1.Preliminary analysis on specific expressed genes in brain tissue of rats with stroke-like episodes induced by complex environmental factors

Objective To investigate the stroke caused by a combination of genetic and environmental factors Though epidemiological studies have provided evidence of genetic influence on the occurrence of human stroke,however,the role of environmental risk factors to the development of stroke is still not well known. Methods Using cold stimuli plus high salt intake as environmental risk factors, we established a hypertension model in rats, for producing a complicated stroke, then, applied a new technique, suppression subtractive hybridization(SSH),to identify the differential genes which were specifically expressed in total rat cerebrum tissues in between two populations, namely the control and the stroke groups. Results Using this SSH approach, totally 288 clones were generated in our study from a subtractive libraries, among them,226 clones were usable and analyzed The average length of this group is (286 6?120 3) bp Among those clones, 126 clones represented sequences with significant identity to the known genes, 78 were matched to existing ESTs in dbEST but not to any known gene sequences, and the remaining 22 were novel transcripts exhibiting no similarity to any known sequences Mitochondrial transcripts were observed at a high rate of 26 5% Mitochondrial genes may play important roles in causes and effects of stroke. Conclusions Our investigation suggests that environmental risk factors may induce an increased sensitivity to stroke through genetic influence Also we will identify the genes responsible for stroke in this rat model

Urothelial carcinoma associated 1 (UCA1) is a kind of long non-coding RNA (lncRNA),which has no capacities for coding proteins.UCA1 over-expresses in diverse tumors,and plays a pivotal role in initiation and progression of tumors.Researches indicate that UCA1 may function as an oncogene in gastrointestinal tumors,such as gastric cancer,colorectal cancer and hepatocellular cancer,and participate in regulating cell proliferation,metastasis and chemo-resistance.
3.An Experimental Study on the Main Pharmacodynamic Actions of Siji Sanhuang Capsule

Objective To compare the main pharmacodynamic actions between Siji Sanhuang Capsules (SSC) and Siji Sanhuang Pills (SSP). Methods Antipyretic, anti-inflammatory and antibacterial effects of SSC and its effect on the intestinal movement in normal mice and constipation mice were observed. Results SSC could relieve fever in rats induced by the fresh beer yeast and 2,4-dinitrophenol, significantly inhibit the auricular swelling induced by xylol and the increased capillary permeability caused by histamine in mice, and promote the intestinal movement in normal mice and constipation mice. In-vitro antibacterial experiments showed that SSC had an obvious bacteriostatic effect on Staphylococcus aureus,typhoid bacillus, Bacillus coli, Shigella flexneri, pseudomonas aeruginosa and Bacillus proteus. The effects were in a dose-dependent manner and the effects of SSC were stronger than those of SSP. Conclusion SSC exerts a Strong antipyretic, anti-bacterial, anti-inflammatory and carthartic effect.
4.Effect of carvedilol on mitochondrial respiratory function of pressure overload induced left ventricular hypertrophy in rats and its mechanism

Objective: To understand the effect of Carvedilol on mitochondrial oxidative phosphorylation function during the development of pressure overload induced left ventricular hypertrophy in rats and its mechanism.Methods: Male SD rats were randomized into 6 groups: 5-and 15-week coarctation of the abdominal aorta(H5,H15),5-and 15-week Carvedilol intervention(HR5,HR15) and 5-and 15-week sham operation(S5,S15).Hemodynamics and ventricular remodeling parameters were measured,and the mitochondrial respiratory function was detected by Clark oxygen electrode.Results: Compared with S5,mitochondrial state 3 and 4 respiration and the oxidative phosphorylation rate(OPR) were increased and the respiratory control rate(RCR) decreased significantly in the H5 group.In comparison with S15,state 3 respiration,OPR and RCR were reduced significantly in the H15group.Carvedilol increased the three parameters and restored them to the level of the S15.Conclusion: Mitochondrial respiratory function decreased during the development of pressure overload induced left ventricular hypertrophy in rats.Carvedilol could protect mitochondrial respiratory function and improve myocardial energy metabolism,which might be a mechanism underlying its protective effect on myocardium.
5.Clinical Observation of Nalmefene Combined with Monosialotetrahexosylganglioside Sodium in the Treat-ment of Patients in Brain Tumor Operation

