Objective To study the levels of matrix metalloproteinase-3(MMP-3),matrix metalloproteinase-9(MMP-9) and inhibitor of metalloproteinases-1(TIMP-1) in patients with acute myocardial infarction(AMI) and mechanism of plaque rupture of coronary atherosclerosis.Methods Sixty-seven patients with AMI were selected,average invasion time was 3 h~5 d.40 cases of non-coronary heart disease were included in control group.The expressions of serum MMP-3,MMP-9,TIMP-1 and the levels of serum cTnI,CK-MB were analyzed by enzyme-linked immunosorbent assay.Results The serum MMP-3,MMP-9 and TIMP-1 levels in AMI group were higher than those in control group,there was significant difference between two groups(P
2.Evaluation of uncertainty in colony forming units on the disinfected skin of nurses in operation room

Objective To develop a feasible method for evaluating uncertainty in colony forming units(CFU)on the disinfected skin of nurses of operation room.Methods A nurse of operation room in a hospital involved the study.Ten spots on her disinfected hand skin were selected for evaluation of uncertainty in the acquired data and analysis of reliability of the evaluation methods,according to Technical Norms for Monitoring the Disinfection and Sterilization Quality in Hospitals in Jiangsu Province and JJF1059-1999 Evaluation and Expression of Uncertainty in Measurement.Results The CFU on the disinfected skin of nurse was(4±2)cfu/cm2, which indicated that the disinfection of the nurse's hands was not qualified.Conclusion The evaluation method by our study is reliable for evaluating the uncertainty in CFU on nurse’s disinfected hand skin.It is simple and fitful for the uncertainty evaluation of hospital disinfection effect.
3.Preparation of paeonol transdermal delivery systems based on proniosomes-based ointment and its pharmacokinetics characters.

The paeonol proniosomes ointment and ordinary ointment were administered to rats. Physiological saline served as perfused solution. The perfusion rate was 5 mL x L(-1) and the microdialysis samples were collected every 20 min intervals. The paeonol concentration in perfused solution was determined by HPLC. Investigation of the pharmacokinetics of paeonol proniosomes ointment and ordinary ointment by the skin-blood synchronous microdialysis coupled with HPLC is reported in this study. The results show that the recovery was (54.80 +/- 1.50)% in vitro and (54.58 +/- 4.61)% in vivo. The results showed that paeonol proniosomes ointment significantly raised the drug concentrations in skin more than the paeonol ordinary ointment. The paeono proniosomes ointment has less drugs into the blood as the ordinary ointments in blood, but its blood drug concentrations were steadier. The paeonol proniosomes ointment may be developed into a new preparation.

4.Expression and clinical significance of Semaphorin4D in non-small cell lung cancer and its impact on malignant behaviors of A549 lung cancer cells.

This study aimed to explore Semaphrin4D (Sema4D) expression and clinical significance in non-small cell lung cancer (NSCLC), and to define the roles and mechanisms of Sema4D in regulating the malignant behaviors of A549 cells by small interfering RNA (siRNA). Firstly, immunohistochemistry revealed that Sema4D was more frequently expressed in NSCLC than in lung benign lesion (P<0.05) and its overexprssion was associated with low differentiation (P<0.05), poor pTNM staging (P<0.05) and occurrence of lymph node (LN) metastasis (P<0.05). Endogenous Sema4D expression was suppressed by Sema4D siRNA in A549 cells overexpressing Sema4D. Protein levels of Sema4D, total Akt and p-Akt were examined by Western blotting. Cell proliferation, migration and invasion abilities were measured by MTT assay and Transwell assay respectively. Results showed that Sema4D siRNA significantly suppressed phosphorylation of AKT in A549 cells, but it did not alter total AKT expression. In addition, efficient down-regulation of SemaD significantly inhibit cell proliferation (P<0.05), migration (P<0.05) and invasion (P<0.05) in A549 cells. These findings suggest that Sema4D might serve as a reliable tool for early prediction of NSCLC poor prognosis. Sema4D could play an important role in promoting tumor proliferation, migration and metastasis in the NSCLC, by influencing the Akt protein phosphorylation. Inhibition of Sema4D may be a useful approach for the treatment of NSCLC.
5.Safety of early nadroparin for prevention deep vein thrombosis in patients with intracerebral hemorrhage

