1.Comparision of STIC-Multiplanar mode and two-dimensional ultrasound in the routine scanning of fetal heart

Objective To explore the clinical application of spatio -temporal image correlation combined with multiplanar mode (STIC-MP) in the routine scanning of normal fetal heart ,and to provide evidence for the application of STIC in the routine scanning of fetal heart in second trimester .Methods One hundred pregnant women at 20-27 weeks′gestation were enrolled.The routine five views of normal fetal heart were examined and recorded by two-dimensional ultrasound,and the volume datasets were acquired with STIC.The time of picture storing and the fetal cardiac structures were recorded respectively .The quality of pictures obtained by both methods were compared by the scoring system .The imaging method of STIC-MP was explored and analyzed.Results Satisfactory gray-scale volume acquisitions were accomplished in all 100 cases.The quality of the four views(four chamber view,left outflow tract,right outflow tract,three vessel-trachea views) derived from volume datasets were comparable to that directly obtained from two-dimensional scanning.No significant difference of visualization rate ( χ2 =3.030,0.337,2.010,0.000,all P>0.05)was found between the STIC-MP(100/100,98/100,98/100,95/99 as 100.0%,98.0%,98.0%,96.0%)and two-dimensional(97/100,99/100,100/100,95/98 as 97.0%,99.0%,100.0%,96.9%).However,the difference of visualization rate was found in the stomach transverse view ( χ2 =9.776,P=0.001).The mean time for the examination and picture saving of fetal cardiac structures by STIC ,mean acquisition time was (1.17 ±0.52) min,which was much shorter than that by two-dimensional scanning (4.20 ±1.35) min (t=0.38,P<0.05).The mean time for obtaining the routine five views of fetal heart and analyzing by STIC and MP were (6.75 ±1.51) min,(1.35 ±0.30)min respectively,which was longer than (4.20 ±1.35)min and (0.83 ±0.22)min by two-dimensional ultrasound(t=11.75,11.75,both P=0.00).Conclusions STIC-MP simplifies the picture-gathering process,decreases the ultrasound exposure upon fetal heart obviously .No significant difference was found between the quality of the five views obtained from STIC-MP and two-dimensional ultrasound.The views of STIC-MP can satisfied the routine scanning of fetal heart ,although the interpretation time of STIC-MP was longer compared with two-dimensional ultrasound.There will be vital significance on the use of STIC in clinical application .
2.Thyroid cancer stem cells and thyroid cancer treatment:theory and applications

BACKGROUND:Thyroid cancer stem cells are one of the reasons for tumor resistance that promotes tumor development. The research of thyroid cancer stem cells has provides a new clinic means for the diagnosis and treatment of thyroid cancer. OBJECTIVE:To overview the discovery, identification, and correlation of thyroid cancer stem cells with thyroid cancer. METHODS:A computer-based online search of PubMed database and Wan Fang database between 1995-01/2014-01 was performed to search related articles with the key words of“thyroid cancer, cancer stem cell, stem cell, cancer suppressor gene”in English and Chinese, respectively. Literatures related to thyroid cancer stem cells were selected;in the same field, the articles published lately in authoritative journals were preferred. RESULTS AND CONCLUSION:A total of 561 literatures were primarily selected, and 57 documents were involved for result analysis according to inclusion criteria. Cancer stem cells have become a focus in the study of carcinogenesis. Stem cells also exist in the thyroid gland and its tumor. At present, there are several ways to isolate and identify thyroid cancer stem cells. The tumor thyroid stem cells are closely related to the occurrence, transfer and treatment of tumors. But whether we can cure thyroid cancer through restraining or eliminating thyroid cancer stem cells is stil unknown that needs further studies.
3.Effect of rotenone on dopamine transporters in the substantia nigra of rats

Objective To investigate the toxic effects of rotenone on dopamine transporter in the substantia nigra of the rat. Methods Rats (male) were divided into normal control group, vehicle control group and rotenone experimental group(1.0mg/kg and 1.5mg/kg). The animals were subcutaneously administrated with the vehicle or rotenone in different dosage once a day for 28 days. One day after last injection, the brain was dissected and the substantia nigra was harvested. Protein expressions of tyrosine hydroxylase and dopamine transporter were determined with the methods of immunohistochemistry and Western blot. The mRNA expressions of dopamine transporter and dopamine receptor were detected with RT-PCR respectively. The activity of Na+-K+-ATPase was assayed biochemically. Results In the experimental groups, both the immune positive neurons of tyrosine hydroxylase and the expression of its protein were significantly decreased in the substantia nigra of the rats after rotenone treatment. The number of dopamine transporter antibody-immune positive neurons were significantly increased in the rats exposed to rotenone. Furthermore, protein and mRNA expressions of dopamine transporter in the experimental group were also obviously up-regulated compared with the normal control group. A decreased activity of Na+-K+-ATPase was observed in the substantia nigra of rats in the rotenone experimental group. In addition, the mRNA expression of D2R was also increased in contrast with that in the control group. Conclusions Rotenone is toxic to dopamine neurons in the substantia nigra of rats. Dysfunction of dopamine transporter may be involved in the mechanism of rotenone toxicity to dopamine neurons.

