2.Effects of Different Mongolian Acupuncture Methods on Neurons Form in Frontal Lobe and NO-cGMP Signaling Pathways in Rats with Chronic Stress Depression

Objective To observe the effects of different Mongolian acupuncture methods on neurons form in frontal lobe and NO-cGMP signaling pathways in rats with chronic stress depression; To explore biological mechanism of different Mongolian acupuncture methods for depression.Methods The male SD rats were randomly divided into control group, model group, fluoxetine group, Mongolian silver acupuncture group, Mongolian acupuncture three modulation methods group, and the combination of acupuncture with fluoxetine group (8 rats in each group). Except for control group, other rats were kept alone. Rats receive different treatment 1 hour before stimulations. Behavior changes were observed after 28 days. Hippocampus and frontal lobe tissues were collected. Frontal lobe neurons form changes were observed through different dyeing methods. The content of NO-cGMP was detected by radioimmunoassay and nitrate reductase method.Results Results of Nissl's staining showed that the neuronal pyramidal cells in the frontal lobe of model group rats arranged disorderedly, morphology was not intact and the number was obviously reduced. Nissl's staining got shallow and the most were blurry. The form of the frontal lobe neurons in Mongolian acupuncture three modulation methods group and Mongolian acupuncture group were good, Nissl's staining was dark blue. HE staining results showed that cytomembrane of frontal lobe neurons in model group rats ruptured, and structure was not clear. some cells fell off and formed into cavities. In Mongolian acupuncture three modulation methods group and Mongolian silver acupuncture group, the frontal lobe cells arranged regularly, cellular level was rich, and the nerve cell membrane was complete. The content of NO-cGMP in frontal lobe and hippocampus tissues was significantly elevated in model group. Mongolian acupuncture, Mongolian acupuncture three modulation methods, fluoxetine, and the combination of acupuncture with fluoxetine treatments all could reverse the changes.Conclusion Different Mongolian acupuncture methods may treat depression through regulating and controlling the expression of NO-cGMP.
3.A cross sectional survey on the prevalence of food intolerance and its determinants in Beijing,China

Objective To study the prevalence of food intolerance among attendants in a general hospital and to learn its related determinants in Beijing,China.Methods An cross sectional survey,from August 1st,2008 to June 30th,2009,was carried out,including 12 766 adults from Health Sciences Center,Chinese PLA General Hospital.Data for epidemiological analysis were entered (double entry) into computer and organized by EpiData 3.0.Non-conditional logistic regression model was used for odd ratio (OR) and 95%CI calculation,with statistic analysis through SPSS 13.0.Results The three leading intolerance food items were egg,crab and milk,and the prevalence rates were 28.5%,24.5% and 24.5% respectively.Prevalence of food intolerance increased along with aging.Levels and the kinds of food intolerance were associated with age,sex and appeared a linear correlation tendency (P<0.05).Prevalence of food intolerance in females (67.3%) was higher than that in males (56.2%).Data from multiple regression analysis showed that,comparing with age group under 40 and 40 years,the OR(95%CI) of age group 41-50,51-60,and above 60 were 1.125(1.027-1.233),1.307(l.176-1.452)and 1.536(1.275-1.849)respectively.Compared to males,the OR (95% CI) of females was 1.602 (1.475-1.741).When compared with normal weights,the OR (95%CI) of people with low weight was 1.772 (1.207-2.602).Conclusion Food intolerance was associated with age,sex and body mass index.Health education should be carried out according to the related characteristics of age and sex.Suggesting that reducing the risk of food intolerance,keeping the suitable weight was necessary.
4.Effect of icariine combined with embryonic stem cells on apoptosis of kidney epithelial cells in contrast-induced nephropathy rat models

