1.Hydrogen Dioxide Hypothermic Plasma Sterilizing System in Operating Room:A study of Application Cost

OBJECTIVE To explore the application of hydrogen dioxide hypothermic plasma sterilizing system(STERRAD 100S) in operating room,and study the cost.METHODS The cost of sterilizing endoscopic instruments in operating room was analyzed.And the method of using medical material was improved as follows: use the non-woven fabrics and transparent packing bags repeatly to increase the utilization efficiency of medical material.RESULTS Compared with the traditional method,the cost of sterilizing instruments by STERRAD 100S decreased 2-3 times,which brought much economic benefit.CONCLUSIONS 100S Can improve the working efficiency,reduce inventory level of endoscopic instruments and sterilizing cost,which can relief patient economic burden and ensure patient interest.
2.Pathogeny Analysis of Herpes Viral Encephalitis

Objective To probe into the relationship between the viral encephalitis and herpes viral infection, to understand the primary patheogeny of herpes viral encephalitis.Methods HSVⅠ-DNA,HSVⅡ-DNA,VZV-DNA,HCMV-DNA,EBV-DNA in CSF from viral encephalitis patients were detected by PCR.Results 82(15 6%) cases of 526 patiens with viral encephalitis were diagnosed as herpes simplex viral encephalitis, of which the age of high morbidity was at 0~10 years old children,9(1 7%) cases as cytomegaloviral encephalitis,7(1 7%) cases as varicella-zoster viral encephalitis and 4(0 7%) cases as EB viral encephalitis.Conclusions Herpes simplex viruses are the primary pathogeny in herpes viral encephalitis, PCR technique is valuable for the diagnosis of herpes viral encephalitis.
3.Effect evaluation and application of predictive nursing in nursing cooperation of living donor liver transplantation

Objective To investigate the effect of predictive nursing on the living donor liver transplantation.Methods Forty patients undergoing living donor liver transplantation were divided into the intervention group and the control group with 20 patients in each group randomly.We gave routine nursing cooperation to the control group while predictive nursing besides routine nursing cooperation to the intervention group.The operation time,cold ischemia time of donor liver,nursing complication,cost of surgery and satisfaction degree of surgery cooperation were recorded.Results Compared with the control group,the operation time,cold ischemia time of donor liver,nursing complication,cost of surgery were all decreased,and the satisfaction degree of surgery cooperation was improved.Conclusions Predictive nursing can improve nursing operation quality,raise the efficiency of nursing and surgery,reduce the incidence of complication,which is beneficial to smooth progress of operation and increase satisfaction degree of doctors and nurses to nursing cooperation.
4.Clinical features of microscopic colitis in the elderly and short-term effect of mesalamine on microscopic colitis

Objective To investigate clinical features of microscopic colitis (MC) in the elderly and short-term effect of mesalamine on MC.Methods Totally 116 elderly patients with MC in our hospital were recruited from January 2011 to December 2011.Clinical manifestations,use of MC related drugs (MCRDs),pathological classifications and short-term effect of mesalazine were collected and analyzed.Results MC patients accounted for 27.6 % in elderly patients with chronic diarrhea,who presented with mild to middle watery or mucous stool,and only 1.7% of MC patients accompanied with severe stomach pain.Collagenous colitis (CC) and lymphocytic enteritis (LC) accounted for 31.9% and 68.1%,respectively in MC patients and 50% of them had MCRDs taking history.There was a significant difference in short-term effective rate between CC group and LC group at 4 weeks after mesalazine treatment (64.9% vs.84.8%,x2 =5.929,P=0.015).The effective rate was significantly related with the relief of pathology in LC group (x2 =5.896,P=0.015).The relapse rate had no significant difference between CC group and LC group at 8 weeks after withdrawal of mesalazine treatment (45.8% vs.34.3 %,P =0.317).Conclusions MC may be one of the main reasons leading to chronic diarrhea in the elderly.Mesalazine has a better effect for the short-term treatment of MC in the elderly,but the long-term maintenance therapy still needs further study.
5.Effects of artificial bone particles in maxillary sinus augmentation in rabbits:An experimental study

Objective: To compare JGW with autogenous bone when used as grafting material in maxillary sinus floor augmentation.Methods: A study was conducted by performing a bilateral maxillary sinus floor augmentation with autogenous bone and JGW(Jin Gu Wei,Golden Bone,bone substitute,Shanghai Xiaobo Tec.) on rabbits.The process of bone formation was evaluated during different periods by imageology and histomorphology methods.Results: At 2nd week the gray scale of JGW was very lower than that of autogenous bone,but no significant difference was found at 12th week.The number of osteoblasts decreased with the time,no significant difference was found at 12th week.The degradation of JGW was relatively faster.Conclusion: JGW is feasible in maxillary sinus floor augmentation.
6.A clinical study of piezosurgery osteotomy in lateral sinus floor augmentation

Objective:To evaluate the clinical effect and safety of piezoelectric osteotomy in lateral sinus floor augmentation.Meth-ods:53 implants were placed in 30 sites after lateral sinus floor augmentation(LSFA)in 29 patients by piezoelectric osteotomy.47 implants were placed simultaneously with LSFA,6 were delayed.6-8 months after implantation,the final fixed prostheses were per-formed.The patients were followed-up at 3,6,and 12th months after restoration.Results:The mean residual bone height(RBH) was 4.46 mm before operation.The mean height of sinus elevation was 7.34 mm after operation.No membrane perforation occurred. Peri-implantitis,peri-implant mucositis and implant mobility were not found during follow-up.X-ray examination showed good os-seointegration in the interface between the implants and bones.The implant survival rate and the operation success rate were 100%. Conclusion:The application of piezosurgery is safe and reliable in lateral sinus floor augmentation.

