1.Effect of reconstruction on medial collateral ligament healing of knee in rabbits

Objective To provide histological evidence for clinic tendon reconstruction on severe medial collateral ligament injury in rabbits. Methods Medial collateral ligaments of two knee joints were completely incised to create own control model of MCL rupture. The operation group intimated the procedure of human being MCL reconstruction with semitendinous tendon. In the 4th,12th,16th,24th weeks, healing of MCL was observed by anatomy and the picrosirius-polarization method. Results In the 4th week, the MCL healing tissue of ex-periment group is more than control group. The average ratios of type Ⅲ collagen to Ⅰ, Ⅲ collagen in the experiment group [(0.263±0.075), (0.235±0.041), (0.210±0.045), (0.197±0.029)] were significantly lower than those in control group [(0.310±0.072), (0.286±0.045), (0.261±0.046), (0.236±0.043) ] in the 4th, 12th, 16th, 24th weeks, respectively (P < 0.05). Conclusions The semitendinons tendon reconstruction can improve MCL healing in early and middle period after injury.
2.A Study on the Relationship between Fatty Liver and Body Mass Index (BMI), Serum Lipid Levels in Old Retired Population

Objective To explore the relationship between fatty liver and body mass index (BMI), serum lipid levels in aging population. Method 748 of retired people (≥45 years) were studied. All participants accepted liver B -mode Ultrasonography examination, height, weight and blood lipid detection. The relationship between Fatty Liver and BMI, serum lipid levels were analyzed. Results The detection rate of Fatter Liver in male and female was 61.8% and 44.6% separately. The detection rate of Fatter Liver was 53.4% in the aging group (45~59 years), while the rate was 51.0% in elderly group (≥60 years). Compared to the 23.07 kg / m2 and 1.58 mmol/L in group without fatty liver, the BMI and serum triglyceride was 26.77 kg / m2 and 2.13 mmol / L in the group suffered the Fatter Lever. Objective To observe the safety and effectiveness of high-dose chemotherapy(HDT) supported by autologous peripheral blood hematopoietic stem cells transplantation(APBHSCT) in the patients of terminal or relapsed malignant tumors.Methods The patients achieved CR or PR after 4 cycles of conventional salvage chemotherapy were divided into two groups randomly.26 patients in the research group were adapted APBHSC+HDC,50 patients in the control group were adapted 2~4 cycles of routine chemotherapy.Results In the research group,hematopoiesis was reconstructed in all patients,8 out of 17 patients who achieved PR after 4 cycles of conventional salvage chemotherapy turned into CR after APBHSC+HDC(CR rate 47.1%).In the control group,33 patients who achieved PR after 4 cycles of conventional salvage chemotherapy were still PR after 2~4 cycles of routine chemotherapy.The median survival period in the research group was 11 months,which was longer than the one in the control group.The survival rates of 2,3 and 4 years in the research group were evidently higher than the ones in the control group(P
4.Mismatch negativity in chronic tension-type headache with and without medication-overuse

Background: It is unknown if medication-overuse headache, clinically similar to chronic tension-type
headache, has pre-attentive problems which may be related to pain or substance abuse. Methods:
Auditory frequency deviance elicited mismatch negativity was recorded from 22 patients with chronic
tension-type headache, 26 with medication-overuse headache from underlying chronic tension-type
headache and 41 healthy volunteers as controls. Their depression and anxiety scores were noted.
Results: There were no signifi cant differences in the N1 latency or amplitude to both standard and
deviant stimuli for the different groups. However, the latency and amplitude of mismatch negativity were
signifi cantly shortened and reduced at Fz, Cz, and Pz in medication-overuse headache as compared to
chronic tension-type headache and normal subjects. Anxiety levels were elevated in chronic tensiontype
headache and medication-overuse headache subjects compared to healthy controls but were not
correlated with mismatch negativity latency or amplitude in a given group.
Conclusions: In medication-overuse headache subjects, the shortened mismatch negativity latency
indicates quick involuntary attention switching to auditory change, while its reduced amplitude
indicates poor accuracy in discriminating early stimuli, which may be related to medication overuse
rather than to the head pain experienced.
5.Influenc of electroacupuncture on urinary retention after abdominal operation

Objective To investigate the effect of prevention and treatment of urinary retention for patients after abdominal surgery using electroacupuncture unit model 6805.Methods 180 cases of ab-dominal surgery patients were randomly divided into three groups, the treatment group(64 patients)were treated with electroacupuneture at the points of sanyinjiao, zhongji, pangguangyu, the control group 1(57patients)Was treated with moxibustion and foment were used in the control group 2(259 patients).The incidence rate of urinary retention Was compared between the three groups. Results The rate of spontaneously emiction was 56.25%in 30 minutes and 35.94%in 40 minutes in the treatment group, the results in the control group 1 and 2 were 22.81% and 15.79%,23.73% and 13.56% respectively. There were significant differences between three groups. Conclusions Electroacupuncture Can effectively prevent and treat urinary retention after abdominal operation.
6.Influence of humidification or non-humidification on oxygen humidity during low-mid volume oxygen inhalation through nasal cannula

