1.Absorbed constituents in rat serum after oral administration of water ex-tract of Simiao Yong’an Decoction

Objective To study the absorbed constituents of water extract of Simiao Yong’an Decoction (SYD)in serum of rats,so as to find out the pharmacodynamics material basis.Methods Using liquid chromatograph-mass spectrometry (LCMS)-ion trap (IT)-time of flight (TOF)method under both positive and negative ion modes of electrospray ionization,the water extract of SYD,rats’serum after oral adminis-tration of SYD and the blank serum were analyzed.Serum samples were pretreated by using methanol.The chromatographic condition consisted of Inertsil ODS-3 column,mobile phase of acetonitrile (A)and water (0.1% formic acid contained)(B)with gradient elution.The compounds absorbed in serum was identi-fied by multiple stage mass spectrum fragments,published reports and standard substances.Results Fi-nally,14 compounds were preliminary identified in rats serum,of which ten compounds were prototypes of SYD and four were metabolites.Nine peaks were validated as harpagide,chlorogenic acid,loganin,swero-side,liquiritin,hyperoside,angoroside C,isochlorogenic acid C and harpagoside.Conclusion Constitu-ents of SYD in serum were generated from compatibility of all herbal medicines.This study would contrib-ute to the further study of pharmacodynamics properties and metabolism of SYD.
2.Dynamic changes of chemical compositions in volatile oil extracted from Menthae Haplocalycis Herba during different harvest periods using HS/SPME-GC-MS

Objective To determine the chemical compositions of volatile oil extracted from Menthae Haplocalycis Herba (mint) collected during different harvest periods by using headspace solid phase micro-extraction coupled with gas chromatography-mass spectrometry (HS/SPME-GC-MS).Methods The volatile oils were extracted by the method of headspace solid phase micro-extraction, then the chemical compositions were determined by using GC-MS and quantified by the area normalization method.Results A total of 47 compositions were detected in Menthae Haplocalycis Herba in mature stage, and 12 of those had been identified with the total content to be 78.23%.By comparing with the relative amount of menthone, l-limonene and l-menthol extracted from the herbal medicine samples collected during different harvest periods, which are the key components in biosynthesis pathway of the herb, the preliminary determination of the best time to harvest was September.Conclusion The effective and rapid method for the determination of volatile oils provided basis for studying the dynamic changes of the chemical compositions of volatile oil extracted from Menthae Haplocalycis Herba and the proper time to harvest.
