2.The effects of phenytoin on the differentiation of rat bone marrow mesenchymal stem cells into vascular endothelial cells

Objective:To investigate the effects of PHT on the differentiation of rat bone marrow mesenchymal stem cells(BMSCs) into vascular endothelial cells(VECs).Methods:rBMSCs were cultured in indirect coculture system with VECs and independent culture system in the presence of phenytoin at 0,20 and 40 μg/ml respectively.After 1 4 d culture the mRNA expression levels of the intercellular adhesion molecule-1 (ICAM-1 ),vascular cell adhesion molecules-1 (VCAM-1 ),vascular endothelial growth factor re-ceptor -2(KDR)and Runt-related transcription factor 2 (RUNX2)were detected by real-time PCR.Results:The expression levels of ICAM-1 ,KDR and RUNX2 were upregulated in rBMSCs by PHT treatment.The expression levels of ICAM-1 ,KDR and RUNX2 in the co-culture groups were higher than those in the independent culture groups of rBMSCs treated by PHT at the same concentra-tion.The expression level of VCAM-1 in co-culture groups was lower than that in the independent culture groups.Conclusion:PHT may promote rBMSCs differentiation into rVECs in the co-culture system.
3.One case of pediatric obstructive sleep apnea hypopnea syndrome with pulmonary hypertension.

The clinical manifestation included snoring and mouth breathing for 2 years, repeated coughing and shortness of breath in action for more than 1 year. Physical examination of oral cavity showed tonsils were in grade III. The endoscopy showed 2/3 of postnaris were blocked by the adenoids. The preoperative ultrasonic cardiogram revealed the right atrial and right ventricular dilatation, pulmonary artery widened. The preoperative polysomnography (PSG) showed apnea-hypopnea index (AHI) was 28.5 events an hour, and the lowest oxygen saturation (LSaO2) was 39%. The patient was diagnosed as severe obstructive sleep apnea hypopnea syndrome with pulmonary hypertension. The postoperative PSG showed the AHI was 11.7 events an hour, and the LSaO2 was 86%. The ultrasonic cardiogram at 5 months after surgery didn't show any abnormalities.

4.Professor He Jialin's Experiences in Treating Leucorrhoea Disease

Objective] Summarize the experience of diagnosis and treatment of leucorrhoea disease by professor He Jialin. [Method] By collecting medical records, from the pathogenesis, syndrome differentiation, to expound the academic viewpoints and clinical experience of professor He Jialin in treating leucorrhoea disease, summarize its academic characteristics on integrating traditional Chinese and western medicines, internal and external treatments and TCM prescription, and exemplify that. [Results] Professor He Jialin believes that the major pathogenesis is dampness,with intrinsic deficiency in spleen and kidney, explicit accumulation of damp and heat.Combined with western medicine examination methods, professor He Jialin treats the disease by invigorating the spleen for damp elimination, nourishing kidneys to consolidate thoroughfare vessel, heat-clearing and detoxicating, each therapeutic principle combined with external therapy receives a good effect. [Conclusion] Professor He Jialin's academic thoughts and clinical experience on syndrome differentiation, application method, addition or subtraction of changes provide a more complete mentality and pragmatic value in treating leucorrhoea disease.

6.RNAi silencing of XIAP gene induces apoptosis and susceptibility to chemotherapy of osteosarcoma cell line MG63

Objective To explore the effect RNAi-mediated inhibition of X-linked inhibit apoptosis protein (XIAP) gene on apoptosis and susceptibility to chemical therapy drugs in osteosarcoma cells and its underlying mechanism.Methods shRNA plasmid of XIAP gene were constructed. MG63 cells were conventionally cultured and divided into untreated group (group A,non-transfection),blank plasmid group (group B,psiRNA-Con transfected),the experimental group (group C,psiRNA-XIAP transfected),and 2 chemical drug interfered groups,non-transfection+0.3 ?mol Dox group (A+Dox group) and psiRNA-XIAP transfection +0.3 ?mol Dox group (C+Dox group). The XIAP expression in MG63 cells was detected by Western blotting. The proliferation of above-mentioned MG63 cells was examined by MTT assay. 7.Recent Advances in RNA Riboswitch

Riboswitch is a novel type of posttranscriptional regulatory elements discovered by Breaker et al. in 2002. It can regulate gene expression by binding directly to small metabolites without the aid of protein molecules. Compared to normal protein-mediated regulation, riboswitch responds to metabolites more rapidly and sensitively. Its discovery opens a new world for RNA research. The recent advances in riboswitch researches were summarized, including crystal structure determination, mechanism and dynamics study, biosensor and antibacterial drug design. Topp et al. successfully reprogrammed E. coli to detect, follow, and precisely localize to a completely new chemical signal by using a synthetic riboswitch. This work provided new ideas for synthetic biology and artificial biology network. The advances in riboswitch 3D structure determination, reaction mechanism and dynamics provide useful information for rational drug design towards new generation of riboswitch-targeting antibacterials.
8.Differential Expression of Synaptophysin in the Developing Rat Cochlea

