Objective To clone and express prokaryotic recombinant plasmid of nucleoside triphosphate hydrolase (NTPase) gene of Toxoplasma gondii, and analyze its antigenicity. Method NTPase gene was amplified by PCR from RH strain of T. gondii and cloned into pGEM-T Easy vector. Positive clones were screened and identified by BglⅡ, HindⅢ digestion and sequenced. The target gene was then subcloned into prokaryotic expression vector pBAD-HisB and transformed into E. coli BL21(DE3). The expressed recombinant protein was purified with Ni-NTA agarose and further analyzed by sodium dodecyl sulfate polyacrylamide gel electropheresis (SDS-PAGE) and Western blotting. Results NTPase-Ⅱ gene was specifically amplified, and the homology of DNA sequence was 100% to that in the GenBank. SDS-PAGE showed that the recombinant NTPase protein with correct molecular weight was expressed highly in E.coli BL21(DE3). Western blotting testified that the purified recombinant protein could be specifically recognized by mouse serum immunized with T. gondii and mouse anti-recombinant protein serum. Conclusion The NTPase-Ⅱ gene has been cloned and expressed in E.coli BL21(DE3), and the purified protein of NTPase-Ⅱ gene displays a specific antigenicity.

Aim To study the anti-itching and anti-hypersensitivity effects of Doxepin hydrochloride cream. Methods The effects of Doxepin hydrochloride cream on the pruritus of skin in mice due to 4-AP and guineapigs due to histamine phosphate were observed. The inhibitory actions of it on the increase of abdominal cavity capillary permeability in mice due to acetic acid and the angiectasis of skin in rats due to histamine were also observed. Its effects on the delayed-type hypersensitivity (DTH) of mice due to 2, 4-dinitrochlorobenzol (DNCB) and the homologous passive anaphylaxis of skin in rats were used to investigate its anti-hypersensitivity actions. Results The application of Doxepin hydrochloride cream (50, 125, 250 mg?kg -1) on the skin of mice had significant inhibitory actions on the pruritus of skin caused by 4-AP, and the application of Doxepin hydrochloride cream (10, 25, 50 mg?kg -1) on the skin of guinea pigs also had significant inhibitory actions on the pruritus of skin caused by histamine phosphate. The application of Doxepin hydrochloride cream (10,25,50 mg?kg -1 and 50,125,250 mg?kg -1) on skin inhibited the increase of skin capillary permeability in rats caused by histamine phosphate and the increase of abdominal cavity capillary permeability in mice caused by acetic acid. The application of Doxepin hydrochloride cream (50, 125, 250 mg?kg -1 and 10, 25, 50 mg?kg -1) on skin also significantly inhibited the delayed-type hypersensitivity in mice due to 2, 4-dinitrochlorobenzol and the homogeneous cutaneous anaphylaxis in rats. Conclusions Doxepin hydrochloride cream can relieve itching, inhibit the increase of capillary permeability caused by inflammatory substances. It also has significant inhibitory actions on TypeⅠ and Type Ⅳ allergic reactions.

China Journal of Traditional Chinese Medicine and Pharmacy 2005;0(06):-
Objective:The topographical mapping technique of cortical evoked potential was used to observe the changes of the functional activities in the primary somatosensory area of cortex (SI) during external stimulation, the immanent relation between external stimulation and topographical mapping of cortical somatosensory evoked potential (CSEP). Methods:33 healthy adult volunteers were under observation. Cerebral electric signal processing system was used to lead CSEP on the scalp. The external stimulation was applied as following:①A soft paintbrush was used to slightly brush the face only. ②Paintbrush was used to slightly brush the lower limb only. ③Paintbrush was used to slightly brush the trunk only. ④Paintbrush was used to slightly brush the lower limb or the trunk or the Gallbladder Meridian (GBM) and passing the upper arm by imitating the speed of meridian transmission phenomenon respectively. Results:①Evoking responses appeared in the foot areas in SI,without meridian transmission phenomenon when brushing the face, lower limb or trunk only. ②Healthy volunteers were offered imitation of sensation conduction along the GBM as well as brushing through the upper arm in addition, while, the results showed that the evoking responses appeared in the foot, the upper arm and the face representation area of SI simultaneously. Conclusion:The external stimulation along the meridian course was able to change the distribution of evoking responses in SI during stimulating acupoint. But the local stimulating on face, lower limb or trunk can not change the distribution of CSEP. The results provided experimental evidence for the supposition that 'activation in sequence in periphery'was the decisive factor for the formation of meridian transmission phenomenon.

Objective :
To explore the mediating effect of sleep quality on mobile phone dependence and loneliness in university students, so as to provide evidence for prevention and intervention of mobile phone dependence.
Methods :
A survey was conducted from December 2019 and January 2020 among the students of Guangzhou Medical University. The general information questionnaire, mobile phone dependence index scale, UCLA loneliness scale and Pittsburgh sleep quality index scale were used to analyze the mediating effects of sleep quality on mobile phone dependence and loneliness.
Results :
A total of 575 questionnaires were distributed and 573 valid ones were collected, with an efficiency of 99.65%. The detection rate of 115 students with mobile phone dependence was 20.07%, and that of 203 students with sleep quality problems was 35.43%. The students scored ( 48.03±6.07 ) points in loneliness, and 405 of them had high level. Mobile phone dependence was positively correlated with loneliness and sleep quality ( r=0.299, 0.385, both P<0.05 ); loneliness was positively correlated with sleep quality ( r=0.553, P<0.05 ). Mobile phone dependence and sleep quality both could positively predict loneliness, mobile phone dependence could positively predict sleep quality, and sleep quality and gender had a significant interaction effect on loneliness ( all P<0.05 ). The mediating effect value of sleep quality on mobile phone dependence and loneliness was 0.290 ( 95%CI: 0.186-0.400 ) in males and 0.131 ( 95%CI: 0.084-0.187 ) in females.
Conclusion
Sleep quality has a mediating effect on mobile phone dependence and loneliness among university students. Male students are susceptible to the negative effects of mobile phone dependence.

Journal of Biomedical Engineering 2015;32(1):235-239
Mechano growth factor (MGF) is an autocrine/paracrine factor and sensitive to mechanical stimulation. MGF can be highly expressed in various soft tissues under physical stimuli, biochemistry stimuli or in damaged situation. MGF may "compensate" the stress for tissue in the processing of tissue repair. MGF can effectively accelerate the repair of the soft tissue by promoting the proliferation, migration and differentiation of cells. This paper summarizes the MGF expressions in different soft tissues and their functions in soft tissue repair. The paper also discusses current problems and challenges in using MGF to repair the soft tissue.
