1. Preparation, identification and application of polyclonal antibodies against human PIWIL2 protein

Objective: To prepare rabbit polyclonal antibody against human P-element-induced wimpy testis like 2 (HIWI2) protein, identify its properties and investigate its distribution in normal and tumor tissues by means of tissue chip. Methods: PIWIL2 peptide was synthesized chemically and conjugated to Keyhole limpet hemocyanin (KLH) as an immunogen. Then the PIWIL2-KLH conjugation was injected into rabbits subcutaneously to produce polyclonal antibodies. The specificity and sensitivity of antibodies were identified by ELISA and Western blotting after purification by affinity chromatography. PIWIL2 was then immuno-stained on the tissue chip to study its distribution in the normal and tumor tissues. Results: Rabbit's antibodies against human PIWIL2 were prepared after the injection of PIWIL2-KLH conjugation subcutaneously. These antibodies were identified as PIWIL2 peptides by ELISA and Western blotting assay. PIWIL2 protein expression was tissue-specific in tumor tissues, with PIWIL2 protein found in the cytoplasm of smooth muscle cells of most normal and tumor tissues. Conclusion: The polyclonal antibodies against human PIWIL2 protein have been successfully prepared, which provides a basis for further study on the role of PIWIL2 in the pathway of miRNA/RNA.
2.The long term result of the acute closed rupture of achilles tendon treated by the short incision and tenocutaneous relaxing suture

[Objective]To evaluate the long term effect of the short incision and tenocutaneous relaxing suture to treat the acute closed rupture of the Achilles tendon.[Method]28 cases were treated by short incision and tenocutaneous relaxing suture from 1996-2005,The scar tissue and blood blocks between the ends of ruptured Achilles tendon were first debrided completely,then treated with percutanneous decmpression suture,the duration of follow up ranged from 2 years to 10 years.The treatment outcome were evaluated by Arner-Lindholm evaluation system and the MRI of post operation.2 cases were lost.[Result]According to the Arner-Lindholm evaluation system and the MRI of post operation,the treatment outcome were excellent in 18,and good in 6,with excellent rate of 92%,No complication including nerve injury and infection were found after operation,except 1 rerupture of Achilles tendon was found during the follow up period.The MRI showed the rupture Achilles tendon were repaired and remodeled very well in all cases.[Conclusion]The short incision and tenocutaneous relaxing suture is an ideal surgical technique,with good Clinical result and less Significant complication.
3.The imaging analysis of intracranial malignant teratomas

Objective To analyse the imaging characters of intracranial malignant teratomas (IMT). Methods The imaging appearances of IMT were analyzed in 15 pathologically proved IMT. MRI were performed on all the 15 cases, CT on 8 of them. Results Thirteen IMT showed as a lobular cyst solid mass with clear borders. The solid portion of IMT displayed iso (6/13) to hypointense(7/13) on T 1WI and iso (2/13) to hyperintense(11/13) on T 2WI. The heterogeneous enhancement was found in 11 cases, homogeneous in 2. Two IMT exhibited as the intrasellar solid lesions involving thalamencephalon. Calcification was found in 8 IMT on CT scan. No fat was demonstrated in the tumor on both CT and MR. Conclusion IMT displays relatively typical appearances on imaging studies. Together with the CT scan, The MR study may make the diagnosis before the operation.
4.Preparation and biomedical application of superparamagnetic iron oxide

BACKGROUND:Superparamagnetic iron oxide has been widely used in biomedical applications, especialy in the field of targeted diagnosis and therapy. OBJECTIVE:To summarize the physicochemical property, preparation, surface modification, product testing and biomedical applications of superparamagnetic iron oxide. METHODS: In order to search relevant articles about the physicochemical property and biomedical applications of superparamagnetic iron oxide from PubMed, Sciencedirect, CNKI and Wanfang databases (from 2000 to 2014), a computer-based search was performed using the keywords of “superparamagnetic iron oxide, preparation, coprecipitation, surface modification, magnetic resonance imaging contrast agent, fluorescent tracing, targeted therapy” in English and Chinese, respectively. RESULTS AND CONCLUSION: The superparamagnetic iron oxide can be produced in the laboratory, and the preparation methods include hydrothermal method, vapor deposition method, mechanical miling method, liquid-phase microwave dielectric heating method, sol-gel method, emulsion method, co-precipitation method. Moreover, the superparamagnetic iron oxide can have different properties by surface modification and be applied in many biomedical fields, such as magnetic resonance contrast agent, fluorescent tracer, nanoparticle targeted therapy, hyperthermia and bioseparation.
6.Clinical detection and identification of seven species human herpesviruses by DNA microarray technique

Objective To establish a method for rapid diagnosis and identification of seven species human herpesviruses infection.Methods Primers and oligonucleotide probes were designed and synthesized based on the highly conserved regions of the DNA polymerase genes in human herpesviruses, namely herpes simplex virus type 1 (HSV-1),HSV-2,varicella-zoster virus (VZV),Epstein-Barr virus (EBV),cytomegalovirus (CMV),and human herpes virus 6 (HHV-6A/6B).DNA microarrays were made by printing the oligonucleotide probes on the special glass slides.A total of 282 blood specimens from children with suspected infection were analyzed by this DNA microarray technique,and the results were compared with those of TaqMan PCR.Results The products of the seven human herpesviruses after PCR amplification could be used to identify the virus species with DNA microarrays.The detection limits were 10 copies/?l for HSV-1,HSV-2,VZV,EBV,CMV,HHV-6A,and HHV-6B,respectively.The assay did not show cross-reaction to the DNA extract of hepatitis B virus,staphylococcus aureus,E.coli,Candia albicans and human genome.Among the 282 samples,59 were positive for human herpesviruses DNA.Compared with those of TaqMan PCR,the sensitivity and specificity of the microarray assay were 96.7% and 99.5%,respectively,and the index of accurate diagnosis was 0.962.Conclusions This DNA microarray for identifying human herpesviruses species is specific and sensitive,and may serve as an efficient technique for simultaneous detection and species identification of human herpesviruses in clinical specimen.
7.One case report of nasal vestibule aggressive fibromatosis.

