1. Microbial transformation of Ursolic acid by Streptomyces griseus subsp. griseus CGMCC 4.18

OBJECTIVE: To modify the structure of Ursolic acid(UA), a pentacyclic triterpene acid, existing a variety of natural plants with extensive biological activities thus to increase the diversity of structure of UA and lay the foundation for screening more active compounds. METHODS: The structure of UA was modified by Streptomyces griseus subsp. griseus CGMCC 4.18 by microbial transformation. RESULTS: A rare hydroxylated at 2 position, product corosolic acid, 2α, 3β-dihydroxy-urs-12-en-28-oic acid which is difficult to obtain by chemical methods and the product 3, 4-seco-ursan-4-hydroxy-12-en-3, 28-dioic acid with cleavage of ring-A were yielded. Interesting, the hydroxylation at C-2 is α configuration that is rare. The structures of product were established based on spectroscopic data and reference compounds. CONCLUSION: The structure diversity of ursolic acid was increased by microbial transformation by Streptomyces. Furteremore, microbial transformation illustrated regional selectivity and high stereo-selectivity.
2.Multi-slice CT Reconstruction in the Diagnosis of Intracranial Foreign Bodies

ObjectiveTo evaluate the role of multi-slice CT(MSCT) reconstruction in the diagnosis of intracranial foreign bodies.MethodsMSCT scans were performed in 16 patients with intracranial foreign bodies. The volumetric data were reconstructed in the workstation. 5 patients were detected by CT angiography (CTA) examination. Imaging results were then compared with findings found during surgery.Results11 foreign bodies located in one brain lobe, 3 passed two brain lobes, 2 located in ventricle. Among them, 6 cases were closed to parietal including 2 cases closed to cerebral arterial circle. 14 patients were operated and extirpated 18 foreign bodies. Compared with surgical findings, the orientative accuracy of reconstruction imaging was 100%, qualitative accuracy was 85.7%.ConclusionMSCT reconstruction may display the intracranial foreign bodies from multi-angles. It is propitious to know the characteristics, location and whole shape of the foreign bodies for the surgeon before the operation, accordingly to make a best operation project.
3.Expression and significance of follicular helper T cells and galactose-deficient IgA1 in children with Henoch-Schönlein purpura.

OBJECTIVE:
To study the role of follicular helper T (Tfh) cells and galactose-deficient IgA1 (Gd-IgA1) in the pathogenesis of childhood Henoch-Schönlein purpura (HSP) and the correlation between them.
METHODS:
A total of 36 children with newly-diagnosed HSP were enrolled. They were divided into two groups: HSP nephritis (HSPN) group with 11 children and non-HSPN group with 25 children according to the presence or absence of HSPN. Another 15 children who underwent physical examination at the outpatient service were enrolled as the healthy control group. Flow cytometry was used to measure the proportion of Tfh cells (CD4CXCR5ICOS) in peripheral blood. ELISA was used to measure the levels of interleukin-21 (IL-21) and interleukin-6 (IL-6) in peripheral blood and the serum levels of IgA1 and Gd-IgA1. A Pearson correlation analysis was used to investigate the correlation of serum Gd-IgA1 concentration with Tfh cells and related factors expression in the children with HSP.
RESULTS:
Both the HSPN and non-HSPN groups had significantly higher proportion of Tfh cells and expression levels of IL-21 and IL-6 in peripheral blood than the healthy control group (P<0.05). The HSPN group had significant increases in the above indices compared with the non-HSPN group (P<0.05). Both the HSPN and non-HSPN groups had significantly higher serum levels of IgA1 and Gd-IgA1 than the healthy control group (P<0.05). The HSPN group had significantly higher serum levels of IgA1 and Gd-IgA1 than the non-HSPN group (P<0.05). In the children with HSP, serum Gd-IgA1 level was positively correlated with Tfh cells proportion and IL-21 and IL-6 levels (P<0.05).
CONCLUSIONS
Tfh cells and related cytokines and serum Gd-IgA1 are involved in the development of HSP/HSPN. Tfh cells may mediate the increased production of Gd-IgA1.