Objective To investigate the differential expression of synaptophysin(SYN)in the developing rat cochlea,to analyze the relationship between the expression of SYN and auditory functional development,and to explore the source of the adenosine triphosphate(ATP)in the cochlea.Methods The immunofluorescence was applied to observe the differential expression of SYN in the cochlea of SD rats on different days(p1,p5,p10,p14 and p28)after parturition.Results No significant stain was observed in the Kolliker organ or the Corti's organ of P1,P5 and the top turn of the cochlea in P10.The SYN expression was turned on the outer spiral bundle,the inner spiral bundle,and the medial wall of the Deiters' cell of the cochlea in P14,P28 and the second or the third turn of Corti's organ of P10.All the spiral ganglion neurons(SGN)were stained in different aged cochlea.Conclusion The expression of SYN was different during development of rat cochlea.This difference was in favor of the configuration constructed between nerve endings and target cells.It also played a key role in the formation of correct auditory coding during auditory system development.The hair cells and the supporting cells,possible the source of ATP in the cochlea,contained ATP that maybe existed in a non-vesicular form or specific staining of non-SYN.
9.The relationship between ultrasonic feature of lower limb peripheral nerve and Toronto clinical scoring ;system in type 2 diabetes mellitus

Objective To evaluate the morphological changes of the small nerves in lower extremity with high-resolution ultrasonography in type 2 diabetes mellitus (T2DM). Methods 84 cases of T2DM patients were divided into four different groups according to TCSS score [group Ⅰ (0-5 point), group Ⅱ (6-8 point), groupⅢ (9-11 point), group Ⅳ (12-19 point)]. The ultrasonic feature of posterior tibial nerve, sural nerve and saphenous nerve were observed to observe the relationship between ultrasound findings and scoring. Results Abnormality rate admitted from ultrasound findings are as follows. In group Ⅳ, nerve abnormality rate was the highest, followed by group Ⅲ, group Ⅱ and group Ⅰ (P < 0.05). The major manifestations are obscure border between spineurium and peripheral tissue , nerve bundle mesh like structure disappearing , enlargement of posterior tibial nerve and sural nerve. There was no statistical difference in irregular abnormality rate (P > 0.05). The saphenous nerve epineurium abnormality increased in group Ⅱ and Ⅲ compared with group Ⅰ and Ⅳ(P <0.05). Abnormality rate of 3 neural ultrasound decreased in line with sural nerve, posterior tibial nerve and saphenous nerve. Conclusions Certain correlation was observed between TCSS and never ultrasonic feature in T2DM patients. The higher TCSS scores is, the greater chance of presence of abnormal sonographic features of nerves will be. The sural nerve abnormality was higher than others.
10.Changes of cytokine levels before and after anti-tuberculosis treatment

Abstract: Objective To analyze the changes of cytokine levels in patients with sensitive pulmonary tuberculosis and drug-resistant pulmonary tuberculosis before and after anti-tuberculosis treatment. Methods The clinical data of 213 pulmonary tuberculosis patients admitted to the Eighth Affiliated Hospital of Xinjiang Medical University from August 2019 to October 2020 were collected, and divided into a sensitive pulmonary tuberculosis group and a drug-resistant pulmonary tuberculosis group, of which 141 were in the sensitive group and 72 in the drug-resistant group , and the pro-inflammatory factors tumor necrosis factor-α (TNF-α), IL-6, IL-17, interferon-γ (IFN-γ) and anti-inflammatory factors IL-10 and IL-4 in plasma before treatment, 3 months and 6 months after treatment were compared. Results Compared with the healthy group, the levels of plasma IL-6 in the sensitive group and the resistant group were significantly increased, difference was statistically significant (P<0.01). The plasma levels of IFN-γ and IL-17 in the sensitive group were increased, and the difference was statistically significant (P<0.05). Compared with before treatment, the levels of IL-6 and IL-4 in the plasma of the sensitive group after 6 months of treatment were lower than those after 3 months of treatment, and the difference was statistically significant (P<0.05), and IFN-γ was significantly decreased (P<0.001). After 6 months of treatment, the IL-10 content was lower than that before treatment, and the difference was statistically significant (P<0.05), and the IFN-γ decreased significantly (P<0.000 1). The levels of IFN-γ at 3 months were higher than those in the resistant group, and the difference was statistically significant (P<0.01). After 6 months of treatment, there was no significant difference in the content of IFN-γ between the sensitive group and the resistant group. The effective rate of CT absorption was 70% in the sensitive group and only 43.33% in the resistant group at 3 months of treatment, and there was no significant difference in the effective rate between the two groups after 6 months of treatment. Conclusions Detection of pro-inflammatory cytokines and anti-inflammatory cytokines in plasma can provide reference for host-directed therapy of pulmonary tuberculosis. Understanding this important feature of cytokines on pathogen-host interactions can provide new insights into the study of virulence mechanisms and may provide new approaches for immune intervention in pulmonary tuberculosis.
