Objective To investigate the change of oxidation system and antioxidation system in mesangial proliferative glomerulonephritis(MsPGN) induced by anti-Thy1.1 antibody,and further to study the intervention of rosmarinic acid(RAD).Methods Anti-THy1.1 serum was produced,and then intravenously injected into rats for establishing an experimental model of MsPGN.The experiment was designed for control with or without RAD,glomerulonephritis with or without RAD,respectively.The activity of superoxide dismutase(SOD) and the content of malondialdehyde(MDA) in tissue homogenate were detected by spectrophotomerty.Results The activity of SOD significantly decreased,while the content of MDA increased in MsPGN.RAD could inhibit oxidation in the mesangial cells.Conclusion Lipid peroxidation participates in MsPGN and RAD can control the changes of the mesangial cells and show the activity of antioxidation.

To study the effects of surfactants on wettability of excipients, the contact angles of six types of surfactants on the surface of two common excipients and mixture of three surfactants with excipients were measured using hypsometry method. The results demonstrated that contact angle of water on the surface of excipients was associated with hydrophilcity of excipients. Contact angle was lowered with increase in hydrophilic groups of excipient molecules. OBJECTIVETo detect the influencing factors for posttraumatic hydrocephalus in patients with severe traumatic brain injuries and provide theoretical reference for clinical treatment.

METHODSRetrospective study was made on 139 patients with severe traumatic brain injuries in our hospital. The patients were divided into two groups: hydrocephalus group and non-hydrocephalus group. Single factor analysis and multiple factor analysis were used to determine the related factors and hydrocephalus. Multiple factor analysis was conducted with logistic regression.

RESULTSPosttraumatic hydrocephalus was found in 19.42% of patients. Age(OR equal to 1.050, 95%CI: 1.012-1.090), decompressive craniectomy (OR equal to 4.312, 95%CI: 1.127-16.503), subarachnoid hemorrhage(OR equal to 43.421, 95%CI: 7.835-240.652) and continuous lumbar drainage of cerebrospinal fluid (OR equal to 0.045, 95%CI: 0.011-0.175) were screened out from nine factors as the influencing factors for posttraumatic hydrocephalus.

CONCLUSIONSRisk factors for PTH are as follows: age, decompressive craniectomy and subarachnoid hemorrhage (SAH). Continuous lumbar drainage of cerebrospinal fluid can greatly reduce posttraumatic hydrocephalus.

To increase the dissolution rate and extent of valsartan, valsartan nanosuspensions have been prepared. Controlled precipitation assisted with sonication is utilized to prepare valsartan nanosuspensions, the concentration of the drug, stabilizer and costablizer had a great effect on the stability of the preparation according to the pre-experiment. So the method of central composite design-response surface is used to optimize the prescription based on the above three factors and the particle size as the response value. The software Origin 8.0 is used to draw the view of the three-dimensional effects and 2D contour map, to get the optimal prescription area. Valsartan nanosuspensions were prepared. The mean diameter and zeta potential are about 216.6 nm and -57.7 mV, respectively. Compared with the microsuspensions and commercial preparation, the dissolution of valsartan nanosuspensions was faster and the bioavailability can be enhanced to some extent.

Objective To evaluate the clinical significance of glycated albumin(GA),a parameter in reflection of recent glycemic control in patients with diabetes mellitus. Methods Four hundred and forty-five patients with type 2 diabetes mellitus who were hospitalized in our hospital from May to November 2006 were enrolled into the study.The fasting plasma glucose(FPG),postprandial 2-hour blood glucose(P2hBG) and glycated hemoglobin(HbA1c) were measured,the enzymatic measurement of GA was conducted and the CGMS was performed.The correlation between GA and the other parameters monitored was analysed. Results The correlation analysis indicated that GA was well correlated with HbA1c(r=0.818,P0.05),respectively for those with HbA1c more than 7.5%,between 6.5% and 7.5%,and less than 6.5%. Conclusion GA is well correlated with HbA1c,especially in those with poor glycemic control for a long time.The correlation between GA and long-term glycemic control is stronger than that between GA and instant plasma glucose or MBG in three days.

OBJECTIVETo discuss the role of gadolinium chloride (GdCl(3)) in lung injury associated with acute necrotizing pancreatitis (ANP).

METHODSExperimental animals were randomized into five groups (n = 18 for each group): normal control group, ANP group, GdCl(3) pretreatment group, ANP GdCl(3) pretreatment group, ANP GdCl(3) treatment group. Rat ANP model was induced by intraductal administration of 3% sodium taurocholate. Alveolar macrophages (AM) were obtained by bronchoalveolar lavage. The blood gas assay, the ratio of wet/dry tissue, protein content of bronchoalveolar lavage fluids (BALF), the myeloperoxidase (MPO) of lung tissue and generation of TNFalpha and NO by AM were evaluated. The apoptosis of AM was checked by agarose gel electrophoresis analysis, transmission electric microscopy observation and cytometry propidium iodide single stained method. The lung tissue was examined by histology.

RESULTSThe parameter of GdCl(3) pretreatment group compared with normal control group had no statistical significance (P > 0.05). The indicators of ANP GdCl(3) pretreatment group and ANP GdCl(3) treatment group were elevated compared with the normal control group and had statistical significance (P < 0.05). But compared to the ANP group, they were all decreased and also had the statistical significance (P < 0.05). The 180 - 200 bp ladder pattern unique to apoptosis in agarose gel electrophoresis and the apoptotic typical morphologic feature in AM by transmission electric microscopy and typical subdiploid peak in DNA content figure could be observed in ANP GdCl(3) pretreatment group and ANP GdCl(3) treatment group, while the other three groups could not.

