1.Influence of collagen coating on the biocompatibility of three-dimensional printed implants

BACKGROUND: Inert biomaterials such as metal usually hold poor biocompatibility and weak bonding force, which is against the effect of dental or bone implants. Therefore, how to improve their biocompatibility has become the research hotspot.OBJECTIVE: To prepare collagen-coated titanium alloy (Ti6Al4V), and to assess its biocompatibility.METHODS: Ti6Al4V served as the matrix, the 3-amino propyl triethoxy silane (KH550) as the crosslinking agent, and the three-dimensional printed titanium alloy coated by collagen type Ⅰ and Ⅱ was prepared,respectively. The coated materials were co-cultured with mouse preosteoblasts MC-3T3-E1 to evaluate its biocompatibility. The effect of different kinds of collagen on the cell differentiation was compared by differential recognition of surface proteins.RESULTS AND CONCLUSION: When the N content on the titanium alloy surface (Φ10 cm) was 8.41%, the cladding quantity of collagen type Ⅰ and Ⅱ was 0.81 and 0.77 mg, respectively. Compared with the bare titanium alloy, the cell adhered well and distributed extensively on the coated titanium alloy,which showed strong viability and fast proliferation.The cells cultured on collagen type Ⅰ coated materials expressed the proteins associated with matrix synthesis, and those on collagen type Ⅱ coated materials expressed the proteins associated with mineralization. These results clarify that the collagen coating can improve the biocompatibility of titanium alloy, and different types of collagens act on different functional proteins.
2.Observation on the efficacy of traditional Qigong exercise combined with Tuina manipulations in treating lower cervical disc herniation

Objective:To observe the efficacy of traditional Qigong exercise combined with Tuina(Chinese therapeutic massage)manipulations in treating lower cervical disc herniation(CDH).Methods:A total of 120 CDH patients who met the inclusion criteria were randomly divided into an observation group and a control group,with 60 cases in each group.Patients in both groups were given the same Tuina manipulations.In addition,patients in the observation group practiced traditional Qigong exercise,and those in the control group did conventional neck exercise.After 8 weeks of treatment,the efficacy of the two groups was observed.Follow-up was conducted 3 months after the end of the treatment to record any recurrence.Results:The total effective rate of the observation group was higher than that of the control group(P<0.05).The improvement in symptom scores was more significant in the observation group than in the control group(P<0.05).There was inter-group statistical significance in comparing the cross-sectional area and fatty infiltration of certain cervical muscles(P<0.05).In addition,the recurrence rate in the observation group was lower than that in the control group(P<0.05).Conclusion:On the basis of the same Tuina manipulation treatment,the efficacy of traditional Qigong exercise in treating CDH is superior to that of conventional neck exercise,with more significant improvements in pain and quality of life and a lower recurrence rate.
3.Compound Xianzhuliye combined with compound ipratropium bromide in the treatment of patients with acute exacerbation of chronic obstructive pulmonary disease

