1.Expression of hypoxia inducible factor-1? in colorectal tumor:a meta-analysis of case-control trials

Objective:To evaluate the studies about expression of HIF-1?in colorectal tumor with meta-analysis.Methods: Search was conducted on the following electronic databases:Chinese Bio-medicine Database,Medline,Embase to identify all the relevant case-control trials.Statistical analysis was performed with RevMan4.2 to investigate heterogeneity. The studies were combined and the publication bias was evaluated. Results: Totally 11 studies were recruited.Compared with normal tissue,colorectal cancer and colorectal adenoma showed a significantly higher expression of hif-1?(P0.05),the pooled OR and 95%CI was 2.20[0.98,4.97]. Conclusion:The expression of HIF-1? exists in the whole process of colorectal carcinoma. There is a significantly higher expression of hif-1?in colorectal cancer and colorectal adenoma than that in non-neoplasm colorectal mucosa,and no difference is found between colorectal cancer and colorectal adenoma.

Objective To explore the influence of RNAi-mediated hypoxia-inducible factor 1? (HIF-1?) silencing on the growth and metastasis of colon cancer in nude mice.Methods A total of 30 6-week-old female nude mice were randomly divided into normal saline group (NS group),negative plasmid group and RNA interference group after injected with SW480 subcutaneously to form a colon carcinoma.shRNA-HIF-1? vector was constructed and injected (50 ?g/time,once per 2 d,for 7 times) into the tumor mass of nude mice of RNA interference group to induce RNAi.Normal saline or blank vector was injected into the corresponding nude mice.The changes of HIF-1? were detected with RT-PCR and Western blot analysis.The size of tumors,metastasis rate and the positive rate of lymphnode were compared among different groups.Results In 15 d after RNAi vector injection,the tumor volume of RNAi group [(273.9?14.7) mm3] was much smaller than those of NS group [(1 845.5?91.2) mm3] and negative plasmid group [(1 842.7?115.7) mm3] (P

Bone tissue engineering is one of the most clinical applicable research areas in tissue engineering. Seed cell is the first step and essential element of construction and application of tissue-engineered bone. In recent years, many inspiring achievements have been gained in the field of seed cell study. In this review, current status and the prospects of study of seed cell source in bone tissue engineering are reviewed.
4.Tannic acid inhibits the proliferation of SW620 cell lines by downregulating TMEM16A expression

Objective To investigate the effects of Tannic acid on the proliferation of human colon cancer SW 620 cell line and the mRNA and protein levels of TMEM16A .Methods Human colon cancer cell line SW620 were divided into the low dose(50 μmol/L) ,high dose(100 μmol/L) ,they were cultured for 48 h or 72 h separately .Control groups were cultured in the medium with DMSO .The proliferation of SW620 cell line was detected by the MTT assay at different time points (48 h or 72 h) .The cell cycle and apoptosis in the Tannic acid-treated groups were detected by flow cytometry .RT-PCR and Western blotting were used to de-termine the mRNA and protein levels of TMEM16A separately .All data were analyzed using the one-way analysis of variance (ANOVA) ,SNK test by the SPSS software .Results Compared with the control group ,the proliferation of SW620 cell line was significantly inhibited after the treatment by Tannic acid at the concentration of 50 μmol/L and 100 μmol/L for 48 h or 72 h(t=15 .35 ,P<0 .01 ;t=22 .32 ,P<0 .01) .Determined by flow cytometry ,after treated with Tannic acid ,the numbers of SW620 cells were inhibited in the G0/G1 phase and the S phase(F=6 782 .1 ,1 509 .3 ,P<0 .01) ,and the numbers of SW620 cells in G2/M phase were not varied in each group(F=1 .37 ,P>0 .05) ,and increased apoptotic rate when compared with control group (F=545 .3 ,P<0 .01) .The value of 3H-TdR and 3H-Leucine incorporation of SW620 cells treated with Tannic acid(100 μmol/L) 48 h and 72 h separately ,were obviously decreased as compared with that of control group (P<0 .05) .In the low dose treated groups (50 μmol/L) ,the mRNA levels in 48 h group and 72 h group were(0 .633 ± 0 .009) and(0 .621 ± 0 .011) ,and in the high dose treated groups (100 μmol/L) ,the mRNA levels in 48 h group(0 .64 ± 0 .15) and 72 h group(0 .63 ± 0 .11) ,were lower than the control group(F=7 .645 ,P< 0 .05) .After treating SW620 with Tannic acid for 48 h and 72 h ,in the low dose groups ,the protein expression of TMEM16A were(0 .68 ± 0 .14) and(0 .65 ± 0 .12) ,and in the high dose groups ,the protein expression of TMEM16A were(0 .64 ± 0 .15) and(0 .63 ± 0 .11) were decreased when compared with the control group (1 .28 ± 0 .06)(F=4 .508 ,P<0 .05) .Conclusion Tannic acid arrested SW620 at G1-S phase and decrease the mRNA and protein expression of TMEM16A .
5.A new method for constructing engineered corneal epithelium based on embryonic stem cells