CONCLUSIONSLung injury associated with ANP could be ameliorated by application of GdCl(3) through inducing apoptosis of AM of ANP.

OBJECTIVETo study the significance of vascular endothelial growth factor (VEGF) and microvascular density (MVD) expression in breast cancer.

METHODSThe expression of MVD and VEGF was studied in 81 patients with primary breast cancer by SP immunohistochemical technique.

RESULTSThere were 49 patients with high VEGF expression (60.5%) and 35 patients with high MVD expression (43.2%). There was significant correlation between VEGF or MVD expression and TNM stage, but not age, tumor size, ER expression or lymph node status. VEGF was significantly related with MVD expression. Univariate analysis showed that patients with low expression of VEGF and MVD had longer disease free survival (DFS) and overall survival (OS). Grouping univariate analysis showed that VEGF had significant correlation with the DFS of all grouped patients and the OS of patients with LN(+) or stage III lesions. MVD had significant correlation with the DFS and OS of LN(+) patients and the DFS of stage III lesion patients. Multivariate analysis showed that MVD was a risk predictor because of its positive statistic value, the higher expression, the shorter survival time of the patients.

CONCLUSIONBoth VEGF and MVD are useful prognostic factors in breast cancer. MVD appears to be a strong and independent biologic marker for breast cancer prognosis.

OBJECTIVESTo assess the effect of freezing on the sperm DNA.

METHODSTo assess the sperm DNA preserved at -80 degrees C by using the single cell gel electrophoresis (SCGE).

RESULTSThere was no statistical difference on the time factor by analysis of variance (ANOVA).

CONCLUSIONSIntegrity of sperm DNA could not be devastated in frozen states.

OBJECTIVETo investigate if the Ribbond polyethylene fiber has the effect of reinforcing polymethyl methacylate.

METHODS28 specimens were fabricated and divided into 3 groups: group of chemical-cured PMMA, group of chemical-cured PMMA reinforced by stainless steel wire and group of chemical-cured PMMA reinforced by Ribbond polyethylene fiber. A three-point bending test was used to measure the flexural strength and flexural modulus of specimens. Then the data were analyzed with a one-way analysis of variance.

RESULTSThe flexural strength of chemical-cured PMMA group was (51.383 +/- 2.761) MPa, the flexural modulus was (1791.2 +/- 113.760) MPa; The flexural strength of stainless steel wire reinforced group was (58.725 +/- 1.218) MPa, the flexural modulus was (2092.76 +/- 120.28) MPa; The flexural strength of Ribbond polyethylene fiber reinforced group was (80.975 +/- 2.58) MPa, the flexural modulus was (2866.53 +/- 107.51) MPa. The one-way analysis of variance showed that the results were significant (P < 0.001). Newman-Keuls method showed that the differences among all groups were significant (P < 0.05).

CONCLUSIONThe Ribbond polyethylene fiber can raise the flexural strength and flexural modulu of polymethyl methacylate.

Objective To explore the protective effects and its mecha-nism of stachydrine on focal cerebral ischemia reperfusion injury in mice . Methods The focal cerebral ischemia reperfusion injury model in mice was established by thread method.KM mice were randomly divided into 6 groups(n＝16):sham operation group , model group, control group, ex-perimental-L, experimental -M, experimental -H groups, mice in each group were given related drugs via intragastric administration before the operation, once a day, lasting for 7 d.Reperime 22 h, the levels of Caspase-3, B-cell lymphoma-2 (Bcl-2), B-lymphoma-2 gene-related X protein (Bax) and adenosine triphosphate (ATP) enzyme activity in each group was detected by enzyme linked immunosorbent assay .Results The relative expression of Bax in model group,control group, experimental -L, experimental -M, experimental -H groups were (4.00 ±0.51 ), (2.84 ±0.48),(2.87 ±0.41),(2.92 ±0.46),(3.12 ±0.59)ng· mL-1,respectively; the relative expression of Caspase-3 in the 5 groups were(13.43 ±0.87),(9.95 ±1.57),(10.02 ±1.55),(10.58 ±1.48),(10.79 ±1.69) pmol· L-1,respectively; the relative expression of Bcl -2 in the 5 groups were (11.94 ±1.40 ),(16.14 ±1.33 ), (15.89 ±1.45),(15.31 ±1.20),(14.78 ±1.17)ng· mL-1,respectively;compared with the model group, the rela-tive expressions of Bax and Caspase -3 in control group, three doses experimental groups markedly reduced while the relative expression of Bcl-2 in the 4 groups markedly increased significantly (all P<0.01).The Na +-K+-ATP en-zyme activity in the model group ,control group, experimental -L, experimental -M, experimental -H groups were (2.82 ±0.56),(4.84 ±1.61),(4.81 ±1.48),(4.50 ±1.22),(4.13 ±1.17)μmolPi· mg -1· h -1,respectively;and Mg2+-ATP enzyme activity in the 5 groups were (2.55 ±0.56), ( 5.25 ±1.86 ), ( 5.15 ±2.37 ), (4.62 ±1.89 ) ,(4.13 ±1.54 ) μmolPi· mg-1· h-1, respectively; comparison between 4 drugs groups and model group,the differences of the factors were significant ( P<0.05 or P<0.01 ) .Conclusion Stachydrine can markedly reduce the lesion of focal cerebral ischemia reperfusion model in mice , and its mechanism may be related to improve energy metabolism disorder , inhibit neuronal apoptosis and improve the microcirculation of the brain .