OBJECTIVE:To explore the clinical efficacy and safety of nalmefene combined with monosialotetrahexosylganglio-side sodium in the treatment of patients in brain tumor operation. METHODS:64 patients in brain tumor operation were randomly divided into observation group and control group,32 cases in each group. Control group received brain cell protection,reducing in-tracranial pressure,anti-infective,vasodilator and other conventional treatment;observation group additionally received 0.7μg/(kg· h) nalmefene combined with 100 mg/d monosialotetrahexosylganglioside sodium from the first day of operation,for continuous 7 d. The plasma cortisol,adrenocorticotropic hormone (ACTH) before and 3,7 d after treatment,changes of serum IL-6,TNF-αand Barthel index (BI) in 2 groups were observed,improvement of neurological function and ADR were observed;Karnofsky (KPS)score was determined after 90 d follow-up used to assess the patient’s ability to take care of themselves. RESULTS:After treatment,plasma cortisol and ACTH content,serum TNF-αand IL-6 levels in 2 groups significantly decreased than before,the dif-ference was statistically significant(P<0.05);cortisol,ACTH and TNF-α levels 3,7 d after treatment,IL-6 level 7 d after treat-ment in observation group decreased more significantly than control group,the differences were statistically significant(P<0.05). After treatment,BI score in 2 groups significantly increased,and observation group increased more significantly than control group,the differences were statistically significant (P<0.05). 7 d after treatment,improvement rate of neurological function was 50.00%,which was significantly higher than that of the control group(18.75%),the difference was statistically significant(P<0.05). There was no significant ADR occurred of 2 groups in the study. Following-up for 90 d,the KPS score in observation group with not less than 70 points accounted for 71.88%,which was significantly higher than the control group(46.87%),the difference was statistically significant (P<0.05). CONCLUSIONS:Nalmefene combined with monosialotetrahexosylganglioside sodium can promote the recovery of patients in brain tumor operation,reduce cortisol,ACTH,IL-6 and TNF-α levels and improve prognosis of patients.
6.Experimental determination of dosimetry parameters for Sinko 125I seed source using thermoluminescent dosimeter

Objective To study the dosimetry parameters of 125I seed source (type Sinko BT-125-1) with thermoluminescent dosimeter (TLD) in the phantom.Methods The new type of phantom was modified to suit to measurement of a common type of 125I seed source.The AAPM TG43 protocol recommended measurements of dose-rate constant (Λ),radial dose function (gL (r)),and anisotropy function (F (r,θ)) have been performed in the phantom with TLD.Results The Λ was 0.928 cGyh-1 U-1.The gL(r) was determined at different radial distances r ranging from 1.0 to 10.0 cm with an interval of 1.0 cm ; and F (r,θ) at angles from 0° to 90° in 10° increments.The gL (r) of 125I seed source showed a difference of 9.6% at the most in comparison to the corresponding values of 125I seed source (type Amersham 6711).The difference in F(2 cm,θ) of 125I seed source and Amersham 6711 was up to 10.2% near the source end.With the phantom the combined standard uncertainty in the whole measurement was less than 6.0%.Conclusions The experimental results exhibit fairly small measurement uncertainties and good self-consistency.It's feasible to measure the dosimetry characters of permanent implant seeds in the modified phantom.
7.Lymph node metastasis around the root of inferior mesenteric artery in rectal cancer

Objective To investigate factors affecting the metastasis of lymph nodes around the root of inferior mesenteric artery(IMA) in rectal cancer,and the significance of root lymph nodes dissection of IMA in radical surgery for rectal cancer.Methods Clinicopathological data of 105 rectal cancer patients undergoing root lymph node dissection of IMA during radical resection in Peking University First Hospital from January 2005 to December 2008 were analyzed retrospectively.Rectal cancer patients without root lymph node dissection of IMA during the same period served as control.Results were compared between these two groups for survival and local recurrence rates.Results The rate of lymph node metastasis around the origin of IMA was 9.5% (10/105).The five-year survival rate in patients with IMA root nodal dissection was 71.3%,and that without was 70.6% (P =0.995),while the local recurrence was respectively 1.9% and 7.4% (P < 0.05).In multivariate analyses,IMA root nodal metastasis occurred more frequently in patients with pT3 and pT4 tumor(Wald =5.764,P < 0.05) and poorly differentiated tumor(Wald =7.818,P < 0.05).Conclusions Root lymph nodes dissection of IMA could not increase five-year survival rate,but it could reduce local recurrence rate in patients with rectal cancer.In radical surgery of rectal cancer,lymphadenectomy of IMA root should be performed in patients with T3 and T4 tumor with poorly differentiated tumor,so as to reduce local recurrence rate.
8.Effects of antihypertensive agent on cardiac hypertrophy in hypertensive rats induced by high salt diet combined with cold stress

