2.Observation on COLIXA3 polymorphism of patients with fluorosis

Objective To study the COLIXA3 gene polymorphism of patients with fluorosis and to explore the pathogenesis of COLIXA3 gene in endemic fluorosis.Methods Fifty one cases of patients with drinking-water borne fluorosis were selected as the case group in Xinzhou city,Shanxi province and 28 cases of healthy people were as the control group.Dental fluorosis was detected by Dean method and skeletal fluorosis was examined by X-ray.COLIXA3 of exon 5 gene product of 103 points was amplified by PCR and the gene locus genotype was sequenced.Results Ten cases of mild dental fluorosis,14 cases of moderate dental fluorosis,15 cases of severe dental fluorosis were detected among the 51 patients.The control group was free of dental fluorosis.All the 51 cases of patients with fluorosis had varying degrees of skeletal fluorosis,mainly osteosclerosis lesions,accounting for 86.27%(44/51 ),and mild skeletal fluorosis patients were all osteosclerosis lesions,and osteosclerosis lesions and multiple skeletal lesions were found among moderate and severe skeletal fluorosis patients in the case group,while control group had no skeletal fluorosis.The differences between genotypes of frequency distribution of AA,Aa,aa of COLIXA3 of case and control groups were not statistically significant [96.08%(49/51 ),3.92%(2/51 ),0.00%(0/51) and 96.43%(27/28),3.57%(1/28),0.00%(0/28),x2 =0.94,P > 0.05].ConclusionsCOLIXA3 gene polymorphism is not significantly correlated to fluorosis.

4.Silencing Filamin A Inhibits the Invasion and Migration of Breast Cancer Cells by Up-regulating 14-3-3σ

Filamin A and 14-3-3-σ are closely associated with the development of breast cancer.However,the exact relationship between them is still unknown.The present study aimed to examine the interaction of filamin A with 14-3-3-σ in the invasion and migration of breast cancer.RNA interference technology was employed to silence filamin A in MDA-MB-231 cells.Real-time PCR and Western blotting were used to detect the expression of filamin A and 14-3-3-σ at mRNA and protein levels,respectively.Double immunofluorescence was applied to show their colocalization morphologically.Wound healing assay and Trans-well assay were used to testify the migration and invasion of MDA-MB-231 cells in filamin A-silenced cells.The results showed that silencing filamin A significantly increased the mRNA and protein levels of 14-3-3σ.In addition,double immunofluorescence displayed that filamin A and 14-3-3σ were predominantly colocalized in the cytoplasm of MDA-MB-231 cells.Silencing filamin A led to the enhanced fluorescence of 14-3-3σ.Furthermore,cell functional experiments showed that silencing filamin A inhibited the migration and invasion of MDA-MB-231 cells in vitro.In conclusion,silencing filamin A may inhibit the invasion and migration of breast cancer cells by upregulating 14-3-3σ.
5. Effect of Zingiberis Rhizoma Recens on Hemorrhoids Model in Rats and Mice

Objective: To explore the effect of Zingiberis Rhizoma Recens(ZRR) on hemorrhoids in mice and rats. Method: Sixty SPF-grade SD rats were divided into blank group, model group, Ma Yinglong Shexiang hemorrhoid ointment group (7.5 g·kg-1), and large and small-dose ZRR paste groups (10, 5 g·kg-1). ZRR paste was applied in Yongquan acupoint to observe the effect of 0.05 mL injection with 75% acetic acid solution on hemorrhoids induced by subcutaneous anus in rats, the levels of interleukin-1β (IL-1β), interleukin-6 (IL-6), tumor necrosis factor-α (TNF-α), nitric oxide (NO) in the serum were detected by enzyme linked immunosorbent assay(ELISA), and the rectal histopathology was detected by hematoxylin-eosin(HE). Sixty SPF-grade KM mice were divided into blank group, model group, MA Ying-long Shexiang hemorrhoid ointment group (7.5 g·kg-1), and large and small-dose ginger essential oil groups (0.06, 0.03 mL, three times a day). ZRR paste was applied in crissum to observe the effect of injection of 20% acetic acid solution 0.05 mL (maintaining for 1 min) on hemorrhoids in mice induced by anus. The degree of local swelling ulcer around the anus and score was observed, levels of IL-1β, TNF-α in the serum were determined by ELISA, and the rectal histopathology was detected by HE staining. Result: In the experimental study on treating hemorrhoids with ZRR paste applied on Yongquan point of rats, compared with normal group, serum levels of IL-1β, IL-6, TNF-α, NO in model group were significantly higher (P<0.01). The pathological injury of rectum was serious. Compared with model group, serum levels of IL-1β, IL-6, TNF-α, NO were decreased in each administration group (P<0.05, P<0.01), and the degree of rectal tissue injury was relieved (P<0.01). In the experiment of external application of ZRR essential oil around the anus for treating hemorrhoids, compared with normal group, perianal swelling and severe ulcer injury were observed in the model group (P<0.01). The serum levels of IL-1β, TNF-α in model group were significantly higher (P<0.01), and the pathological injury of rectum was serious. Compared with model group, each administration group showed the alleviation in perianal pathological conditions (P<0.01), the reduction in serum IL-1β, TNF-α levels (P<0.01), and the relief in the extent of rectal tissue injury (P<0.01). Conclusion: External application of ZRR can effectively inhibit perianal swelling and ulcer degree, with a good therapeutic effect on hemorrhoids model in rats and mice.
6.Pressure ulcer prevention in acute care using the pressure ulcer bundle of care.

