1.Effect of Bu Yang Huan Wu Decoction on ischemia reperfusion injury after limb replantation in rats

BACKGROUND:Traditional Chinese medicine plays a specific role in the clinical diagnosis and treatment of ischemia reperfusion injury. OBJECTIVE:To investigate the effect of Bu Yang Huan Wu Decoction on ischemia reperfusion injury in rats after replantation of of lower limbs, and evaluate the protective effect and mechanism of ischemic tissue. METHODS:A total of 120 Wistar rats were randomly divided into four groups:normal control (intragastric administration of saline after anesthesia), Bu Yang Huan Wu Decoction (intragastric administration of 10 mL/kg Bu Yang Huan Wu Decoction immediately after reperfusion), edaravone (intravenous administration of 10 mL/kg edaravone immediately after reperfusion), and blank control (intragastric administration of saline) groups. At 24 hours after reperfusion, the myeloperoxidase, lactate dehydrogenase, nitricoxide synthase, inducible nitricoxide synthase, constitutive nitricoxide synthase level in the blood were detected. At 1 week after reperfusion, the activity of superoxide dismutase in gastrocnemius tissue, nitrogen monoxidum and malondialdehyde, and the load of Ca2+were assayed. RT-PCR semiquantitative analysis was performed to detect bcl-2 mRNA and bax mRNA transcriptional levels in the gastrocnemius.RESULTS AND CONCLUSION:Bu Yang Huan Wu Decoction protected the rats from ischemia reperfusion injury by reducing the production of oxygen free radicals, reinforcing the activity of superoxide dismutase, lessening the calcium overload, decreasing the production of nitrogen monoxidum, up-regulating the expression of anti-apoptotic gene bcl-2, down-regulating the expression of pro-apoptotic gene bax, and attenuating the injury of skeletal muscle after ischemia reperfusion injury. Bu Yang Huan Wu Decoction has significant protection on cerebral ischemia reperfusion injury in rats after replantation of severed limb, which may be related to the scavenge of free radicals, inhibition of skeletal cells apoptosis, and attenuation of reperfusion injury.

3.Research of placental vascular distribution and clinical outcome in monochorionic twins

Objective To study the placental vascular distribution of monochorionic (MC) twins with twin-to-twin transfusion syndrome (TTTS) or hirth weight discordance.Methods Twenty-eight MC placentas were injected in Peking University Third Hospital between Feb.2010 and Feb.2011.The vascular distribution type (parallel,crossed,mixed and monoamniotic),the anastomosis of vessels and the placental sharing were recorded.The outcome of pregnancy and the placental characteristics of birth weight discordance (birth weight discordance ≥ 20%) in non-TTTS MC twins were analyzed.Results (1) The outcome of pregnancy:the miscarriage or gestational weeks of 28 MC twins were 20 to 38 weeks (median of 35 weeks).Six cases were TTTS,3 of which received fetoscopic laser occlusion of communicating vessels (FLOC).There were 48 live births,with an average birth weight of (2036 ± 623) g.(2) Type of placental vascular distribution:in the 28 MC placentas,number of parallel,crossed,mixed and monoamniotic type of placental vascular distribution were 4 (14%),14 (50%),6 (21%) and 4 (14%) cases,respectively.No parallel type was found in TTTS.There was no significant difference of vascular anastomosis or unequal placental sharing among the different placental vascular distribution types (P > 0.05).(3) Characteristics of placental vascular distribution in birth weight discordance twins:there were 20 non-TTTS MC twin pregnancies,all of which got live births of both babies.Birth weight discordance equal to or more than 20% was found in 6 pairs of newborns,while birth weight discordance less than 20% was found in the rest 14 cases.Ratio of unequal placental sharing was significantly different between the two groups (P < 0.01).There was no significant difference of umbilical cord insertion,placental vascular distribution and anastomosis in the two groups (P >0.01).Conclusions Vascular distribution type of MC twins might be related to TTTS.Unequal placental sharing is a risk factor of birth weight discordance in non-TTTS MC twins.
4.Effect of CoCl2 on induction hypoxia inducible factor-1α in mice hippocampus depend on time delay

