2.An analysis of ocurrence and prognosis related factors of renal dysfunction in patients with multiple myeloma.

Objective To analyse the occurrence and prognosis related factors of renal dysfunction in patients with multiple myeloma(MM).Methods During Janaury 1991 and Janaury 2007 seventy-four cases of MM in Department of Hematology,Gernal Hospital of Tianjin Medical University were enrolled in this study.The occurrence,the risk factors and prognosis were analyzed.Results The incidence of renal dysfunction(RD)in MM was 56.8%.Age,hypertention,hemoglobin,serum ALB,GLO,calcium,phosphonium,?2-MG,the percentage of plasma and immature plasma cells in bone marrow,Durie-Salmon stage and type of MM were associated with the incidence to RD with MM by monovariam analysis.Hypertention,serum ALB and ?2-MG were associated with the incidence of RD with MM by multivariant analysis.ALB was the protected factor and the other two were risk factors.The renal function improved rapidly in the patients who received CR or blood transfusion for severe and moderate anemia.Compared with patients of normal renal funtion,RD patients survived shorter and had higher early death rate.Conclusion Hypertention and high tumor burden are risk factors of renal dysfunction in MM.Effective chemothemapy and supportive treatment helped renal function recovery.
3.Clinical analysis of autoimmunic hemocytopenia complicated with thrombosis.

Objective To probe the clinical features and possible mechanisms of autoimmunic hemocytopenia complicated with thrombosis.Methods From 2007 to 2008,the clinical and laboratory data of seven patients with autoimmunic hemocytopenia complicated with thrombosis in the hematology department General Hospital tianjin medical university were analysed.Results One of the patients was with Evans syndrome,and another six patients with immuno-related pancytopenia.Three were complicated with lower extremity Veins embolism;two with cerebral vessel and pulmonary artery embolisms respectively;one with splenic artery and coronary artery embolisms respectively.Among them,three were complicated embolism of two parts;six patients had higher D-Dimer level.They were treated with glucocorticoids,cyclosporin A,and immunoglobulin for immunic hemocytopenia and with anti-coagulation therapy(aspirin、heprin and warfarin) for thrombosis.Seven patients' hemogram,signs and symptoms improved gradually since they received immunosuppression and anticoagulation therapy.The symptoms of thrombosis disappeared two weeks later.The degree and extent of thrombosis also alleviated;at the same time,the function of blood coagulation recovered partly or completely.Conclusion Thrombosis is a rare but severe complication of autoimmunic hemocytopenia.Promptly anti-coagulation and immunosuppressive therapies can get ideal therapeutic effects.

5.Treatment of declining in ovarian reserve by shen-reinforcing and menstrual cycle-regulating therapy combined with western medicine: a clinical observation.

OBJECTIVETo observe the clinical effect of Shen-reinforcing and menstrual cycle-regulating therapy (SRMCRT) combined with Western medicine (WM) in treating decline in ovarian reserve (DOR).
METHODSTotally 78 patients with DOR were assigned to the traditional Chinese medicine combined WM group (abbreviated as IM group, 40 cases), and the WM group (38 cases) according to random digit table method. Patients in the WM group were treated with hormone replacement therapy, while those in the IM group additionally received SRMCRT. The therapeutic course for all was 3 consecutive months. The therapeutic efficacy was compared between the two groups. The serum levels of follicle stimulating hormone (FSH), FSH/luteinizing hormone (LH), and estradiol (E2), as well as the development of sinus follicles were compared between before and after treatment in the two groups.
RESULTSThe therapeutic effective rate was 92.5% in the IM group, higher than that of the WM group (73.68%, P < 0.05). The serum levels of FSH, FSH/LH, and E2 decreased (P < 0.05) and the number of the sinus follicle increased (P < 0.05) in the two groups after treatment. Besides, IM was superior in decreasing serum levels of FSH and FSH/LH, and increasing the number of the sinus follicle (P < 0.05).
CONCLUSIONSSRMCRT was an effective method for treating ROD. IM was superior in decreasing serum levels of FSH and FSH/LH, and increasing the number of the sinus follicle.

