1.The effects of DEHP on morphology and function of progenitor Leydig cell in rat.

OBJECTIVETo investigate the effects and mechanisms of diethylhexylphthalate (DEHP) on morphology and function of progenitor Leydig cells (PLC) in rats.
METHODSTwenty pregnant SD rats were randomly divided into 4 groups ( n = 5): normal control group, DEHP low dose group , middle dose group, and high dose group, which were treated from postnatal day (PND) 1 to PND 21 of the pubs with DEHP at the doses of 0, 10, 100, 750 mg/(kg · d) in 0.5 ml of corn oil by gavage respectively. At the end of the treatment, the male pups were killed and blood samples were collected for determination of serum testosterone concentration by chemiluminescence method. The body weight, testis weight and anogenital distance (AGD) were measured. The morphology of PLC was observed by light and transmission electron microscopy. The protein expression of steroidogenic acute regulatory protein(StAR) in PLC was determined by immunohistochemistry. The mRNA expression of insulin-like growth factor-I (IGF-I) in the testis was assayed by real-time PCR.
RESULTSCompared with normal control group, the serum testosterone and AGD of male pubs from the middle and high dose groups were declined significantly (P < 0.01), the testis weight and body weight from high dose group were decreased significantly (P < 0.01), while the testis weight increased in the low dose group (P < 0.05). Under light microscope, PLC showed hyperplasia and cluster aggregation in the low dose group and focal hyperplasia in the middle and high dose group. The spermatogenic cells in seminiferous tubules showed decrease, apoptosis and unfix in the high dose group. Under transmission electron microscope, the PLC showed decreased lipid droplets, smooth endoplasmic reticulum and mitochondriae in the treated group. The mRNA expression of IGF-I increased in the low dose group, and the protein expression of StAR decreased in the middle and high dose group.
CONCLUSIONLactating exposure to DEHP may interfere with the synthesis of testosterone of PLC in male pubs, the decrease of StAR and the damage of PLC may be involved in it.

2.Effect of tanshinone ⅡA sodium injection in the treatment of portal hypertension patients with esophageal varices

Objective To investigate the clinical efficacy of sodium tan-shinone ⅡA in the treatment of portal hypertension patients with esopha-geal varices and its impact on angiotensinⅡand hemodynamic.Methods (P <0.05).The incidence of adverse reactions in treatment group and control group were 3.33%(2/60), 10.00%(6/60), with significant difference (P<0.05).Conclusion The clinical efficiency of tanshinone ⅡA sodium on patients with esophageal varices and portal hypertension was significant , with low incidence of adverse reactions.

4.Investigation of anxiety and depression of osteosarcoma patients and its influence factors

Objective To study the incidence of anxiety and depression among patients with osteosarcoma so as to analyze its influencing factors.Methods Between January 2010 and December 2011,94 hospitalized patients with osteosarcoma were investigated with self-rated anxiety scale ( Self-rating Anxiety Scale,SAS) and self-rated depression scale (Self-rating Depression Scale,SDS),then analyzed the influencing factors.Results And the incidence of anxiety and depression was 27.7% and 44.7%,respectively.The total score of SAS and SDS of patients with osteosarcoma (44.95 ± 11.11) and (50.22 ± 12.40),was significantly higher than that of the national norm[ ( 33.80 ± 5.90 ),(41.85 ± 10.57 ) ; t =6.878,4.629,respectively;P<0.01].Logistic analysis showed that gender,disease stage and education were the influencing factors for anxiety and depression of patients with osteosarcoma.Conclusions Patients with osteosarcoma are easily have the mental problems of anxiety and depression,so psychological interventions,social support and necessary medicine treatment are necessary for osteosarcoma patients with anxiety and depression.

OBJECTIVETo explore association genetic polymorphism of XPD with chromosomal damage in workers exposed to radiation.
METHODS182 workers exposed to radiation for at least one year with chromosomal damage were selected as cases based on a general health examination for all workers exposed to radiation in Tangshan city. The control group without chromosomal damage was matched to case by age (within 5 years), sex, work unit, type of exposed to radiation, cumulate serve length (within 1 year) according to 1:1. The micro whole blood cultivation was used for the chromosome analysis. The chromosome aberration type and rate were observed and counted. The polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) was used to examine the genotype of three XPD loci (751, 312 and 156).
RESULTSThe frequency of XPD 751 AA in cases was higher than that in controls (P < 0.05). The frequency of 751 allele in case group was statistically higher than that in the control groups (P < 0.05). No statistical difference was found in the frequencies of XPD 312 genotype and allele between the case and control group (P > 0.05). 156 mutant gene type in case group was higher than that in the control groups. The frequency of 156 A allele in case group were higher than that of the control groups (P < 0.05). The frequency of genotype with both 751AA and 156CA or 751AA and 156AA was higher in cases than that of controls (P < 0.05).
CONCLUSIONXPD 751AA genotype is a possible risk factor for radiation-induced chromosomal damage. XPD 156 mutant gene type is a possible risk factor for radiation-induced chromosomal damage. Individuals with both XPD 751AA and 156 (CA+AA) genotypes are susceptible to radiation-induced chromosomal damage. No association of XPD 312 polymorphism with radiation-induced chromosomal damage is found.