Objective To observe the effects of CoC12 treatment on the expression of Hypoxia-inducible factor-1(HIF-1α) in mice hippocampus at different time point.Methods Balb/c mice were injected with CoCl2 and the change of HIF-1 α was detected by western blot and immunofluorescence and confocal laser scanning microscope at different time point(0h,1h,2h,3h,4h,5h and 6h) after injection.Results The relative protein level of HIF-1α was 0.135 ±0.01,0.572 ±0.01,0.595 ±0.03,1.09 ±0.03,1.30 +0.04,1.275 ±0.03,0.947 ±0.03respectively at different time point after the injection.The HIF-1α protein level reached its peak value at 4 h and decreased at 5h and 6h.Fluorescence intensity of HIF-1α was 13.33 ± 3.42,30.95 ± 7.86,46.50 ± 9.65,61.50± 10.02,88.30 + 15.69,71.39 ± 11.28,67.41 ± 10.78 respectively at different time point after the injection.The HIF-1α fluorescence intensity also reached its peak value at 4 h and decreased at 5h and 6h.Conclusion Time dependent HIF-1α accumulation was in close correlation with the CoCl2.
5.Application of left femoral neck radioactive uptake ratio in 99Tcm-MDP bone scan for evaluation of osteoporosis

Objective To evaluate the value of 99Tcm-MDP uptake by left femoral neck for diagnosing osteopomsis.Methods A total of 58 cases (23 males,35 females,mean age:(66.15±8.45) years) with spondyloarthmpathies from May to December of 2012 were selected.Serum concentrations of type Ⅰ collagen telopeptide (sCTX-1) and bone ALP (BALP) were determined.All patients underwent dual-energy X-ray absorptiometry (DXA) to detect bone mineral density (BMD).According to the T scores,patients were divided into 2 groups:normal group (NG) (T>-1.0) and osteoporosis group (OG) (T≤-2.5).99TcmMDP bone scan was further performed.The average radioactive ratio of the left femoral neck to the medial soft tissue of left femur (T/N) was measured.Data differences between the 2 groups were compared by twosample t test and Pearson correlation analysis.Results According to BMD,13 patients (7 males,6 females) were included in NG and 28 patients (10 males,18 females) were included in OG.The mean ages of OG and NG were significantly different ((68.82± 10.41) years vs (62.46± 11.77) years; t =3.560,P<0.05).The BMD of left femoral neck in OG was significantly lower than that in NG ((0.67±0.08) g/cm2 vs (0.91±0.10) g/cm2 ; t=9.917,P<0.01).Although BALP level of OG was significantly higher than that of NG ((35.92±11.58) U/L vs (22.38±6.34) U/L; t=-3.397,P<0.05),no significant difference was observed on sCTX-1 between the 2 groups (t=-0.463,P>0.05).T/N ratio of OG (11.63±6.22) was higher than that of NG (9.74±4.44) (t =-3.027,P< 0.05).There were significant correlations between the T/N ratio of the left femoral neck and the sCTX-1 and BALP concentrations (r=0.376,0.483,both P<0.01).No correlations between the T/N ratio of the left femoral neck and age,BMI and BMD were observed (r=-2.031,-0.017,0.134,all P>0.05).Conclusion The uptake ratio of the left femoral neck in 99Tcm-MDP bone scan could evaluate the metabolism of bone,and it is useful for the early diagnosis of osteoporosis.
6.The Radiosensitizing Effect of Resveratrol on Hopypharyngeal Carcinoma Cell Line FADU and its Effect on the Cell Cycle.

OBJECTIVETo study the radiosensitizing effect of resveratrol on hypopharyngeal carcinoma cell line FADU in vitro.
METHODSHypopharyngeal carcinoma cell line FADU was cultured in in vitro DMEM. Its inhibition on cell proliferation was detected using cytotoxicity test (MTT assay). The cell survival curve was drawn using clone formation to obtain sensitive enhancement ratio (SER). Changes of the cell cycle and cell apoptosis were analyzed using flow cytometry (FCM).
RESULTSResults of MTT showed the inhibition of resveratrol on FADU cells increased along with its concentrations (P < 0.05). Results of clone formation indicated the surviving fraction at 2 Gy (SF2) was 0.717 ± 0.062 in the irradiation group, and 0.426 ± 0.035 in the resveratrol plus irradiation group (with SER ranged 1.684 ± 0.178) with statistical difference (P = 0.007). Results of FCM showed that after radiation of 4 Gy radiation, cells at G2/M phase arrest increased, but cells at G1 decreased. After radiation of resveratrol for 24 h, cells at G1 decreased, but cells at G2/M phase and S phase arrest increased. When 4 Gy radiation combined resveratrol was used, cells at G2/M phase arrest significantly increased, but cells at G1 significantly decreased. The apoptosis rate was 1.94% ± 1.65% in the control group, 4.56% ± 0.92% in the irradiation group, 2.03% ± 1.46% in the resveratrol group, and 23.11% ± 7.22% in the resveratrol plus irradiation group. There was statistical difference between the resveratrol plus irradiation group and the rest 3 groups (P < 0.05).
CONCLUSIONResveratrol could enhance the radiosensitivity of hypopharyngeal carcinoma FADU cells in vitro possibly by inducing cell apoptosis and causing changes in the cell cycle distribution.