Objective:To investigate the clinical effect of compound Xianzhuliye combined with compound ipratropium bromide in the treatment of patients with acute exacerbation of chronic obstructive pulmonary disease. Methods:160 patients with acute exacerbation of chronic obstructive pulmonary disease who received treatment in Zhejiang Putuo Hospital from December 2019 to December 2020 were included in this study. They were randomly assigned to receive either routine treatment combined with compound ipratropium bromide (control group, n = 80) or routine treatment, compound ipratropium bromide and compound Xianzhuliye in combination (observation group, n = 80) for 14 successive days. Curative effects, blood gas analysis indexes (arterial partial pressure of oxygen, oxygenation index and arterial partial pressure of carbon dioxide), respiratory mechanics indexes (peak airway pressure, airway resistance and mean airway pressure), changes in lung function [percentage of predicted forced expiratory volume in 1 second forced expiratory volume in one second (FEV 1%), FEV 1/forced vital capacity], inflammatory factors (C-reactive protein, interleukin-6 and procalcitonin) before and after treatment were compared between the control and observation groups. Results:Total effective rate in the observation group was significantly higher than that in the control group [90.00% vs. 76.25%, χ2 = 5.391, P < 0.05]. After treatment, arterial partial pressure of oxygen and oxygenation index in the observation group were (62.17 ± 6.19) mmHg and (258.83 ± 25.45) mmHg, respectively, which were significantly higher than those in the control group [(53.86 ± 5.46) mmHg, (210.93 ± 23.12) mmHg, t = 9.005, 12.460, both P < 0.05]. After treatment, arterial partial pressure of carbon dioxide in the observation group was significantly lower than that in the control group [(46.89 ± 5.25) mmHg vs. (57.12 ± 4.12) mmHg, t = 13.711, P < 0.05]. After treatment, peak airway pressure in the observation group was significantly higher than that in the control group [(37.87 ± 3.25) cmH 2O vs. (30.82 ± 4.26) cmH 2O, t = 11.768, P < 0.05]. After treatment, airway resistance and mean airway pressure in the observation group were (6.75 ± 1.82) cm H 2O/L/s and (7.34 ± 1.30) cm H 2O, respectively, which were significantly lower than those in the control group [(9.82 ± 2.10) cm H 2O/L/s and (9.12 ± 1.23) cm H 2O, t = 9.881, 8.896, both P < 0.05). After treatment, FEV 1%, FEV 1/ forced vital capacity in the observation group were (67.31 ± 4.56)% and (61.23 ± 3.67)%, respectively, which were significantly higher than those in the control group [(58.98 ± 3.67)%, (52.18 ± 4.68)%, t = 12.729, 13.610, both P < 0.05). After treatment, C-reactive protein, interleukin-6 and procalcitonin in the observation group were (8.86 ± 1.53) mg/L, (65.87 ± 12.90) ng/L and (0.15 ± 0.04) μg/L, respectively, which were significantly lower than those in the control group [(19.90 ± 3.56) mg/L, (109.32 ± 19.76) ng/L and (0.34 ± 0.07) μg/L, t = 25.484, 16.469, 21.079, all P < 0.05). Conclusion:Compound Xianzhuliye combined with compound ipratropium bromide in the treatment of acute exacerbation of chronic obstructive pulmonary disease can acquire good clinical curative effects through improving blood gas analysis indexes, respiratory mechanics indexes and lung function, and reduce inflammatory reactions.
4.Survey on the depression status of the frontline healthcare professionals in the prevention and control of the risk of imported COVID-19 cases

ObjectiveTo study the prevalence of depression and the related influencing factors among the frontline healthcare professionals in the prevention and control of risk of imported COVID-19 cases, so as to provide references for improving their mental health status. MethodsBased on Wenjuanxing platform, a total of 984 frontline healthcare professionals participating in the screening and treatment of imported COVID-19 cases in Beijing Xiaotangshan hospital were assessed using Patients’ Health Questionnaire Depression Scale-9 item(PHQ-9). ResultsA total of 770 questionnaires were collected with a valid rate of 78.3%, among which 344 cases (44.7%) had depressive symptoms. Logistic regression analysis showed that great changes in work content (OR=2.206, P<0.01), sleep deprivation (OR=2.359, P<0.01), six hours or less of sleep (OR=2.032, P<0.05), focusing on epidemic information more than 1 hour per day (OR=1.095, P<0.05) were risk factors for depression, and regular exercise (OR=0.473, P<0.01) was a protective factor for depression relief. ConclusionThe frontline healthcare professionals in the prevention and control of risk of imported COVID-19 cases suffer a high risk of depression, while measures including scientific shift system, adequate sleep, less attention to the epidemic, and regular exercise may be effective in maintaining their physical and mental health.
5.Development of a fluorescence resonance energy transfer (FRET) immunoassay for the measurement of serum insulin.

Objective To investigate the fluorescence resonance energy transfer (FRET) effect between dylight (DL) and AuNP (AuNP), and to construct a new fluorescence immunoassay for insulin in combination with the immunocompetition method. Methods Insulin antigen (Ag) and insulin antibody (Ab) were conjugated with DL and AuNP respectively to form DL-Ag conjugate and AUNp-AB conjugate. A novel fluorescence immunoassay for insulin was developed on the basis of FRET effect and the immune competition response between them. Then the performance of the method was evaluated and its application in actual samples was explored. Results The fluorescence immunoassay showed high sensitivity (0.015 ng/mL), short measurement time (4 min) and good specificity. It was successfully used in the measurement of serum insulin, and the recovery was between 96.9% and 121.1%. Conclusion FRET effect between AuNP and DL can be applied to develop a fluorescence immunoassay for the measurement of serum insulin.
