1.Significance of serum cytokines concentrations and APACHE scores in evaluating the illness state for Critical patients

Objective To investigate the clinical significance of serum cytokines concentrations and A- PACHE scores in evaluating the illness state for critical trauma patients.Methods A clinical prospective self-control trial was performed,in which 36 patients admitted to ICU by SIRS were enrolled.Objects were divided into mild and severe trauma group according to APACHE score.The TNF-?and IL-6 concentrations were determined on the 1st, 3rd and 5th day of admission,the APACHE score were assessed at the same time.Statistic analysis was performed according to this group.Results The TNF-?concentrations decreased continuously in the following days while IL-6 decreased from the 7th day in the mild trauma group.In the severe trauma group the TNF-?and IL-6,APACHE score concentrations kept increasing.There was a significant difference of TNF-?and IL-6 concentrations between severe trauma and mild trauma group.Conclusion Dynamic measurement of TNF-?and IL-6 concentrations with APACHE score provide great help to evaluate the illness state and predict the prognosis.
2.Clinical study of methyl prednisolone shock therapy for acute spinal cord injury

Objective To assess the efficacy and adverse reaction of methyl prednisolone for the treatment of patients with acute spinal cord injury ( ASCI ) .Methods Sixty cases with ASCI were retrospectively reviewed and analyzed in our hospital from 2010 to 2014.Thirty-two sub-jects in control group only received hemostasis, dehydration, and nerve nutrition and so on, based on which the twenty-eight subjects in treat-ment group received high dose of methyl prednisolone 30 mg? kg-1? h-1 through intravenous infusion less than 15 minutes and 45 minutes later, 5.4 mg? kg-1? h-1 methyl prednisolone was infused in 24 hours.The clinical efficacy and adverse reactions were evaluated by American Spinal Injury Association ( ASIA) score before treatment and six weeks after treat-ment.Results The somatosensory and somatomotion scores were all improved after treatment in two groups(P<0.05), and both scores in the treatment group were much higher than those in the control group after treatment ( P<0.05).But the gastrointestinal reactions found in the treat-ment group were much more than those in the control group ( P<0.05). Conclusion Methyl prednisolone shock therapy can relieve spinal cord compression and reduce secondary neuronal damages, thereby it can improve the function of the spinal nerves, but with higher incidence of complications.
3.Preliminary study on the detection of the SARS-CoV specific target cDNA fragments by multiplex PCR.

The multiplex polymerase chain reaction (PCR) technique was applied to detect the SARS-CoV (severe acute respiratory syndrome-associated coronavirus) specific target cDNA fragments in the present study. The target cDNA fragments of SARS-CoV were synthesized artificially according to the genome sequence of SARS-CoV in GenBank submitted by The Chinese University of Hong Kong, and were used as simulated positive samples. Five primers recommended by World Health Organization (WHO) were used to amplify the fragments by single PCR and multiplex PCR. Three target cDNA fragments (121, 182 and 302 bp), as well as the three different combinations of any two of these fragments, were amplified by single PCR. The combination of these three fragments was amplified by multiplex PCR. The results indicated that the multiplex PCR technique could be applied to detect the SARS-CoV specific target cDNA fragments successfully.

4.Observation on therapeutic effect of electroacupuncture on obesity induced by antipsychotics.

OBJECTIVETo find out an effective therapy for obesity induced by antipsychotic agents.
METHODSOne hundred and one cases of obesity who were being treated with antipsychotic agents were randomly divided into an observation group and a control group. The observation group were treated with electroacupuncture at Quchi (LI 11), Zusanli (ST 36), Fenglong (ST 40), Shangjuxu (ST 37) and Xiajuxu (ST 39), once a day for 8 weeks; and no treatment was given to the control group. The Brief Psychiatric Rating Scale (BPRS) was used for assessment of therapeutic effect and the Treatment Emergent Symptom Scale (TESS) for adverse effects.
RESULTSThe clinically effective rate of the observation group (54.9%) was superior to the control group (10.0%) with a significant difference between the two groups (P<0.05). The BPRS score-reducing rate was 24.92% and 28.62% in the both groups respectively, and with no adverse effects.
CONCLUSIONElectroacupuncture has a good therapeutic effect on obesity induced by antipsychotic agents, and it improves the patient's compliance with no adverse effect.

5.Expression of nm23 H(1) gene in childhood acute lymphoblastic leukemia and the relationship between nm23 H(1) expression and immunophenotype.

OBJECTIVETo study the expression of nm23-H(1) gene in children with acute lymphoblastic leukemia (ALL) and the relationship between nm23-H(1) expression and immunophenotype.
METHODSnm23-H(1) expression was measured by semiquantitative RT-PCR in children with ALL (newly diagnosed, n = 40; remission, n = 32; relapse, n = 16; refractory, n = 3). Twenty normal children served as the control group. The relationship between nm23-H(1) expression and immunophenotype was evaluated.
RESULTSThe expression of nm23-H(1) in the newly diagnosed ALL group was significantly higher than that in the control (p<0.01) and the remission groups (p<0.01). There was no difference in the nm23-H(1) expression between the remission and the control groups. The expression of nm23-H(1) in the relapse group was significantly higher than that in the control (p<0.01) and the remission groups (p<0.01), and similar to that in the newly diagnosed ALL group. The three children with refractory ALL had higher nm23-H(1) expression. Both the positive rate and expression of nm23-H(1) in children with T-lineage ALL were higher than in children with B-lineage ALL (p<0.05).
CONCLUSIONSThe expression level of nm23-H(1) varies with the stages of ALL. Newly diagnosed, relapsed and refractory ALL children have higher nm23-H(1) expression. High nm23-H(1) expression may be associated with a poor prognosis and relapse. A higher expression of nm23-H(1) in children with T-ALL may be contributed to a low remission rate and a poor prognosis.

