1.Relationship between Serum Advanced Glycation End Products Levels of Diabetic Mothers and Adverse Cardiovascular Function of Newborn Infants

Objective To explore the relationship between serum advanced glycation end products(AGEs) levels in gestation diabetic mother(GDM) and cardiovascular function of newborn infants.Methods Sixty mid-gestation GDM and 72 late-gestation GDM fulfilling the inclusion criteria were recruited,72 mid-gestation and 80 late-gestation mothers with no pregnancy complications were collected as controls.Fasting blood glucose and serum AGEs levels were analyzed in each group.Clinical data of GDM and their babys were collected.Accor-ding to cardiovascular function of neonates,these neonates were divided into 2 groups:normal neonate group with normal cardiovascular function and anormal neonate group with anormal cardiovascular function.Maternal serum AGEs levels,blood glucose between mid-gestation groups and late-gestation groups were compared.Factors which affected the prevalence of complications of fetal outcome in GDM were analyzed.Results 1.Mid-gestation and late-gestation GDM groups had higher serum AGEs levels and fasting blood glucose compared with those of their respective controls(Pa0.05).3.Abnormal fetal outcome in GDM had significantly higher maternal serum AGEs levels than that in controls with normal fetal outcome(P

Objective To investigate the effect of atorvastatin calcium on serum endothelin-1(ET-1), polyamine oxidase(PAO), heart-type fatty acid binding protein(H-FABP), vascular endothelial growth factor(VEGF), 100β, inflammatory cytokines and nerve function in patients with acute cerebral infarction.Methods According to the random number table, 113 patients were randomly divided into two groups (n=61) and control group (n=52).The control group received conventional treatment methods, and observation group received atorvastatin calcium on the basis of control group. The treatment course was two weeks.Serum ET-1, PAO, H-FABP, VEGF, S100β, inflammatory cytokines and NIHSS score were compared between two groups before treatment, 7d and 14d after treatment.Results The serum levels of ET-1, PAO, H-FABP after 7d, 14d treatment of observation group was significantly lower than that of control group, respectively (P<0.05).The VEGF level of observation group after 7 d, 14 d treatment of observation group was significantly higher than that of control group, respectively (P<0.05).The S100βlevel after 7 d, 14 d treatment of observation group was significantly lower than that of control group, respectively (P<0.05).The hs-CRP, IL-8, TNF-αlevels after 7 d, 14 d treatment of observation group was significantly lower than that of control group, respectively (P<0.05).NIHSS score after treatment of observation group was significantly lower than that of control group (P<0.05).Conclusion The atorvastatin calcium can improve neurological function in patients with brain injury through reducing serum ET-1, PAO, H-FABP and S100βlevels, promote angiogenesis through increasing VEGF expression, and alleviate inflammation and ischemia-reperfusion injury through reducing inflammatory cytokines, thereby promote neurological functional recovery.

5.Preparation and preliminary application of monoclonal antibody against Vp1 protein of chlamydiaphage ΦCPG1

Objective To express recombinant Vp1 protein of chlamydiaphage φCPG1, prepare monoclonal antibody against Vp1 protein and utilize it to screen clinical isolates of Chlamydia trachomatis. Methods The Vp1 protein was obtained by prokaryotic expression, and monoclonal antibody against this protein was prepared by hybridoma technique. ELISA and Western blot were used to identify monoclonal antibodies. Then,the monoclonal antibody was prepared in quantity by injecting hybridoma cells into the abdominal cavity of BALB/C mice, and purified by using protein G affinity chromatography. Clinical isolates of Chlamydia trachomatis were screened for the chlamydiaphage by immumofluorescence assay using the prepared monoclonal antibody.Results Purified Vp1 protein was obtained. The monoclonal antibody against Vp1 protein was gained after 3times of sub-clone and consistently identified as IgG1. Three hybridoma cell strains that stably secreted monoclonal antibody were generated. Chromosome analysis of hybridoma cells showed that the mean number of chromosome was 96, most of them were telocentric and a few were submetacentric. The titer of purified monoclonal antibody was more than 1: 12 800. Twenty clinical isolates were screened by using the monoclonal antibody and no positive results were obtained. Conclusions The monoclonal antibody against Vp1 protein of chlamydiaphage φCPG1 is successfully prepared, while no chlamydiaphage is detected by immumofluorescence assay using the prepared antibody in 20 clinical isolates of Ct.

