTnI).Baseline left ventricular ejection fraction(LVEF)was determined by cardiac echo.RESTULTS: Coronary collaterals were detected in 38 patients(as CC group),another 213 patients without coronary collaterals were served as control group.Serum levels of cTnI in patients in CC group was significantly higher than in control group((0.91)?(1.13) vs(0.29)?(0.23)(ng?ml~(-1)),P(0.05)).Serum levels of cTnI in patiemets of CC group was significantly lower than those of control group((0.29)?(0.23) vs(0.91)?(1.13)(ng?ml~(-1)),P
10.Inhibition of bosentan on cultured cardiomyocyte hypertrophy from neonatal mice

AIM: To observe the role of endothelin antagonist bosentan that inhibits cultured neonatal mice cardiomyocyte from endothelin-1 (ET-1) induced cell hypertrophy. METHODS: Digested by trypsin, neonatal mice cardiomyocytes were isolated and cultured in 4 groups. With a group of regular culture cells as control, other three groups were respectively treated by bosentan 10 -8 mol?L -1, ET-1 10 -10 mol?L -1 andET-1 10 -10 mol?L -1+bosentan 10 -8 mol?L -1 which were initially administered to the culture medium. The cell growing was observed carefully through micrography. 72 hours later, the level of atrial natriuretic peptide (ANP) and ET-1 in the media of the 4 groups were determined by method of radioimmunoassay. RESULTS: The values of ANP in different groups were: 6.46? 0.38 ?g?L -1 in control, 4.87? 0.56 ?g?L -1 in bosentan group, 13.60? 1.12 ?g?L -1 in ET-1 group and 9.42? 0.58 ?g?L -1 in ET-1+bosentan group. Also by micrographic observation, endothelin induced cell hypertrophy was proved, and the hypertrophy could be reversed by bosentan. CONCLUSION: Endothelin induces hypertrophy of cultured neonatal mice cardiomyocytes, but it can be inhibited by endothelin antagonist of bosentan.
