1.Construction and expression of the eukaryotic coexpression plasmid of Mycobacterium tuberculosis Ag85B and MPT64 gene

Objective:To construct an eukaryotic coexpression plasmid containing Mycobacterium tuberculosis Ag85B and MPT64 gene.Methods:Mycobacterium tuberculosis Ag85B and MPT64 gene were cloned into pBudCE4.1 to construct recombinant plasmid pBud85B-MPT.The recombinant plasmid was transfected into COS-7 cells,and its expression of Ag85B and MPT64 was assesed by RT-PCR.Results:Mycobacterium tuberculosis Ag85B and MPT64 were detected in transfected COS-7 cells.Conclusion:The recombinant plasmid pBud85B-MPT can express Ag85B and MPT simultaneously in COS-7 cells which provides the basis for further investigation of DNA vaccine against tuberculosis.

Social media,such as WeChat and microblog,have become the platform for domestic college and university libraries to popularize and provide information service for their teachers and students.The use of social media,public account,menu functions,information pushing of WeChat and running of official miroblog in domestic medical college and university libraries were investigated,which showed the problems in running of social media in medical college and university libraries.Measures for the solution of such problems were put forward.The fact that social media are expected to become the ubiquitous carrier of information service in medical college and university libraries and help medical college and university libraries to improve their core competitive power.

4.From the qi annotation in Xiaozhenjie to qi view in the ancient time.

Xiaozhenjie (Miraculous Pivot Chapter 3: Annotation of Fine Needle) is the earliest annotation of Jiuzhen Shieryuan (Miraculous Pivot Chapter 1: Nine Needles and Twelve Yuan-Primary Acupoints). There are particularly 24 annotations of qi in the chapter. Based on the original literature, the styles of qi annotationsl were divided into three categories. By comparing with the original text, semantic features of qi annotations were reasoned. In reference to the understandings and notes of qi annotations given by the scholars of different dynasties, the origin of qi connotation in the ancient time was clarified so as to provide the historical evidence to the for-mation of this unique "qi language" and "qi worldview". It presents the instructive significance to understand and interpret "qi" for the scholars in the modern time.

6.Advances of immunologic dysregulation mechanism in biliary atresia

Biliary atresia(BA) is a destructive inflammatory obliterative cholangiopathy of neonates affecting both intrahepatic and extrahepatic bile ducts.BA is more common in east Asia.Genetic susceptibility,viral infections,and immune dysregulation may be related to BA,but immune dysregulation may play a key hole in the pathogenesis of BA.A current view of the pathogenesis of BA is that it may involve both a primary perinatal hepatobiliary viral infection and a secondary generation of an autoimmune-mediated bile duct epithelial injury.The etiology of BA is unknown,but there is evidence for the involvement of immunologic dysregulation mechanisms,which will be discussed in this review.
7.Further understanding of the complications after orthotopic liver transplantation and their interventional management

The majority of complications due to orthotopic liver transplantation (OLT)need interventional treatment. For arterial and venous complications after OLT, we have established clear and definite interventional therapeutic mode. However, it has been the difficult point as well as the weak point for interventional radiologists to treat the biliary complications. With the technical improvement and deep-going understanding, a part of biliary complications of OLT can be relieved or even finally be cured with percutaneous biliary drainage(PTBD) and endoscopic nasobiliary drainage(ENBD). For some patients, PTBD and ENBD seem to be the main palliative therapeutic means, which might accompany the patients for a lifetime. Now, it is an urgent demand to formulate the clinical therapeutic guidelines for the treatment of biliary complications after OLT.

The management of frailty in elderly patients with chronic kidney disease (CKD) has been receiving increasing attention.Frailty in elderly patients with CKD has a high incidence and is used for risk prediction of adverse outcomes,including death,hospitalization,falls,dialysis and so on.A range of pathophysiological changes caused by CKD,aging and multimorbidity work together to promote the development of frailty.Treatment for CKD patients with frailty includes mainly nutritional support,exercise,drug therapy and comprehensive intervention.This article reviews recent research progress concerning frailty in elderly CKD patients.
9.Reduced expression of Diceri is associated with poor prognosis in patients with nasopharyngeal carcinoma.

OBJECTIVE:
Dicerl plays an important role in generation of microRNA, the purpose of this study was to evaluate Dicerl expression and its prognostic value in nasopharyngeal carcinoma (NPC).
METHOD:
The protein expression of Dicerl was examined by immunohistochemistry in 276 NPC specimens, and the mRNA levels of Dicerl were analyzed by qRT-PCR in 56 NPC and 11 nasopharyngitis tissues. Cox regression analysis was used to identify independent prognostic factors, and a prognostic score model was constructed for survival prediction.
RESULT:
Expression of Dicerl was downregulated in NPC tissues at both the mRNA and the protein levels, and there was a notable positive correlation between the expression levels of Dicerl mRNA and protein. Low Dicerl expression was positively correlated with distant metastasis (P<0. 01) and death (P<0. 05). In addition, low expression of Dicerl was significantly associated with poorer overall survival (HR = 2. 32, 95% CI: 1. 30 ~ 4. 14, P<0. 01) and poorer distant metastasis-free survival (HR = 2. 56, 95% CI: 1. 39 ~ 4. 74, P<0. 01). Furthermore, multivariate analysis showed that low expression of Dicerl and tumor-node-metastasis (TNM) stage were independent prognostic indicators for NPC patients. A prognostic score model combining the Dicerl expression and TNM stage had a better prognostic value than the TNM stage alone model or Dicer) expression alone model (P< 0. 05).
CONCLUSION
Dicerl was downregulated in NPC tissues at both the mRNA and the protein levels, and low expression of Dicerl could be served as novel prognostic biomarker for NPC patients.