Objective: To investigate the changes of adhesion molecules in peripheral blood in patients with recurrent cerebral infarction. Objective:To construct a recombinant eukaryotic expressive vector of pEGFP-N3-M-IL-2(88 Arg,125 Ala),and to study the expression of this gene in the Glioma cell line U 87,and to detect its antitumor activities of the fusion protein M-IL-2(88 Arg,125 Ala).Methods:The target fusion gene M-IL-2 (88 Arg,125 Ala) was amplified by PCR from pPICZαA/M-IL-2 (88 Arg,125 Ala) and cloned into pEGFP-N3 vector after digestion to construct recombinant eukaryotic expressive vector pEGFP -N3-M-IL-2( 88 Arg,125 Ala).And then recombinant plasmid pEGFP-N3-M-IL-2(88Arg,125Ala) was transfected into Glioma cell U87 by LipofectamineTM2000 immediately after it was confirmed by restrictive enzyme analysis and sequencing .RT-PCR and Western blot were used to confirm expression of the fusion gene in the U87.Prohibitory effect of recombinant M-IL-2(88Arg,125Ala) protein on U87 was assessed by CCK-8 assay.Results:Restrictive analysis and sequence analysis revealed that M-IL-2(88Arg,125Ala) fusion gene was cloned into the vector pEGFP-N3 suc-cessfully,fusion gene M-IL-2(88Arg,125Ala) could express in U87 cells and could inhibit the growth of U87 cells.Conclusion:The eu-karyotic expression plasmid pEGFP-N3-M-IL-2( 88 Arg,125 Ala) was constructed and expressed in U 87 cells successfully ,the fusion protein could inhibit the growth of U87 cells.We laid a foundation for further research of gene M-IL-2(88 Arg,125 Ala).

Multilocus sequence typing (MLST) is a molecular genotyping method based on nucleotide sequencing. The procedure of this method characterizes isolates of bacterial species using the DNA sequencing of multiple housekeeping genes(usually seven). For each housekeeping gene, the different sequences present within a bacterial species are assigned as distinct alleles.For each isolate, the alleles at each of the loci define the allelic profile or sequence type (ST). MLST has the advantages of being robust (based on genetic data) and electronically portable to generate data that allow rapid and global comparisons between different laboratories. In this paper, the principle, method, data analysis, application, advantages and flaws of MLST are introduced.

OBJECTIVETo establish a method for GC fingerprint determination of the chemical constituents in Herba Asari.
METHODGC and GC-MS were used to optimize the fingerprint determination method, and identify the main peaks in the GC fingerprint.
RESULTA preferable method for GC fingerprint determination of the chemical constituents in Herba Asari was established.
CONCLUSIONA general acquaintance of the chemical constituents in Herba Asari can be obtained by using the preferable GC fingerprint determination method, which is useful for quality evaluation of the crude drug of Herba Asari.

Two new species of Fulvifomes are described from specimens collected in rainforests of Nonggang Nature Reserve of southern China, based on morphological characteristics and molecular phylogenetic analysis of the internal transcribed spacer (ITS) and nuclear large subunit ribosomal DNA (nLSU) sequences. Fulvifomes nonggangensis sp. nov. is characterized by perennial, sessile and solitary basidiocarps, applanate pileus, small cystidioles of 9.9–15.4 Â 2.9–3.5 lm, large pores of 5–6 per mm, a dimitic hyphal system, and broadly ellipsoid basidiospores of 4.3–5.3 Â 3.3–4.2 lm. F. tubogeneratus sp. nov. is characterized by perennial, sessile, and imbricate basidiocarps, a duplex context, small pores of 7–8 per mm, a dimitic hyphal system, and ovoid to subglobose basidiospores of 5.72 Â 5.00 lm.

10.Simultaneous liver-kidney transplantation in polycystic kidney and hepatic disease with kidney and liver failure.

OBJECTIVETo review the clinical experiences concerning simultaneous liver-kidney transplantation in polycystic kidney and hepatic disease with kidney and liver failure.
METHODSThis study involved 8 cases of simultaneous liver-kidney transplantation in polycystic kidney and hepatic disease with kidney and liver failure. There were 5 male and 3 female patients, aged from 41 to 67 years old with a mean of 52.8 years old. Six cases transplanted kidney after liver with orthotopic liver transplantation, and 2 cases transplanted liver after kidney with piggy-back liver transplantation. The acute rejections, complications, liver function, kidney functions, and survival rates of patient/liver/kidney were recorded.
RESULTSWithin the follow-up of 28 to 65 months, all 8 patients are still alive with normal liver and kidney functions: 2 living more than 5 years, 2 living more than 4 years and 4 living more than 2 years. 2 cases of pleural effusion and 1 case of pneumonia were complications after operation, which had been cured successfully. No acute rejection of allograft was observed.
CONCLUSIONSSimultaneous liver-kidney transplantation is a safe and effective treatment for polycystic kidney and hepatic disease with kidney and liver failure.
