Objective To establish allergic rhinitis(AR) model in rats to observe the changes in IL-5,IL-12 and IL-18 in serum of AR rats,and explore the influence of glucocorticoid on IL-5,IL-12 and IL-18 expression,and to elucidate further the pathogenesis of AR and the mechanism of glucocorticoid.Methods Thirty-six rats were divided into three groups:normal control(NC) group,AR group and dexamethasone intervention(Dex) group(12 each).Animals in AR and Dex group were sensitized with egg albumin,and in NC group were treated with normal saline as control.After sensitization and reproduction of AR model,rats in AR group received no treatment,while those in Dex group were treated with dexamethasone(5mg/kg,once a day) for 5 days.The changes in behavior was examined,and pathology of nasal mucosa were observed with HE staining,and the expressions of IL-5,IL-12 and IL-18 in serum were determined with ELISA.Results Compared with NC group,the frequency of sneesing and nose-scratching,and the expressions of IL-5 and IL-18 were increased obviously(P

OBJECTIVETo evaluate in vitro effect of abnormal savda munziq (ASMq) on the proliferation and apoptosis of human hypertrophic scar fibroblasts (HSFs).
METHODSHSFs were divided into six groups to receive different treatments as group A (blank control group), group B-E (ASMq in different concentration), and group F(5-Fu). Each group contains six specimens. The HSFs were cultured in vitro. After culture for 48 hours, the CCK8 test and flow cytometry methods were used to detect the proliferation, cell cycle and apoptosis.
RESULTSThe proliferation of HSFs in the B, C, D and E groups was inhibited at G2/M period, while it was inhibited at G0/S period in group F (P < 0.05). The inhibition effect of ASMq (0.1-1.0 mg/ml) on the fibroblasts enhanced in a concentration-dependent manner. Flow cytometry analysis with annexin V-FITC and PI staining confirmed the apoptotic. When HSFs were exposed to ASMq at 1.0 mg/ml (group E) for 48 h, the percentage of apoptotic cells increased to (43.7 +/- 2.58)%, which was significantly higher than that of blank control group (2.2 +/- 0.59)%. The induced apoptosis effect was also increased in a concentration-dependent manner.
CONCLUSIONASMq has a inhibitory effect on the proliferation and an enhancement effect on the apoptosis of fibroblast. ASMq could be used as an effective drug for treatment of hypertrophic scar.

5.Analysis of AGXT gene mutation in a primary hyperoxaluria type Ⅰ family

Objective To describe the clinical characteristics,and to analyze the AGXT gene mutation in three siblings with primary hyperoxaluria type I (PHI).Methods AGXT gene mutation was analyzed by direct sequencing analysis in this family,and the minor allele status was also tested.One hundred unrelated healthy subjects were also analyzed as controls.Results Three mutations in AGXT were identified in each of three patients including two novel heterozygous missense mutations and one previously reported variant.One mutation was a methionine to leucine substitution at position 49 (p.M49L,c.145A > C) in exon 1,one was an asparagine to isoleucine transition at codon 72 (p.N72I,c.215A > T) in exon 2,and another was a heterozygous nonsense mutation at codon 333 (p.R333*).Both p.M49L and p.R333* occured in cis configuration with the minor allele IVS1 +74 bp.Conclusions Two novel mutations are identified probably in association with PHI,however their pathogenicity and potential molecular mechanisms should be explored by further investigations.This is the first investigation on mutant gene analysis of PHI in China.
6.Inhibitory effects of carbon nanotube-PAMAM-anti-survivin oligonucleotide compounds on proliferation of HepG2 cells

Objective:To investigate the efficiency of carbon nanotube(CNT)-PAMAM mediated entrance of anti-survivin oligonucleotide into HepG2 cells,and its effects on the proliferation of HepG2 cells.Methods:CNT-PAMAM-anti-survivin oligonucleotide compounds were prepared and characterized by AFM and 1% agarose gel electrophoresis analysis.TEM was used to observe the distribution of CNT-PAMAM-ASODN compounds in HepG2 cells.CNT-PAMAM-ASODN compounds were added into the medium and co-cultured with HepG2 cells for 24 h,48 h,72 h,and 96 h at 37℃,5% CO_2.MTT method was used to detect the effects of ASODN and CNT-PAMAM-ASODN on the proliferation of HepG2 cells.Results:CNT-PAMAM-ASODN compounds were successfully synthesized via AFM and agarose gel electrophoresis.TEM showed that the compounds were located in the cytoplasm.When CNT-PAMAM-ASODN(1.0 ?mol/L)and ASODN(1.0 ?mol/L)were used for a 48 h culture,the inhibitory rates of HepG2 cells were(45.97?4.28)% for CNT-PAMAM-ASODN compounds group,(9.33?0.85)% for ASODN group,and(6.37?0.69)% for CNT-PAMAM group.CNT-PAMAM-ASODN compounds at 1.5 ?mol/L inhibited HepG2 cells by(70.22?7.25)%,and the inhibitory effects were in a time-and concentration-dependent manner.There was statistical difference between experiment group and control group(P

