1.Clinical research of phakic posterior chamber intraocular lens implantation for high myopia

Objective To investigate the clinical efficacy and safety of phakic posterior chamber intraocular lens (ICL) implantation for high myopia. Methods Twenty-four eyes of 12 patients with high myopia were treated with ICL implantation. The best corrected visual acuity, intraocular pressure and lens changes shape between post-operative and pre-operative were compared. Results Successful implantations were done in all patients. All eyes had a significant increase in uncorrected visual acuity. Exactly improved visual function in patients with high myopia status. The intraocular pressure was ( 14.26 ± 2.13) mm Hg(1 mm Hg = 0.133 kPa) of pre-operative , and was ( 14.79 ± 2.77) mm Hg at 1 year after post-operative, the intraocular pressure was stability after the operation. UBM showed that there was a certain gap between ICL and the lens. During the follow-up period,no complications such as cataract,chronic uveal infusion were found. Conclusion Phakic posterior chamber ICL implantation is effective,safe and reliable method for the correction of high myopia.
2.Discussion on the cultivation of new health inspection and quarantine professional talents under the background of internationalization

On the background of internationalization for higher education,we have reformed the educational model for bachelor students majoring in public health laboratory science and quarantine.The exploration and pilot study are focused on educational principles,curriculum planning,teaching patterns,experiments and practice,and teachem training.The four teaching strategies have been firstly recommended as follows:laying the strongest foundation of chemistry,expanding quarantine-related curricula for increasing quarantine capability,making full use of strength of clinical laboratory medicine,emphasizing English application.The curricula structure is refined and more social supporting resources have been got to make globalization to cultivate more health inspection and quarantine talents with multidisciplinary knowledge,abilities and strong adaptability.
3.Electrophysiological study of V535M hERG mutation of LQT2.

This study examined the current changes of human ether-a-go-go-related gene (hERG) mutation derived from a LQT2 Chinese family with a highly penetrating phenotype. Mutation was identified and site-directed mutagenesis was performed to induce the mutation in wild-type (WT) hERG. WT hERG and mutated V535M were cloned and transiently expressed in HEK293 cells. At the 48th and 72nd h after transfection, membrane currents were recorded using whole cell patch-clamp procedures. An A>G transition at 1605 resulting in replacement of V535M was identified. Compared to WT, V535M mutation significantly decreased tail currents of hERG. At test potential of -40 mV after depolarizing at +50 mV, tail current densities were 83.35±7.06 pA/pF in WT and 50.38±7.74 pA/pF in V535M respectively (n=20, P<0.01). Gating kinetics of hERG revealed that V (1/2) of steady-state inactivation shifted to negative potential in the mutant (V (1/2,V535M): -61.81±1.7 mV vs. V (1/2, WT): -43.1±0.71 mV). The time constant of recovery from inactivation was markedly prolonged in the mutant compared to WT among test potentials. V535M hERG mutation demonstrated markedly decreased tail current densities, which suggests that V535M is a new loss-of-function mutation of hERG channel responsible for LQT2.
4.Growth of Schwann cells in silk fibroin scaffolds with different pore sizes

BACKGROUND: The growth behaviors of cells on the biomaterials scaffold may be affected by the topography, pore size, wettability, porosity and other factors.OBJECTIVE: This research is aimed to observe the growth and proliferation of Schwann cells in silk fibroin scaffolds with different pore sizes. METHODS: Two kinds of silk fibroin scaffolds with different pore sizes were prepared, including a large pore size scaffold (pore size 50-60 μm) and a small pore size scaffold (pore size 10-20 μm). Schwann cells (R3 [33-10ras3]) served as seed cells and incubated in 37 ℃, 5% CO2 incubation box. When cells filled up the culture bottle bottom and formed a dense monolayer, they were digested and the cell concentration adjusted, then Schwann cells were seeded onto the surface of the porous silk fibroin scaffolds with different pore sizes. After seven days of co-culture, the growth and proliferation of Schwann cells were observed under scanning electron microscope.RESULTS AND CONCLUSION: The growth of Schwann cells on the surface of silk fibroin scaffolds with different pore sizes was varied. On the surface of small pore size scaffold (10-20 μm), the cell density was low, while the phenotype of cells was bipolar, cells arranged in parallel or linked as the cell chains. On the surface of large pore size scaffold (50-60 μm), more cells could be seen, but most of the cells were in the shape of single sphere, cells clustered on the surface of the porous scaffold or aggregated as a bunch of grape at the bottom of pores. Only few cells were bipolar and lied on the ridge between the pores. The result showed that the pore size of porous silk fibroin scaffolds is an influential factor for the growth and adhesion of Schwann cells. Schwann cells are conducive to grow on the scaffolds with pore size larger than cell body diameter.
5.The effects of proanthocyanidins on antidepressant-like behaviors and the adrenal gland in chronic stress rats