The patient had found local uplift on the left nasal vestibule area for 3 years. An 2.0 cm x 1.0 cm mass was found on the left nasal vestibule area, which had tough texture, clear boundaries and no tenderness. In the operation, the tumor was found located between the left maxilla and major alar cartilage with no capsule and unclear boundary. The tumor is aggressive fibromatosis, which has invasive growth in adjacent muscle tissue. Immunohistochemical results: Ki-67, Vimentin(+), SMA(+), CD31(+), CD34(+), CK(-), Desmin (-), S-100(-), LCA(-).

8.Research of correlation between MRI fluid-attenuated inversion recovery hyperintense vessel sign and prognosis in acute middle cerebral artery infarction

Objective To investigate the significance of MRI T2 fluid-attenuated inversion recovery hyperintense vessel sign (FLAIR HVS)in clinical prognosis evaluation of the patients with acute middle cerebral artery irnfarction.Methods The data in 57 inpatients with acute middle cerebral artery infarction in our hospital from Aug.2013 to Aug.2015 were retrospectively analyzed.All cases were performed the intact MRI examination(ineluding FLAIR,DWI and MRA)and CTA.The infarct volume with DWI,national institute of health stroke scale(NIHSS)score and modified Rankin Scale(mRS)score on 30 d after discharge were performed the comparative analysis.Results Fifty-seven cases of middle cerebral artery occlusion were divided into the distal HVSgroup and non-distal HVS group(8 cases in proximal HVS group,21 cases in HVS negative group).The infarction volume of DWI sequence,NIHSS scores at admissiom and discharge and mRS score on 30 d after discharge in the distal HVS group were superior to those in the non-distal HVS group(P<0.05).Conclusion MRI-T2 FLAIR sequence HVS has certain reference value in the prognosis evaluation in the patients with middle cerebral artery occlusion.
9.Effect of ginkgolide B on expression of Foxg1 gene and proliferation of cells in brain tissue of newborn rats with hypoxic-ischemic brain damage

Objective To observe the effects of ginkgolide B (GB) on mRNA expression of foxgl and proliferation of cells in brain tissue of newborn rats with hypoxic-ischemic brain damage (HIBD).Methods A total of 128 clean 7-day-old healthy SD rats were randomly divided into sham operation group,the model group,the low-GB dose and the high-GB dose treatment groups.Classic Rice method were used to establish HIBD models in the latter 3 groups.Four hours after operation,and GB in dose of 5 mg/kg and 10 mg/kg was given to rats in the low and the high dose treatment groups by intraperitoneal injection postoperatively,once a day for 5 days,while sham operation and model groups were treated with equal physiological saline.All groups were respectively sacrificed on 3 d,7 d,14 d,28 d respectively.Quantitative real-time fluorescent polymerase chain reaction was employed to detect expression of Foxg1 gene.Then the number of 5-bromodeoxyuridine positive cell in subgranular zone was investigated by immunolluorescent stairning.Results The Foxg1 mRNA expression was observed 3 days after HIBD,peaked on 7th day,and then declined gradually; the levels of Foxg1 mRNA in the 2 treatment groups were higher than that of the HIBD group (all P < 0.01) ; The expression of Foxgl at 7 d,14 d,28 d,in high-dose group were higher than those in the low-dose group (all P < 0.01).The number of 5-bromodeoxyuridine positive cell was increased after HIBD,and the levels in the low-and the high-dose treatment groups were all higher than that of the model group (all P < 0.05) ; the number of positive cell in high-dose treatment groups were higher than that in the low-dose treatment groups (P < 0.05).Conclusions GB can promote the expression of Foxg1 gene and improve the proliferation of cells in Brain tissue after HIBD,which shows more significant efficacy in high-dose group than in low-dose group.
10.Effects of chemotherapy on free radicals in the rat glioma

AIM: To observe the role of free radicals in the inhibitory effect of chemotherapy on glioma cells. METHODS: C6 glioma cells were cultured in vitro , and treated with carmustine (B), teniposide (V), or/and nimodipine (N). Furthermore, the glioma-bearing rats were treated with B plus N, B+V+lisplatin (D)+N, or B+V+D+N+angiotensin Ⅱ. The MDA content and superoxide dismutase (SOD) activity in the culture supernatant and cortical brain tissue were assayed. RESULTS: B, V and N significantly decreased MDA content and SOD activity in the supernatant of glioma cell culture and C6 glioma cells. Chemotherapy reduced MDA content and increased SOD activity in the cortical brain tissue of tumor-bearing rats, with highest efficiency in B+V+D+N+angiotensin Ⅱ group. The survival time of tumor-bearing rats in B+V+D+N+angiotensin Ⅱ group was longer than that in other chemotherapy group. CONCLUSION: The antitumor effects of combined chemotherapy may be involved in the free radical metabolism. [
