1.Influence of scanning position of 3.0T magnetic resonance on the detection rate of rotator cuff injury

Objective To study the correlation between scan orientation and the detectability of rotator cuff injury.Methods Different scan orientations of magnetic resonance imaging of the rotator cuff injury were analyzed in high magnetic field in 103 patients.Results The supraspinatus tendon and signal abnormities were obtained in 103 patients,the infraspinatus tendon and signal abnormities in 53 patients,subscapular tendon and signal abnormities in 18 patients,knolls tendon and signal abnormities in 15 patients.The oblique coronal MR imaging was clearest in supraspinatus tendon,while at the axial images was clearest in infraspinatus tendon.Conclusion Rotator cuff and the surrounding tissue injury could be clearly scanned in high magnetic field,the detectability of rotator cuff injury depended on different scan orientations of magnetic resonance imaging.
2.Analysis of the phenotypes and the function of CD56+T cells during primary HIV-1 infection

Objective To investigate the changes of phenotypes and function of CD56+T cells during primary HIV-1 infection and their relationship with disease progression.Methods Peripheral blood mononuclear cells (PBMCs) were collected from 53 subjects with primary HIV-1 infection and 31 HIV-1-negative healthy subjects.The percentages of CD56+T cells and the expression of several phenotypic markers on CD56+T cells including CD16, CD161, NKB1, NKG2A, NKp46, NKG2D, NKG2C and CD158a were analyzed by flow cytometry.IFN-γand TNF-αreleased by CD56+T cells with and without K562 stimulation and the levels of cytotoxic molecular CD107a were measured.Results The percentages of CD56+T cells in patients with primary HIV-1 infection were significantly lower than those of healthy subjects (P=0.025). The levels of CD56+T cells were negatively related to the viral loads in plasma samples ( P=0.021, r=-0.316).Compared with healthy subjects, the expression of CD16 (P=0.003), CD161 (P=0.023), NKB1 (P=0.023) and NKp46 (P=0.021) on CD56+T cells were decreased in patients with primary HIV-1 infection.The levels of NKB1 were positively related to the CD4+T cell counts ( P=0.007, r=0.364), but were negatively related to the viral loads in plasma samples (P=0.030, r=-0.299).Sponta-neous secretion of IFN-γand TNF-αby CD56+T cells and the expression of CD107a were dramatically in-hibited in patients with primary HIV-1 infection as compared with healthy subjects ( all P<0.001 ) . Moreover, the killing ability of CD56+T cells against K562 target cells was weakened in patients with prima-ry HIV-1 infection as the levels of IFN-γ-, TNF-α-and CD107a-producting CD56+T cells were significantly decreased (P<0.001 for IFN-γand TNF-α, P=0.016 for CD107a).Conclusion Inhibited expression and altered phenotypes of CD56+T cells were identified during primary HIV-1 infection.Lower levels of cy-tokines and cytotoxic molecular were also detected, indicating the dysfunction of CD56+T cells appeared dur-ing early stage of HIV-1 infection and was associated with disease progression.
3.Sentinel surveillance and analyze for the detection of respiratory infection in children:nasopharyngeal viral etiolo-gy in Nanxiang, Shanghai during 2007 to 2013

Absract: Objective To report the result of annual monitoring and analysis of nasopharyngeal virus in children with respiratory tract infections in Nanxiang, Shanghai District. Methods Nasopharyngeal secretions were collected from 4389 children with acute respiratory tract infection in outpatient department from January 2007 to September 2013, 9 common respiratory viruses were analyzed by Multiplex RT-PCR, including inlfuenza virus (FLU), parainlfuenza virus (PIV), respiratory syncytical virus (RSV) , adenovirus (ADV), human bocavirus(HBOV), human coronavirus(Cov), enterovirus(EV), human metapneumovirus(HMPV), and rhinovirus(HRV). The same analysis was done in 123 asymptomatic children during the same period. Results The positive rate of detected respiratory viruses in children with respiratory tract infections in nasopharyngeal secretions were 34.8% (1526/4389), including FLU 10.3% (453/4389), RSV 7.3% (320/4389), PIV 6.2%(274/4389), ADV 3.3%(146/4389), HBOV 2.7%(118/4389), EV 2.5%(110/4389), Cov 2.4%(105/4389), HRV 1.6%(72/4389), HMPV 1.5%(67/4389);two and more combined respiratory viral infection were found in 273 cases (6.2%). The virus detection
rate between age groups was signiifcantly different (χ2=41.91, P<0.001). The school-age group had the lowest positive rate of 23.4%and the positive rates in other three groups were all higher than 35.0%. The infant group had the higher positive rate of RSV and HRV. FLU detection rate in school-age group was 13.6%. Respiratory viruses in children with asthmatic disease has high detection rate. RSV infection rate was the highest 14.8%(30/204) in the asthmatic disease group, followed by HBOV 13.8% (28/204). In nasopharyngeal secretions of 123 asymptomatic children, virus-positive detection rate of 6.5% (8/123), which showed signiifcant difference from that in respiratory virus infection group (χ2=42.60, P<0.001). Conclusions In seven consecutive years of testing, the inlfuenza virus and respiratory syncytial virus play an important role in children with respiratory tract infections in this region. The detection rate of virus showed difference between different age groups and a higher detection rate of RSV in infants with respiratory tract infections was observed. The overall detection rate of virus was decreased with the increase of age excluding the inlfuenza virus.
4.Study on effects of Tripterygium wilfordii polycoride in resisting macrophage inflammation and regulating inflammation via TLR4/NF-κB.

