3.Expression of survivin in human dermal hemangioma and its relationship with caspase-3

Objective To investigate the expression of survivin and its relationship with that of caspase-3 in two stages of hemangioma and normal skin, and to explore the role of survivin in the patho-genesis of hemangioma. Methods A total of 50 cases of human dermal hemangioma and 8 eases of normal skin were analyzed for expression of survivin and caspase-3 by immunohistochemistry. Results The posi-tive ratio and average light density of survivin were obviously higher in proliferative hemangioma (0.2449±0.0135,0.7246±0.0747) than that in involutional hemangioma (0.1648±0.0217,0.5592±0.1601) and normal skin (0.1789±0.0126,0.4626±0.0961) (P<0.01). On the contrary, the expression of easpase-3 was up-regulated in involutional hemangioma (0.2386±0.0175, 0.4378±0.0593). Analysis of data also showed that there was a negative correlation between the average light density of survivin and caspase-3 (r=-0.95,P<0.01). Conclusions Survivin plays an important role in the pathogenesis of hemangioma and has a negative relationship with easpase-3.
4.Serum proteomic spectra of esophagial carcinoma patients and a corresponding esophagial carcinoma diagnostic model

Objective:To examine the serum proteomic spectra of human esophagial carcinoma by matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS),so as to set up a diagnostic model of esophagial carcinoma and to investigate its clinical value. Methods:Thirty-two esophagial carcinoma patients and 28 healthy controls were obtained from Fourth Affiliated Hospital of Hebei Medical University during May to September of 2008. Serum protein was extracted by weak cation exchange (WCX) protein chip system,and proteomic spectra was examined by MALDI-TOF MS. The obtained data were analyzed by ZUCI-protein chip data analyze system (ZUCI-PCDAS) and an esophagial carcinoma diagnostic model was established by genetic arithmetic (GA) combined support vector machine (SVM). The above 60 samples were randomly divided into training set and blinding test set,with training set including 21 esophagial carcinoma patients and 19 healthy controls and blinding test set including 11 esophagial carcinoma patients and 9 healthy controls,so as to examine the specificity and sensitivity of this diagnostic model. 5.Evaluation of the toxicity of a unitary package peracetic acid disinfectant in rats

Objective To evaluate the use safety of a unitary package peracetic acid ( PAA) disinfectant through acute and subacute toxicity experiments in rats.Methods This study was conducted according to the Technical Standard For disinfection ( Version 2002 ) .In the acute toxicity experiment, 60 healthy Wistar rats were randomly divided into different groups of PAA given by single intragastric gavage.Symptoms of poisoning of the animals were observed to calculate the half lethal dose ( LD50 ) .In the subacute toxicity experiment, 40 healthy Wistar rats were randomly divided into three dose groups and negative control group.The rats were given 33-342 mg/kg BW PAA for 28 days.At the end of test, the body weight, the organ-to-body weight ratio and the hematological and biochemical indices were determined, and pathological examination was performed.Results The acute oral LD50 of female rats was 1470 mg/kg BW, and the LD50 of male rats was 1710 mg/kg bw.The results of subacute toxicity test showed that the body weight, the organ-to-body weight ratio and the hematological and biochemical indices had no significant difference between the dose groups and control group.No obvious abnormality was observed in gross anatomy and histopathological examination.Conclusions The results of the acute toxicity test prove that PAA has a low toxicity level, and no significant subacute toxicity was observed at the exposure doses.
6.Effects of celecoxib on tumor growth and cell apoptosis in human triple-negative breast cancer xenografts in nude mice

Objective To evaluate the effects of celecoxib on tumor growth and cell apoptosis in human triple-negative breast cancer (TNBC) xenografts in nude mice.Methods Human TNBC MDA-MB-231 cells were inoculated subcutaneously into BALB/c nude mice.The mice (n=32) were then randomly divided into 4 groups,the control group and the celecoxib group (receiving 25,50,100 mg·kg-1·d-1 respectively).At the end of the study,tumor tissues were collected and tumor volume was measured.Cell apoptosis was determined by flow cytometry (FCM) analysis.NF-κB p65 and pS0 protein levels were measured by immunohistochemistry.Caspase-3 and survivin protein levels were detected by western blotting.Results celecoxib at dose of 25,50 and 100 mg·kg-1·d-1 inhibited the tumor growth significantly,compared with the control group.FCM results showed that apoptotic rates were (13.58±3.16) % and (21.91±4.75) % in moderate and high dose of celecoxib-treated group,compared with (3.15±1.73) % in control group (t =6.736,P < 0.05;t =12.151,P < 0.05).p65 expressions were 79.3 %,46.7 % and 23.9 % in low,moderate and high dose of celecoxib-treated group,compared with 89.7 % in control group (x2 =3.312,P < 0.05; x2 =10.785,P < 0.05;x2 =15.900,P < 0.05).Besides,western blotting analysis demonstrated that celecoxib significantly downregulated survivin expression,while upregulated the active form of caspase-3 expression.Conclusion Celecoxib could suppress TNBC tumor growth and induce cell apoptosis,which might be partially associated with inactivation of p65 and downregulation of survivin.
7.The influence of L-glutamate and carbachol on burst firing of dopaminergic neurons in ventral tegmental area.