A study was conducted to determine the effectiveness of the pressure ulcer bundle of care (PUB) in preventing pressure ulcers among patients in acute care. The pre and post-test quasi-experimental design was utilized to predict a model of preventing pressure ulcer in acute care setting. Thirty acute care patients with moderate risk for pressure ulcers were randomly selected to receive the following five PUB interventions: assessment of pressure ulcer risk, repositioning, head elevation, heel elevation, and frequent diet monitoring. Pressure ulcer risk was assessed using the Braden risk assessment scale before and after PUB interventions. This scale assesses important aspects of ulcer formation according to six subscales: sensory perception, moisture, mobility, physical activity, nutrition, and friction/shear. Profile of the patients according to age, sex, and length of hospital stay was described using frequency and percentage distribution. Bundle compliance, as measured by performance of the five interventions was described using mean scores and standard deviations. The t-test was used to determine the differences in pressure ulcer risk or occurrence between pre- and post-intervention phases. Multiple linear regression analysis was used to determine the relationship of Pressure Ulcer Risk Assessment Scores (PURAS) to the PUB, and to identify the predictor(s) of PURAS among the four interventions in the PUB. Statistical significance was considered at the .05 level. Pressure ulcer risk scores of patients improved significantly from "mild risk" to "not a risk" post-PUB (p=<0.001). Head elevation, heel elevation, and diet monitoring were found to be predictors of pressure ulcer risk scores after PUB interventions. Repositioning was not significantly associated with pressure ulcer risk scores of patients after PUB interventions. The three predictor model revealed the PUB interventions were able to account for 52% of the variance in pressure ulcer risk scores, which indicates a strong significant relationship between patients receiving PUB and their improvement in pressure ulcer risk. In conclusion, the pressure ulcer bundle of care intervention is effective in prevention of pressure ulcers in patients at risk. Nurses should adopt the provision of bundle of care intervention(s) to enhance patient safety and quality of care.

7.Effects and safety of facemask use on healthy adults during exercise: a systematic review and meta-analysis protocol.

BACKGROUND:
Facemasks are used to minimize SARS-CoV-2 spread during the COVID-19 pandemic. However, facemask use during exercise is
associated with possible adverse effects.
OBJECTIVES:
To compare the effects of facemask use vs. non-facemask use on subjective responses, COVID-
19 incidence, and physiologic changes in healthy adults during exercise.
METHODS:
The systematic review (PROSPERO registration number:
CRD42022296247) will follow the PRISMA-P guidelines and use electronic databases Science Direct, PubMed, Google Scholar, Herdin, and
EbscoHost. This will cover randomized parallel groups or randomized crossover studies investigating tolerability, physiologic effects, and the
impact on SARS-COV2 incidence of commercially-available cloth, surgical, or FFR/N95 facemasks compared to no-facemask conditions during
exercise among healthy adults, including studies published from the earliest date to January 31, 2022. Outcomes of interest will be facemask
tolerability in 10 domains of comfort and objective cardiopulmonary, gas exchange, and metabolic responses. Mean differences (MD) or
standardized mean differences (SMD) with a 95% confidence interval (CI) will be calculated overall and for subgroups using RevMan software
(version 5.4.1). Pooled and subgroup estimates will be calculated using random-effects meta-analysis. The chi-squared test, I2 statistics, and visual
analysis will assess heterogeneity. The GRADEpro will determine the certainty of the level of evidence.
EXPECTED RESULTS
An evidence-based
recommendation using GRADE on the changes attributed to facemask use during exercise will be available. This will be useful for organizations
when developing appropriate guidelines for exercising while mitigating the risk of SARS-CoV-2 transmission. Future researchers may use this study
when redesigning comfortable facemasks without compromising filtration capability.
8.Relationship between NK Cell Reconstitution and aGVHD after Allo-HSCT.

OBJECTIVETo investigate the relationship between NK cell count/activity and acute graft-versus-host disease (aGVHD) in patients receiving allogeneic hematopoietic stem cell transplantation (allo-HSCT).
METHODSA total of 26 patients who had undergone allo-HSCT from January to July 2015 were enrolled in this study. The NK cell count/activity in the peripheral blood of recipients on day 30 after allo-HSCT were monitored by using 4-color flow cytometry. The incidence of aGVHD in patients was evaluated by clinical manifestation combinating with related pathologic indicators, and the relationship between NK cell count/activity and aGVHD were analyzed.
RESULTSIn the aGVHD group and the no-aGVHD group, the NK cell count and activity on days 30 after allo-HSCT were 655±216 cells/µl vs 1169±372 cells/µl(P=0.002) and 7.3±3.6% vs 9.0±3.6% (P=0.008). In the II-IV grade aGVHD group and the 0-I grade aGVHD group, the NK cell count/activity were 617±220 cells/µl vs 1081±399 cells/µl (P=0.001) and 4.2±1.7% vs 8.3±3.5%(P=0.001). As compared with the 0-I grade aGVHD group, patients in the II-IV grade aGVHD group had higher relapse rate (57% vs 5%)(P=0.010) , lower 1-year progression-free survival(PFS) rate (43% vs 84%)(P=0.010).
CONCLUSIONNK cell count/activity on day 30 after allo-HSCT were closely relates with aGVHD, which may be a potential marker for aGVHD and can provide a new target for aGVHD therapy.