Objective:To investigate the effect of icariin combined with embryonic stem cells on the renal epithelial cells in the contrast-induced nephropathy (CIN) rats and its possible mechanism,and to provide experimental evidence for clinical application of icariin.Methods:Fifty SD rats were randomly divided into,control group,model group,icariin group,stem cell group and combination group (n =10).The rat models were established by tail vein injection of INDO,L-NAME and diatrizoate.The serum creatinine (Scr) levels of the rats were detected before and after modeling.The expressions of p38MAPK and caspase-3 mRNA and protein were detected by semi-quantitative RT-PCR and Western blotting method.The apoptotic rate of renal epithelial cells was detected by flow cytometry.Results:The levels of Scr of rats were significantly increased 48 h after combination contrast agent injection,suggesting that the CIN models were successfully set up.Compared with model group,the Scr level of the rats in group was significantly decreased (P<0.05).compared with model group the expression levels of p38MAPK and caspase-3 mRNA and protein in icariin group and stem cell group and in combination group were decreased (P<0.05).The apoptotic rate of renal epithelial cells in combination group was significantly lower than those in icariine and stem cell groups (P<0.05).Conclusion:Icariine combined with embryonic stem cells has a protectice effect on the renal epithelial cells in the CIN rats,and its mechanism may be related to the inhibition of the related factors in the apoptosis pathways.
5.Effect of chemical microenvironment after traumatic brain injury on temperature-sensitive umbilical cord mesenchymal stem cells.

OBJECTIVETo simulate the chemical microenvironment of injured brain tissue, and to explore the effect of this chemical microenvironment on temperature sensitive umbilical cord mesenchymal stem cells (tsUC).
METHODSRat models of traumatic brain injury (TBI) were made by fluid percussion injury, and then the brain tissue extracts of the injured regions were acquired. Human umbilical cord mesenchymal stem cells (UC) were isolated and cultured, and the tsUC were obtained through the infection of temperature-sensitive Simian 40 Large T- antigen (ts-SV40LT) retrovirus. After that, both the two kinds of cells were cultured on the polyacrylamide gels which mimicking the elastic modulus of brain. Four groups were included: UC cultured under normal temperature (UC group), UC cultured added brain tissue extract under normal temperature (UC plus extract group), tsUC cultured under mild hypothermia (tsUC group), and tsUC added brain tissue extract under mild hypothermia for 3 days, then normal temperature for 4 days (tsUC plus extract group). After 24 hours, the apoptosis level was checked. Cell growth and morphological changes in each group were given dynamic observation. Seven days later, cell immunofluorescences were implemented for examining neural differentiation level.
RESULTSCompared with UC plus extract group, the apoptosis and proliferation in UC plus extract group were significantly reduced (P < 0.01) and increased (P < 0.01) respectively. Cell immunofluorescence showed that the both GFAP and Neuron positive cells were significantly enhanced in UC plus extract group than those in tsUC plus extract group.
CONCLUSIONtsUC combining with mild hypothermia could significantly reverse injury induced cell apoptosis, improve cell proliferation and neural differentiation under chemical microenvironment after brain injury, which confirmed the adaptation and resistance of tsUC under mild hypothermia after TBI.

6.Design of a universal robot system for health monitoring and medical services.

An autonomous robot system that is able to provide real time health monitoring and simple medical services is realized in this design. The system applies multiple sensors to monitor ordinary healthcare parameters, which is then transmitted via ZigBee network. In case of emergency, real time medicine delivery service is provided by the robot, applying a novel Received Signal Strength (RSS) based location method in ZigBee network. The system combines healthcare monitoring, data transmission and real time location into several portable wireless nodes, with the advantage of cost effectiveness, low power and easiness for configuration. Field tests under indoor environment have proved the stability and practicability of this design.

7.Simultaneous determination of melamine, ammelide, ammeline, and cyanuric acid in milk and milk products by gas chromatography-tandem mass spectrometry.

OBJECTIVETo develop an analytical method for simultaneously qualitative and quantitative determination of melamine and triazine-related by-products including ammelide, ammeline, and cyanuric acid in milk and milk products by gas chromatography-tandem mass spectrometry (GC-MS/MS).
METHODSMelamine and triazine-related by-products namely ammelide, ammeline and cyanuric acid in the samples were extracted in a solvent mixture of diethylamine, water, and acetonitrile (10:40:50, V/V/V). After centrifugation, an aliquot of the supernatant was evaporated to dryness under a gentle stream of nitrogen gas, and then melamine and triazine-related by-products were derivatized using BSTFA with 1% TMCS. The derivatives of melamine and its analogues were determined by gas chromatography/tandem mass spectrometry using multiple reactional monitoring (MRM) with 2, 6-Diamino-4-chloropyrimidine (DACP) being used as an internal standard.
RESULTSThe linear detectable ranges were from 0.004 mg/kg to 1.6 mg/kg for melamine, ammelide, ammeline, and cyanuric acid with a correlation coefficient no less than 0.999. The recovery rates of the four compounds in spiked blank milk powder at concentrations 0.5, 1, 2 mg/kg were between 61.4%-117.2%, and the relative standard deviation was no more than 11.5% (n=6). The detection limits of melamine, ammelide, ammeline and cyanuric acid in milk powder were 0.002 mg/kg with a ratio of signal to noise of 3.
CONCLUSIONThis GC-MS/MS method for simultaneous determination of melamine, ammelide, ammeline, and cyanuric acid in milk and milk products is sensitive and specific.