Burst firing of dopaminergic neurons in ventral tegmental area (VTA) induces a large transient increase in synaptic dopamine (DA) release and thus is considered the reward-related signal. But the mechanisms of burst generation of dopaminergic neuron still remain unclear. This experiment investigated the burst firing of VTA dopaminergic neurons in rat midbrain slices perfused with carbachol and L-glutamate individually or simultaneously to understand the neurotransmitter mechanism underlying burst generation. The results showed that bath application of carbachol (10 μmol/L) and pulse application of L-glutamate (3 mmol/L) both induced burst firing in dopaminergic neuron. Co-application of carbachol and L-glutamate induced burst firing in VTA dopaminergic cells which couldn't be induced to burst by the two chemicals separately. The result indicates that carbachol and L-glutamate co-regulate burst firing of dopaminergic neuron.

9.Effect of Low Molecular Weight Heparin Calcium Combined Compound Danshen Injection on Perinatal Outcomes of Nephrotic Syndrome Patients with Early Onset Severe Pre-eclampsia.

OBJECTIVETo observe the effect of low molecular weight heparin calcium (LMWHC) combined Compound Danshen Injection (DI) on nephrotic syndrome patients with early onset severe preeclampsia.
METHODSTotally 80 nephrotic syndrome patients with early onset severe pre-eclampsia were randomly assigned to four groups voluntarily, i.e., Group A (22 cases, treated by magnesium sulfate), B (19 cases, treated by magnesium sulfate plus LMWHC), C (21 cases, magnesium sulfate plus DI), D (18 cases, magnesium sulfate plus LMWHC and DI). Umbilical arterial S/D ratios, amniotic fluid index (AFI), prolonged gestational age, placenta weight, neonatal weight, and Apgar score were compared among the four groups.
RESULTSCompared with before treatment in the same group, umbilical arterial S/D ratios decreased in the four groups (P <0. 05). AFI decreased in Group A, while it increased in Group B, C, and D (P<0. 05). Compared with Group A at the same time point, umbilical arterial S/D ratios decreased, and AFI increased in Group B, C, and D (P <0. 01 , P <0. 05). Prolonged gestational age and neonatal weight were increased in Group B, C, and D (P <0. 01, P <0. 05). Placenta weight were increased in Group B and D (P <0. 05). Apgar scores at 1 and 5 min were improved in Group D (P <0. 05). Compared with Group B and C at the same time point, umbilical arterial S/D ratios decreased, and AFI increased in Group D (P<0. 05). Compared with Group B, prolonged gestational age and placenta weight were decreased in Group C, but prolonged gestational age and placenta weight were increased in Group D (P <0.05). Compared with Group C, prolonged gestational age, placenta weight, and neonatal weight were increased in Group D (P <0. 05).
CONCLUSIONTreatment of nephrotic syndrome patients with early onset severe pre-eclampsia by LMWHC combined DI could prolong gestational ages, obviously improve prenatal outcomes, with better effect obtained than using any of them alone.

10.The change of serum neuron enolase level and ambulatory electroencephalogram after initial polytherapy in patients with newly diagnosed infantile spasm

Objective To investigate the efficacy of initial polytherapy in the patients with newly diagnosed infantile spasm by evaluating the ambulatory electronecephalogram (EEG) profiles and serum neuron specific enolase ( NSE ) level.Methods Data from 108 cases of newly diagnosed infantile spasm from Sep.2007 to Nov.2010 in the Department of Pediatrics,the First Hospital of Jilin University,were respectively analyzed for the EEG profiles and serum NSE level.The patients were treated with multiple drugs.Results ( 1 )After treatment,EEG showed normal shape in 56 cases ( 62.2% ),sporadic sharp and slow waves without hypsarrhythmia in 32 cases( 35.6% ),and hypsarrhythmia in 2 cases (2.2%).The overall effective rate was 97.8%.(2) Serum NSE in 35 seizure-free cases were examined at the time points of hospital admission,48 hours and one month after seizure cessation,respectively.NSE level was significantly decreased at 48 hours after seizure control than that at hospital admission ( t =6.54,P < 0.05 ) ; NSE at one month after seizure free was further decreased compared with that at admission point ( t =9.29,P < 0.05 ).Conclusion Polytherapy for infantile spasms can ameliorate the abnormal EEGs.NSE level is decreased after seizure cessation.Intelligence is improved 6 months after seizure free and polytherapy may improve the prognosis of the children with infantile spasms.
