1.Clone of apoptosis-related genes in prostate cancer cell by means of an improved subtractive hybridization

Objective To set up an apoptosis model of prostate cancer cell line and to clone and study the apoptosis related genes. Methods An apoptosis model of prostate cancer cell line DU-145 has been set up through induction by all transretinoic acid (ATRA).During the process of cell apoptosis the apoptosis-related gene was cloned by means of improved PCR-based subtractive hybridization from the apoptosis prostate cancer cell line DU-145 prostate cancer cells. Results During the process of DU-145 cell apoptosis,c-erb B-2 expression,TNF genes and some unknown apoptosis-related gene were observed.This has been accepted by Genebank,the accession number being AF174394. Conclusions ATRA-induced apoptosis of DU-145 cells is a complex process with multiple genes involved,some of which being unknown yet.
2.Anti-proliferation effect of~(103) Pd radioactive stent in micro-porcine model with coronary restenosis

Objective To investigate the effectiveness, dose-effect and time-effect relationships for inhibiting neointimal proliferation by 103 Pd radioactive stent, and the safety of the radioactive stent. Methods By angiogaphic, morphometric and histopathologic analysis, we compared 1?000 ?Ci, 500 ?Ci and 100 ?Ci 103 Pd stents with non-radioactive stents on 5 weeks and 12 weeks after stenting in a micro-porcine coronary model of restenosis. Results The neointimal areas of 500 ?Ci and 100 ?Ci 103 Pd stents were significantly smaller than control stents. 1?000 ?Ci stents and control stents had similar neointimal areas. The neointimal areas of 500 ?Ci stents were significantly smaller than 100 ?Ci stents. Compared with control stents, 500 ?Ci stents reduced the neointimal areas by 49% and 50% respectively on 5 weeks and 12 weeks after stenting. Compared with 5 weeks after stenting, the effect of inhibiting neointimal proliferation by 100 ?Ci stents was reduced by 56% (32% vs 14%) on 12 weeks. The neointimal areas on both edge of 100 ?Ci stents were significantly larger than control stents. The neointimal areas on both edge of 1?000 ?Ci stents and 500 ?Ci stents were similar with control stents. No evident radiation damage had been found in the experimental animals who had 103 Pd stent. Conclusion There may be an effective dose window for 103 Pd radioactive stent in inhibiting neointimal proliferation. The effect of inhibiting neointimal proliferation was dose dependent. 103 Pd radioactive stent with appropriate activities may effectively, persistently inhibit neointimal proliferation with no adverse radiation sequelae. These results suggest that 103 Pd radioactive stent prevent restenosis. These data may be useful in predicting safe and effective activity for 103 Pd radioactive stent.

6.Analysis of thrombus risk factors for routine blood test indicators in outpatients during the large-scale SARS-CoV-2 outbreak period

Abstract: Objective To investigate the impact of SARS-CoV-2 virus infection on the risk of thrombosis in COVID-19 outpatient patients with mild and regular symptoms. Methods Outpatient patients during the SARS-CoV-2 large-scale infection period after the policy adjustment for COVID-19 in Beijing in 2022 were selected as the observation group, and the dynamic zero-clearing period before the policy adjustment and outpatient patients during the 2022/2021/2020 period were taken as the three control groups. The patients with physiological factors that may increase the risk of coagulation, such as thrombotic diseases, malignant tumors, female pregnancy and other physiological factors, were excluded. Pediatric patients under 14 years old were also excluded. Age was expressed as median (interquartile). The changes in blood routine, fibrin/fibrinogen degradation products, and D-Dimer in Beijing outpatient patients were studied with statistical method and data analysis techniques. Results Compared with the control groups, the observation group showed a statistically significant decrease in red blood cells (RBC), hemoglobin (Hb), and hematocrit (HCT) levels, and an increase in monocytes (MONO) and platelet (PLT) counts, all showed statistically significant differences (P<0.0001). The proportion of fibrinogen degradation product (FDP) and D-Dimer of observation group exceeding the range increased significantly. Compared with the three control groups, the number of outpatient fibrinogen degradation products (FDP) in the observation group of patients aged 50 years and verage number of patients under 50 years old in the observation group with D-Dimer exceeding the threshold increased by more than 48.98%, and the monthly average number of patients with D-Dimer exceeding the threshold in patients aged 50 or older increased by 346%-998%. Conclusions The results of this study suggest that outpatient patients with mild or regular SARS-CoV-2 infection are also at risk for thrombotic events, and monitoring blood coagulation indicators such as D-dimer is recommended to avoid the sudden onset of thrombosis-related fatal complications .
7.Preliminary study of microsatellite instability and loss of heterozygosity in thymic squamous cell carcinoma