6.Effect of toosendanin on invasion and migration of human ovarian cancer cells

AIM:To investigate the effect of toosendanin(TSN)on invasion and migration abilities of human ovarian cancer cells and the related mechanism.METHODS:The human ovarian cancer cell lines CAVO-3 and SKVO-3 were treated with TSN at different concentrations.The cell viabilty at 12,24,48,72 and 96 h after TSN treatment was measured by CCK-8 assay.Scratch wound healing assay and Transwell assay were employed to measure the invasion and migration abilities of CAVO-3 cells.The protein expression of nuclear factor-κB(NF-κB)p65,E-cadherin,N-cadherin,vimentin and Snail was determined by Western blot.RESULTS:TSN significantly inhibited the viability of CAVO-3 and SKVO-3 cells(P<0.05 ).Compared with control group ,the migration and invasion abilities of CAVO-3 cells in TSN group decreased significantly(P<0.05).In addition,the expression of NF-κB p65 and E-cadherin protein increased no-tably,followed with N-cadherin,vimentin and Snail protein decreased significantly(P<0.05).However,the inhibitor of NF-κB BAY11-7082 reversed the impact above.Compared with TSN group ,the migration and invasion abilities in TSN +BAY11-7082 group increased significantly(P<0.05).The protein expression of E-cadherin also decreased notably ,fol-lowed with the protein expression of N-cadherin,vimentin and Snail increased significantly(P<0.05).CONCLUSION:TSN inhibits the invasion and migration abilities of human ovarian cancer cells ,which is related to the inhibition of epitheli-al-mesenchymal transition process mediated by NF-κB/Snail signaling pathway.
7.Evaluation of coronary artery in-stent restenosis using 64-slice CT

Objective To evaluate the accuracy of 64-slice CT in the diagnosis of coronary in-stent restenosis (ISR).Methods Fourteen patients with 37 implanted coronary stents were examined by both 64-slice CT angiography (CTA) and conventional coronary angiography(CCA).The diagnosis of ISR was evaluated by two methods(visual inspection and the measurement of the in-stent contrast attenuation)on CTA.The accuracy of the two methods in the diagnosis of ISR was compared with reference to CCA.Results ISR(>50%)was found on CCA in 11 stents.CTA with visual inspection and with measurement of in-stent CT attenuation correctly detected ISR in 2 and 3 stents respectively with reference to CCA.The sensitivity, specificity,positive predictive value and negative predicitive value of the two methods were 18%,69%, 20%,67% and 27%,81%,38%,72%,respectively.There was no significant difference (P>0.05) between the accuracy of two methods.Conclusion The ISR was very difficult to diagnose by 64 slice CT, but the high specificity of 64-slice CT study implied an important role in excluding ISR.
8. Outcomes of reduced-intensity conditioning allogeneic hematopoietic stem cell transplantation for 10 patients with myelofibrosis

Objective:
To evaluate the efficacy of reduced-intensity conditioning allogeneic hematopoietic stem cell transplantation (RIC-allo-HSCT) for patients with myelofibrosis (MF).
Methods:
The clinical data of 10 patients with myelofibrosis (MF) who underwent RIC-allo-HSCT.
Results:
Of all 10 patients, 6 were male and 4 women, with a median age of 28.5 (22-54). Using fludarabine/busulfan plus total body irradiation (FB+TBI) pretreatment scheme based. Hematopoiesis reconstitution was achieved in 9 patients (90%). The median time of neutrophil and platelet engraftment was 13.5 (10-22) day and 16.5 (13-40) day, respectively. Acute GVHD occurred in 4 cases while chronic GVHD in 5 cases. The prospective OS for 3 years was (90.0±8.5)% after a median follow-up time of 17 months. Transplant related mortality was 1 case.
Conclusion
RIC-HSCT with FB+TBI is a feasible and effective alternative for MF patients.
9.Measurement and correlation analysis of vermilion and facial skin color

BACKGROUND: Prosthesis simulation is critical for maxillofacial defects caused by maxillofacial tumor and trauma. Few studies have been reported on the vermilion color, much less the correlation between facial skin color and vermilion color. OBJECTIVE: To collect thecolorimetric values of human vermilion and facial skin, so as to determine the colorimetric value range and its relevance.. METHODS: The colorimetric values of vermilion and facial skin in 202 volunteers were measured by Japanese Konica Minolta CM-700d spectrophotometer, L* (brightness), a* (from –a* (green) to +a* (red)), b* (from –b*(yellow) to +a* (blue)) values were surveyed and calculated, and then analyzed with CIELAB color system. The correlation betweencolorimetric values of the vermilion and cheek skin color was analyzed.RESULTS AND CONCLUSION: For the vermilion color in 202 volunteers, L* value was 53.25±3.42, a* value was 11.19±1.70, and b* valuewas 8.77±2.12; for the cheek skin color, L* value was 61.87±3.59, a* value was 7.21±1.29, and b* value was 12.98±1.64. L* value of thevermilion was negatively correlated with its a* value, positively correlated with its b* value, positively correlated with L* value of cheek skin andnegatively correlated with a* value of the cheek skin. a* value of the vermilion was positively correlated with a* value of the cheek skin. b*value of the vermilion was positively correlated with b* value of the cheek skin. L* value of the cheek skin was positively correlated with b*value of the vermilion, and negatively correlated with its a* value. a* value of the cheek skin was negatively correlated with b* value of thevermilion, and negatively correlated with its b* value. In summary, we preliminarily determine the colorimetric value range of the facial skin andvermilion, as well as their relevance, which provide a basis for prosthesis production and choice of colors in clinic.