Objective To discuss influence of the humidification or non-humidification on oxygen humidity during low-mid volume oxygen inhalation through nasal cannula.Methods A total of 160 patients with low-mid volume oxygen inhalation through nasal cannula for≥12 hours were randomly assigned to the humidified group and the un-humidified group with 80 patients in each.Recording the chases of oxygen humidity from the humidifying bottles.at the same time the indoor air humidity was also observed.Influence of the humidification or non-humidification on oxygen humidity during low-mid volume oxygen inhalation through nasal cannula was observed.Results There was not significant difference in oxygen humidity between the two groups.Conclusions Routine humidifying is not necessary in low-mid volume oxygen inhalation through nasal cannula.
7.A comparison study on the respiratory symptoms between patients receiving low-to-mid flow humidified and dry oxygen by nasal cannula

Objective To explore the effect of humidification of oxygen delivered by nasal cannula on the respiratory symptoms. Method A total of 540 patients receiving low-to-mid flow oxygen therapy (<4L/min) by nasal cannula for more than 12 hours were assigned to receive humidified (n=235) or dry (n=305)oxygen. While 226 patients receiving a low-to-mid flow oxygen therapy for more than 24 hours received humidified or dry oxygen respectively. The patients' respiratory symptoms were observed and recorded. Results Dryness over naso-pharyngeal region was the only symptom reported by patients in this study. There was no significant difference on the rate of dryness over naso-pharyngeal region between patients receiving humidified oxygen and patients receiving dry oxygen (P>0.05) either in the same patients when receiving dry and humidified oxygen (P>0.05). Conclusion Routine humidification is not necessary in low-to-mid flow oxygen therapy by nasal cannula.
8.Risk factors and prevention of lower extremity lymphedema after treatment for cervical cancer

Cervical cancer is a common malignant tumor of female reproductive system. The treatment of cervical cancer is based on surgery and radiotherapy (or concurrent chemoradiation). Lower extremity lymphedema (LEL) is a frequent complication after cervical cancer treatment, which significantly affects the quality of life of patients. Both pelvic surgery and radiation for cervical cancer can lead to LEL. The risk factors for LEL are complicated and involving characteristics regarding patient (age, comorbidities, lifestyle, etc.), tumor [International Federation of gynecology and Obstetrics (FIGO) stage, lymph node metastasis, etc.], and treatment (number of resected lymph nodes, removal of circumflex iliac nodes, adjuvant therapy, etc.). Comprehensive measures are proposed to prevent cervical cancer patients from LEL, and further investigations in terms of effectiveness are warranted.

Copy number variants (CNVs) are common causes of human genetic diseases. CNVs detection has become a routine component of genetic testing, especially for pediatric neurodevelopmental disorders, multiple congenital abnormalities, prenatal evaluation of fetuses with structural anomalies detected by ultrasound. Although the technologies for CNVs detection are continuously improving, the interpretation is still challenging, with significant discordance across different laboratories. In 2020, the American College of Medical Genetics and Genomics (ACMG) and the Clinical Genome Resource (ClinGen) developed a guideline for the interpreting and reporting of constitutional copy number variants, which introduced a quantitative, evidence-based scoring framework. Here, we detailed the key points of interpreting the copy number gain based on the guideline, used six examples of different categories to illuminate the scoring process and principles. We encourage a professional understanding and application of this guideline for the detected copy number gains in China in order to further improve the clinical evaluation accuracy and consistency across different laboratories.

10.Analysis of the characteristics of SPTB gene variants among 16 children with Hereditary spherocytosis.

OBJECTIVE:
To analyze the clinical characteristics and spectrum of SPTB gene variants among 16 Chinese children with Hereditary spherocytosis (HS) and explore their genotype-phenotype correlation.
METHODS:
Sixteen children who were diagnosed with HS at the Affiliated Hospital of Capital Institute of Pediatrics from November 2018 to July 2022 were selected as the research subjects. Genetic testing was carried out by whole exome sequencing. Candidate variants were verified by Sanger sequencing and subjected to bioinformatic analysis and prediction of 3D structure of the protein. Correlation between the SPTB genotypes and clinical phenotypes was analyzed using Chi-squared test.
RESULTS:
The male-to-female ratio of the HS patients was 6 : 10, with the median age being 7-year-and-10-month. Clinical features of the patients have included anemia, reticulocytosis and gradual onset of splenomegaly. Mild, moderate and severe anemia have respectively occurred in 56.25% (9/16), 31.25% (5/16) and 12.50% (2/16) of the patients. SPTB gene variants were detected in all patients, among which 10 were unreported previously and 7 were de novo in origin. Loss of function (LOF) variants accounted for 93.75% (15/16). Only one missense variant was detected. Eleven, 4 and 1 of the variants had occurred in the repeat domain, CH1 domain, and dimerization domain, respectively. There was no significant correlation between the type or domain of the SPTB gene variants with the clinical features such as severity of anemia (x² = 3.345, P > 0.05). All of the variants were predicted to be pathogenic or likely pathogenic based on the guidelines from the American College of Medical Genetics and Genomics.
CONCLUSION
Mild to moderate anemia are predominant clinical features of the HS children harboring a SPTB gene variant, for which LOF variants are the main mutational type. The clinical feature of HS is unaffected by the type of the variants.
