1.Interferon Effect of Retinoic Acid on Lung Damage of Neonatal Rats Suffered from Hyperoxia

Objective To observe influence of retinoic acid on the lung damage and expression of Connexin26(Cx26) of neonatal rats suffered from hyperoxia.Methods Twenty-four 3-day SD rats were randomly divided into three groups:retinoic acid+ hyperoxia,hyperoxia,air group;rats in the group of hyperoxia were suffered 7 days hyperoxia (the level of oxygen was above 950 mL/L),air group was instead of air.All rats were weighed,value of radical alveolar counts(RAC) and microstructure of the lungs were observed by HE staining under light and electron microscope respectively;and the expression of Cx26 in the lungs was determined by immunohistochemistry.Results Compared with air group,weight,value of RAC were both decreased obviously in hyperoxia,retinoic acid+hyperoxia groups[(13.53?1.27),(13.38?1.29),(17.37?0.89)g,F=30.19 P=0;11.15?1.33,12.49?1.47,13.94?0.98,F=9.59 P=0),while the expression of Cx26 increased [(13.68?1.28)%,(17.82?1.72)%,(19.69?1.77)%,F=29.36 P=0].Conclusion Retinoic acid has a protective effection from damaged lung of neonatal rats suffered from hyperoxia,and it may be related to the alteration of expression of Cx26 protein in the lungs.
2.Retrospective analysis on treating chronic renal failure by herbal decoction

Objective:To make clear the present situation of treating chronic renal failure(CRF) by TCM in our hospital. Methods: Adopting a retrospective survey of medical records in a row. Results: In total of 76 cases of patients,74 cases took herbal decoction. 19 prescriptions were used which focused on Shenqidihuang Decoction 37 cases (50.00%), Huanglianwendan Decoction 9 cases (12.16%) and Dangguishaoyao Powder 7 cases (9.46%). Herbs such as Radix Salviae Miltiorrhizae,Radix et Rhizoma Rhei,Radix Achyranthis Bidentatae and Rhizoma Coptidis were the main medicines. Herbal medicine accompanied with Shenqidihuang Decoction were Cortex Eucommiae, Radix Acanthopanacis Bidentatae(62.16%), Pericarpium Citri Reticulatae, Fructrs Amomi,Radix Aucklandiae, folium perillae, caulis perillae (59.46%) and Rhizoma Coptidis, Caulis Bambusae in Taeniam, fortune eupatorium herb (48.57%) respectively. With the main methods of tonifying qi and enriching yin, activating blood and detoxifing and regulating the middle energizer. The herbal decoction had a good effect on CRF patients in the near future. Conlusion: The prescriptions used in our hospital had the characteristics of giving consideration to deficiency and excess, takes both deficiency and excess, tonification and discharge, and paying attention to spleen. The prescriptions had some effect on CRF in the near future, but the long-term effect need further observation.
3.A novel zebrafish vascular injury model for asseessing drug efficancy of Yangxue Qingnao granules

OBJECTIVE To assess the efficancy of Yangxue Qingnao granules on simvastatin-induced vascular injury model in zebrafish. METHODS Since statins can inhibit vascular development in zebrafish,in this study,we developed a novel animal model using 1 to 3 day post-fertilization larval zebrafish by optimizing the doses and duration of simvastatin exposure.Five pro-angiogenic drugs with a variety of mechanisms were tested to validate the newly developed zebrafish model. Zebrafish was treated with different concentration of Yangxue Qingnao granules( 62.5,125 and 250 μg·mL-1)for 2 d then tested for the area of subintestinal vein vessels. RESULTS Vascular regeneration promoting effect of five pro-angiogenic drugs (calycosin, astragaloside, chlorogenic acid, ferulic acid and Panax Notoginseng Saponins)were 8-48%,24-51%,35-58%,28-75% and 37-69%,respectively.In 62.5,125 and 250 μg·mL-1Yangxue Qingnao granules group,vascular regeneration promoting effect were 21% (P>0.05), 84%(P<0.01) and 53%(P<0.01). CONCLUSION Our results demonstrate that the zebrafish vascular injury model validated in this study could be used for in vivo angiogenesis studies and drug screening and for assessing pro-angiogenic drugs with different mechanisms.Yangxue Qingnao granules could promote the vascular regeneration in zebrafish.

