2.Preparation and in Vitro Release of Risperidone Injectable Biodegradable Implants

OBJECTIVE:To prepare risperidone injectable biodegradable implants and study its in vitro release. METHODS: Polylactide-co-glycolide copolymer(PLGA)was chosen for carrier and N-Methyl pyrolidone(NMP)as solvent to prepare risperidone injectable biodegradable implants. The percentage of accumulative drug release within 30 days,the change in molecular weight of copolymer and superficial opening diameters of copolymer in function time were observed. RESULTS:Drug release curve of risperidone injectable biodegradable implants for 30 days was stable with small burst. The percentage of accumulative drug release was 89.01% and the percentage of burst accounted for 13.8% at the first 24 hours. The average molecular weight of copolymer decreased to about 10 000 from 43 000 after 30 days and the size of the superficial openings became larger and larger. CONCLUSION:Risperidone injectable biodegradable implants can release continuously and stably in vitro for 30 days.

AIM: Hydroxymethylglutaryl CoA (HMG-CoA) reductase inhibitors, such as simvastatin, have been shown to reduce atherosclerotic cardiovascular morbidity and mortality by mechanisms unrelated to its lipid-lowering effect. Several studies have shown that simvastatin induces apoptosis in a varieties of cell lines including vascular smooth muscle cells (VSMC). The aim of this study was to investigate the signal pathways involved in apoptosis induced by simvastatin. METHODS: Cultured VSMC were treated with simvastatin. Calpain activity was determined by measuring Ca 2+ ionophore-specific calpain substrate (suc-LLVY-AMC), caspase-3 activation was detected by Western blot, and apoptotic changes were distinguished by annexin Ⅴ binding and DNA laddering. 4.An analysis of values of massage to facilitate the rehabilitation of the women in puerperium

Objective To discuss the valuable influence of the total-body massage on the facilitation of the rehabilitation for the women in puerperium.Methods Both Chinese traditional medical total-body massage and the biological information theories and its methodology were employed in dealing with 50 cases of puerperants without contraindications,who had labored naturally 12 hours before and who had received cesarean sections 48 hours before. The results were compared with the control group in which conventional preventive health care for puerperants was adopted.Results Notable differences exist in the aspect of lochia,defecation,lactation and emotions between the two groups.Conclusion Significant benefits can be obtained from the massage to facilitate the rehabilitation of the women in puerperium while promoting their quality of lives and broadening the coverage of services of the preventive health care for the puerperants.
5.Clinical Significance and Influence on Glucocorticoid Receptor Resulted from Tripterygium Wilfordii Hook F for Children with Nephrotic Type of Henoch-Schonlein Purpura Nephritis

Objective To explore the mechanism of tripterygium wilfordii hook f(TWHF)for Henoch-Schonlein purpura nephritis(HSPN,nephrotic type)in children.Methods All the children with HSPN(nephrotic type)were divided into 3 groups casually.Method of radioactive pairs conjugating analysis was used to determine the numbers of glucocorticoid(GC)receptors(GCRs).Results were statistically analysed respectively.Results GCRs tested in all the patients with HSPN(nephrotic type)before treatment were statistically lower than those in control group of normal children;GCRs in all the patients tested after 1 week treatment with GC were lower than before treatment.Three weeks later,GCRs in patients treated with TWHF increased much higher than them without TWHF.Following up the recurring times of all the patients 3 months to 5 years,they were much more in patients treated without TWHF treatment than in patients treated with it.Conclusions TWHF treatment on HSPN(nephrotic type)is obviously effective.It can not only decrease the dosage of GC,but decrease the recurring times.One of the mechanism may be TWHF can resist the down-regulation GC to GCR,and enhance the effect of GC.

8.Changes of Serum Zinc Level of Rotaviral Enteritis in Children and Therapeutic Effect of Zinc

Objective To explore the change of serum zinc level of children with rotavirus(RV) enteritis and efficacy of zinc supplementation on children with rotaviral enteritis.Methods One thousand one hundred and eighty-three healthy children were enrolled in this study,who were divided into different groups,and the zinc levels of children in every different age group were detected.These levels were used as normal levels of children in Dongguan district.One hundred and forty-three diarrhea patients in this 2 years were enrolled.Confirmed dia-gnosis was depended on the RV test in patients′ stool.The children with diarrhea were randomly divided into zinc treatment group (n=72) and conventional therapy group (n=71).The serum zinc levels in RV enteritis was analyzed.The patients in conventional therapy group were received diarrhea conventional therapy inchuded smectite and fluid infusion and so on;in addition to diarrhea conventional therapy,zinc gluconate 20 mg/d (10 mg/d below 6 months of age) for successive 14 days were given the patientsin zinc treatment group.The different efficacy of the 2 groups and the changes of serum zinc in treatment group were analyzed.Results Compared with normal children,the ones suffering from RV enteritis had lower serum zinc level(P0.05).Conclusions The ones suffering from RV enteritis have lower serum zinc level.In addition to diarrhea conventional therapy,giving zinc treatment has better effect on enhancing curative effect of RV enteritis.On the other hand,the suggestion of zinc supplement made by WHO is also suitable for Chinese children.

10. The role of liquid biopsy of cerebrospinal fluid in the diagnosis and treatment of central nervous system tumors

The central nervous system tumor is a common tumor seen both in children and adult. It is difficult to remove by surgery, and its mortality rate is high. Early diagnosis and real-time monitoring of this tumor are difficult due to drawbacks of the current laboratory tests (imaging, cell morphology, cytogenetics), such as low sensitivity, low positive rate and difficulty in sampling. The cerebrospinal fluid, which can circulate rapidly between ventricle and spinal cord pool, is fully in contact with the central nervous system, thus containing a great number of metabolic materials from the central nervous system. For this reason, the cerebrospinal fluid is an ideal sample for detecting central nervous system tumors. Compared with the conventional biopsy, the liquid biopsy of cerebrospinal fluid has many advantages, such as less-invasive, sampling successively, reflecting the progression of disease immediately and adjusting the treatment regimen flexibly. This paper will briefly review the molecular targets of liquid biopsy such as DNA, RNA and circulating tumor cells, elaborate the role of novel technologies including next-generation sequencing and digital PCR in the diagnosis and treatment of the central nervous system tumors.(
