1.Effects of the total extract of astragalosides on the growth, AFP secretion and γ-GT activity of human hepatoma cells

AimTo study the effect of the total extract of astragalosides(TEA) on the growth, AFP secretion and γ-GT activity of human hepatoma cells invitro.Methods The human hepatoma HepG2 and Bel-7404 cells were cultured with TEA 5~160 mg · L-1 for 6 days. Anti-hepatoma activity was measured by MTT method. AFP was assaysed by ELISA and γ-GT was determined by enzyme-substrate method. Results The growth of the human hepatoma HepG2 and Bel-7404 cells and the secretion of AFP of HepG2 cells were reduced significantly by TEA 5~160 mg· L-1. The γ-GT activity of HepG2 cells was inhibited and its ALB content was increased by TEA 20, 40 and 80 mg · L-1. The γ-GT activity of Bel-7404 cell was inhibited and its ALB content was increased byTEA 40 and 80 mg · L-1 Conclusion These results suggest that TEA has inhibitory effects on the growth, AFP secretion and γ-GT activity of human hepatoma cells.
2.Effects of total extract of astragalus(TEA) on the proliferation and the production of collagen of hepatic stellate cells

Aim To study the effects of total extract of astragalus(TEA) on activating the primary cultured hepatic stellate cells(HSC) and synthesizing collagen. Methods Livers of adult rats were perfused by pronase E and collagenase in situ, HSC and Kupffer cell (KC) were isolated by centrifugation with 18% Nycodenz. The subcultured HSC were induced by Kupffer cell conditioned medium(KCCM) and were incubated with 5~ 80 mg?L -1 TEA for 6 days. The proliferation of the cells was measured by 3H TdR and the production of collagen by 3H Proline.Results TEA (10~ 40 mg?L -1 ) could suppress the proliferation of hepatic stellate cells and TEA(40~ 80 mg?L -1 ) could suppress the production of collagen. Conlusion The suppression of hepatic stellate cell proliferation and the collagen's production by the TEA may be one of the mechanisms for depressing the hepatic fibrosis by the TEA.
3.Monitoring of Serum Phenytoin Concentration in 130 Cases and Analysis of the Cases of Intoxication

OBJECTIVE: To study the relationship among the dosage, serum concentration and clinical effect of phenytoin(PHT) in treating epileptic patients .METHODS: 130 steady -- state serum concentrations of PHT were determined by fluores-cence polarization immunoassay(FPIA) in 70 patients with or without other drugs used in combination .RESULTS: The indi-vidual difference among the patients who received routine dosage of PHT was very significant. Intoxication occurred ex-tremely often in patients whose PHT serum concentrations were over 30?g/ml .CONCLUSION: It is necessary to perform TDM(Therapeutic Drug Monitoring) and individualize the use of PHT,so that the safe and effective use of drug can be ensured.
4.Study on the Adsorption of Disposable Transfusion Connective Tube and Infusion Needle for Single Use only to Nitroglycerin

OBJECTIVE:To study the adsorption of disposable transfusion connective tube and infusion needle for single use only to nitroglycerin in intravenous injection via minipump.METHODS:The intravenous injection via minipump was im-itated;the concentration change of nitroglycerin during the injection process was determined by the high performance liquid chromatography(HPLC)and ultraviolet spectrophotometry.RESULTS:Disposable transfusion connective tube and infusion needle for single use only had significant adsorption to nitroglycerin with an mean adsorption rate at(73.88?2.05)%within8hours.CONCLUSION:It is unsuitable to use those disposable transfusion connective tubes and infusion needles for single use only that have strong adsorption to nitroglycerin in the intravenous injection of nitroglycerin via minipump.
5.Quality Investigation of Compound Danshen Injection from Different Manufactures

OBEJECTIVE:To study quality differences of compound danshen injections of different batch numbers from different manufactures.METHODS:The fingerprints of compound danshen injections were obtained through gradient elution by HPLC;The contents of danshensu,protocatechualdehyde and savianolic acid B in samples of different batch numbers and different manufactures were determined,and the insoluble fine particle,protein,tannin,resin etc.in samples were studied ac-cording to the specification of China Pharmacopoeia2005.RESULTS:The fingerprints of compound danshen injections were specific;Great differences were found in the contents of danshensu,protocatechualdehyde and salvianolic acid B of the samples from different manufactures,quality discrepancies were found between some of the related substances in some of the prepara-tions and the specifications.CONCLUSION:Quality differences were found in compound danshen injections from different manufactures,therefore,the quality control standards of compound danshen injections should be enhanced and unified,and with regard to the controlling of the insoluble fine particles,the technics and techniques of which should be improved and given a further study.
6.Clinical Observation on Regulating Effect of Pravastatin on Blood Lipid and Blood Pressure in Patients with Coronary Heart Disease

OBJECTIVE:To explore the regulating effect of pravastatin on hyperlipidemia and hypertension in patients with coronary heart disease.METHODS:80 patients with coronary heart disease were randomly divided into 2 groups,pravastatin treatment group(pravastatin,20 mg,qd,n=42)and placebo control group(placebo,qd,n=38).The course of treatment was 4 weeks in both groups,serum total cholesterol(TC),triglycerides(TG),high density lipoprotein cholesterol(HDL-C)were assayed,and systolic blood pressure(SBP)as well as diastolic blood pressure(DBP)were measured before and after 4 weeks of treatment.RESULTS:There was a significant decrease of serum TC and TG levels,as well as DBP in treatment group after treatment,as compared with control group,while HDL-C level increased significantly(P
7.Detection of Serum Benzodiazepines Concentrations in 149 Cases of Unconfirmed Drug Overdose

OBJECTIVE:To find the quick detection method for serum benzodiazepines concentrations.METHODS:Using flurosence polarization immunoassay method to detect the serum concentration of benzodiazepines(BZDs)in149patients who were suspicious of receiving drug overdose in emergency unit.RESULTS:We got positive results in104samples from149pa?tients,but the levels of BZDs concentration were not coincident with the state of drug overdose.CONCLUSION:The method used in our report is a quick and useful assay for patients with unconfirmed drug overdose in emergency unit,but caution should be taken in the result interpretation to doctors.