OBJECTIVETo explore the therapeutic effect and the mechanism of acupuncture in reducing body weight.
METHODSFifty cases of simple obesity were randomly divided into an acupuncture group and a drug control group, 25 cases in each group. The patients in the acupuncture group were treated with body acupuncture at Tianshu (ST 25), Guanyuan (CV 4), etc, electroacupuncture and ear point tapping and pressing therapy (Shenmen, Endocrine, etc. were selected); the patients in the control group were treated with oral administration of sibutramine. The serum leptin level before and after treatment were determined and the therapeutic effect in reducing body weight was assessed.
RESULTSThe total effective rate was 88.0% in the acupuncture group and 80. 0% in the control group, with no significant difference between the two groups (P>0.05); after treatment, the serum leptin level in both the two groups decreased significantly (P<0.01), and the decreasing amplitude in the acupuncture group was superior to that in the control group (P<0.05).
CONCLUSIONAcupuncture therapy has a definite therapeutic effect in reducing body weight, and the improvement of serum leptin level of the patient is possibly one of the mechanisms of acupuncture in reducing body weight.

Açaí (Euterpe oleracea) emerged as a source of herb has a long history in South America, which was approved by the Ministry of Health used in China and it has been introduced planting in Guangdong and Taiwan. This article summarized applied history of Açaí and its present status in China. Did theoretical study on the Chinese herbal properties of Açaí based on the Chinese traditional philosophical culture to analysis the function and symptom preliminary, combining with used for medical recordation, chemical component, biological activity. 9.Comparative study on effects of puerarin and granulocyte colony-stimulating factor in treating acute myocardial infarction.

OBJECTIVETo compare the effects of puerarin and granulocyte colony-stimulating factor (G-CSF) in treating acute myocardial infarction (AMI) and on the size of infarcted area and cytokines.
METHODSSeventy-nine patients with anterior AMI were randomly divided into three groups, they were treated with conventional Western medical treatment, but to the puerarin group (PG) and the G-CSF group (GCG) puerarin and G-CSF was given additionally, respectively. The infarcted size, plasma G-CSF, matrix metalloproteinase-9 (MMP-9), serum interleukin-6 (IL-6) and tumor necrosis factor-alpha (TNF-alpha) were detected before and after treatment.
RESULTSThe infarcted size was positively correlated to the levels of G-CSF, MMP-9, IL-6 and TNF-alpha before treatment ( r = 0.45, 0.42, 0.44 and 0.42, P<0.01 ). The infarcted size in the PG and the GCG decreased on the 28th day (P<0.01), the level of G-CSF, MMP-9, IL-6 and TNF-alpha in the PG on the 7th day all decreased (P<0.05), but these indexes in the GCG increased (P<0.05), while those in the control group were unchanged (P>0.05).
CONCLUSIONPuerarin and G-CSF are effective in decreasing infarcted size, but their effects on cytokines are different entirely.

10.An in vitro study on the incidence of the second mesiobuccal canal in the mesiobuccal root of the first and second maxillary molars.

OBJECTIVETo investigate the incidence of the second mesiobuccal canal (MB2) in the mesiobuccal root of the maxillary first and second molars in Chinese population using three techniques, including the clearing technique, spiral CT scanning and serial root sections.
METHODSA total of 216 extracted human first and 334 second maxillary molars were randomly divided into two groups respectively: group A and B. The teeth in group A were cleared. The specimens in group B were subjected to spiral CT scanning, and then the roots were cross-sectioned every 1 mm from the root apex. Under the Dental operating microscope (DOM), the incidence of MB2 were recorded.
RESULTS(1) The incidence of MB2 in the first and the second maxillary molars were 81.48% and 49.70% respectively by clearing, and 77.78% and 47.31% from S-CT scanning, 88.89% and 53.89% respectively from serial root section. The occurrence of MB2 in maxillary first molars was statistically higher than in maxillary second molars (P < 0.05, chi square test). (2) There was no significant difference among the three approaches for detecting the MB2 canal (P > 0.05, chi square test).
CONCLUSIONSBoth the maxillary first molars and the second molars have high incidence of MB2.