Acute myeloid leukemia (AML),the most common disease in acute leukemia,is a highly heterogeneous invasive hematological disease.The t(8;21)(q22;q22) translocation is the most common chromosomal translocation in AML,generating AML1-ETO fusion gene and encoding AML1-ETO fusion protein.This article summarizes the two-hit hypothesis in AML occurrence,the pathogenesis of t(8;21)AML,all features involved in t(8;21)AML,and the function of the components in AML1-ETO fusion protein,providing important basic information for the treatment and prognosis of t(8;21)AML.Meanwhile,this article also summarizes the progress of next generation sequencing technique in leukemia,providing a new technique for the accurate therapy of (8;21)AML.
5.Study on effect of aqueous extracts from aconite on "dose-time-toxicity" relationships in mice hearts.

OBJECTIVETo study the effect of single administration of aqueous extracts from aconite on "dose-toxicity" relationship and "time-toxicity" relationship of mice hearts, through changes in electrocardiogram (ECG) and serum biochemical indexes.
METHODMice were grouped according to different drug doses and time points, and orally administered with water extracts from aconite for once to observe the changes of mice ECG before and after the administration, calculate visceral indexes heart, liver and kidney, and detect levels of CK, LDH, BNP and CTn-I in serum.
RESULTAccording to the "time-toxicity" relationship study, at 5 min after oral administration with aqueous extracts from aconite in mice, the heart rate of mice began rising, reached peak at 60 min and then slowly reduced; QRS, R amplitude, T duration and amplitude and QT interval declined at 5 min, reduced to the bottom at 60 min and then gradually elevated. The levels of CK, LDH, BNP and CTn-I in serum elevated at 5 min and reached the peak at 60 min, with no significant change in ratios of organs to body at different time points. On the basis of the "dose-toxicity" relationship, with the increase in single dose of aqueous extracts from aconite, the heart rate of mice. QRS, T duration and amplitude and QT interval declined gradually, and levels of CK, LDH, BNP and CTn-I in serum slowly elevated, with a certain dose dependence and no significant change in ratios of organs to body in mice.
CONCLUSIONSingle oral administration of different doses of aqueous extracts from aconite could cause different degrees of heart injury at different time points, with a certain dose dependence. Its peak time of toxicity is at 60 min after the administration of aqueous extracts from aconite.

7.Construction of Pseudomonas sp. M18 pqsR Mutant and Its Regulation on Plt Biosynthesis

Pseudomonas sp. M18 is one of plant growth-promoting rhizobacteria capable of producing two kinds of anti-fungal agents: phenazine-1-carboxilic acid (PCA) and pyoluteorin (Plt). The pqsR gene, which encodes a LysR family member PqsR, was amplified from chromosomal genome of strain M18. Using the homologous recombination technique, a chromosomal pqsR inactivated mutant strain M18PRG was constructed in Pseudomonas sp. M18. To study the effect of pqsR gene on Plt biosynthesis, the dynamic curves of Plt production by strains M18 and M18PRG was measured in KMB media. As a result, Plt production of the pqsR mutant was three to four folds higher than that of its parent strain M18. The Plt production was restored to the wild-type level when strain M18PRG was complemented with pqsR gene in trans. The regulation of pqsR gene on Plt production was further confirmed by the pltA′-′lacZ translational fusion analysis. These results indicate that pqsR gene negatively controls the Plt biosynthesis. Additionally, by analyzing the growth curves of wild type strain M18 and pqsR mutant, wecan readily find that PqsR has a negative influence on cell growth. It was also shown that the production of red pigments in strain M18 required the expression of pqsR gene. In conclusion, the data presented in this study clearly demonstrate that PqsR acts as a global regulator involved in many physiological activities in Pseudomonas sp. M18.
8.Clinical results following microsurgical discectomy: comparison of microscope and loupes