To investigate the effect of Tripterygium wilfordii polycoride (TWP) on LPS-induced macrophage inflammatory response, particularly the inhibitory effect on inflammatory factors TNF-α and IL-1β and the regulatory effect on inflammation via TLR4/NF-κB. The MTT method was adopted to test the effects of tested drugs, TWP, dexamethasone (DXM) and azathioprine (AZA) on cell growth to define the appropriate concentration. LPS was used to induce the inflammatory reaction in mouse RAW264. 7 cell lines. The Elisa kit was adopted to test the release level of TNF-α and IL-1β. The Western blotting was applied to test the protein expressions of TNF-α and IL-1β. The RT-PCR was adopted to test the expressions of TLR4 and NF-κB. According to the results, TWP could inhibit the release of macrophage inflammatory factors TNF-α and IL-1β in a dose dependent manner. All of TWP groups showed a weaker efficacy than that of the DXM group. But the TWP high dose group revealed a better effect on TNF-α and equal effect on IL-1β compared with the AZA group. TWP show an equal or better effect in down-regulating TLR4 and NF-κB p65 expressions in a dose dependent manner than DXM and AZA. In conclusion, TWP could inhibit TLR4 and NF-κB p65, which may be related to the down-regulation of TLR4 and NF-κB p65 receptor expressions.

5.Perform analyse on the predictive model of ischemic cardiovascular diseases in Qingdao.

OBJECTIVETo explore the clinical usage of the methods and tools of the 10-year's risk estimation of ischemic cardiovascular disease (ICVD) in Chinese.
METHODSThe risk of ICVD in 2287 middle-aged Qingdao people was evaluated by the methods and tools of the 10-year's risk estimation of ICVD in Chinese, which was developed by Cardiovascular Institute, Chinese Academy of Medical Sciences.
RESULTS(1) 98.16% of male and 99.39% of female had a 10-year absolute risk of ICVD less than 10%. 0.19% of male and 0.15% of female had a 10-year absolute risk of ICVD higher than 20%. (2) In the low risk group (absolute risk < 10%) detection rate of abnormal SBP, FBG, TC, BMI was 100%, 20.8%, 75%, 87.5% respectively. While in the high risk group (absolute risk > or = 20%) detection rate of abnormal SBP, FBG, TC, BMI was 7.31%, 3.4%, 37.74%, 59.26% respectively.
CONCLUSIONSThe prediction models and simplified tools for estimating 10-year-risk of ICVD in Chinese can predict satisfactorily the occurrence of cardiovascular disease in Qingdao area.

6.Quality control of plant extract.

The current situation of plant extract in domestic and international market was analyzed in the paper. The quality control of 20 plant extracts which have reasonably good sales in USA market was compared and analyzed. The analysis of the quality control of six plant extracts indicated that there were two main reasons leading to the varied quality specifications among different suppliers. One reason was that the plant species utilized by different companies were different. The other reason was that the extraction processes were different among different production plants. Comparing with the significant international suppliers of plant extracts, the product quality of Chinese companies were not satisfactory. It was suggested that chromatography and chromatographic fingerprint techniques should be applied to improve the quality control standard of plant extract in our country.

7.Experimental navigation and deuteroexhaustive exercise suppress the function of the hypothalamic-pituitary-testicle axis in rats.

OBJECTIVETo study the effects of experimental navigation and deuteroexhaustive exercise on the serum levels of testosterone (T), corticosterone (CORT), luteinizing hormone (LH) and follicle stimulating hormone (FSH).
METHODSThirty-six male SD rats were randomly divided into an experimental navigation group (Group A), which underwent 180 min wearing floating with psychological stress, a deuteroexhaustive exercise group (Group B), which were subjected to 120 min intensive running on the treadmill after the accomplishment of the same procedure as Group A, and a control group (Group C). Blood samples were obtained at the end of the experiment to determine the T, CORT, LH and FSH of the rats.
RESULTSCompared with Group C, serum T was statistically decreased in Group A and B (P < 0.05), while CORT was increased slightly in Group A and significantly in Group B (P < 0.05). A statistically lower level of serum LH was observed in Group B (P < 0.05), but no significant differences were found in serum FSH among the three groups.
CONCLUSIONStresses of experimental navigation and intensive exercise suppress the function of the hypothalamic-pituitary-testicle axis in rats.

