2.Analysis and application of the images from the segmenting CT in the space of nasal endoscope

Objective To analyze the images from the segmenting CT in the space of nasal endoscope and to observe the benefits in applying the method in endoscopic sinus surgery. Methods Five patients with inverted papilloma and ten patients with chronic rhinosinusitis were enrolled. The images from the coronal CT were put in the ideal model of the nasal cavity, which had been designed by SketchUp software package, and the space of the model in the 0° rod-lens telescope was simulated and observed according to definite elevation angle between the endoscope and bottom of nasal cavity and the distance of the endoscope entering the nasal cavity. The surgical plan was designed and compared with the models of the intraoperative corresponding vision. Results The anatomical form of the images from the coronal CT and the directions of the space in the visual field of the endoscope were correlative with the elevation angle between the endoscope and bottom of nasal cavity and the distance of the endoscope entering the nasal cavity. Based on the elevation angle and the entering distance of the endoscope, the images from the CT scan in the space of nasal endoscope were analysed according to the order, which were consistent with the findings in the intraoperative corresponding vision, providing the accurate directions of the space for the endoscopic sinus surgery. Conclusions The changes of the images of the CT scan in the space of the endoscope were consistent with the perspective law. To know the perspective law could grasp the directions of the space in the visual field of the endoscope and ensure the safety of the endoscopic surgery. The method was simple and feasible. The benefits of this technique were confirmed by clinician.
3.Study of the key points and safety measures during perioperative period in patients with obstructive sleep apnea hypopnea syndrome

Objective To study the safety measures during perioperative period in patients with obstructive sleep apnea hypopnea syndrome (OSAHS) , to minimize uvulopalatopharyngoplasty (UPPP) major complications. Methods The complications and other relative information of 1446 OSAHS patients treated with UPPP were analyzed. Complications of 1004 OSAHS patients adopted standardized management measures after 2004 and 442 OSAHS patients who performed UPPP before 2003 were compared. Results Among the 1446 cases of OSAHS surgery, there were 49 cases (3. 39% ) of difficult intubation; 8 cases (0.55%) intraoperative mild arrhythmia; 13 cases (0. 90%) of surgery dangerous situations after extubation; 19 cases (1. 31%) of intraoperative primary hemorrhage; 11 cases (0. 76%) of mild arrhythmia during the postoperative observation period; 21 cases (1.45%) of uncontrolled hypertension immediately after surgery; 32 cases of secondary hemorrhage a week after surgery. All complications were cured and no death occured. Compared with those operations before 2003, complications were significantly reduced with enforcement of standardized management of perioperative measures since 2004. The data had significant difference by χ2 test( P <0.05). Conclusions There are five key points during the periperative period of OSAHS surgery: preoperative examination, intubation, surgery, postoperative extubation, and postoperative care. Risk factors and complications could be prevented effectively in the above five important processes.
4.Observation on therapeutic effect of acupuncture, moving cupping and blood-letting puncture on chloasma.

OBJECTIVETo compare therapeutic effects of acupuncture, moving cupping, blood-letting puncture and medicine on chloasma.
METHODSOne hundred and seventy-six cases of chloasma were randomly divided into two groups. The acupuncture, moving cupping, blood-letting puncture group was treated with acupuncture at Ashi points, Xuehai (SP 10), Sanyinjiao (SP 6), moving cup at The Governor Vessel and The Urinary Bladder Channel on the back and then blood-letting puncture at Dazhui (GV 14), Feishu (BL 13), Geshu (BL 17), Xinshu (BL 15), Ganshu (BL 18) and nearby more severe purple stasis regions, and the medication group was treated with oral administration of Vit E and Vit C. Their therapeutic effects were observed after 3 courses.
RESULTSThe total effective rate was 96.7% in the acupuncture, moving cupping, blood-letting puncture group and 51.2% in the medication group with a significant difference between the two groups (P < 0.05).
CONCLUSIONThe therapeutic effect of acupuncture, moving cupping, blood-letting puncture on chloasma is better than that of the medicine.

5.Gene transfection of NGF and/or Noggin affecting the differentiation of bone marrow-derived mesenchymal stem cells

Objective To explore the feasibility of exogenous NGF and/or Noggin gene transfecting into the bone marrow-derived mesenchymal stem cells (BMSCs) and observe the differentiation of BMSCs modified by NGF and/or Noggin.Methods BMSCs were isolated from SD rat and purified by adherent method and these cells were identified by their phenotypical properties and their abilities of differentiating into adipocytes.Ad-GFP-NGF and/or Ad-GFP-Noggin were transfected into BMSCs.The protein expressions of NGF and/or Noggin were detected using immunocytochemistry and Western blotting.The differentiations of gene modified BMSCs were observed by immunohistochemistry.Result The cells selected by adherent method had basic phenotypical properties of BMSCs and could differentiate into adipocytes.BMSCs without transfection and those transfected with Ad-GFP expressed low level of NGF without Noggin expression.All gene modified BMSCs could express NGF and/or Noggin.After transfection, BMSCs could differentiate into cells having neuronal morphology and expressing NF (H).The combined transfection group had the highest ratio of NF(H)+ cells among all the groups.Conclusion BMSCs can be isolated and purified from rat bone marrow by adherent method.Ad-GFP-NGF and/or Ad-GFP-Noggin can transfect BMSCs safely and the transfected cells can express those proteins persistently and efficiently.NGF and Noggin can induce BMSCs into neuron-like cells in vitro and when they exist simultaneously,the differentiation is further enhanced.
6.Brain-derived neurotrophic factor prevents against amyloid beta protein-induced impairment of hippocampal in vivo long-term potentiation in rats.