Objective To observe the effects of antihypertensive agents on renin angiotensin aldosterone system, blood pressure and myocardial hypertrophy induced by high salt diet plus cold stress. Methods Fifty male Wistar rats weighting 200~250 g were divided into 5 groups: control, saline, captopril, betaloc and nitrendipine. The rats treated with dugs or saline were fed with high salt diet(8% NaCl) and subjected to cold stress 4 hours a day. Blood pressure and body weight were measured once a week. Rats were sacrificed and then the gross weight of hearts was measured 8 weeks later. Renin activity(RA), angiotensinⅡ(AngⅡ) and aldosterone(Aldo) in the left ventricular wall were determined by radioimmunoassay. 9.Expression and clinical significance of serum microRNA-100 in gastric cancer patients

Objective: To investigate the expression of microRNA-100 (miR-100) in the serum of gastric cancer patients and its clinical significance. Methods: At The First Affiliated Hospital of Bengbu Medical College, 40 gastric cancer patients who underwent surgery with complete follow-up data and 40 healthy controls were selected as research subjects. Total miRNA was isolated from the se-rum samples of the cancer patients. A stable and sensitive detection method for the absolute quantification of miR-100 was established. The serum levels of miR-100 in the patients and healthy controls were tested according to this novel method. Differences of the miR-100 expression in the serum samples of the patients and healthy controls were analyzed using statistical analysis. The correlation between miR-100 expression levels and the clinicopathological features of gastric cancer was also analyzed. Results: The expression level of miR-100 in the serum was significantly higher in cancer patients[(2.78±1.92) fmol/L]than in the healthy controls[(0.19± 0.15) fmol/L, P<0.01]. The receiver operating characteristic curve of miR-100 indicated that serum miR-100 has satisfactory specificity and sensitivity for the diagnosis of gastric cancer (AUC=0.985). Further analytical results indicated that no significant differences were found among the expression levels of serum miR-100 with respect to the gender, age, tumor diameter, infiltrating depth, differentiation degree, frequency of lymph-node metastasis, and TNM stage (P>0.05). Conclusion: Serum miR-100 testing may be helpful in the diag-nosis of gastric cancer.
10.CT Features of Solid Components in Pulmonary Mixtured Ground-glass Opacity

Purpose To evaluate the CT features and pathological manifestations of the solid components of mixture ground-glass opacity (GGO) in adenocarcinoma in situ (AIS), minimally invasive adenocarcinom (MIA) and invasive adenocarcinoma (IAC), to analyze the qualitative diagnosis value of solid components of mixture GGO in the diagnosis of AIS, MIA and IAC, to provide reference for the selection of clinical treatment.Materials and Methods Eighteen patients with AIS, 53 patients with MIA and 28 patients with IAC (the maximum diameter smaller than 2 cm) proved by surgery and pathology with CT features appearing as mixture GGO were retrospectively analyzed, CT features of the solid components in three groups were analyzed and compared with pathology.Results The solid components in AIS mainly appeared as punctiform or polygon, with extensive distribution, solid nodules were usually single (17 cases, 94.44%), located in the middle of the lesion (14 cases, 77.78%), with clear binderies (16 cases, 88.89%) and the same density with vessels in the same axis (13 cases, 72.22%); the majority of solid components in MIA appeared as circular or elliptical (33 cases, 62.26%), less than or equal to 5 mm (48 cases, 90.57%), with eccentric or multi-point distribution (45 cases, 84.90%), the boundaries were less sharp (40 cases, 75.47%), with slightly lower density than that of the vasculars in the same level (34 cases, 64.15%); the solid components in IAC mainly appeared as irregular lesions (21 cases, 75.00%), lager than 5 mm (24 cases, 85.71%), with eccentric growth (20 cases, 71.43%) and less sharp boundary (15 cases, 53.57%), the integration of multiple nodules could also be observed. There were statistically significant differences in the CT features of solid components within the lesions among the three groups (P<0.01).Conclusion It is possible to predict the pathological typing and the prognosis of pulmonary mixture GGO in a certain extent according to the different CT features of the solid components in it, and to guide clinical treatment principles.
