BACKGROUNDAlthough flunarizine has been widely used for migraine prophylaxis with clear success, the mechanisms of its actions in migraine prophylaxis are not completely understood. The aim of this study was to investigate the effects of flunarizine on tetrodotoxin-resistant Na(+) channels and high-voltage activated Ca(2+) channels of acutely isolated mouse trigeminal ganglion neurons.
METHODSSodium currents and calcium currents in trigeminal ganglion neurons were monitored using whole-cell patch-clamp recordings. Paired Student's t test was used as appropriate to evaluate the statistical significance of differences between two group means.
RESULTSBoth tetrodotoxin-resistant sodium currents and high-voltage activated calcium currents were blocked by flunarizine in a concentration-dependent manner with the concentration producing half-maximal current block values of 2.89 µmol/L and 2.73 µmol/L, respectively. The steady-state inactivation curves of tetrodotoxin-resistant sodium currents and high-voltage activated calcium currents were shifted towards more hyperpolarizing potentials after exposure to flunarizine. Furthermore, the actions of flunarizine in blocking tetrodotoxin-resistant sodium currents and high-voltage activated calcium currents were use-dependent, with effects enhanced at higher rates of channel activation.
CONCLUSIONBlockades of these currents might help explain the peripheral mechanism underlying the preventive effect of flunarizine on migraine attacks.

OBJECTIVENephrotic syndrome (NS) is characterized by marked urinary excretion of albumin and other intermediated-size plasma proteins such as transferrin. The aim of this study was to determine the changes of serum iron and transferrin and the relationship between the serum and urinary transferrin.
METHODSThe indexes related to iron metabolism, including serum iron, ferritin, transferrin, total iron-binding capacity, transferrin saturation and hematological parameters (Hb, MCV, MCH), and urinary transferrin were measured in 37 children with NS before treatment and at the remission stage. Thirty-five age-matched healthy children served as controls.
RESULTSSerum iron levels (18.8 +/- 3.8 micromol/L) in NS patients before treatment were significantly lower than in the healthy controls (22.2 +/-3.8 micromol/L) and those measured at the remission stage (21.0 +/- 3.5 micromol/L) (P < 0.01). Serum transferrin levels in NS patients before therapy (1.9 +/- 0.3 g/L) also decreased compared with those in the healthy controls (3.1 +/- 0.5 g/L) and those measured at the remission stage (2.9 +/- 0.6 g/L) (P < 0.01). In contrast, serum total iron-binding capacity and transferrin saturation were noticeably higher in NS patients before treatment than those in the healthy controls (total iron-binding capacity 56.4 +/- 9.2 micromol/L vs 50.7 +/- 6.8 micromol, P < 0.01; transferrin saturation 55.7 +/- 9.2 % vs 46.4 +/- 8.2%, P < 0.01) and were also higher than those measured at the remission stage (51.9 +/-7.7 micromol/L and 47.4 +/- 13.3%) (P < 0.01). Serum transferrin positively correlated to serum albumin (r = 0.609, P < 0.01) and negatively correlated to urinary transferrin (r = -0.550, P < 0.01) in NS patients before treatment.
CONCLUSIONSSerum iron and transferrin levels markedly decreased in NS patients, which may be partially related to the urinary loss of transferrin.

Targeted to the junior nursing students in our university, we applied experiential teaching in the course of Obstetrics and Gynecology Nursing to carry out humanistic care infiltration education, which mainly included three parts: aesthetic activities in theoretical class, experiential activities in experimental class and enlightenment after class. After class, compared with the nursing students who were educated by traditional teaching methods, the quality of humanistic care of nursing students who accepted experiential teaching was improved more obviously. It is suggested that the application of experiential teaching is effective in infiltrating humanistic care education in nursing professional courses, which is also in line with the development of nursing discipline.

8.Venipuncture indication system based on an ultra-miniature side-view optical coherence tomography endoscopic probe
International Journal of Biomedical Engineering 2024;47(5):430-435
Objective:To develop a venipuncture indication system based on an ultra-miniature side-view optical coherence tomography (OCT) endoscopic probe. This system enables real-time scanning and imaging of the tissue around the tip of the intravenous needle, which is used to determine whether the intravenous needle has penetrated the blood vessel. Methods:An ultra-miniature side-view OCT needle probe was fabricated and placed into the stem of an elongated 30G hypodermic needle to make the OCT endoscopic probe. The endoscopic probe was inserted into the stem of an intravenous needle and the tip of the probe was allowed to probe out of the intravenous needle. This allows the OCT endoscopic probe to be driven by the rotating collar to scan and image the tissue around the tip of the intravenous needle. Imaging experiments were performed using mouse blood and non-blood tissues (skeletal muscle and liver) to verify system function. Results:Blood did not produce OCT signals at most locations except for a weak OCT signal at locations near the scanning window of the endoscope probe (i.e., the top of the image). The intensity of the OCT signal generated by it decayed rapidly with the increase in imaging depth. At a depth of 300-400 μm from the scanning window, the OCT signal has almost completely disappeared. Strong OCT signals appeared in the images of skeletal muscle. Its signal intensity decayed with the increase of imaging depth, but a more obvious OCT signal could be observed at a depth of more than 900 μm from the scanning window. The distribution of OCT signal intensity within skeletal muscle was very heterogeneous. The connective tissue embedded in the skeletal muscle showed stronger OCT signals than the surrounding skeletal muscle, while there was almost no OCT signal in the muscle at the spaces located between the muscle fiber bundles. A strong OCT signal was observed in the images of liver tissue. The attenuation of this signal with increasing imaging depth was more pronounced. At a distance of 600-700 μm from the scanning window, the OCT signal had almost completely disappeared. The intensity distribution of the OCT signal in the internal image of the liver tissue was relatively uniform. Conclusions:The proposed venipuncture indication system is able to image the tissue around the tip of the injection needle instantly, and provide a more reliable indication of whether the needle has penetrated the blood vessel during clinical venipuncture.