Objective To construct engineered corneal epithelium from embryonic stem cells (ESCs) using Rock inhibitor combined with hypoxia-normoxia culture condition. Methods Human ESC line H1 was induced to differentiate into epithelial-like cells by addition of retinoic acid (RA) and bone morphogenetic protein 4(BMP4) in the differentiation medium under the adherent culture condition. The ESCs derived epithelial-like cells were expanded in the mixed medium of SHEM and KSFM with the mixture ratio of 1 : 2 with or without Rock inhibitor Y27632. The H1 derived epithelial-like cells were seeded on the denuded ammonic membrane to construct engineered corneal epithelium under hypoxia,normoxia and hypoxia-normoxia culture conditions,respectively. The inducted effect of ESCs into epithelial-like cells,the expansion ability of the epithelial-like cells and the characteristics of the constructed engineered corneal epithelium were evaluated by morphological observation, real-time fluorescent quantitative polymerase chain reaction (RTFQ-PCR) and immunofluorescence technology. Results Compared with the control group,the relative expressions of ESCs marker Oct4 mRNA, Notch signaling pathway related factors Notch1 and Jagged1 mRNA,and Wnt signaling pathways related factors c-myc and Cyclin D1 mRNA were significantly reduced, and the relative expressions of cutaneous ectoderm markers p63 and K18 mRNA were significantly increased at day 8 after induction in the induced group,with significant differences between them (t =14.63,20.15,93.50,11.60, 19.30,18.44,22.63;all at P<0.05). Compared with the without Y27632 group,the relative expressions of p63 and K14 mRNA,Notch signal pathway receptor Notch1 and Jagged1 mRNA were significantly increased,and Wnt signaling pathways downstream targeted gene c-myc and CylinD1 mRNA were significantly decreased at day 8 after induction in the Y27632 group,with significant differences between them (t =20.29,59.22,2.90,39.59,5.32,10.14;all at P<0.05),and the relative expression of K18 mRNA in the two groups was not significantly changed(t=1.38,P>0.05). The ESCs derived epithelium and constructed under hypoxia-normoxia culture condition showed more obvious stratification and tighter cell arrangement in comparison with those cells cultured in consistent hypoxia culture condition or normoxia culture condition. Epithelial markers Pan-CK and K18 as well as epithelial progenitor cell markers p63 and K14 expressed in the whole cell layers of the ESCs derived epithelium constructed under hypoxia-normoxia culture condition. Conclusions The addition of Y27632 enhances the proliferation ability of H1 derived epithelial cells and actives Notch signaling pathway and inhibits Wnt signaling pathway. The culture and construction in the expansion medium with Y27632 under the hypoxia-normoxia culture condition can promote the stratification of H1 derived engineered corneal epithelium.
6.The correlation between preoperative serum GGT level and the prognosis in liver cancer patients undergoing living donor liver transplantation