Objective To assess the safety of early subcutaneous injection of a low-dose low molecular weight heparin (LMWH) nadroparin for prevention of deep vein thrombosis (DVT) in patients with spontaneous intracerebral hemorrhage (sICH).Methods The patients with sICH who early using nadroparin or lower limb intermittent pneumatic compression (IPC) for prevention of DVT were enrolled.A nadroparin group continuously injected nadroparin 0.4 ml/d subcutaneously for 10 days at day 4 after admission and an IPC group used lower limb IPC.Head CT was reexamined and hematoma volume changes were evaluated at day 3,5,and 14 after admission.The hemorrhagic events during the course of treatment were documented,and the lower limb DVT was examined by color Doppler sonography.Results A total of 94 patients with acute sICH (n =41 in the nadroparin group,n =53 in the IPC group) who early use of nadroparin or IPC for prevention of DVT were enrolled.Fourteen patients had lower limb DVT,5 (12.2%) of them were in the nadroparin group and 9 (17.0%) of them were in the IPC group.However,there was no significant difference in the incidence of DVT between the two groups (x2 =0.418; P =0.518).During the treatment,no patient experienced increased intracranial hematoma and rebleeding.Conclusion Early subcutaneous injection of low-dose nadroparin for the prevention of DVT in patients with sICH is safe.
6.Effects of periodontal health and related factors on the oral health-related quality of life in type 2 diabetic patients with chronic periodontitis.

OBJECTIVETo evaluate the oral health-related quality of life (OHRQOL) in type 2 diabetic patients with chronic periodontitis.
METHODSA total of 80 type 2 diabetic patients with chronic periodontitis who had at least 16 teeth and never accept any periodontal therapy in latest one year were recruited. The clinical periodontal index such as probing depth, attachment loss (AL), modified bleeding index, plaque index were obtained with a Williams type periodontal probe. All accepted structured questionnaire interview including the evaluation of OHRQOL.
RESULTSThe geriatric oral health assessment index (GOHAI) results demonstrated good content validity, construct validity and responsibility. The periodontal index had positive correlation with the GOHAI scores. Individuals with self-unsatisfied of oral health status, using hypoglycemic drugs, accompanied with serious gingival inflammation and higher AL had relatively poor OHRQOL.
CONCLUSIONThe OHRQOL of type 2 diabetes with chronic periodontitis and the negative experiences of items listed in GOHAI questionnaire were affected by self-conscious of oral health and periodontal inflammation status.

7.A study on expression of platelet derived growth factor a in periodontal tissue during orthodontic tooth movement with rotating pulsed magnetic field

Objective To investigate the effects of rotating pulsed magnetic field on platelet derived growth factor a (PDGF-A) expression in periodontal tissue during tooth movement. Methods 30 white rabbits were random divided into 6 groups with 5 rabbits each group,including groups of 1,3,5,7,14 and 21 days. Under anesthesia condition by 2% pentobarbital sodium,the stainless coil springs were fixed between the first maxillary molar and the incisor producing the force of 80g. The experimental group was treated by rotating pulsed magnetic field and the force, the control group was only treated by t he force. The expression of PDGF-A was half-quantitatively investigated through immunohistochemical analysis. Results The expression of PDGF-AA in the experimental group enhanced apparently compared with that in the control group. There were significant differences among the 5,7, and 14day groups (5. 28 ± 0. 14 vs 2. 03 ±0. 18,7.63±0.27 vs 2. 84 ±0. 12,3.52 ±0. 16 vs 1.65 ±0.03;8.10±0.13 vs 4. 30 ±0. 21,13. 27 ±0. 31 vs 6.47 ± 0.15,5.66 ± 0.22 vs 3. 15 ± 0. 27, P < 0. 05). The expression of PDGF-AA of 7 day group was higher than that of other groups. Conclusion Rotating pulsed magnetic field promotes the expression of PDGF-A in the periodontal tissue and alveolar bone remodeling.
8.The relationship between serum uric acid and fasting blood glucose in men and women with newly diagnosed type 2 diabetes

Objective To explore the relationship between serum uric acid and fasting blood glucose ( FBG) in newly-diagnosed type 2 diabetes in regard to different genders. Methods In 282 patients of newly diagnosed type 2 diabetes(208 males and 74 females), detection of serum FBG, uric acid, creatinine, transaminases, blood lipids, etc were determined. FBG<7. 0 mmol/L was found in 118 cases (89 males and 29 females);7. 0 mmol/L≤FBG≤9. 0 mmol/L in 114 cases ( 88 males and 26 females ); FBG>9. 0 mmol/L in 50 cases ( 31 males and 19 females). Results (1)Serum uric acid showed statistically significant difference in regard to sex[Male:(325. 82±92. 26)μmol/L, Female:(292. 26±76. 19)μmol/L, P<0. 01]. (2) FBG and serum uric acid were negatively related(r=-0. 316, P<0. 01). (3) FBG and serum uric acid were negatively related both in male(r=-0. 320, P<0.01)andinfemale(r=-0.291,P<0.05). (4)Multiplestepwiseregressionanalysisshowedthatthethreegroups of FBG and serum uric acid showed regression relationship in male and female patients(P<0. 01). Conclusion With the increased FBG, serum uric acid was decreased in both males and females. Therefore, serum uric acid might serve as a potential biomarker of glucose metabolism.
9.Analysis of relevant factors for delayed cerebral edema and plasma matrix metalloproteinase 9 after intracerebral hemorrhage