6.miR-22 inhibited glioma cells proliferation by targeting

Background and purpose:miR-22 has been reported to be down-regulated in gastric cancer, lung cancer, colorectal cancer, and breast cancer. However, its expression in glioma was still poorly known. This study aimed to explicit whether miR-22 suppresses cell proliferation by targeting MTDH, thus to reveal molecular mechanism that miR-22 functions as a tumor suppressor in glioma. Methods:Quantitative real-time polymerase chain reaction (qRT-PCR) was conducted for detecting the expression of miR-22 in gliomas and normal brain tissues. MTDH 3’UTR-luciferase vector was constructed and dual-luciferase reporter gene assay was employed to examine the effect of miR-22 on luciferase activity. U251 cells were transfected with miR-22 mimics, and MTDH siRNA as for postive control, then Western blot was performed to detect the expressions of MTDH protein. The proliferation ability of U251 cells was evaluated by MTT assay. Results:miR-22 was down-regulated in glioma tissues. Glioma patients with relatively high expression of miR-22 showed lower mortality compared with low expression of miR-22 by using Kaplan-Meier survival curves. We demonstrated miR-22 could bind to the 3’ untranslated region (UTR) of MTDH and inhibited the luciferase activity. Western blot showed that the expression of MTDH protein was inhibited by restored miR-22 or siR MTDH in U251 cells. Overexpression of miR-22 or siR MTDH inhibited the proliferation of U251 cells. Conclusion:miR-22 suppresses cell proliferation by targeting MTDH in glioma.

8.Serodiagnosis of trichinellosis by ELISA using recombinant nudix hydrolase of Trichinella spiralis

Trichinella spiralis nudix hydrolase (TsNd) gene encoding a 46 kDa protein was
expressed in Escherichia coli and the potential of recombinant TsNd protein (rTsNd) as an
antigen for the serodiagnosis of trichinellosis was investigated by ELISA and compared with
those of ELISA with T. spiralis muscle larval excretory–secretory (ES) antigens. The sensitivity
of both ELISA was 100% (30/30), for the detection of anti-Trichinella IgG antibodies in sera of
the experimentally infected mice, and the specificity of rTsNd-ELISA and ES-ELISA was 100%
(54/54) and 98% (53/54), respectively (P>0.05). Serum anti-Trichinella antibodies were firstly
detected by rTsNd-ELISA at 14 days post infection (dpi), then continued to increase with a
detection rate of 100% at 36 dpi. The anti-Trichinella antibody levels at different times after
infection were statistically different (P<0.05). The results showed that the rTsNd might be a
potential candidate antigen for specific serodiagnosis of trichinellosis. But, it needs to be
further evaluated with sera of the patients with trichinellosis and other helminthiasis.

10.Expression and significance of follicular helper T cells and galactose-deficient IgA1 in children with Henoch-Schönlein purpura.

OBJECTIVE:
To study the role of follicular helper T (Tfh) cells and galactose-deficient IgA1 (Gd-IgA1) in the pathogenesis of childhood Henoch-Schönlein purpura (HSP) and the correlation between them.
METHODS:
A total of 36 children with newly-diagnosed HSP were enrolled. They were divided into two groups: HSP nephritis (HSPN) group with 11 children and non-HSPN group with 25 children according to the presence or absence of HSPN. Another 15 children who underwent physical examination at the outpatient service were enrolled as the healthy control group. Flow cytometry was used to measure the proportion of Tfh cells (CD4CXCR5ICOS) in peripheral blood. ELISA was used to measure the levels of interleukin-21 (IL-21) and interleukin-6 (IL-6) in peripheral blood and the serum levels of IgA1 and Gd-IgA1. A Pearson correlation analysis was used to investigate the correlation of serum Gd-IgA1 concentration with Tfh cells and related factors expression in the children with HSP.
RESULTS:
Both the HSPN and non-HSPN groups had significantly higher proportion of Tfh cells and expression levels of IL-21 and IL-6 in peripheral blood than the healthy control group (P<0.05). The HSPN group had significant increases in the above indices compared with the non-HSPN group (P<0.05). Both the HSPN and non-HSPN groups had significantly higher serum levels of IgA1 and Gd-IgA1 than the healthy control group (P<0.05). The HSPN group had significantly higher serum levels of IgA1 and Gd-IgA1 than the non-HSPN group (P<0.05). In the children with HSP, serum Gd-IgA1 level was positively correlated with Tfh cells proportion and IL-21 and IL-6 levels (P<0.05).
CONCLUSIONS
Tfh cells and related cytokines and serum Gd-IgA1 are involved in the development of HSP/HSPN. Tfh cells may mediate the increased production of Gd-IgA1.