8.Application of network biology on study of traditional Chinese medicine.

With the completion of the human genome project, people have gradually recognized that the functions of the biological system are fulfilled through network-type interaction between genes, proteins and small molecules, while complex diseases are caused by the imbalance of biological processes due to a number of gene expression disorders. These have contributed to the rise of the concept of the "multi-target" drug discovery. Treatment and diagnosis of traditional Chinese medicine are based on holism and syndrome differentiation. At the molecular level, traditional Chinese medicine is characterized by multi-component and multi-target prescriptions, which is expected to provide a reference for the development of multi-target drugs. This paper reviews the application of network biology in traditional Chinese medicine in six aspects, in expectation to provide a reference to the modernized study of traditional Chinese medicine.

9.Protein array technology applied in high throughput monoclonal antibody generation.

UNLABELLEDTo reduce the huge labor-cost in the screening in traditional monoclonal antibody generation, We established a new system for monoclonal antibody generation integrating with protein array. BALB/c mice were immunized by eight recombinant proteins respectively, and the positive hybridoma cells were obtained by cell fusion and ELISA screening. All the eight kinds of positive hybridoma cells were mixed, cloned, screened by protein array, and definite dilution cloned.
RESULTS175 single cell clones were obtained by complex cloning, and 119 of those were positive clones. Then 8 positive cell lines were generated by the following 2 rounds definite dilution cloning. By comparing with the traditional method, we got 8 monoclonal antibodies using the combined protein array screening and multiplex cloning method in 1 cycle, and fewer amounts of antigens were used. As a result, the combined protein array and multiplex cloning method could be used as an economical, rapid and simple tool applying in high throughput monoclonal antibody generation.

10.Research on optimization of preventive maintenance path for electric medical equipment based on multi-criteria decision analysis

Objective:To optimize the preventive maintenance path of electric medical equipment by adopting multi-criteria decision analysis(MCDA),and to verify its optimization effect.Methods:The preventive maintenance paths for electric medical equipment were determined through literature research and management group discussions.An expert meeting was organized to discuss,determine,and demonstrate the preventive maintenance path evaluation criteria,and the weight of the criteria was set.Each preventive maintenance path was scored by experts based on criteria,the score of each maintenance path was calculated using the mean and sorted to form the optimal path.767 electric medical equipment of 7 categories in clinical use in the 960th Hospital of the PLA Joint Logistics Support Force from 2021 to 2022 were selected,conventional preventive maintenance management(referred to as conventional management mode)and preventive maintenance path management optimized by MCDA method(referred to as MCDA management mode)were adopted respectively according to different management modes.The changes in indicators such as failure rate,maintenance time,quality inspection pass rate and maintenance cost of electric medical equipment were compared between the two management models.Results:The failure occurrence rate of electric medical equipment of the MCDA management mode was 8.71%(67/767),which was lower than that of the conventional management mode,the difference was statistically significant(x2=3.960,P<0.05).The equipment maintenance time of the MCDA management mode was(2.24±1.17)days,which was lower than that of the conventional management mode,the difference was statistically significant(t=2.360,P<0.05).The quality inspection qualification rate of the MCDA management model was(96.57±2.74)%,which was higher than that of the conventional management mode,the difference was statistically significant(t=4.342,P<0.05).The average maintenance cost of equipment of the MCDA management model accounted for 2.37%of its assets,which was lower than that of the conventional management mode,the difference was statistically significant(x2=4.261,P<0.05).Conclusion:The MCDA method can provide a quantitative structural model for the optimization of preventive maintenance paths for electric medical equipment,and the optimized preventive maintenance paths can achieve efficient management of electric medical equipment,and focusing on the training of maintenance personnel's technical level can increase the self-repair rate and reduce the failure rate of medical equipment.
