Objective To investigate the expression patterns of major bone growth factors and bone morphogenetic protein-2 (BMP2) in the distracted calluses following transfected gene during mandibular distraction osteogenesis in a rabbit model. Methods Bilateral mandibular osteotomies were performed in New-Zeland rabbits. After a latency of 3 days, the mandibles were elongated using distractors for 7 days. After the completion of distraction, the rabbits were randomly divided into 5 groups.Three animals each were sacrificed at the end of the delay phase,at 7,14,and 28 days after completion of distraction, respectively. The lengthened mandibles were harvested and processed for immunohistochemical detection of BMP2 expression, the mean optic densities and integral optical density of BMP2 positive cells were measured by computerized image analyzer. Results Elevated cellular expression of BMP2,in the distraction gap,was observed following mandibular distraction.BMP2 staining was mainly located in inflammatory cells, and the connective tissues arrounding the new bone.Their strongest expression was the 7th day, some of those growth factors expressed weakly or negatively. Conclusions Electroporation-mediated gene transfection can promote BMP2 expression effectively, which plays an important role in cell differentiation and proliferation during distraction osteo genesis. The BMP2 stimulates extracellular matrix synthesis, induces the proliferation and differentiation of fibroblasts and osteoblasts, which then promotes the new bone formation and repair.

An effective RT-PCR method was developed to detect exogenous gene xylB transcript levels in Zymomonas mobilis CP4. Total RNAs without genomic DNA contamination were purified from wild-type and gene engineering strains, and were quantified to the same concentration. Then, cDNAs synthesis and PCR analysis of these samples were conducted by reverse transcription PCR. The optimal number of cycles was determined by observing amplification profile of target gene xylB and internal control gene 16s rRNA, and relative expression levels of xylB in various samples were analyzed by RT-PCR. The results indicated that the xylB transcript was not be detected in CP4, however that could be found in recombinant strains, in which xylB transcription abundance was similar. The enzyme assay furthermore confirmed that effective ex-pression of the target gene. The method provided a useful and rapid tool for detecting transcript levels of target genes from various samples of Z. mobilis.

Objective:To explore the influence of different emotional states on college students' attention control ability in single and dual attention control tasks. Methods:A total of 28 college students from Xinjiang Normal University were selected as participants, and emotional pictures were used to activate the positive, negative and neutral emotional states of the participants, and the integrated paradigm of " double-choice Oddball+ Stroop" was used to investigate their attention control ability in different emotional states. The experiment was designed by 3 (emotional states: neutral mood, positive emotion and negative emotion) × 4 (task types: baseline task, single response inhibition, single interference inhibition and double attention control task), and the E-prime software was used to record the participant's reaction time and error rate. SPSS 21.0 software was used to perform repeated measurement ANOVA on the reaction time and error rate of 28 participants. Results:The interaction between emotional state and task type was significant( F(6, 22)=2.70, P<0.05). The main effect of emotional state was not significant ( F(2, 26)=0.79, P=0.461). The main effect of task type was significant ( F(3, 25)=28.46, P<0.001). The differences in average reaction time between baseline tasks, single interference suppression tasks, single response suppression tasks, and dual attention control tasks were statistically significant (all P<0.001). Simple effect analysis showed that in the single response inhibition task, the reaction time in positive emotional state ((499.62±72.09) ms) and negative emotional state ((521.52±86.39) ms) were significantly longer than that in the neutral emotional state((475.43±76.38) ms, both P<0.05), and that in the negative emotional state was significantly longer than that in the positive emotional state ( P<0.05). There was no significant difference between emotional states in the baseline task, single interference suppression task, and double attention control task (all P>0.05). Conclusion:In the single attention control task, both positive and negative emotional states have emotional interference effect on the response inhibition ability of college students, and the interference effect of negative emotional state is more sighificant. In the dual attention control task, the emotional interference effect disappeared.

OBJECTIVE:
To compare the clinical application and health economic values of non-invasive prenatal testing (NIPT) and second trimester serum screening (STSS).
METHODS:
A retrospective analysis was carried out on 54 026 singleton pregnant women undergoing NIPT and STSS from March 1, 2018 to December 31, 2019 in Changsha Maternal and Child Health Care Hospital. For pregnant women with high-risk results of NIPT, prenatal diagnosis and follow-up of pregnancy outcomes were conducted. The data was grouped to 4 screening models, and their cost-benefit was analyzed.
RESULTS:
The sensitivity, specificity and positive predictive value of NIPT were all higher than STSS. Screening models 1 to 4 have prevented the birth of 71, 29, 52 and 54 patients with Down syndrome, respectively. The safety index of screening models 1 to 4 were 0.0036, 0.3944, 02215 and 0.1281, respectively. When the price of NIPT was decreased to 600 RMB, the cost-benefit of the screening models 1 to 4 was 0.46, 0.65, 0.44 and 0.40 million RMB, respectively.
CONCLUSION
NIPT has a better detection performance than STSS. When the price of NIPT is 600 RMB, screening model 1 has the best screening effect and the highest accuracy, safety index and health economical value.

Journal of Xi'an Jiaotong University(Medical Sciences) 2021;42(6):935-940
【Objective】 To investigate the effects and potential mechanisms of neurodegenerative lesions in male mice caused by ozone exposure. 【Methods】 We divided 23 C57BL/6N male mice aged 8 to 9 months into control group (clean air group, 11) and ozone group (1 mg/m 3, 4h/d, 12). After 8 weeks of continuous ozone exposure, the Morris water maze experiment was used to detect the mice’s learning and memory ability, HE dyeing to observe pathological changes in hippocampal tissue cells, and immunoprinting tests to detect the expression levels of Tau, p-Tau and α-synuclein proteins in the cerebral cortex tissue. 【Results】 After 8 weeks of ozone exposure, the mice’s spatial learning and memory ability were impaired to a certain extent, the incubation period decreased with time, and the two lines were separated, but the difference was not statistically significant. Ozone exposure caused changes in the morphology of the mice’s hippocampal tissue cells, disorders in the arrangement of hippocampal neuron, and nuclear wrinkles, and significantly increased levels of p-Tau and α-synuclein protein expressions in cerebral cortex tissues (P<0.01), but there was no statistical significance in the total Tau expression level. 【Conclusion】 Ozone exposure leads to the loss of learning and memory in mice, changes in hippocampal neurocellular pathology, and increased expression levels of neurodegenerative variable-related proteins.
