1.Effect of Rosmarinic Acid on Antioxidative Systems in Mesangial Proliferative Glomerulonephritis in Rats

Objective To investigate the change of oxidation system and antioxidation system in mesangial proliferative glomerulonephritis(MsPGN) induced by anti-Thy1.1 antibody,and further to study the intervention of rosmarinic acid(RAD).Methods Anti-THy1.1 serum was produced,and then intravenously injected into rats for establishing an experimental model of MsPGN.The experiment was designed for control with or without RAD,glomerulonephritis with or without RAD,respectively.The activity of superoxide dismutase(SOD) and the content of malondialdehyde(MDA) in tissue homogenate were detected by spectrophotomerty.Results The activity of SOD significantly decreased,while the content of MDA increased in MsPGN.RAD could inhibit oxidation in the mesangial cells.Conclusion Lipid peroxidation participates in MsPGN and RAD can control the changes of the mesangial cells and show the activity of antioxidation.
2.The delivery mechanism of micro-porous osmotic pump tablets.

To investigate the delivery mechanism of micro-porous osmotic pump tablets ( MPOP), taking tramadol hydrochloride ( TR) as the model drug, tramadol hydrochloride micro-porous osmotic pump tablets (TR MPOP) were prepared with compressible starch as diluent, cellulose acetate as coating material, polyethylene glycol 400 as pore-forming agents. The equilibrium solubility and osmolality of TR were determined. The effects of fillers in tablet cores, coating levels, and osmotic pressures of release media on expansion behavior of preparations were described. The influences of the category, osmolality, and pH value of release media, release methods, and release conditions on release curves of tablets were evaluated. Based on several models, the delivery pattern of TR MPOP was fitted. The equilibrium solubility in water and osmolality of TR were (775.8 +/- 17.7) g x L(-1) and 4.036 Osmol x kg(-1), respectively. During the drug-release period, it was observed that the tablets expanded markedly in response to the expansion characteristics of compressible starch and the osmotic pressure difference across the membrane. When osmotic pressure of release media increased, the significant change of the equilibrium solubility of TR was not found, but the release rates of TR MPOP decreased significantly. The delivery rate was not influenced by the pH of release mediums, dissolution methods and paddle stirring rates. The drug release profile conformed to the model of zero order in 8 h. The pore-forming agents were dissolved in release medium, which caused micro-pores. The expansion of tablets made the size of micropores bigger, and then the drug-releasing pores were obtained. It was proved that the drivers of drug delivering from TR MPOP were mainly the difference of osmotic pressure, and secondly the difference of solubility. TR MPOP were the controlled-release preparation.

3.Determination of salidroside and p-tyrosol in Hongjingtian for injection(freezing-dry) by SPE-HPLC.

OBJECTIVETo assay salidroside and p-tyrosol in Hongjingtian for injection (freezing-dry).
METHODSamples were purified by Sep-Pak C18 column and salidroside and p-tyrosol were determined by HPLC with Irregular-H C18 (4.6 mm x 250 mm, 5 microm), and eluted with a mobile phase of methanol-acenitonitrile -0.06% phosphate (10: 10:80). The flow rate was 1.0 mL x min(-1), the detection wavelength was set at 275 nm and the column temperature was maintained at 30 degrees C.
RESULTThe calibration curves were linear in the range of 2.24-22.4 microg for salidroside (0.999 7) and 0.856-8.56 microg for p-tyrosol (0.999 6), the average recovery was 101.3%, 99.8% respectively.
CONCLUSIONThe method is convenient, rapid, accurate and reliable.

4.The relationship between arsenic in drinking water and skin lesions in endemic arsenism area in Shanyin County of Shanxi Province

Objective To investigate the relationship between arsenic in drinking water and skin lesions in endemic arsenism area in Shanyin County of Shanxi Province,in order to provide epidemiologic data for further arsenism research.Methods One hundred and eighty-nine endemic arsenism patients and 59 controls were randomly selected in 17 endemic amenism countries in Shanyin County of Shanxi Province.The content of arsenic in drinking water which wa8 collected indoom was half-quantitatively screened by a kit made by Chinese Center for Disease Control and Prevention,then quantitatively determined by HPLC-ICP-MS.Patients of endemic arsenism were diagnosed by "The Standard of Diagnosis for Endemic Amenism"(WS/T 211-2001).Results There were 64.9% (87/134)samples above the arsenic level(50μg/L)of drinking water and the median value of arsenic in drinking water was 91.43 μg/L in 134 water samples.The OR(95%CI)value between arsenic in drinking water and hyperkeratosis,hyperpigmentation,depigmentation was 2.46(1.22-4.94),3.34(1.50~7.44)and 2.86(1.50-5.46),respectively.The prevalence of hyperkeratosis,hyperpigmentation and depigmentation increased,as the arsenic in drinking water increased(≤10,≤50,≤200,>200μg/L),especially in>200μg/L group(OR=6.15,13.96,11.41,P<0.05).The arsenic level in drinking water of Ⅲ degree of depigmentation patients(318.300μg/L)was higher(P<0.05)than that of 0,Ⅰ and Ⅱ degree groups(86.670,131.800,1 10.590μg/L,P<0.05).Conclusions Shanyin County is a medial arsenic pollution area. Arsenic in drinking water is considered as a risk factor of skin lesion. The degree of skin lesions increased,as the arsenic in drinking water increased.
5.Diagnosis and treatment of pancreatic trauma.