7.Regulating effects of novel CpG chitosan-nanoparticles on immune responses of mice to porcine paratyphoid vaccines.

OBJECTIVETo study the regulating effects of a novel CpG oligodeoxynuleotide and the synergistic effect of chitosan-nanoparticles (CNP) with CpG on immune responses of mice, which were used to develop a novel immunoadjuvant to boost immune response to conventional vaccines.
METHODSA novel CpG ODN containing 11 CpG motifs was synthesized and its bioactivities to stimulate the proliferation of lymphocytes of pig in vitro were detected. Then it was entrapped with CNP prepared in our laboratory by the method of ionic cross linkage, and immunized Kunming mice were co-inoculated with paratyphoid vaccine. The peripheral blood was collected weekly from the tail vein of inoculated mice to detect the contents of IgG, IgA, IgM, and specific antibody against salmonella as well as the levels of interleukin-2 (IL2), IL-4, and IL-6 by SABC-ELISA assay. The numbers of leucocytes, monocytes, granuloytes, and lymphocytes were calculated separately using the routine method. The experimental mice were orally challenged with virulent salmonella 35 days after inoculation.
RESULTSThis CpG ODN could remarkably provoke the proliferation of lymphocytes of pig in vitro in contrast with the control (P < 0.05). Compared with those of the control, immunoglobulins, including IgG, IgA, IgM, and specific antibodies to paratyphoid vaccine, increased significantly in sera from the CpG or CpG-CNP-vaccinated mice (P < 0.05). IL-2, IL-4, and IL-6 increased remarkably in sera from immunized mice (P < 0.05). The leucocytes, monocytes, granuloytes, and lymphocytes of the mice immunized with CpG or CpG-CNP were also increased in number (P < 0.05). After the challenge, these immunity values were elevated in the mice vaccinated with CpG or CpG-CNP. The immunized mice all survived, while the control mice fell ill with evident lesions with diffuse hemorrhage in stomach, small intestine, and peritoneum.
CONCLUSIONSCpG ODN entrapped with CNP is a promising effective immunoadjuvant for vaccination, which promotes humoral and cellular immune responses, enhances immunity and resistance against salmonella by co-administration with paratyphoid vaccine.

8.Expression change and significance of proliferating cell nuclear antigen in mice hepatocellular carcinoma

Objective To investigate the dynamic change and signifi-cance of proliferating cell nuclear antigen ( PCNA ) expression of mice which were induced into hepatocellular carcinoma ( HCC ) by diethylni-trosamine ( DEN ) .Methods Mice continuously induced cancer with DEN containing drinking water for 24 weeks .The mice were killed in the induction of cancer at different time points.The expression of PCNA protein and nucleic acid in mice liver with HCC were evaluated by Western blot and reverse transcription polymerase chain reaction,respec-tively.Results The expressions level of PCNA protein and nucleic acid in the liver tissue in mice after induced into HCC by DEN increased gra-dually.They reached a peaked at 24th week, and there was significant differences between them ( P<0.05 ).Conclusion There is a signifi-cant dynamic change of PCNA expression in the liver tissue in mice during the progression of HCC which were induced by DEN.It suggest that PCNA might play an important role in the incidence and development of HCC.
9.Analysis the expression change of hepatic glycogen in hepatocellular carcinoma in the mice induced by N-nitrosodiethylamine