Objective To establish a sandwich enzyme-linked immunosorbent (ELISA) method for early diagnosis of hepatitis C. Methods Immunization of Balb/c mice with hepatitis C core antigen were prepared by genetic engineering. Mouse monoclonal antibodies (McAb) to anti-HCV-core Ag were obtained. A sandwich ELISA kit for detecting HCV-core Ag was developed by using four strains of anti-HCV-core Ag McAb. One hundred and twenty five serum specimens with increased ALT but negative for anti-HCV, anti-HIV, and anti-Tp tests were tested. Results Nine of the 125 specimens were positive for HCV-core Ag. Conclusion The double sandwich ELISA kits for detecting HCV-core Ag may be useful for the early diagnosis of hepatitis C .
9.Inhibition of MicroRNA-92a Improved Erectile Dysfunction in Streptozotocin-Induced Diabetic Rats via Suppressing Oxidative Stress and Endothelial Dysfunction

Purpose:
To determine whether microRNA could be a therapy target of erectile dysfunction (ED) and the underlying mechanisms.
Materials and Methods:
Eight-week-old fasting male SD rats were intraperitoneally injected with streptozotocin to construct diabetic rat models. Diabetic ED rats were treated with miRNA-92a inhibitor. The cavernous nerves were electrically stimulated to measure the intracavernous pressure and mean arterial pressure of rats in each group. After the detection, the penile cavernous tissues are properly stored for subsequent experiments. Rat aortic endothelial cells were used in in vitro studies.
Results:
The expression of miR-92a was significantly increased in the corpus cavernosum of Streptozocin (STZ)-induced diabetic rats and injection of miR-92a antagomir into the corpus cavernosum of diabetic rats significantly increased eNOS/NO/ cGMP signaling pathway activities, cavernous endothelial cell proliferation, endothelial cell-cell junction protein expression and decreased the levels of oxidative stress. These changes restored erectile function in STZ-induced diabetic rats. Moreover, in vitro study demonstrated that the miR-92a expression increased significantly in endothelial cells treated with high glucose, inhibiting AMPK/eNOS and AMPK/Nrf2/HO-1 signaling pathways in rat aortic endothelial cells via targeting Prkaa2, causing endothelial dysfunction and overactive oxidative stress, miR-92a inhibitor can improve the above parameters.
Conclusions
miRNA-92a inhibitor could exert an inhibition role on oxidative stress and endothelial dysfunction to improve diabetic ED effectively.
10.Clinical significance of standardized pathological examination of specimens for en bloc transurethral resection technique with Hybrid Knife to treat NMIBC

Objective To evaluate the pathological stage,the presence of detrusor muscle and the clinical significance for standardized examination of specimens for en bloc transurethral resection technique with Hybrid Knife to treat NMIBC (ERBT) compared with conventional TURBT.Methods This was prospective randomized controlled study.This study was approved by the Ethics Committee of Tongji Hospital,Tongji Medical College of Huazhong University of Science and Technology (TJ-IRB20180604),and patients all signed informed consent.The clinical study registration number of this study:NCT03221062.The margin of the tumor was recognized and marked by with Hybrid Knife (0.5 cm away from the normal mucosa).Then water was injected into the submucosa and form a water pad,with a circular cutting layer by layer (0.5 cm away from the marked position),reaching the detrusor muscle in depth.After complete resection,the tumor was removed by specimen bag.Specimens for ERBT cohort were given standard handling.Resected specimen of ERBT stretched with pins on foam and its margin was stained.The basement of specimen was also stained.Total specimen sectioned into appropriate pieces for histological assessment in the department of pathology.TURBT cohort performed traditional surgical methods and pathological examination.All patients received postoperative intravesical instillation according to their pathology.Imaging and cystoscopy were performed every 3 months.The primary study end-point was the quality of resection,including the pathological stage and the presence of DM.Secondary outcomes were:short-term tumour recurrence rate (18 month),feasibility,and safety.Results From January 2017 to October 2017,109 patients were enrolled.51 patients underwent ERBT,and 58 patients underwent TURBT.The clinical characteristics of the patients in each cohort,such as average age,gender,average BMI,smoking history,the mean number of lesions and tumour size had no significant differences (P > 0.05).The operation of 109 cases was completed successfully.There was no statistical difference between the operative time and the postoperative bladder irrigation time.Major intraoperative or postoperative complications (Clavien ≥ Ⅱ) did not occur in all of the patients.The percentage of T1 staging was higher in the ERBT cohort vs.TURBT cohort [21/51 (41.2%) vs.13/58 (22.4%),P =0.035],of which ERBT cohort accurately detected 9 cases (42.8%) of T1b patients,significantly higher than TURBT cohort (2 cases,15.4%) (P =0.096).All the ERBT samples showed the presence of DM (100.0%),while there was only 77.4% in TURBT cohort (P < 0.05).Mean follow-up (20.3 ± 3.1) months (ranged from 18 to 24 months).Recurrence rate were 8.9% (4/45) in ERBT cohort vs.22.2% (12/54) in TURBT cohort (P=0.059).Conclusions ERBT with Hybrid Knife for treatment NMIBC is a safe,effective,and provides high-quality specimens compared to TURBT.More high-risk NMIBC patients,especially T1 b patients,can be detected obviously by pathologist with the standardized treatment of specimens.