Objective To investigate the frequency of micresatellite instability (MSI) and loss of heterozygosity (LOH) and select sensitive loci for studying microsatellite DNA imbalance in thymic squamous cell carcinoma. Methods 5 microsat-ellitc polymorphism markers and extrated DNA were selected from 9 specmiens of paired thymie squamous cell carcinoma/nor-real tissues. MSI and LOH in the specmiens of thymic carcinoma and relevant pericancerou tissues were detected by polymerase chain reaction (PCR) followed by 6% polyacrylamide gel electrophoresis(PAGE) with silver staining. Results MSI or LOH was detected in 9 thymic carcinoma tissues. The frequency of MSI or LOH was 66.7% (6/9) at loci of D6S1708, 33.3% (3/9) at TP53, 33.3% (3/9)at DM, 33.3% (3/9)at D11S988 and 0% (0/9)at D8S136, LOH at D6S1708 (5/6) was a common genetic alteration. DI1S988 had only LOH alteration. Conclusion D6S1708, TP53, DM, and D11S988 are sensi-tive loci for studying microsatellite DNA imbalance in thymic squamous cell carcinoma. Microsatellite DNA imbalance may play a certain role in occurrence and development of thymic squamous cell carcinoma, and the relationship between MSi or LOH.The linicopathological features of thymic squamous cell carcinoma needs further investigation.
8.Effects of Low Load Exercise on Gait and Balance in Patients with Parkinson's Disease

Objective To explore the effects of low load exercise on the gait and balance in patients with Parkinson's disease. Methods 12 inpatients with Parkinson's disease from May to August, 2015 accepted low load exercise on Power Rehabilitation System 14 times in 2 weeks, with the medication as before. They were assessed with 3D gait analysis and Berg Balance Scale before and after treatment. Results The step length, stride length and walking speed improved after treatment (P<0.05), and improved more in the seriously affected limbs. There was no difference in cadence, stride time, step time, single support, double support, opposite foot off and opposite foot contact after treatment (P>0.05). The scores of Berg Balance Scale improved after treatment (P<0.05). Conclusion The low load exercise can improve the gait and balance in patients with Parkinson's disease.
9.Detection of Early Cervical Cancer Patient’s Peripheral Blood CK19 mRNA before Surgery and its Clinical Significance

Objective To investigate the detection of early cervical cancer patient's peripheral blood CK19 mRNA and its clinical significance. Methods 30 early (ⅠA2~ⅡA) cervical cancer patients from the department of gynecological tumors in the third affiliated hospital of Kunming Medical University were selected as experimental group, 15 patients with uterine myoma and 15 healthy volunteers as negative control group,15 advanced (ⅡB~ⅢB) cervical cancer patients as positive control group. Peripheral blood samples before receiving radical resection of experimental group,and the same samples before treatment of control groups were collected. The change of relative expression level and positive rate of peripheral blood CK19 was analyzed using qRT-PCR technique, and its correlation with clinicopathological parameters and prognosis was further analyzed. Results There was significant difference in relative expression level of CK19 mRNA in the peripheral blood between experimental group and control groups ( <0.001) . And the positive rate of CK19 in the peripheral blood from 30 patients with early cervical cancer before operation in the experimental group was 30% (9/30) . Furthermore, CK19 mRNA positive rate before surgery was correlated with lymph node metastasis, muscularis invasion and pathologic risk factors for postoperative ( <0.05) . There were no significant correlation between CK19 mRNA positive rate and other clinical and pathological features such as age,clinical stage (FIGO),tumor cell differentiation,tumor size,pathological type and etc ( > 0.05) . Conclusions CK-19 mRNA is a specific and suitable molecular marker for the detection of circulating tumor cells in early cervical cancer. Detection of peripheral blood CK19 mRNA expression before receiving radical hysterectomy in patients with early cervical cancer can also make up for the disadvantage of other methods such as the imaging, tumor marker detection. Most important of all, it can provide the basis for choosing a proper adjuvant therapy post-operation and estimating the relapse and prognosis.
10.The mediating effects of rumination on the relationship between negative life events and depressive symptoms in treatment-naive depressed patients

Objective To explore the mediating effects of rumination and its subtypes on the relationship between negative life events and depressive symptoms in treatment-naive depressed patients.Methods Sixty-two treatment-naive depressed patients completed the center for epidemiologic studies depression scale (CESD),rumination response scale (RRS),and life event scale (LES).Results ① The scores of CESD were positively correlated with total score of RRS,brooding score of RRS,and negative LES score (NLES) in depressed patients (r=0.66,P< 0.01;r =0.50,P< 0.01;r =0.51,P< 0.01).The total score of RRS and brooding score of RRS were positively correlated with NLES(r=0.45,P<0.01;r=0.47,P<0.01).② The total score of RRS played a mediating role between NLES and CESD (indirect effect=0.12,95% CI =0.04 ~ 0.23).③The brooding of RRS played a mediating role between NLES and CESD (indirect effect =0.07,95% CI=0.02~0.15).Conclusion The RRS and the brooding of RRS exert mediation effects on the relationship between NLES and CESD in patients with depression.