Objective To analyse the change of hepatic glycogen in the process of hepatocellular carcinoma development in N -nitrosodiethy-lamine(DEN) -induced mice, and investigate the relationship between the hepatic glycogen content and the different stage of liver cancer . Methods There were forty clean and healthy male mice .They were di-vided in two groups ,the normal group ( n=3 ) and the model group ( n=37).After adapting environment in one week ,the group of normal mice were sacrificed and all the model animals were administrated by solution of DEN (30 μg? mL-1 ) in the carcinoma-induced period.Every three mice were sacrificed in the this group induced by DEN in 4 , 8 , 12 , 16 , 20 ,24 weeks(n=21).The obtained liver tissues divided in seven groups and embedded by paraffin were cut at thick piece .After staining with pe-riodic acid-Schiff ( PAS) the hepatic glycogen were observed .In order to obtain the mean optical density , samples of each group were disposed by the software Motic Images Advanced 3.2.Lastly.Analysis of variance ( ANOVA) test was followed by individual comparison by Student -New-man-Keul test using SPSS V16.0.Result The contents of hepatic gly-cogen in normal group , 8-week and 12-week groups were decreased successively and significantly , and the 16-week were least .The con-tents of hepatic glycogen in 16-week compared with 20-week and 24-week groups were increased successively and significantly . Conclusion In the process of development of hepatic carcinoma induced by DEN , the regular of hepatic glycogen′s change was decreasing gradually at the prior period ,and then the level of hepatic glycogen increased later .
10.Comparison of clinical effect and muscle injury imaging between oblique lateral lumbar interbody fusion and transforaminal lumbar interbody fusion in the treatment of single-segment degenerative lumbar spinal stenosis.

OBJECTIVE:
To compare the efficacy and muscle injury imaging between oblique lateral lumbar interbody fusion (OLIF) and transforaminal lumbar interbody fusion (TLIF) in the treatment of single-segment degenerative lumbar spinal stenosis.
METHODS:
The clinical data of 60 patients with single-segment degenerative lumbar spinal stenosis who underwent surgical treatment from January 2018 to October 2019 was retrospectively analyzed. The patients were divided into OLIF groups and TLIF group according to different surgical methods. The 30 patients in the OLIF group were treated with OLIF plus posterior intermuscular screw rod internal fixation. There were 13 males and 17 females, aged from 52 to 74 years old with an average of (62.6±8.3) years old. And 30 patients in the TLIF group were treated with TLIF via the left approach. There were 14 males and 16 females, aged from 50 to 81 years old with an average of (61.7±10.4) years old. General data including operative time, intraoperative blood loss, postoperative drainage volume, and complications were recorded for both groups. Radiologic data including disc height (DH), the left psoas major muscle, multifidus muscle, longissimus muscle area, T2-weighted image hyperintensity changes and interbody fusion or nonfusion were observed. Laboratory parameters including creatine kinase (CK) values on postoperative 1st and 5th days were analyzed. Visual analogue scale(VAS) and Oswestry disability index(ODI) were used to assess clinical efficacy.
RESULTS:
There was no significant difference in the operative time between two groups(P>0.05). The OLIF group had significantly less intraoperative blood loss and postoperative drainage volume compared to the TLIF group(P<0.01). The OLIF group also had DH better recovery compared to the TLIF group (P<0.05). There were no significant differences in left psoas major muscle area and the hyperintensity degree before and after the operation in the OLIF group (P>0.05). Postoperativly, the area of the left multifidus muscle and longissimus muscle, as well as the mean of the left multifidus muscle and longissimus muscle in the OLIF group, were lower than those in the TLIF group (P<0.05) .On the 1st day and the 5th day after operation, CK level in the OLIF group was lower than that in the TLIF group(P<0.05). On the 3rd day after operation, the VAS of low back pain and leg pain in the OLIF group were lower than those in the TLIF group (P<0.05). There were no significant differences in the ODI of postoperative 12 months, low back and leg pain VAS at 3, 6, 12 months between the two groups(P>0.05). In the OLIF group, 1 case of left lower extremity skin temperature increased after the operation, and the sympathetic chain was considered to be injured during the operation, and 2 cases of left thigh anterior numbness occurred, which was considered to be related to psoas major muscle stretch, resulting in a complication rate of 10% (3/30). In the TLIF group, one patient had limited ankle dorsiflexion, which was related to nerve root traction, two patients had cerebrospinal fluid leakage, and the dural sac was torn during the operation, and one patient had incision fat liquefaction, which was related to paraspinal muscle dissection injury, resulting in a complication rate of 13% (4/30). All patients achieved interbody fusion without cage collapse during the 6- month follow-up.
CONCLUSION
Both OLIF and TLIF are effective in the treatment of single-segment degenerative lumbar spinal stenosis. However, OLIF surgery has obviously advantages, including less intraoperative blood loss, less postoperative pain, and good recovery of intervertebral space height. From the changes in laboratory indexes of CK and the comparison of the left psoas major muscle, multifidus muscle, longissimus muscle area, and high signal intensity of T2 image on imaging, it can be seen that the degree of muscle damage and interference of OLIF surgery is lower than that of TLIF.
