Objectives: To explore the epidemiological distribution of body mass index(BMI),waist to hip ratio(WHR) and abdominal circumference in population of Nanjing,Jiangsu Province. Methods: BMI, WHR and abdominal circumference were measured on 3 445 healthy cases aged 18~90 of Nanjing. Epidemiological distribution of BMI,WHR and abdominal circumference were analyzed according to sex and age. 2.Isobaric tags for relative and absolute quantitation for preliminarily screening of differential proteins in hippocampus of Ts65Dn mice

Objective To explore the molecular mechanism of hypophrenia induced by Down syndrome (DS).Methods Ts65Dn mice were used as DS animal model.Three female mice and three male mice of three to twelve weeks old were mated.Among the 17 first-generation mice alive,five mice remained Ts65Dn trisomy and 12 mice were normal.Five Ts65Dn mice and five normal mice were selected randomly as Ts65Dn group and control group,and bred till 16 to 18 weeks old for experiments.Differential proteins in hippocampus of mice were tested by isobaric tags for relative and absolute quantitation (iTRAQ).Expressions of the differential proteins in Ts65Dn group were detected compared with those in control group.Results A total of 2805 proteins were identified in hippocampus of Ts65Dn group and control group,and significant differences were observed in the expressions of 374 proteins.Compared with those in control group,expressions of 195 proteins increased and 179 reduced in Ts65Dn group.Sorted by P value from low to high,the seven proteins with the lowest P value were uncharacterized protein C2orf47 homolog,isoform 2 of filamin A-interacting protein 1-like,zinc finger protein,isoform 1 of pericentriolar material 1 protein,SEC23 interacting protein,BAG family molecular chaperone regulator 3 and serpin H1.Conclusions Differential proteins are observed in hippocampus of Ts65Dn mice,perhaps closely correlating to neurological defects.The new technology of iTRAQ helps to screen and identify differential proteins in hippocampus.
3.Clinical application and evaluation on therapeutic effects of mandibular reconstruction with free fibula muscle flaps

Objective To investigate the application of vascularized free fibula flaps in reconstruction of mandibular defect,and to evaluate the survival rate and repair effect of the method.Methods 16 patients with mandibular tumor,having a desire to reconstruct mandible,and their systemic state could tolerate the long time operation, were selected to reconstruct mandibular defect by vascularized fibula flaps,of which 12 were male,4 female,aged 24-66 years old,average 45.2 years old.The resection of primary tumor and free fibula flaps were simultaneously conducted,then the fibular flaps were shaped according to the defect location and size of the mandible,and were fixed with reconstructive titanium plate for repair and reconstruction of mandible. The survival of flaps was determined by skin color, texture, and skin temperature of the flaps. The reconstruction effects were evaluated through the patients’surface like photos and X-ray picture of mandible before and after operation.Results All of flaps were survived and no serious complications were found. The complications of the supplied sites were skin tension and wound dehiscence, which were healed by dressing. The mandibular reconstruction effects were good through 2 or more persons’double blind evaluation.Conclusion Free fibula flaps have high survival rate and good results in mandibular reconstruction,and they can meet the needs of various types of mandibular defect repair.
4.Study on influence of RNAi gene silence on chemosensitivity of human digestive system neoplasms

Objective To investigate the influence of RNAi gene silence on the chemosensitivity of human digestive system neo-plasms .Methods Esophageal cancer EC109 cells and gastric adenocarcinoma SGC-7901 cells were selected and divided into the transfection group and the blank control group for culturing .Two cell lines in the transfection group were transferred by siRNA and silenced the expression of Survivin ,giving cisplatin 5 mg/L to co-culture for 72 h .The blank control group was given the conven-tional culture and the same dose of cisplatin for co-culture for 72 h .The in vitro proliferation rate of cells in the two groups were measured by MTT and their sensitivities to the chemotherapy drug cisplatin were compared .Results After siRNA silencing the ex-pression of Survivin in SGC-7901 and EC109 cell lines ,the sensitivity of cells to cisplatin drugs was increased .MTT results sugges-ted that the proliferation activity of cells was significantly lower than that in the non-silence cell lines ,the difference showing statis-tical significance(P<0 .05) .Conclusion Adopting RNAi gene silencing technique can interfere the expression of Survivin gene ,in-hibit the proliferation of tumor cell ,and at the same time significantly improve the sensitivity of tumors to chemotherapy .
5.Expression of ceruloplasmin in Down syndrome babies and their mothers