OBJECTIVETo improve diagnosis and therapeutic efficacy for pancreatic trauma.
METHODSWe retrospectively analyzed 71 cases of pancreatic injuries received in our department from 1987 to 2004. Different surgical procedures were performed according to different patterns of pancreatic injuries. Among them, 31 cases were defined as Grade I or II injury according to the pancreatic organ injury score developed by American Association for the Surgery of Trauma and were performed pancreas debridement and drainage; 26 cases belonged to Grade III injury and were performed distal pancreatectomy plus external drainage; 10 cases of Grade IV injury in whom 8 were performed distal Roux-en-Y pancreaticojejunostomy and 2 were performed Whipple operation; 4 cases of Grade V injury in whom 1 was performed restoration of duodenal damage, suture of proximal pancreatic laceration and distal Roux-en-Y pancreaticojejunostomy, and 3 were performed Whipple operation.
RESULTSSixty-six cases were cured, of whom 14 had postoperative complications, and 5 cases died. The causes of death were of pancreatic fistula in 2 cases, upper gastrointestinal bleeding in 1 case, ARDS in 1 case, and serious abdominal infection in 1 case.
CONCLUSIONSPreoperative diagnosis for pancreatic trauma is rather challenging. Prompt explorative laparotomy is still a reliable diagnostic method for pancreatic trauma. In order to improve curative rate, different surgical procedures should be undertaken according to different sites and grades of pancreatic traumas.

6.Modulating drug loading and release profile of beta-cyclodextrin polymers by means of cross-linked degree.

The purpose of the present study is to use beta-cyclodextrin polymers (beta-CDP) with different cross-linked degree (CLD) to form inclusion complexes with ibuprofen and examine the effects of structural and compositional factors of beta-CDP on its drug loading and release behaviors. A series of beta-CDP with different CLD were synthesized and characterized by Fourier Transform Infrared Spectroscopy (FT-IR) and 13C NMR spectrum. The beta-CDP was systemically characterized for the relation between the CLD of beta-CDP and the drug loading and release as well. The results of FT-IR and 13C NMR showed that similar peak-shaped vibration of beta-CDP and beta-CD implies that the polymer keeps the original characteristic structure of beta-CD. The CLD of the beta-CDP played a critical role in the drug loading and release, increasing the CLD resulted in reduction of drug loading, but increase in drug release.

Thirty-eight individuals with normal glucose regulation and thirty-nine newly diagnosed type 2 diabetic patients were observed by continuous glucose monitoring system(CGMS)for three days.The mean blood glucose,mean amplitude of glycemic excursions(MAGE),and absolute means of daily differences(MODD)were calculated in each subject.The results suggested that the amplitude of glyeemic excursions revealed by CGMS could be used to evaluate the quality of glycemic control in type 2 diabetic patients.

To study the effects of surfactants on wettability of excipients, the contact angles of six types of surfactants on the surface of two common excipients and mixture of three surfactants with excipients were measured using hypsometry method. The results demonstrated that contact angle of water on the surface of excipients was associated with hydrophilcity of excipients. Contact angle was lowered with increase in hydrophilic groups of excipient molecules. The sequence of contact angle from small to large was starch < sodium benzoate < polyvinylpyrrolidone < sodium carboxymethylcellulose < sodium alginate < chitosan < hydroxypropyl methyl cellulose
9.The preparation and kinetic study on enzymatically-controlled drug release of isotretinoin/amylose inclusion complex.

The inclusion complex of isotretinoin was prepared by sealed-control temperature method and amylose was used as carrier. The formation of inclusion complex was confirmed by powder X-ray diffraction and DSC. The equation of enzymatically-controlled drug release was established by kinetic theory, and the release characteristic of drug was confirmed by using the kinetic equation. The results show that the drug release was attributed to first order reaction without alpha-amylase. However, with alpha-amylase, the drug release was an acceleration process by the effect of both dissociation and enzymatic hydrolysis simultaneously. The research indicates that drug release from the inclusion complex was modulated by the addition of alpha-amylase.

10.The potential effect of proanthocyanidins on the stability of resin-dentin bonds against thermal cycling.

OBJECTIVETo investigate the potential effect of proanthocyanidins (PA), a natural cross-linker, on the stability of resin-dentin bonds against thermal cycling.
METHODSTen percent, 15% PA-based preconditioners, and 5% glutaraldehyde were prepared for the transient pretreatment of demineralized dentin before bonding. Specimens without pretreatment were used as negative controls (n = 4 teeth for each group). Microtensile bond strength, failure mode, micromorphologies of resin-dentin interface and the collagen degradation of bonded specimens after thermal cycling were evaluated.
RESULTSAfter thermal cycling, the microtensile bond strength values of resin-dentin bond in groups pretreated with 15% PA for 120 s and 60 s [(23.09 ± 3.19) and (21.88 ± 3.49) MPa] were significantly higher than that in control group [(15.47 ± 3.78) MPa] (P < 0.05). Mixed fractures were the most prevalent failure mode. Specimens with pretreatment presented compact hybrid layer, while some narrow gaps were found in hybrid layer of non-treated specimens. Collagen biodegradation rates in groups with pretreatment were significantly lower than that in control group (P < 0.05). Among them, specimens pretreated by 15% PA preconditioner for 120 s exhibited the lowest biodegradation rates [(0.316 ± 0.019) mg/g].
CONCLUSIONSThe application of natural cross-linker PA on demineralized dentin reduced the bond degradation against aging by thermal cycling, and can be helpful to create more durable bonds to dentin.