Objective Toinvestigatetheclinicalsignificanceofplasmamatrixmetalloproteinase9 (MMP-9)intheformationofdelayedcerebraledemaafterintracerebralhemorrhage.Methods The clinical data of 107 patients with spontaneous intracerebral hemorrhage treated with conservative medical treatment were analyzed retrospectively. According to the clinical features and imaging changes,they were divided into either a delayed cerebral edema group (case group n=39)or a non-delayed cerebral edema group (control group n =68 ). The plasma MMP-9 level was detected with enzyme-linked immunosorbent assay within 24 h after onset. The patients performed head CT scan again at day 7 and 14 after admission. The changes of hematoma and edema volume were detected. All the possible factors associated with the formation of delayed cerebral edema were firstly analyzed by the univariate analysis. Univariate analysis showed that the variables with significant differences were enrolled into multiple logistic regression analysis. Results TheplasmaMMP-9levelofthedelayedbrainedemagroupwassignificantlyhigherthanthatof the control group,they were 189 ± 51 and 118 ± 27 mg/L respectively (P<0. 01). The result of univariate analysis showed that age,history of smoking,blood glucose level,baseline hematoma volume,and National Institute of Health stroke scale (NIHSS )score on admission might be associated with the formation of delayed cerebral edema after intracerebral hemorrhage. Logistic regression analysis showed that MMP-9 level (OR,9. 745,95%CI 6. 754-15. 466,P<0. 01),baseline hematoma volume (OR,2. 411,95%CI 1. 190-2. 728,P =0. 018),blood glucose level on admission (OR,1. 327,95%CI 1. 133 -1. 850,P =0.004),and NIHSS score (OR,1. 867,95%CI 1. 272-2. 364,P=0. 020)were the independent risk factorsfortheformationofdelayedcerebraledemaafterintracerebralhemorrhage.Conclusion Theamount of bleeding,NIHSS score,and hyperglycemia are the risk factors for the formation of delayed cerebral edema in patients with spontaneous intracerebral hemorrhage,while high MMP-9 level on admission indicated that the risk of the formation of delayed cerebral edema is high.
10.Effect of berberine combined with bone marrow mesenchymal stem cells on energy metabolism of HUVECs damaged by high glucose

Aim To investigate the effects of berberine combined with bone marrow mesenchymal stem cells ( BM-MSCs) on the energy metabolism of human um-bilical vein endothelial cells ( HUVECs ) under the condition of high glucose. Methods ①The state of cell reproduction and cell proliferating activity were de-termined by MTT assay. ②The cell cycle was detected by flow cytometry ( FCM ) . ③DNA damage of cells was measured by comet tail assay. ④The contents of ATP, ADP and AMP were determined by high perform-ance liquid chromatography ( HPLC ) and the level of energy charge ( EC) was calculated. ⑤The expression of CCR and COX mRNA was detected by reverse tran-scription-polymerase chain reaction ( RT-PCR) . ⑥The expressions of COX and CCR were detected by Western blot. Results ①The proliferating activity of HUVECs declined apparently and the proliferation decreased af-ter high glucose intervention. Meanwhile, the quantity of cells during S +G2 dropped dramatically and there was certain degree of damage to DNA. The berberine and BM-MSCs respectively improved the proliferating activity and the proliferation in different degrees, in-creased the quantity of cells during S+G2 and promo-ted the repair of DNA ( P <0 . 01 ) , and so did the combination of the two, with a better effect than each of them alone. ②After high glucose intervention and the damage caused, the content of both ATP and ADP of HUVECs was reduced, and EC level also declined significantly, while the content of AMP increased. The berberine and BM-MSCs respectively up-regulated the content of ATP and ADP ( P<0. 01 ) , and so did the combination of the two, with a better effect than each of them alone. ③After high glucose intervention and the damage caused, the expression of COX, CCR mR-NA and protein decreased obviously. Yet, all of the three gained a dramatic increase when the berberine, BM-MSCs or the combination of the two were added ( P<0. 01 or P <0. 05 ) , among which the combination worked more effectively. Conclusions The berber-ine, BM-MSCs and the joint use of the two could im-prove the energy metabolism of HUVECs, which had been damaged by high glucose, probably because the berberine and BM-MSCs could up-regulate the expres-sion of COX, CCR mRNA and protein, which leads to the hydrolyzation of glucose oxide and thus the im-provement of blood environment and the enhancement of glucose's supply and intake of HUVECs. Then, here comes the final result: the improvement of the energy metabolism of damaged vascular endothelial cells by high glucose.