8.Observation of the morphological changes of corneal nerve in experimental herpes simplex keratitis in rabbit eyes

Objective To observe the morphological changes of corneal nerve in experimental herpes simplex keratitis (HSK) in rabbit eyes. Methods Inoculation of the scarified cornea with herpes simplex virus-Ⅰ(HSV-Ⅰ)leads to herpetic infection of cornea. The HSK was detected by slit lamp and polymerase chain reaction(PCR), and the changes of corneal nerves in various periods with methylene blue vital staining and ultrastructure of corneal nerve were studied. Results Under light microscope, fewer corneal nerve bundles were observed, scattered with lower density of nerve fiber at center of cornea on the 4th day after the infection. At days 7 and 14, the seriously damaged nerve fiber was intermittent. The neuraxon became shorter with little polarity and the density of nerve fiber was extremely low. At day 30, the density was still low while the nerve fiber was approximately normal. Under electromicroscope, at day 4, the lamellar sheath of nerve fiber in the epitheliums appeared intermittent, and the neuroplasm of endings was partly lysed. During day 7 to day 14,neuroplasm was damaged and became vacuolar. The mitochondria swelled with vacuolar crest, and then were destroyed and lysed. The nerves in stroma were also injured. On day 30, neurilemma sheath was still intermittent with the decrease of microfilament and racuole. Conclusion HSK leads to the damage of nerve and the decrease of nerval density, and the damage of corneal nerve repairs very slowly.
9.Temporal Pattern of DNA Breaks in Human Ovarian Cancer Cells after Exposure to Nanosecond Electric Pulses.

This study aims to explore the temporal pattern of DNA breaks induced by nanosecond electric pulses (nsEP) in cisplatin-sensitive and cisplatin-resistant human ovarian cancer cells. Human ovarian cancer cells A2780 (cisplatin-sensitive subline) and C30 (cisplatin-resistant subline) were exposed to nsEP. Sham exposed groups were shame exposed to nsEP. Cell viability was determined using CCK-8 assay after 0 h, 4 h, 8 h, 12 h and 24 h, respectively, and the percentage of dead cells was calculated. The DNA break was detected with the alkaline single cell gel electrophoresis (comet assay), and the 75th percentiles of TL (tail length), TM (tail moment) and OTM (Olive tail moment) were measured. Cell viability displayed an early decrease and late increase, with the valley value seen at 8 h. Percentages of cell death and comet-formed in A2780 cells were higher than those in C30 cells (P < 0.05) at 8 h, respectively. TL, TM and OTM in C30 cells were less than those in A2780 cells (P < 0.05). The percentage of comet-formed correlated with that of cell death in either A2780 (r = 0.997, P < 0.05) or C30 (r = 0.998, P < 0.05) cells. DNA breaks induced by nsEP in cisplatin-sensitive cells differred from that in resistant cells, and DNA break resulted in fraction of cell death.

10.Expression of Blimp1 mRNA in the PBMCs of patients with RA and the corresponding regulation of IL-21

Objective To Investigate the expression of IL-21 and Blimp1 mRNA in Rheumatoid arthritis ( RA) patients and the influence on the expression of Blimp 1 in peripheral blood mononuclear cells (PBMCs) of RA patients after IL-21 stimulated; To further explore the mechanism of IL-21 and blimp1 in the pathogenesis of RA.Methods Case control study.The samples of peripheral venous blood from 50 RA patients of department of rheumatology of The First Affiliated Hospital of Zhengzhou University and 50 healthy people were collected respectively , then the plasma and PBMCs was separated.IL-21 in plasma was measured by ELISA; the correlation between patients clinical index DAS 28, anti-CCP antibody and IL-21 was analyzed.Blimp1 mRNA of patients′PBMCs was detected by qPCR; PMBCs were isolated from RA patient and then cultured in vitro.Blimp1 mRNA level was measured by qPCR and the ratio of CD 20 positive B cell and the ratio of CD138 positive cells in all groups were detected by flow cytometry after 72 h stimulated by IL-21 and CD40L.Results IL-21 content in RA patient blood plasma (130.51 ±11.35)ng/L was significantly higher than that in healthy control (25.46 ±6.05)ng/L, t=5.39,P<0.05.Besides, IL-21 level also had a close relativity with patients DAS28(r=0.658) and anti-CCP antibody (r=0.674, P=0.039 and 0.035).In addition, the expression level of Blimp1 mRNA in RA patient PMBCs (1.321 ± 0.11)was higher than that in healthy control group (1.000 ±0.000), Z=-2.48, P<0.05.While after IL-21 and/or CD40L stimulation, Blimp1 mRNA of IL-21 group and CD40L+IL-21 group(1.084 ±0.029, 1.157 ±0.028)were higher than those of control (1.000 ±0.000)(P=0.002,P=0.001), moreover the expressive level of Blimp1mRNA of CD40L+IL-21 group was higher than that of control group (t=4.862, P=0.02).Compared to negative control group , the ratio of CD20 positive B cells [2.42 ±0.35, 2.63 ± 0.33, 6.35(4.85,6.57),F=278.363,P<0.001] and the ratio of CD138 positive cells(0.474 ±0.110, 0.668 ±0.120, 0.955 ±0.170,F=49.01, P<0.001) in CD40L group, IL-21 group and CD40L+IL-21 group were much higher and the differences among CD 40L+IL-21 group with CD40L group and IL-21 group were statistically significant.Conclusion IL-21 could promote the level of Blimp 1 mRNA in peripheral blood mononuclear cells in RA patient; IL-21 and CD40L could co-promote B cell maturation though regulating Blimp1 mRNA expression and eventually participate in RA pathogenesis.