OBJECTIVETo explore the effects of brain-derived neurotrophic factor (BDNF) pretreatment on beta amyloid protein (Abeta) induced impairment of in vivo hippocampal long-term potentiation (LTP) in the CA1 region of rats.
METHODSThirty-six adult male SD rats were randomly divided into six groups (n = 6): control, Abeta25-35, BDNF, (0.02 microg, 0.1 microg, 0.5 microg) BDNF + Abeta25-35. A self-made hippocampal local drug delivery catheter and a parallel bound stimulating/recording electrode were used to deliver drugs/stimulation and record field excitatory post-synaptic potentials (fEPSPs) in the hippocampal CA1 region of rats. High-frequency stimulation (HFS) was used to induce in vivo LTP.
RESULTS(1) Abeta25-35 (2 nmol) injection into CA1 region of rats did not affect the baseline fEPSPs, but inhibited the HFS-induced LTP significantly (P < 0.01). (2) Hippocampal CA1 injection of BDNF (0.1 microg) alone did not affect the baseline fEPSPs and HFS-induced LTP. (3) Compared with Abeta25-35 alone group, the averaged amplitude of LTP in BDNF (0.1 microg and 0.5 microg) plus Abeta25-35 groups significantly increased at 0 min, 30 min, and 60 min after HFS (P < 0.01), indicating that pretreatment with BDNF effectively protected against the Abeta,25-35 induced depression of LTP in a dose-dependent manner.
CONCLUSIONIntrahippocampal injection of BDNF can protect against the Abeta25-35-induced LTP impairment, suggesting that the up-regulation of BDNF in the brain could maintain the normal hippocampal synaptic plasticity and may contribute to the improvement of learning and memory in Alzheimer's (AD) disease patients.

7. The plasma level of amyloid-β is associated with cognitive decline: a two years follow-up study in Xi′an rural areas

Objective:
To explore the relationship between plasma amyloid-β (Aβ) and cognitive decline during 2 year follow-up in a population-based cohort in Xi′an rural areas.
Methods:
The study was conducted in Qubao village in Xi′an suburbs cognitively normal residents over 40 years old were recruited from October 2014 to March 2015 and given a face-to-face standardized interview. Mini-mental state examination (MMSE) was employed to evaluate the global cognitive function, and quantification of plasma Aβ was measured by sandwich enzyme-linked immunosorbent assay (ELISA) at baseline. Two years later, MMSE was tested at the end of study. Then logistic regression was performed to analyze the relationship between baseline Aβ and cognitive change during 2 year follow-up.
Results:
A total of 1 020 participants completed the study, among whom 223 subjects (21.9%) presented MMSE scores decline (defined as MMSE scores decreased ≥2 points). Compared with those without decline, participants in the MMSE decline group were older (

9.Construction of the recombinant adenovirus vector bearing dual-survivin shRNA.

OBJECTIVETo construct a recombinant adenovirus vector bearing dual-survivin short hairpin RNA (shRNA).
METHODSDual-survivin shRNA was designed and synthesized respectively, both inserted into adenovirus DNA. The recombinant adenovirus vector was confirmed via both sequencing and restriction digestion analysis, and then linearized and transfected into the HEK 293 cell line to generate recombinant adenoviruses.
RESULTSThe recombinant adenovirus vector was constructed and the target sequence was obtained.
CONCLUSIONThe construction of the recombinant adenovirus vector provides a basis for the research of potential gene therapy for prostate cancer.

10.Morphologic observation of oral cancer cells cocultured with mesenchymal cells in vitro.

OBJECTIVETo study the morphologic and growing alterations of oral cancer cell line Tca8113 before and after cocultured with tumor stromal fibroblasts (TSF) and normal stromal fibroblasts (NSF) respectively, and evaluate the influence of mesenchymal cells on tumor cells.
METHODSTSF and NSF were isolated and cultured. To observe the morphologic change of Tca8113 cells after cocultured with TSF and NSF respectively.
RESULTSWhen cocultured with NSF, the Tca8113 cells proliferated as rapidly as monocultured to form colonies, while the NSF proliferated slowly to form pieces and then joined each other to form network. The NSF network segmented and surrounded the colonies of cancer cells so that the cancer cells shrank, turn round, broke away from the bottom and floated into the medium. The cancer cells proliferated actively but they were elbow out entirely in the end. TSF proliferated slowly when cocultured with cancer cells, projected several branched protrusions. The cancer cells proliferated along the two sides of protrusions of TSF, or projected short protrusions to connect the body or protrusions of TSF, and overlaid the protrusions gradually, finally, cover the body. In the end, TSF melt away, and the cancer cells took on the figure of TSF.
CONCLUSIONThe results do suggest that, oral cancer cell line Tca8113 are restrained when coculture with NSF, but are promoted when with TSF.