Objective To investigate the effects of proanthocyanidins on depressant-like behaviors and the structure of adrenal gland in chronic unpredictable mild stress (CUMS) rats. Methods Rats were randomly divided into 6 groups:control group, stressed group (CUMS + vehicle), three treatment groups (CUMS + proanthocyanidins 25,50,100 mg·kg-1,respectively) ,and imipramine group (CUMS + imipramine 10 mg·kg-1). Used the CUMS model in rats to investigate the effects of chronic oral administration (21 days) of proanthocyanidins and imipramine (ip) on the open-field;and forced swimming and sucrose consumption tests and the ratio of adrenal gland/body weight,and its thickness were examined by HE stain. Results Compared with control group, rats subjected to CUMS exhibited increased ratio of adrenal gland /body weight ( P < 0. 01), less sucrose consumption( P<0.01) and inhibited in the open-field test( P<0.01) as well as more despair time in the forced swimming test( P<0.01). While compared with stressed group,treatments with proanthocyanidins (25,50,100 mg·kg-1, po ,21 days) could significantly improve the activities in open-field test ((39.6±3.4) vs (49±4.5), (52.6±3.7),(54.1±1.8) ;all P<0.01) and sucrose consumption( (5.8±2.5)ml vs (8.1±3.3)ml,(8.5±4.1) ml, (9.2±2.6) ml; P < 0.05, P < 0.05, P < 0.01 respectively); Meanwhile, it could reduce the duration time in forced swimming test significantly( (103.5±10.2)s vs (83.7±8.8)s,(75.8±5.9)s,(67.2±6.5)s; all P<0.01) as well as thickness of the adrenal gland(P<0.05, P<0.01).Conclusions This study suggests that the proanthocyanidins (25,50,100 mg·kg-1) has an antidepressant-like effects in CUMS rats. The antidepressant actions of proanthocyanidins, in some degrees, may be related with the regulation of the adrenal gland's structure.
6.Clinical analysis of five cases of fulminant type 1 diabetes mellitus

The clinical data of 5 cases of fulminant type 1 diabetes mellitus were analyzed retrospectively.This disease was characterized by abrupt onset and severe diabetic ketoacidosis.The mean duration from the appearance of hyperglycemic symptoms to first hospital visit was 3.4 days.The mean plasma glucose level was 47.7 mmol/L,but the mean value of HbA_(1C) level was 6.8% at first visit.The mean fasting serum C-peptide was 40.0 pmol/L,and mean serum postprandial C-peptide was 68.0 pmol/L at onset.β-cell function did not recover after 3-26 months of follow-up.
7.Assessment Method of Remnant α-1, 3-galactosyle Epitopes in Animal Tissue-derived Biomaterials.

The aim of this study was to establish an assessment method for determining α-Gal (α-1, 3-galactosyle) epitopes contained in animal tissue or animal tissue-derived biological materials with ELISA inhibition assay. Firstly, a 96 well plate was coated with Gal α-1, 3-Gal/bovine serum albumin (BSA) as a solid phase antigen and meanwhile, the anti-α-Gal M86 was used to react with α-Gal antigens which contained in the test materials. Then, the residual antibodies (M86) in the supernatant of M86-Gal reaction mixture were measured using ELISA inhibition assay by the α-Gal coating plate. The inhibition curve of the ELISA inhibition assay, the R2 = 0.999, was well established. Checking using both α-Gal positive materials (rat liver tissues) and α-Gal negative materials (human placenta tissues) showed a good sensitivity and specificity. Based on the presently established method, the α-Gal expression profile of rat tissues, decellular animal tissue-derived biological materials and porcine dermal before and after decellular treatment were determined. The M86 ELISA inhibition assay method, which can quantitatively determine the α-Gal antigens contained in animal tissues or animal tissue-derived biomaterials, was refined. This M86 specific antibody based-ELISA inhibition assay established in the present study has good sensitivity and specificity, and could be a useful method for determining remnant α-1, 3Gal antigens in animal tissue-derived biomaterials.

8.Comparative analysis via data mining on the clinical features of Western medicine and Chinese medicine in diagnosing rheumatoid arthritis.

OBJECTIVETo compare the clinical characteristics of traditional Chinese medicine (TCM) and Western medicine (WM) in diagnosing rheumatoid arthritis (RA).
METHODSA total of 85 clinical RA related messages were enacted and classified into 5 sets as pathological locations, quantitative diagnosis, symptomatic descriptions, general status and environment factors. The respective frequency of their presence in the TCM or WM data sets for RA diagnosis collected from MEDLINE and China National Knowledge Infrastructure (CNKI) were analyzed statistically by Chi-square test, and the relationship of some TCM diagnostic factors/conditions with the RA related biological factors was analyzed by co-occurrence-based literature approach of mining.
RESULTSThere was significant difference between the diagnostic pattern of WM and TCM (P < 0.01). Compared with that of WM, TCM diagnosis on RA paid more attention to environmental factors and symptomatic descriptions, which showed definite association with the cytokines and neuro-endocrine factors in RA.
CONCLUSIONExamination of environmental factors and symptomatic descriptions for RA diagnosis is one of the important characteristics of TCM treatment in accordance to syndrome differentiation, it has its potential biologic basis. A novel approach is proposed for exploring the characteristics of TCM in diagnosis and observation.

10.Study on Effects of Chromium on Heavy Metal Removing Strains

Effects of Cr 6+ concentration and culture time on four heavy metal removing strains,stability of these four strains removing Cr 6+,configuration changes inside or outside their cells,effects of Cr 6+ on soluble reductive sugar inside their cells,and effect of several factors on these strains had been studied,and the Cr 6+ resistance mechanisms of these strains have been discussed elementarily. The results showed that the Lethality of these strains caused by Cr 6+ was similar with one another, namely, increasing at first, then decreasing, and finally increasing again as culture time passed. Acclimatization of Candida sp. was better than Sporobolomycetaceae sp.,and the Cr 6+ resistance of Sporobolomycetaceae sp. 7-3 was the best of the four. The research also demonstrated that the metabolic activity of these strains had been influenced by Cr 6+ in a certain extent. Scanning electron microscopy,transmission electron microscopy,and atomic force microscopy observations approved that removal of Cr 6+ by Candida sp. was depended on both surface adsorption and intracellular accumulation. Effects of Cr 6+ concentration, pH, culture time, nitrogen source, carbon source and adsorption time on these strains are not the same.