Objective To explore the relationship between the serum gamma glutamyl transpeptidase (GGT) level and the prognosis of liver cancer patients undergoing living dornor liver transplantation.Methods A retrospective analysis was made on 139 liver cancer cases receiving living donor liver Tx from Apri 2005 to Oct 2015 in Sichuan University Huaxi Hospital.GGT cut-off value of 71 U/L was calculated from the ROC curve method (sensitivity was 72.2%,specificity was 59.0%).According to the values of the cut-off patients were divided into high GGT group (73 cases) and low GGT group (66 cases),chi-square test was used to judge its correlation with capillary tumor emboli and Kaplan Meier method to determine the overall survival rate difference.Results The tumor vascular invasion,AFP level,tumor size were in disfavour for higher GGT group (P <0.05).The 1,3,5 year's survival rates were 59.0%,44.5%,39.7%,for high GGT group,and in low GGT group it was 78.6%,63.6%,63.6%,P =0.010).Conclusions The liver cancer patients with high preoperative serum GGT values have poor long-term prognosis after receiving living donor liver transplantation.
7.Application of patient-controlled intravenous analgesia of dezocine combined with sufentanil in burn patients after surgery.

OBJECTIVETo evaluate the efficacy and safety of patient-controlled intravenous analgesia (PCIA) of dezocine combined with sufentanil in burn patients after escharectomy or tangential excision followed by autologous skin grafting.
METHODSSixty burn patients hospitalized in Department of Burns and Plastic Surgery of our hospital from February 2011 to December 2013, conforming to the study criteria and going to have escharectomy or tangential excision followed by autologous skin grafting, were divided into sufentanil group (S, n = 30) and dezocine+sufentanil group (DS, n = 30) according to the random number table. Patients in group S were given 150 mL normal saline containing 2.5 µg/kg sufentanil citrate and 6 mg tropisetron after skin grafting for 48 hours. Patients in group DS were given 150 mL normal saline containing 0.25 mg/kg dezocine, 1.5 µg/kg sufentanil citrate, and 6 mg tropisetron for 48 hours. Visual Analog Scale (VAS), Bruggrmann Comfort Scale (BCS), and Ramsay Sedation Scale were used to evaluate the sedative effect or analgesic effect, and their scores were recorded at administration hour (AH) 2, 6, 12, 24, and 48. The times of efficient injection and incidence of adverse effect within the 48 AH were recorded. Data were processed with analysis of variance for repeated measurement, t test, chi-square test, and Fisher's exact test.
RESULTSThere were no obvious differences in the scores of VAS and BCS between two groups at each time point (with t values from -0.426 to 0.864, P values above 0.05). The scores of Ramsay Sedation Scale in group S at AH 2, 6, 12, 24, and 48 were respectively (3.2 ± 0.6), (3.2 ± 0.5), (3.3 ± 0.7), (3.2 ± 0.4), and (3.3 ± 0.4) points, which were higher than those in group DS [(2.4 ± 0.6), (2.5 ± 0.5), (2.4 ± 0.6), (2.4 ± 0.4), and (2.4 ± 0.5) points, with t values from 5.302 to 8.391, P values below 0.001]. The times of efficient injection within the 48 AH was 6.8 ± 0.7 in group S and 6.5 ± 0.9 in group DS, showing no significantly statistical difference (t = 1.260, P > 0.05). Respiratory depression was not observed in both groups; the incidence of pruritus was the same, and that of urine retention was similar between the 2 groups within the 48 AH (with P values above 0.05). Within the 48 AH, the incidence of nausea and vomiting in group S was 26.7% (8/30), which was obviously higher than that in group DS (6.7%, 2/30, P < 0.05); the incidence of drowsiness in group S was 20.0% (6/30), which was significantly higher than that in group DS (no patient, P < 0.05).
CONCLUSIONSDezocine combined with sufentanil can provide effective postoperative analgesia with little adverse effect for PCIA in burn patients after escharectomy or tangential excision followed by autologous skin grafting, therefore it can be widely used.
