[Objective] To observe the effect of Pueraria Lobata isoflavones (PLI) on neuropeptide-Y-induced (NPY) vascular smooth muscle cell (VSMC) proliferation. [Methods] Experiment system for VSMC was set up as 4 groups: control group (with DME culture fluid, free of blood serum; group A), model group (with NPY culture fluid; group B), control + PLI group (with PLI culture fluid; group C) and model + PLI group (with NPY and PLI culture fluid; group E). Quantitative fluoroimmunohistochemistry techniques were applied to examine the mean fluorescent values of proliferating cell nuclear antigen (PCNA), platelet derived growth factor (PDGF) and C-myc expression. [Results] The levels of PCNA, PDGF and C-myc expression in model group were higher than control group (P

AIM: To investigate the effects of pueraria lobata isoflavones (PLIs) on neuropeptide-Y(NPY)-induced myocardiac cell hypertrophy in vitro. METHODS: Rat cardiomyocytes cultured in vitro were randomized to control, NPY, NPY+PLIs and PLIs group. The protein synthesis in cardiomyocytes was assessed by [~3H-Leu] uptake, C-jun mRNA by RT-PCR and CaN activity by histochemistry. RESULTS: The rate of [~3H-Leu] uptake, the C-jun mRNA expression and the CaN activity of myocardiac cells in 100 nmol/L NPY group were higher than those in control (P

Duck circovirus(DuCV),a potential immunosuppressive virus, was investigated in Southern China from March 2006 to December 2009 by using a polymerase chain reaction(PCR)based method. In this study,a total of 138 sick or dead duck samples from 18 different farms were examined with an average DuCV infection rate of~35%. It was found that ducks between the ages of 40~60 days were more susceptible to DuCV. There was no evidence showing that the DuCV virus was capable of vertical transmission. Farms with positive PCR results exhibited no regularly apparent clinical abnormalities such as feathering disorders, growth retardation or lower-than-average weight. The complete genomes of 9. strains from Fujian Province and 1 from Zhejiang Province were sequenced and analyzed. The 10 DuCV genomes, compared with others genomes downloaded from GenBank, ranged in size from 1988 to 1996 base pairs, with sequence identities ranging from 83.2% to 99.8%. Phylogenetic analysis based on genome sequences demonstrated that DuCVs can be divided into two distinct genetic genotypes,Group I(the Euro-USA lineage)and Group II(the Taiwan lineage),with approximately 10.0% genetic difference between the two types. Molecular epidemiological data suggest there is no obvious difference among DuCV strains isolated from different geographic locations or different species, including Duck,Muscovy duck,Mule duck,Cheery duck,Mulard duck and Pekin duck.

Objective To observe the efficacy and safety of liver decanoic sulfonic sodium in the treatment of elderly patients with acute coronary syndrome (ACS).Methods84 ACS patients over 70 years were randomly divided into the treatment and control groups. Two groups were treated on the basis of general, treatment 40 cases of liver of dibutyl sebacate with sodium sulfonated added 2.5 mg subcutaneous injection parumbilical, day 1,course 8 d ; control group 44 cases combined with low molecular weight heparin calcium 0.4 ml,2 times/day 1 time every 12 hours, according to body weight adjusted dose,Parumbilical shot in course of 8 days, observing the clinical effect of two groups during the treatment,as well as 4 weeks and bleeding during the treatment the incidence of cardiovascular events. Results The clinical effects of treatment and control groups was no significant difference (95% and 91%,P >0.05) ; None of the two sets of death, myocardial infarction and recurrent malignant ventricular arrhythmia; none of the two sets of bleeding occurs, treatment of minor bleeding rates were significantly lower than those of control groups (7.5% and 25%,P < 0.05).Conclusion The study of dibutyl sebacate with sodium heparin and low molecular weight heparin calcium in the treatment of ACS are valid, but the former which significantly reduce the incidence of minor bleeding.Liver decanoic sulfonic sodium for acute coronary syndrome ACS)anticoagulation can not only effectively reduce cardiovascular events, but also significantly reduce bleeding risk, regardless of the patient's age, gender, renal function and risk stratification, especially for bleeding in high risk elderly patients with safe and clinical promotion.

This study examined the change of p16(INK4a) and PNCA protein expression in myocardium after injection of hIGF-1 gene modified skeletal myoblasts into post-infarction rats. HIGF-1 gene modified skeletal myoblasts (hIGF-1-myoblasts) were injected into hind limb muscles of 18 post-infraction rats (experimental group). Primary-myoblasts were injected into 18 post-infraction rats (control group) and 12 non-infarction rats (sham group). Expression of p16(INK4a) and PCNA protein in myocardiums were separately detected immunocytochemically 1, 2 and 4 weeks after the inuection. The level of hIGF-1 and rIGF-1 protein in serum and myocardium were detected by enzyme-linked immunosorbent assay (ELISA). Compared with the sham group, the percentage of p16(INK4a) and PCNA positive cells reached a peak after 1 week in the control group and the experimental group (P<0.01). Moreover, the percentage of p16(INK4a)-positive cells in the experimental group was lower than in control group whereas the percentage of PCNA-positive cells was lower in the control group than in the experimental group (P<0.01). The percentage of p16(INK4a)-positive cells in the experimental group and the percentage of PCNA-positive cells in the control group were close to that in the sham group from the 2nd week (P>0.05). ELISA analysis disclosed that the myocardium level of rIGF-1 protein increased gradually in the controls and especially in the experimental group (P<0.01). The serum level of rIGF-1 decreased significantly in post-infraction rats, but these conditions were improved in the experimental group (P<0.01). The hIGF-1 protein in serum and myocardium were detected from the 1st week to the 4th week in the experimental group. Statistical analysis revealed significant associations of myocardium level of hIGF-1 protein with expression of p16(INK4a) and PCNA protein (r=-0.323, P<0.05; r=0.647, P<0.01). It is concluded that genetically hIGF-1-myoblast provides a means for constant synthesis and release of hIGF-1. It could not only improve the expression of rIGF-1 and PCNA protein in myocardium, but also suppress the expression of p16(INK4a) protein for 30 days in post-infraction rats. Myoblasts-mediated IGF-1 gene therapy may provide a new alternative for the clinical treatment of heart failure.
