Objective: To establish the HPLC specific chromatograms of the ethyl acetate layer in ten batches of effective parts of Filifolium sibiricum and to determine the contents of five components. Methods: The analysis of effective parts of F. sibiricum was performed on a Thermo AcclaimTM120 C18 column (250 mm × 4.6 mm, 5 μm) with acetonitrile (B)-PBS (A) (0.1 mol/L sodium dihydrogen phosphate-2% glacial acetic acid, 1∶1) as the mobile phase in a gradient elution mode, the detection wavelength was set at 360 nm, the flow rate was 0.8 mL/min, and the column temperature was 35 ℃. Results: The specific chromatograms of F. sibiricum effective parts were established and ten common peaks were designated. Among them, five components including isorientin, isovitexin, isoquercitrin, luteoloside and isorhamnetin-3-O-β-D-glucose all showed good linear relationship within the ranges of 0.018—0.108, 0.066 8—0.400 8, 0.088—0.528, 0.118 4—0.710 4 and 0.017 6—0.105 6 μg, respectively. The average recovery was 98.67%, 97.93%, 98.95%, 99.81%, and 97.33% with the RSD value at 1.10%, 0.93%, 1.10%, 0.62%, and 1.48%, respectively. Moreover, the similarity of the eight batches of samples was above 0.9 in the ten batches of medicinal herbs, the similarity of the two batches of which was 0.688 and 0.695, indicating that its content was lower and the difference was greater. In addition, there were significant differences in the content of five components in each harvest time. The content of flavonoids in medicinal herbs was higher with high flower percentage. It was suggested that the content of flavonoids in F. sibiricum was related to the flower percentage of harvest period. Conclusion: The HPLC specific chromatograms of the F. sibiricum effective parts were established and the common characteristic peaks were determined, which could be used for quality control of the F. sibiricum.

The combined administration of traditional Chinese medicine (TCM) aims at comprehensive adjustment of body based on the theory of TCM and the theory of Chinese medicine property. The natures and tastes of TCM are the core of the theory of TCM property. The combined administration of natures and tastes of TCM is one of the important theories of prescription compatibility. The objective of study on the combined administration of natures and tastes of prescriptions according to symptoms of disease is to clarify the compatibility mechanism of prescriptions. The study on the compound compatibility of TCM under the guidance of theory of TCM focuses on the relationship between the composition, dosage and compatibility of TCM by using modern high-tech means. It demonstrates the effective combination of TCM theory and modern technology, and the inheritance and innovation of TCM theory. The study of the effect and mechanism of compatibility of natures and tastes of TCM under the guidance of TCM theory is helpful for the analysis of the compatibility effect and mechanism of TCM based on the pharmacological effect of natures and tastes of TCM. The correlation between the pharmacological effect of natures and tastes of TCM and the pharmacological effect of components were studied by modern informatics, which is beneficial to promote the development of theory of TCM compound. The study of the compatibility between natures, tastes and component of TCM shall pay attention to the combination of pharmacological effects of natures, tastes and component of TCM, so as to define the scientific connotation of the compatibility of TCM, and make full use of the characteristics and advantages of TCM. The methods and pharmacological effects of the combined administration of TCM compounds are reviewed to provide the theoretical basis for the development of new drugs and clinical application.

There are some small molecules with potential allergenicity in traditional Chinese medicine injections. They are lack of immunogenicity due to their small molecular weight, but they can lead to allergic reactions when they were coupled with appropriate vectors. Therefore, how to couple small molecule semi-antigens with vectors to prepare complete antigens with immunogenicity and reactogenicity is the key for screening small molecular allergenic substances out of traditional Chinese medicine injections. In terms of semi-antigen characteristics of traditional Chinese medicine injections, vector selection and application, coupling method and complete antigen purification and identification, the author introduces the latest research situations of artificial antigen and antibody preparation technology, the advance in experimental studies on screening of allergenic substances in traditional Chinese medicine injections, as well as the application prospect of immuno-chip technology in studies on allergenic substances in traditional Chinese medicine injections, with the aim of providing new experimental thoughts and methods for safety control of traditional Chinese medicine injections.

Objective To investigate the uptake mechanism of HepG2.2.15 cells to the nanoparticles co-loaded with syringopicroside and hydroxytyrosol (SH-NPs). Methods The nanoparticles were prepared by using a nanoprecipitation method with mPEG-PLGA as nano-carrier co-loaded with syringopicroside and hydroxytyrosol. The uptake mechanism of HepG2.2.15 cells to SH-NPs was studied by fluorescence microscopy and flow cytometry using fluoresceineisothiocyanate (FITC) as a fluorescent marker. Results With colchicine as the inhibitor, the incubation time ranged from 0.5 to 24 h, the percentage of positive cells increased from 1.9% to 56.4%; When the drug concentration was 125, 250 μg/mL and 500 μg/mL, the positive cell percentages were 4.9%, 3.4% and 3.9%. With chloroquine as the inhibitor; the incubation time ranged from 0.5 to 24 h, the percentage of positive cells increased from 7.4% to 55.4%; When the drug concentration was 125, 250 and 500 μg/mL, the percentage of positive cells was 19.5%, 22.5% and 27.6%. Conclusion Colchicine and chloroquine have an inhibitory effect on HepG2.2.15 cells uptake, and the uptake of SH-NPs in HepG2.2.15 cells was positively correlated with drug concentration and incubation time. It can be concluded that the uptake mechanism of HepG2.2.15 cells to SH-NPs was nonspecific adsorption endocytosis.
