BACKGROUND:Currently, there are few reports on the effects of Caspase inhibitors combined with Cocktail protease inhibitors on isolation and purification of islet cel s. OBJECTIVE:To explore the effects of Caspase inhibitors and Cocktail protease as protectors on islet cel s during isolation and purification process. METHODS:New-born pigs were selected to separate, purify and culture the islet cel s, and after 24 hours of culture, the specimens were divided into three groups:blank control group, experimental group A (Caspase inhibitors and Cocktail protease were added only during digestion process), and experimental group B (Caspase inhibitors and Cocktail protease were added both during digestion and culture processes). AO-EB staining was used to qualitatively observe cel morphology and apoptosis. Flow cytometry was employed to quantitatively detect cel viability and apoptosis. RESULTS AND CONCLUSION:Percentages ofβcel s were 66.91%in blank control group, 84.58%in the experimental group A, and 87.15%in the experimental group B. The proportions of living cel s, apoptotic cel s and dead cel s were 56.52%, 16.15%, 21.25%in the blank control group, 62.27%, 14.66%, 14.47%in the experimental group A, and 73.09%, 6.83%, 10.28%in the experimental group B, respectively. These findings indicate that in the process of digestion and culture in vitro, Caspase inhibitors and Cocktail protease inhibitors can both obviously decrease the loss of cel s, and increase the percentage of islet beta cel s.

This study is designed to obtain recombinant human acetylcholinesterase (rhAChE) and apply it in screening acetylcholinesterase inhibitors. The rhAChE was overexpressed in HEK293 cells transfected by plasmid of pCMV-AChE with the cationic liposome and rhAChE was found to be secreted into cell culture medium. AChE activity was assayed according to modified Ellman method to obtain kinetic parameters. IC so50 values for donepezil compounds of rhAChE were calculated to determine their activities of inhibition. The results showed that Km value was 151.9 micromol.L-1 donepezil inhibited rhAChE in a mixed competitive-noncompetitive way (Ki= 16.03 nmol.L-1, Ki = 18.36 nmol.L-1) and that most new compounds tested exhibited high activities of inhibition on rhAChE. The study suggests that rhAChE is available to be applied in screening AChE inhibitors in vitro.

Objective To study the constituents in water-extracts from Flos Lonicerae. Methods Compounds were isolated and purified by using various column chromatography such as D101, Sephadex LH-20, and silica gel, etc. Their structures were identified on the basis of physical and spectral data. Results Seven secoiridoid glycosides were obtained and identified as vogeloside (Ⅰ), 7-epi-vogeloside (Ⅱ), secologanic acid (Ⅲ), sweroside (Ⅳ), secoxyloganin (Ⅴ), secologanoside (Ⅵ), (E)-aldosecologanin (Ⅶ). Conclusion Among them, compounds Ⅲ and Ⅵ are firstly obtained from the plants in Lonicera L. The structure of compound Ⅶ is rare in nature so far.

Objective To study between serum uric acid and early morning blood pressure of the different degrees of Obstructive sleep apnea syndrome(OSAHS)patients.Methods134 cases of OSAHS were divided into mild,midrange and severe groups according to apnea-hypopnea index(AHI),and 23 healthy people as normal control group.serum uric acid and early morning blood pressure were measured among four groups.ResultsSerum uric acid levels in mild,midrange,severe OSAHS groups and the control group were(392.10?88.22)?mol/L,(460.14?118.86)?mol/L,(537.63?134.11)?mol/L,(304.36?80.12)?mol/L,there were significant differences among four groups.there were significant differences between severe OSAHS and the others in blood pressure,as well as between OSAHS midrange group and the control group.ConclusionSerum uric acid in OSAHS patients increased with increasing degree of AHI,the disorder of serum uric acid possiblly contribute to abnormal blood pressure in OSAHS patients.

Objective: To evaluate the impact of fine particulate matter ( PM2.5 ) on respiratory diseases and symptoms of community residents in Chun’an County, so as to provide the basis for air pollution treatment strategies. Methods: Using the cluster random sampling method, the permanent residents in Qiandaohu Town of Chun’an County were investigated from 2017 to 2018. The demographic information, respiratory diseases and symptoms were collected by using Surveillance Plan for the Impact of Air Pollution ( Haze ) on health ( 2016 Edition ). The air pollutants and meteorological data were collected through Qiandaohu station of Hangzhou Ecology and Environment Monitoring Center and Hangzhou Meteorological Information Center. The effects of PM2.5 on respiratory diseases and symptoms of residents were analyzed with generalized estimating equation. Results: Totally 1 181 people aged 6 months to 95 years were recruited, including 557 ( 47.16% ) males and 624 ( 52.84% ) females. Acute nasopharyngitis (common cold) and tracheitis/tonsillitis occurred most frequently, with 203 cases, accounting for 3.44%. The median of daily average concentration of PM2.5 was 24 μg/m3, with the standard exceeding rate of 2.80%. The results showed that PM2.5 increased the risk of acute nasopharyngitis ( common cold ), tracheitis/tonsillitis, cough, expectoration, runny nose, sore throat and nasal congestion ( lag 3 days, OR: 1.015-1.022, 95%CI: 1.001-1.037 ); the effect of PM2.5+PM10 ( OR: 1.020-1.040, 95%CI: 1.006-1.070 ) and PM2.5+O3 ( OR: 1.017-1.024, 95%CI: 1.005-1.035 ) was greater than that of PM2.5 alone on respiratory diseases and symptoms. Conclusion Atmospheric PM2.5 exposure in Chun’an County increases the risk of respiratory diseases and symptoms among community residents.

