1.Diffusion Weighted Imaging in Evaluating the Prognosis of Colorectal Cancer Patients with Regional Lymph Node Metastasis

PurposeDiffusion weighted imaging (DWI) can significantly improve the diagnosis of non-enlarged lymph node metastasis. This study aims to evaluate the correlation between the DWI findings and the prognosis, and to identify prognostic factors.Materials and Methods Forty-seven patients with colorectal cancer underwent MRI scan including DWI sequence before surgery. Imaging ifndings were compared with the pathologic results to determine the metastatic lymph nodes (DWI positive) or non-metastatic lymph nodes (DWI negative). Postoperative disease-free survival and overall survival for 5 years of the patients with DWI positive and DWI negative lymph nodes were compared. Correlation between the prognosis and the related factors were investigated including regional DWI-positive lymph nodes, short axis diameter and long axis diameter of the largest DWI-positive lymph node, and number of DWI-positive nodes.Results Of 47 patients,10 (21%) patients had regional DWI-positive lymph nodes showed high signal intensity on diffusion-weighted images. The patients with regional DWI negative lymph nodes had a signiifcant better ifve-year disease-free survival and overall survival (P<0.05). The short axis diameter of the largest DWI-positive lymph node was correlated with distant metastasis (AUC=0.77,P<0.05). The short axis diameter and long axis diameter of the maximum metastatic lymph nodes were correlated with overall survival (AUC=0.84 and 0.75,P<0.05). Five-year disease-free survival and overall survival of the patients with short axis diameter of the largest DWI-positive lymph node ≤9 mm were higher than the patients with lymph node short axis diameter >9 mm (P<0.05). Five-year disease-free survival and overall survival of the patients with long axis diameter of the largest DWI-positive lymph nodes ≤11 mm were higher than the patients with long axis diameter >11 mm (P<0.05). Five-year disease-free survival of the patients with all DWI positive lymph nodes resected was higher than the patient without DWI-positive lymph nodes resected (P<0.05).Conclusion The patients with regional DWI-negative lymph node had a better prognosis. Of the patients with DWI-positive lymph nodes, the patients with smaller lymph nodes have better prognosis than who have larger lymph nodes.
2.Protein stability regulators screening assay (Pro-SRSA):protein degradation meets the CRISPR-Cas9 librar y

The regulation of protein stability is a fundamental issue for biophysical processes, but there has not previously been a convenient and unbiased method of identifying regulators of protein stability. However, as reported in the article entitled“A genome-scale CRISPR–Cas9 screening method for protein stability reveals novel regulators of Cdc25A,”recently published in Cell Discovery, our team developed a protein stability regulators screening assay (Pro-SRSA) by combining the whole-genome clustered regularly interspaced short palindromic repeats Cas9 (CRISPR–Cas9) library with a dual-lfuorescence-based protein stability reporter and high-throughput sequencing to screen for regulators of protein stability. Based on our ifndings, we are conifdent that this effcient and unbiased screening method at the genome scale will be used by researchers worldwide to identify regulators of protein stability.
3.A Case of Cerebral Infarction Caused by Tumor Emboli from the Site of.

Multiple cerebral infarction developed in a 36-year-old woman with adenocarcinoma of the right lung with direct cardiac invasion. Neurological examination and brain MRI showed findings of ordinary infarction. Transesophageal echocardiography showed left atrial pedunculated mass which was the potential source of embolism. We report a rare case who had such multiple spontaneous tumor emboli large enough to result in clinically detectable cerebral infarction.

4.Effects of aqueous extract of Hedysarum austrosibiricum on metabolism of oxyen free radicals in subacute aging mice caused by D-galactose.

OBJECTIVETo investigate the anti-aging effect of aqueous extract of Hedysarum austrosibiricum cultivated in Xin-jiang.
METHODSubacute aging model in mice was established by D-galactose (D-gal) and activities of SOD and GSH-PX, contents of MDA in brain and liver tissues, activities of MAO in brain tissue and immune indexes were determined.
RESULTAqueous extract of H. austrosibiricum Xinjiang markedly increased the activities of SOD and GSH-PX, significantly decreased contents of MDA in brain and liver tissues. MAO activities in brain tissue were also decreased. It also elevated the spleen and thymus indexes.
CONCLUSIONAqueous extract of H. austrosibiricum might have anti-aging effect, which is implemented by eliminating oxyen free radicals, rising activities

5.Study on limit detection of flavones in diterpene ginkgolides meglumine injection materials by LC-MS and HPLC-DAD.

Limit test of flavones in diterpene ginkgolides meglumine injection materials by UV-Vis and HPLC-DAD method was studied in this essay. The HPLC-DAD method has lower LOD (about 1% of the UV-Vis), that is, the sensitivity is higher than UV-Vis method. Through the analysis of the kinds of flavonoids ingredients in the samples by LC-MS, the three compounds with highest contents are kaempferol, quercetin and isorhamnetin. Kaempferol, quercetin and isorhamnetin were chosen as reference compounds for HPLC analysis, and the HPLC separation analysis was carried on an Agilent Eclipse plus C18 column (4.6 mm x 250 mm, 5 μm) with methanol and water containing 0.4% phosphoric acid (50: 50) as mobile phase, and the flow rate was 1.0 mL x min(-1). The detection wavelength was set at 360 nm. This method has good specificity, precision and reproducibility. The LODs of quercetin, kaempferide and isorhamnetin were 27.6, 22.3, 29.5 μg x L(-1). The average recovery was 87.9% (RSD 3.3%), 91.7% (RSD 3.1%), 88.3 (RSD 1.3%) for quercetin, kaempferide and isorhamnetin, respectively. Based on the 10 batches of sample results and sensitivity of different HPLC, the content of total flavonoids ingredients of diterpene ginkgolides meglumine injection materials was limited no more than 2 x 10(-5). This method is simple, quick and has good maneuverability, and could be used to the limit test of flavonoids in the diterpene ginkgolides meglumine injection materials.