ObjectiveTo Compare the clinical results between microscope and loupes which used in microsurgical discectomy.MethodsA prospective randomized controlled trial of 93 patients who had undergone microsurgical discectomy from January 2007 to December 2010 was performed.Clinical results were assessed by comparing the following parameters between patients who had undergone the surgery by microscope and loupes:length of stay,hospitalization cost,operative time,estimated blood loss,Japanese Orthopaedic Association (JOA) score and JOA recovery rate,Odom's standard.ResultsForty-nine patients underwent surgery by microscope,and forty-four patients underwent surgery by loupes.Eighty patients received outpatient or telephone follow-up.The follow-up period was 6.17 to 52.90 months with an average of (29.64±13.05) months,and the follow-up rate was 86.02%.According preoperative data,the two groups didn't differ with respect to age,gender,level of radiculopathy,or preoperative JOA score and JOA recovery rate.No statistically significant differences were identified in postoperative JOA score and JOA recovery rate,length of stay,hospitalization cost,length of follow-up,or relapse rate.Statistically significant differences were identified in operative time,estimated blood loss,and follow-up JOA score and JOA recovery rate.Conclusion Microscope can provide relatively more clear and comfortable vision for the surgery.It can short the operative time,decrease blood loss,reduce the potential risk of nerve injury,and retain more normal tissue,which can ensure better clinical results.
9.Effect of total lumbar disc replacement on the treatment of discogenic low lumbar pain: preliminary outcomes.

BACKGROUNDLumbar pedicle screw fixation and fusion are major procedures for treating discogenic low back pain (DLBP). However, due to its advantages of preserving the segmental motion and biomechanical simulation, artificial total lumbar disc replacement (TDR) is increasingly popular.
METHODSFrom 2007 to 2010, 68 DLBP patients were enrolled. TDR were performed on 34 patients and the other 34 controls underwent the traditional fixation procedure. Qualitative and quantitative evaluations were followed including the changes in range of motion (ROM) and interpedicle height (IPH) at the posterior intervertebral body of operated level, in 6 and 12 months, and 3 years, postoperatively.
RESULTSQualitative results showed satisfying improvement in the two groups after 6 and 12 months, respectively, and the inter-group differences were significant (P > 0.05). The results of ROM and IPH have shown significant differences between the TDR and spinal fusion groups (P < 0.05).
CONCLUSIONWith similar clinical improvements as the traditional spinal fusion method, TDR offered significantly better ROM and intervertebral foramen height.

10.Expression of erythropoietin and its receptor in the brain of newborn rats suffering from fetal distress.

OBJECTIVETo study the expression of erythropoietin (EPO) and its receptor (EPOR) in the brain of newborn rats suffering fetal distress.
METHODSA model of fetal distress was prepared by ligating bilateral uterine arteries of the rats with full-term pregnancy for 10 minutes before cesarean sections. The expression levels of EPO and EPOR in the brain of newborn rats were detected by reverse transcription polymerase chain reaction (RT-PCR) and Western blot at 0, 2, 6, 12, 24, 48, 72 hrs and 7 days after birth. Serum EPO levels were measured using ELISA simultaneously. The newborn rats born by cesarean sections which were not subjected to uterine artery ligation were used as the control group.
RESULTSThe expression of EPO protein and mRNA in brain tissues in the fetal distress group increased significantly compared with the control group 2, 6 and 12 hrs after birth (P<0.05). The expression of EPOR protein and mRNA in brain tissues in the fetal distress group increased significantly compared with the control group 2, 6, 12, 24 and 48 hrs, and 3 days after birth (P<0.05). Serum EPO levels in the fetal distress group were significantly higher than in the control group 2 hrs after birth.
CONCLUSIONSThe EPO and EPOR levels in the brain increase quickly after birth in newborn rats suffering from fetal distress. The EPOR is high expressed for a longer time than EPO. This can provide a basis for the treatment of neonatal brain damage induced by fetal distress by exogenous EPO.