8.The construction of attenuated Tiantan recombinant vaccinia virus vector with IFN-gamma receptor gene deletion.

OBJECTIVEB8R gene encodes a secreted protein with homology to IFN-gamma receptor, which neutralizes the antiviral and immunological regulation activities of IFN-gamma. To improve the safety of vaccinia virus vector, an attenuated recombinant vaccinia virus with the B8R gene deletion from Tiantan vaccine strain (VTT) was constructed.
METHODSThe transfer vectors were generated by joining B8R left flank, B8R right flank, vv promoter, LacZ, multicloning site and pBRSK fragments. The recombinant viruses VTTdeltaB8RLacZ (VTT with B8R deletion and LacZ insertion) were constructed by homologous recombination.
RESULTSThe B8R deletion mutants were confirmed by dot blot with B8R gene probe and PCR amplification. The replication ability of VTTdeltaB8RLacZ strain in vitro was similar to that of the VTT. The skin lesions formed by VTTdeltaB8RLacZ (10(6) pfu) were significantly smaller and healed faster than those formed by VTT when injected intradermally to the rabbits,and no visible ulceration occurred. Meanwhile LacZ in VTKgpedeltaB8RLacZ was expressed stably.
CONCLUSIONSThe attenuated vector with B8R gene deletion improves the safety of recombinant vaccinia virus vaccine B8R locus may be used as a new site for insertion of foreign genes in vaccinia virus vector.

9.Finite element analysis on scoliosis lumbosacral vertebral structure and its deformation trend

Objective To study the correlation between the deformation and displacement trend of the lumbosacral vertebra (L1-S1) for two typical scoliosis spines under vertical loads,so as to provide the mechanical basis of treatment and prevention of scoliosis in clinic.Methods The X-ray computed tomography (CT) images of two typical scoliosis spines (Lenke-4AN type and Lenke-5CN type) were converted into 3D models,and their finite element models were then established and verified.The internal stress distribution and displacement variation of the models were calculated by the finite element software;the correlation between the lumbosacral vertebral structure and displacement of the spine was analyzed.Results Under the same boundary conditions and load cases,the stress and displacement for two kinds of lumbosacral vertebral models showed different trends.Due to its leftleaning and forward convex bending deformation as well as the relatively large lordosis angle (60°) and smaller left-leaning angle (17.37°),the Lenke-4AN type lumbosacral spine produced slightly small forward convex displacement (8.18 mm) and relatively large left-leaning displacement (0.97 mm).The Lenke-5CN type lumbosacral spine showed left-leaning and forward convex bending deformation as well,with relatively large lordosis angle(59°) and left-leaning angle (26.97°),so it produced more severe left-leaning displacement (20.65 mm) andforward convex displacement (9.22 mm).Conclusions The deformation trend of lumbosacral vertebra is closelyrelated to its structural characteristics,and different scoliosis lumbosacral vertebral structures will cause the corre-sponding deformation trend.The research findings are important for the prevention and treatment of scoliosis.
10.An investigation on knowledge, attitude, behavior about nutrition among elderly residents in 2 communities in Wenzhou City

Objective To investigate the knowledge, attitude, behavior about Dietary Guidelines for Chinese (2007) among elderly residents in 2 communities in Wenzhou City, and to exoplore the appropriate method for dietary intervention for elderly people. Methods A questionnaire survey was carried out in 784 people aged 50 years and above in two communities in Wenzhou from December, 2014 to January, 2015 about Dietary Guidelines for Chinese (2007) .The awareness of knowledge, attitude and behavior about nutrition were recorded, and multiple linear regressions were taken to evaluate the impression of knowledge. Results The awareness rate of knowledge was 58.28%, and the awareness about 'Which is the healthiest way to cook?' was the hot question (89.29%) while 'Which disease can be prevented by eating iodized salt?' was not (0.89%) . And 53.06% of respondents interested in nutrient knowledge, and 14.54% of them thought they had learned the knowledge about nutrition, and 39.54% of them thought knowledge had impact on health, and 76.53% of them would like to change their unhealthy dietary behavior. About 50.89% of respondents got knowledge about nutrition from TV or radio programs. Intake frequency about vegetables and fish were 88.01% and 62.63%respectively, while intake frequency about milk and milk products and beans and their products were 21.43% and 15.43%respectively. Multiple linear regression promoted that, female, a high degree of education, less number of offspring, people who had married, people whose income came from social insurance had positive impact on the awareness rate of knowledge. Conclusion The awareness rate of knowledge among elderly people was low, and intake frequency about milk and milk products and beans and their products were efficient. But they would like to change their unhealthy behavior. Long-term education about nutrition should be taken for elderly people.
