1.Research progress of treating Osteoporosis with single Chinese medicine of activating blood and dissolving stasis

In recent years,pharmacological researches of Chinese medicine found that the single Chinese medicines of activating blood and dissolving stasis,such as Drynaria and Bidentata and Salvia and Panax notoginseng had very good effects of resisting osteoporosis,significantly improving bone mineral density(BMD)and bone mineral content(BMC),promoting bone formation,and preventing bone absorption.Based on the documentations from home and abroad,the overview had analyzed the current situation for providing new ideas and methods of osteoporosis prevention and treatment.
2.Research on the Relationship between CRP,HCY and TCM Syndrome Types of Diabetes Mellitus with Hymeperuricemia.

Objective To study the value of CRP and HCY in each TCM type of T2DM with HUA and provide the basis for classification and treatment.Methods 103 cases of T2DM and HUA were divided into four Chinese medicine syndrome groups including deficinecy syndrome of wind-cold and damp-heat phlegm accumlatoin and blood stasis syndrome deficinecy syndrome of Qi and Yin deficinecy syndrome of Yin and yang.Results The differentiation of TCM symptom among four groups was uncorrelated with the level of CRP and Hcy.But the serum CRP and HCY levels increased slighly as compared with those of normal control group.Conclusion The elevation of CRP and Hcy is closely correlated with phlegm accumlation and blood stasis syndrome in patients of T2DM with HUA.As an inflammatoy biomarker,CRP and Hcy may be regard as two of the microcosmic indicators of toxin in Chinese medicine.They can be used as two important indexes of TCM syndrome differentiation of T2DM with HUA.
3.Expression of a protein elicitor pebC1 from Botrytis cinerea in Pichia pastoris.

In order to express PebC1 in Pichia pastoris, the pebC1 sequence was amplified from genome Botrytis cinerea BC-4-2-2-1 by PCR and subcloned into the Pichua pastoris expression vector pPIC9K to generate pPIC9K-pebC1. The recombinant plasmid was linearized by Bgl II and transformed into Pichia pastoris GS115 by electroporation. Recombinant Pichia pastoris GS115/pPIC9K-pebC1 was screened by MD and G418-YPD plates and further confirmed by PCR. The protein expression was induced by methanol and analyzed by SDS-PAGE. SDS-PAGE analysis showed a special band about 39 kDa and western blotting indicated a good antigenicity of the expressed protein. Bioassay results showed that the recombinant protein PebC1 can induce resistance to gray mould disease of cucumber and Arabidopsi thaliana.

4.Expression of peaT1 gene from Alternaria tenuissima in Pichia pastoris and its function.

In this study, peaT1 gene was subcloned into the Pichia pastoris expression vector pPIC9K, which contained both the methanol-inducible promoter and the transcription terminator of the AOX1 gene, resulting the plasmid pPIC9K-peaT1. The recombinant plasmid was linearized by Sal I or Bgl II and transformed into P. pastoris GS115 by electroporation method. Recombinant strain was screened by Minimal Dextrose Medium and further confirmed by PCR. The gene was in frame integrated into the Pichia genome through homologous recombination, resulting the recombinant strain. Regulated by the alpha-Factor, promoter of AOX1 gene and termination signal of yeast genomic, the recombinant protein was expressed and secreted into the supernatant. The SDS-PAGE analysis indicated that the apparent molecular weight of target protein was about 35 kD. Bioassay results showed that the inhibition rate of the expressed protein against TMV was 30.37%. The supernatant was collected and then purified by anion exchange chromatography. This protein can promote seedling growth of wheat obviously.

5.Short-Term Changes of Graft Viscosity after Anterior Cruciate Ligament Reconstruction

Objective To study the short-term variation patterns of graft viscosity after anterior cruciate ligament reconstruction (ACLR) surgery. Methods Six male New Zealand rabbits were selected. The ACLR animal model of unilateral knee was made with Achilles tendon as the graft. The experimental rabbits were euthanized 15 days after ACLR surgery, with removal of the graft, healthy anterior cruciate ligament (ACL) and Achilles tendon. The cross-sectional area and viscosity coefficient of the graft were measured, and the creep experiments were carried out under equilibrium conditions of 0.1 MPa and 1 MPa, respectively. The viscosity coefficent was calculated. Variation patterns of graft viscosity were summarize. The grafts were compared with healthy ACL. Results The cross-sectional area of the graft increased slowly within 15 days after ACLR surgery. The viscosity of ACL and graft changed nonlinearly. The viscosity coefficient was quite different under different stresses. The viscosity coefficient of the graft decreased with the time after ACLR surgery, which was more obviously under the condition of low stress. Conclusions The results are helpful to guide the implementation of early postoperative rehabilitation plan after ACLR surgery .