Tibetan Herbal medicine has its own complete theory based on five sources doctrine. And the theories of "Liuwei", "Baxing" and "Shiqi Gongxiao" formed the basic core components of the property theory of Tibetan medicine. However, books and literature of Tibetan medicine have never been systematically expounded and discussed about it specially which thus will limit the further development of Tibetan medicine theory. In this thesis, we firstly introduced three basic core components of the property theory-the "Liu Wei", "Baxing", and "Shiqi Gongxiao" and their interactions as well. At the same time, the links and similarities between the theory of Tibetan medicine and Chinese medicine theory were compared. The job of the thesis done above is to lay the foundation for further systematic reveal and development of Tibetan medicine theory.

7.Expression, purification of Staphylococcus aureus Efb protein and preparation of its functional antibody

Objective:To clone and express the Staphylococcus aureus Efb(extracellular fibrinogen-binding protein) protein in Escherichia coli, to purify the expression product and prepare its functional antibody and to detect the functions of Efb protein for further studies on S.aureus infection.Methods: Efb gene was amplified by PCR using S.aureus NCTC-8325 genome DNA as template and cloned into the recombinant expression vectors pET28a. E.coli BL21(DE3) with the plasmid was induced with IPTG for protein production. The protein was purified by Ni~(2+) affinity chromatography. The function of Efb protein was determined by complement activity assay and inhibition ELISA.The polyclonal antibodies were prepared by immunizing the animals. Results: The purified recombinant Efb was obtained, which could inhibit the CH50 and AH50 effectively. The functional poly-antibodies of Efb were prepared.Conclusion:Efb could inhibit the classical pathway and alternative pathway of complement activation, and the antibodies against to Efb could block the inhibition of the classical pathway of complement activation induced by Efb.
8.Ligand fishing: a strategy for rapidly screening bioactive compounds from organism extracts

Natural products are valuable resources for discovering new drugs. So far, screening bioactive compounds from organism extracts is still an important and challenging task. Traditional biometric guided method involves repeated fractionation steps and bioactivity tests, which are time-consuming, labor-consuming, and inefficient. Ligand fishing is a bioanalysis method for screening ligands from complex organism extracts based on intermolecular affinity interactions. It has the characteristics of strong specificity, high efficiency, and less requirement for sample pretreatment. In this review, we summarize the classification of ligand fishing strategy and its application in enzyme inhibitors screening. Finally, the development prospects of this technology are forecasted.
9.Discussing tuina for atlantoaxial subluxation in adults based on the theory of 'tendon-bone balance'

According to Chinese medicine, the atlantoaxial joint is a composite joint composed of tendons and bones, and the stability of the joint depends on the 'tendon-bone balance' involving tendons, ligaments, atlas and axis. Multiple causes of 'tendon off-position, joint subluxation' will lead to joint 'tendon-bone imbalance', which will evolve into atlantoaxial subluxation (AAS), endangering human health. Chinese therapeutic massage (tuina) is a very effective treatment for AAS in adults, but conventional manipulations are prone to ineffectiveness or accidents due to neglect of the causal relationship of the 'tendon-bone imbalance' and inappropriate manipulations. Compared with conventional manipulations, the rational choice of modified manipulations under the guidance of 'tendon-bone balance' theory is more effective and less risky, and more worthy of clinical promotion. From the 'tendon-bone balance' theory, we considered the shortcomings of conventional manipulations, and introduced several modified manipulations that have their own strengths in 'tendon smoothing' and 'bone setting', in order to provide new ideas for treatment of AAS in adults.