6.Establishment of recombinase polymerase amplification assay for five hemorrhagic fever-related viruses

Objective To establish a one-step recombinase polymerase amplification (RPA) method for pathogen screening and rapid detection in the field targeting for five hemorrhagic fever related viruses (Zaire ebola virus, Sudan ebola virus, Marburg virus, Lassa virus and Yellow fever virus). Methods The specific nucleic acid (NA) fragments of each virus were selected as target genes by genome sequence analysis, and the primers and probes for RPA assays were designed according to the sequence. A series of diluted template genes were used for RPA detection to determine the sensitivity. The hemorrhagic fever-related viral nucleic acids were used for RPA detection to determine the specificity. The amplification experiments were carried out at different temperature ranging from 37℃ to 42℃ to validate the reaction temperature range. Results The RPA reaction systems of the five hemorrhagic fever viruses could effectively amplify the target genes, the sensitivities were between 1.5×102 and 1.5×103 copies. No cross reactions existed with the other hemorrhagic fever-related viral genes. Meanwhile, RPA assay could effectively amplify the target genes at 37-42℃. Conclusion The isothermal RPA assays of five hemorrhagic fever viruses are established, which may amply target genes fast and react at a wide temperature range, and be potentially useful for in field pathogens detection.
7.Therapeutic effect of sea buckthorn oil on allergic contact dermatitis in mice

Objective To study the therapeutic effect of sea buckthorn oil on allergic contact dermatitis(ACD) in mice. Methods A total of 60 ICR mice were randomly divided into 6 groups: blank control group, model control group,three groups of high-dose, medium-dose and low-dose sea buckthorn oil(100%, 50%, 25%, ad us. ext.), respectively,and positive control group(compound dexamethasone acetate cream), with 10 mice in each group. The mouse model of ACD was established by sensitization and stimulation with 2,4-dinitrochlorobenzene(DNCB),and injuries in the ear skin of the mice were scored and used as the indicator of successful establishment of the model. The degree of ear swelling and the spleen index of the mice in each group were measured at 48 h after stimulation. Hematoxylin and eosin staining was used to observe the pathological changes in the mouse ear skin. Results Measurement of the degrees of ear swelling showed that the high-dose sea buckthorn oil group(9 mL/kg,ad us. ext.)obviously inhibited the DNCB-induced ear swelling in the mice, with a significant difference compared with the model group(P < 0.01)and non-significant difference compared with the positive control group(P> 0.05). The result of the spleen index assessment showed that sea buckthorn oil at each dose may play an immune-regulatory role by promoting the development of immune organs. The pathological examination showed that epidermis thickening of the ear tissue,epidermal and dermal edema and infiltration of lymphocytes were reduced, dermal vasodilatation and congestion were alleviated, and inflammatory cell infiltration was inhibited in the three groups of different doses of sea buckthorn oil compared with the model control group. Conclusions Sea buckthorn oil has an obvious inhibitory effect on the DNCB-induced allergic contact dermatitis in mice. However, its related mechanisms remain to be further investigated.
8.A clinical study into the vestibular function and therapy of patients with chronic positional symptoms after acute vestibular syndrome

Objective:To investigate the status of the vestibular function of the patients with chronic positional symptoms after peripheral acute vestibular syndrome (AVS) and the curative effect of the vestibular rehabilitation therapy (VRT). Method:Using caloric test (CT), head shaking nystagmus test (HST), cervical vestibular evoked myogenic potentials as well as ocular vestibular evoked myogenic potentials to estimate the function of semicircular canal and otolith organs. The patients with normal VEMPs are divided as Group A. Otherwise are as Group B. Both groups are treated with VRT. The curative effect is estimated by vestibular symptom index (VSI) and Berg balance scale (BBS). Result:Thirty-three of 37 patients (86.5%) had an abnormal result of CT and HST, with 23 of these patients (65.7%) had an abnormal of both test. Twenty-two patients (59.5%) were in Group A and 15 (40.5%) in Group B. Before the therapy, Group B had a higher score of the balance and dizziness symptoms of VSI (P<0.05), and Group A had a higher score of the BBS (P<0.05). After the therapy, the VSI scores of both groups dropped and scores of the BBS raised. Conclusion:Patients with chronic positional symptoms after peripheral AVS have dynamic vestibular lesions to different extents. Those with otolith organs lesions tend to have a worse function of balance. Nevertheless, patients have a better off after VRT.

10. Study on microscopic marginal extensions of malignant liver tumors based on tumor types and ultrasonic characteristics

Objective:
To study the relationships between microscopic marginal extensions and tumor types, and ultrasonic characteristics of malignant liver tumors.
Methods:
Two-dimensional ultrasonography, contrast-enhanced ultrasonography and ultrasound-guided biopsy of hepatic tumors and surrounding hepatic tissues were performed in 78 patients with malignant liver tumor. Pathological microscopic extensions were observed after hematoxylin-eosin staining of biopsy specimens.
Results:
The microscopic marginal extension rates of hepatocellular carcinoma (HCC), intrahepatic cholangiocarcinoma (ICC) and metastatic liver cancer (MLC) were 62.5%(30/48), 87.5%(7/8), and 91.0% (20/22), respectively. For tumors with well-defined and poorly-defined two-dimensional ultrasound boundary, microscopic marginal extension rates were 50%(14/28), and 86%(43/50).