ObjectiveTo investigate the relationship between ceruloplasmin expression and Down syndrome (DS). MethodsDifferential protein expression in serum of six mothers with DS fetuses and six mothers with healthy fetuses was detected by two-dimensional electrophoresis and matrix assisted laser desorption ionization-mass spectrum,the results were confirmed by Western blot.The levels of serum ceruloplasmin in 11 mothers with DS fetuses,10 mothers with healthy fetuses,11 DS newborns and 10 healthy babies were detected by enzyme-linked immunosorbent assay.The difference between the two groups was compared by two-independent samples t test. ResultsTwenty-nine differential proteins were found in the serum of the mothers with DS fetuses; among which ceruloplasmin increased significantly compared with that in mothers with healthy fetuese with density ratio of 5.43 (t=2.7102,P<0.05).Western blot results showed that the expression of ceruloplasmin in maternal serum with DS fetuses (0.95 ± 0.24) was higher than that of normal mothers (0.37±0.14) (t=2.9521,P<0.05) ; while the expression of ceruloplasmin in DS babies' serum (0.74±0.03) was lower than that of normal newborns (0.89±0.06)(t=-2.9515,P<0.05).The expression of ceruloplasmin in serum of mothers with DS fetuses [(346.5± 111.8) ng/ml] was higher than that of normal mothers [(248.6478.3) ng/ml] (t=2.301,P<0.05) ; while the expression of ceruloplasmin in DS babies' serum [(166.1 ±55.0) ng/ml] was lower than that of normal newborns [(244.0±36.0) ng/ml] (t=-3.873,P<0.01). ConclusionsAbnormal maternal and neonatal serum ceruloplasmin level might relate to DS.
7.Studyies on the Breeding and Cultivation of L-Lactic Acid Producing Strain

In order to obtain higher L-lactic acid yield industrial strain, the original strain Rhizopus oryzae PW352 was mutated by means of N+ ions implantation and a mutant strain Rhizopus oryzae RE3303 was obtained. Its lactic acid yield was increased by 75% than that of the original one. The acid producing condition was optimized by orthogonal design. The concentration of L-lactic acid reached to 131~136g/L and the conversion rate of glucose was as high as 86%~90% under the optimum condition.

The incidence of male infertility increased year by year, which seriously affects family welfare and social development. This article reviews and sorts out recent years’ studies on acupuncture treatment for male infertility. The results show that acupuncture treatment has produced a good clinical effect on this disease. Some progresses have been made in clinical acupuncture point selection, operating methods, complex treatment and therapeutic mechanism. The clinical effect and the mechanism of action of acupuncture treatment for this disease have been deeply explored from symptom differentiation and classification, acupoint selection and prescription, acupoint stimulation mode, and combined acupuncture and moxibustion therapy so as to provide new ideas for clinical acupuncture treatment and basic study.

ObjectiveTo investigate the serum prostate-specific antigen (tPSA),serum free PSA to total PSA ratio (f/tPSA),prostate volume (PV) and prostate-specific antigen density (PSAD) in early prostate cancer (PCa) diagnosis.MethodsRetrospective analysis was performed on serum PSA values and related test results from 252 cases of BPH patients and 49 patients with PCa.Prostate volume (PV) was measured by transrectal ultrasound (TRUS),and the f/tPSAand PSAD values were calculated.The differences of serum tPSA,f/tPSA,PV,and PSAD between BPH and PCa group were compared,the area under the ROC curve was used to evaluate these four indicators for its diagnostic sensitivity and diagnostic specificity.ResultsThe values of tPSA,PSAD in PCa group were significantly higher than BPH group ( P <0.05),while the values of f/tPSA,PV in PCa group were significantly lower than BPH group ( P <0.01orP <0.05).The ROC area showed that serum tPSA(0.8013),f/tPSA(0.7390),PV(0.5613) had lower diagnosis value than PSAD(0.9214) in early prostate cancer ( PSAD > tPSA > f/tPSA > PV).When the upper limit of normal PSA was set to take 4ng/ml,the sensitivity was 91.49%,diagnostic specificity was 51.05%.When the f/tPSA threshold set to 0.16,the diagnostic sensitivity was 57.78%,diagnostic specificity was 78.72%.When PSAD threshold was set to 0.15,diagnostic sensitivity was 88.24%,diagnostic specificity was 81.52%.ConclusionsPSA,f/tPSA and PSAD are indicators for biopsy or followup in early diagnosis of prostate cancer.In particular,the diagnostic value of PSAD has higher sensitivity and specificity than PSA and f/tPSA in the diagnosis of prostate cancer.
