1.A Study on the Peripheral Anticholinergic Effect of Minaprine.

The authors studied ED50 of bethanechol on the contractilities of the smooth muscles isolated from various organs of rat under the presence of atropine (a classical competitive blocker of cholinergic muscarinic receptor) or minaprine (a newly developed antidepressant drug) to investigate the peripheral anticholinergic effect of minaprine. The results were as follow: 1) There was no significant difference between ED50 of bethanechol in the control group and that under the presence of minaprine 10⁻⁸M and 10⁻⁷M in the smooth muscles isolated from the duodenum. 2) There was no significant difference between ED50 of bethanechol in the control group and that under the presence of minaprine 10⁻⁸M and 10⁻⁷M in the smooth muscles isolated from the ascending colon. 3) There was significant difference between ED50 of bethanechol in the control group and that under the presence of minaprine 10⁻⁸M and 10⁻⁷M in the smooth muscles isolated from the urinary bladder (P<0.01) 4) There was significant difference between ED50 of the atropine 10⁻⁸ and minaprine (10⁻⁸M) in the smooth muscles isolated from the urinary bladder (P<0.05).

6.Role of Sodium Ion in Renal Transport of p-Aminohippurate in vitro.

The effect of sodium on p-aminohippurate (PAH) transport kinetics was studied in isolated rat kidney slices in an attempt to define the role of sodium ion in renal organic acid transport. 1. In normally metabolizing renal slices, Na+ increased the Vmax of PAH influx without changing the Michaelis constant (Km). On the other hand, the effIux of preaccumulated PAH was reduced as the Na+ concentration increased. 2. In metabolically impaired renal slices, Na+ had no apparent effect on the influx and efflux of PAH. These results may indicate that Na+ is important for the energy transducing reaction in the PAH transport process.

10.Stem cell maintenance in a different niche.

To overcome the difficulty of controlling stem cell fate and function in applications to regenerative medicine, a number of alternative approaches have been made. Recent reports demonstrate that a non-cellular niche modulating the biophysical microenvironment with chemical factors can support stem cell self-renewal. In our previous studies, early establishment was executed to optimize biophysical factors and it was subsequently found that the microgeometry of the extracellular matrix made huge differences in stem cell behavior and phenotype. We review here a three-dimensional, non-cellular niche designed to support stem cell self-renewal. The characteristics of stem cells under the designed system are further discussed.