The purpose of this study is to investigate the enantioselectivity of norgestrel (NG) transdermal permeation and the potential influence of linalool and lipids on the enantioselectivity. In vitro skin permeation studies of NG across the excised rat skins were performed with Valia-Chien diffusion cells, and the permeation samples were analyzed by enantioselective HPLC. The possible enantioselective permeation of NG across intact rat back skin and lipids extracted rat back skin and the influence of linalool were evaluated. The skin permeation rate of dl-NG was two times higher than that of l-NG when donor solutions (EtOH/H2O 2 : 8, v/v) containing l-NG or dl-NG. It may be mainly attributed to the solubility discrepancy between enantiomer and racemate. The enantioselective permeation of dl-NG across intact rat skin was observed when the donor solutions containing dl-linalool. The permeation flux of l-NG was 22% higher than that of d-NG. But interestingly, the enantioselective permeation of dl-NG disappeared under the same experimental condition except that the lipid extracted rat skin was used. Attenuated total reflection-fourier transform infrared spectroscopy analysis of stratum corneum showed that the wave number for asymmetric CH2 stretching vibrations of lipids treated with dl-linalool was greater than that of the control. The results indicated that the enantioselective permeation of NG may be contributed by the interaction between dl-linalool and lipids. More than half of lipids were composed of ceramides. The stereospecific interaction maybe existed among chiral enhancer (linalool), lipids (ceramides) and/or chiral drugs (NG).

Objective: The study is to explore daily violence exposure and its association with campus bullying, to provide theoretical basis for positive development of middle school students. Methods: Questionnaire survey was conducted by using Violence Exposure Scale, Normative Beliefs about Aggression Scale, Middle School Students’ Self-control Scale, and Middle School Students’ Campus Bullying Scale. During Aug. to Oct. 2019, 1 372 middle school students were selected by the convenient sampling method as subjects of study from 2 junior high schools and 3senior high schools in Xinxiang. Results: The total score in daily violence exposure was (34.22±12.09). The scores of violence exposure, traditional bullying and cyberbullying in female were lower than in male(t=-2.60--6.32, P<0.05). The scores of violence exposure, traditional bullying and cyberbullying in junior high school students were higher than senior high school students(t=4.59-7.50, P<0.05). The relationship between violence exposure and normative beliefs about aggression, traditional bullying, cyberbullying were positive (r=0.20, 0.44, 0.51, P<0.01). The relationship between violence exposure and self-control was negative (r=-0.29, P<0.01) . The relationship between normative beliefs about aggression and traditional bullying, cyberbullying were positive (r=0.28, 0.22, P<0.01). The relationship between normative beliefs about aggression and self-control was negative (r=-0.38, P<0.01). Violence exposure indirectly affects traditional bullying/cyberbullying through normative beliefs about aggression. The effect of normative beliefs about aggression on the traditional bullying/cyberbullying of middle school students is reduced with the increase of self-control. Conclusion Normative beliefs about aggression plays an intermediary role in violence exposure and traditional bully/cyberbullying, and self-control regulates the relationship between them.

Objective: To explore mediating effect of shame on parent-child attachment and bullying among high school students, and to provide the basis for school bullying prevention among high school students. Methods: From September to November 2018, a total of 1 149 students’ bullying behavior from 4 high schools in Shenyang and Xinxiang, were investigated with the Inventory of Parents and Peer Attachment, Middle School Students’ Bullying Scale and Middle School Students’ Shame Scale. Results: About 31.07% of the subjects involved in school bullying. The report rates of bullying among boys, high school students of second grade, only child, students from rural area were higher (P<0.01). Mother-child attachment was negatively correlated with traditional and cyber bullying(r=-0.14, -0.21, -0.20, P<0.01). Father-child attachment was negatively correlated with traditional and cyber bullying(r=-0.19, -0.21, -0.19, P<0.01). Shame was positively correlated with traditional bullying and cyber bullying (r=0.20, 0.19, P<0.01). Shame mediates mother-child attachment, traditional and cyber bullying, accounting for 11.78% and 15.70% of the total effects. Shame mediates father-child attachment, traditional and cyber bullying, accounting for 15.17% and 17.43% of the total effect. Conclusion Mother-child and father-child attachment have direct effect on school bullying, and indirect effect on school bullying through shame.

Objective: To explore possible mediating effects of stressful life events and Internet game intention in the relationship between role attachment and Internet game addiction,and to provide a theoretical reference for preventing Internet game addiction among middle school students. Methods: During Sept. to Dec. 2018, 1 869 middle school students were selected from 8 middle schools form Liaoning and Henan Province for this investigation through stratified random cluster sampling. Internet Gaming Role Attachment Scale, Adolescent Internet Game Addiction Scale, Adolescent Stressful Life Events Checklist and Adolescent Internet Game Intention Scale was administered. Results: The prevalence of Internet game addiction was 5.00% in middle school students with boys(6.07%)signficantly higher than girls(3.57%)(χ2=5.29，P<0.01). Positive associations were found between Internet game addition with role attachment, stressful life events, Internet game intention (r=0.34-0.58, P<0.01). The results of Bootstrap test showed that stressful life events and Internet game intention had a chain intermediary effect between role attachment and Internet game addiction, and the chain intermediary effect accounted for 6.31% of the total effect. Conclusion Role attachment may affect the occurrence of Internet game addiction through stressful life events and Internet game intention. Helping middle school students to properly cope with stressful life events and Internet game intention may be beneficial for Internet game addiction prevention and intervention.