7.Application of Time-intensity Curve Analysis of Contrast-enhanced Ultrasonography in Differential Diagnosis of Malignant and Benign Breast Masses

Objective To evaluate the application value of time-intensity curve analysis of contrastenhanced ultrasonography (CEUS) in differential diagnosis of malignant and benign breast masses.Methods From January 2015 to September 2015,30 patients with one lesion confirmed by surgical pathology were examined by CEUS.Then time-intensity curve analysis was performed.Results 16 lesions were found in the benign group and 14 lesions were in the malignant group.Five parameters of benign and malignant group were statistically analyzed,including Perfusion slope rate (C),intensity of peak (A),Subsidence slope rate (K),time of peak value (T) and area under the curve (Area).The perfusion slope rate (C) of benign and malignant group is [(0.24 ± 0.41) vs (0.67 ± 0.39)].There was no statistical difference between the two groups in intensity of peak (A),Subsidence slope rate (K),time of peak value (T) and area under the curve (Area),except perfusion slope rate (P<0.05).Conclusion Time-intensity curve analysis of CEUS has certain effect on differential diagnosis of malignant and benign breast masses.
8.Application of Double Contrast-enhanced Ultrasonography in the Diagnosis of Gastric Cancer

Objective To investigate the clinical value of double contrast-enhanced ultrasonography (DCUS) for the diagnosis of gastric cancer.Methods A retrospective analysis was performed to review the DCUS data of 26 patients which were diagnosed as gastric cancer by pathology in the third affiliated hospital of Kunming Medical University from October 2014 to July 2015.The analysis results were compared with postoperative pathology to get accuracy rates.Results The located accuracy,qualitative accuracy and accuracy at T phase of DUCS was 100% (26/26),100% (26/26) and 88.5% (23/26),respectively.Compared with color doppler flow imaging (CDFI),the qualitative accuracy of DUCS was much higher (P < 0.05).The accuracy of DUCS at T phase was higher than that of CDFI with no statistical significance (P > 0.05) Conclusion DUCS has an important application value in the diagnosis of gastric cancer.
9.Clinical observation on acupoint massage plus Vitalstim electrical stimulation for deglutition disorder after stroke

Objective: To observe the effect of acupoint massage plus Vitalstim electrical stimulation on deglutition function and surface electromyography (SEMG) of deglutition muscle groups. Methods: A total of 60 patients with deglutition disorder after stroke were selected and divided into an electrical stimulation group, a massage group and an integrated group according to the random number table method, with 20 cases in each group. Patients in these three groups were given the same routine rehabilitation training for deglutition. In addition, patients in the electrical stimulation group were given extra Vitalstim electrical stimulation, patients in the massage group were given extra acupoint massage on the head, face and neck, and patients in the integrated group were given extra acupoint massage plus Vitalstim electrical stimulation. Fujishima Ichiro food intake level scale (FILS) was scored before and after treatment. The swallowing duration and maximal amplitude of masseter muscle in SEMG were evaluated before and after treatment. Results: After treatment, the FILS score and the maximal amplitude of recruitment potential generated by muscular contraction of masseter muscle group in the three groups were higher than those before treatment (all P<0.05), and the swallowing duration of masseter muscle group was shortened compared with that in the same group before treatment (all P<0.05). After treatment, the FILS score in the integrated group was higher than that in the electrical stimulation group and the massage group (both P<0.05). The swallowing duration of masseter muscle group measured by SEMG was lower than that in the electrical stimulation group and the massage group (both P<0.05), while the maximal amplitude was higher than that of the electrical stimulation group and the massage group (P<0.05). After treatment, there were no significant differences in the FILS score, swallowing duration and maximal amplitude of masseter muscle group between the electrical stimulation group and the massage group (all P>0.05). Conclusion: Both acupoint massage and electrical stimulation can improve the deglutition function in patients with deglutition disorder after stroke, and improve the coordination and flexibility of masseter muscle. The integration of the two is more effective.