Objective To observe the expression of leptin(Lep) receptor (LepR) and aquaporin 2 (AQP2) in the kidneys of obesity-related hypertensive rats ( OHR ) and to explore the mechanism of Lep resistance and water metabolic disorders in them.Methods OHR( model group) were induced by high-fat diet.Normal Wistar rats were chosen as normal control and hypertensive rats(SHR) as positive control.The serum level of triglycerides(TG), total cholesterol(TC), Lep, vasopressin ( AVP ) , angiotensinⅡ( AngⅡ) and β2-microglobulin (β2-MG ) was measured by enzyme linked immunosorbent assays ( ELISA) and renal morphology was observed by HE staining.The density of LepR and AngⅡtype 1( AT1) in the kidney was observed by immunohistochemistry.mRNA And protein expression of LepR and AQP2 in the kidney was assayed by real-time polymerase chain reaction and Western blotting.Results Compared with normal rats,the TG, TC, Lep, AVP, AngⅡand β2-MG of the model group were significantly increased (P<0.05), and protein and mRNA expression of LepR and AQP2 in the kidney were up-regulated (P<0.05).Compared with SHR group, TG, TC and Lep in serum of the model group were significantly increased (P<0.05).The concentrations of AVP,β2-MG and Lep was linearly related(R2 =0.87,R2 =0.95).Conclusion Water metabolic disorder and Lep resistance may be involved in the kidney injury of OHR, which may be one of the important pathogeneses of obesity-related hypertension.
6.Autologous hematopoietic stem cell transplantation for systemic lupus erythematosus 10-year follow-up

Objective To investigate the long-term efficacy of autologous stem cell transplantation for systemic lupus erythematosus (SLE). Methods Long-term follow up of 48 SLE patients with autologous stem cell transplantation were studied. All patients were followed up for 10 years. Among the patients, 24 cases were treated with purified CD34+ cells transplantation and 24 cases were treated with non-CD34+ cell transplantation. Comparison between groups was performed by x2 test. Results Among 5 dead patients, 4 died of transplantation related complications including 3 cases treated with CD34+ transplantion. The survival rate of those patients with more than 10 years duration of lupus was 90%(43/48). Among 43 patients, 7 had disease flare, 6 were treated with non-CD34+ cell transplantation. Eight patients went to college, 26 returned to normal life and 4 of them had children. Conclusion The long-term effect of SLE treated with autologous hematopoietic stem cell transplantation and anti-thymocyte globulin (ATG) is good. The recurrence of CD34+ transplant patients is lower than those treated with non-CD34+ transplantation. The quality of life in SLE patients treated with transplantation is better than those treated with conventional therapy.
7.A study of in vivo two-photon imaging of cerebravascular CO2 reactivity in mice

Objective To examine cerebrovascular reactivity to CO2 inhalation in mice. Methods In vivo Two-Pho?ton imaging technique was used to record the reaction of cerebral cortical vessels including penetrating artery, surface vein and capillary in 5 male C57 mice after CO2 inhalation under a thinned-skull cranial window. Nitric oxide syntheses inhibitor L-NAME and Prostaglandin syntheses inhibitor Indomethacin were used to block different vasodilator pathways, respectively. Results Different mouse cortical vessels displayed different degrees of dilation to 1-minute 5%CO2 inhala?tion. The penetrating artery exhibited the most obvious dilation (45.01%±4.45%). L-NAME intervention significantly di?minished cerebravascular CO2 reactivity(P<0.05). Indomethacin significantly attenuated the dilation of artery but not capillary comparing with L-NAME intervention(P<0.05). Conclusions Different vessels react differently to CO2 inhala?tion in which postaglandins and NO signal pathways are involved.
8.Isolation and Transient Expression Assay of PEAMT Gene Promoter from Salicornia europaea

Phosphoethanolamine N-methyltransferase (PEAMT) is a key enzyme that catalyzes the synthesis of phosphocholine, which is an important precursor of phosphatidylcholine and glycine betaine. A 1249bp 5'-flanking region of phosphoethanolamine N-methyltransferase gene was isolated by anchored PCR, based on the cDNA sequence of PEAMT from halophyte Salicornia europaea. The transcription start site was identified as A and localized at 301bp upstream of the ATG according to the results of RLM-RACE. In SePEAMT promoter region, many potential cis-acting elements were predicted by PlantCARE and PLACE programs. Aside from the basal transcriptional elements TATA-box and CAAT-box, some stress-responsive motifs such as ABRE, HSE and LTR were found. In addition, some pollen-specific activation-related elements were also present in this region. Binary expression vector was constructed by fusing SePEAMT promoter with GUS gene and designated as pPro. The pPro was transferred into tobacco by Agrobacterium-medicated transformation and transient GUS expression analysis indicated that SePEAMT promoter could drive strong GUS expression.
9.A case control study on risk factors for female thyroid tumor in Hohhot of Inner Mongolia

Objective To study the major risk factors for female thyroid tumor in Hohhot of Inner Mongolia and to provide a scientific basis for prevention and treatment of the disease.Methods A case control study was carried out,190 female patients who had suffered thyroid tumor and had been operated on in the Affiliated Hospital of Inner Mongoha Medical College and Inner Mongolia Hospital from March 2010 to September 2011 were selected.Meanwhile,190 female patients who were excluded from being suffering this disease were chosen as controls.A formal questionnaire was made,including the following contents:the general situation,the life style and behavior habit,women's physiological and reproductive situation,job and life factors,personal disease,and medicine and genetic factors.Then,trained investigators who had at least got the bachelor degree interviewed all the patients in the two groups and the doctors in the office of ultrasound and pathology made the check-up.All data made up a database;statistical description and analysis was done with software SPSS 17.0.Through Logistic regression analysis,the relationship between all factors and female thyroid tumor was analyzed in order to find major risk factors related to female thyroid tumor in Hohhot.Results The results of Logistic regression analysis showed that the delay of the first pregnancy,the higher Body Mass Index,too much preserved food and seafood in the diet,higher level of education,negative live event were apparently related to female thyroid tumor (OR =1.527,22.729,2.271,4.303,1.117,1.489,all P < 0.05).Conclusions The female thyroid tumor in Hohhot is a result of comprehensive effect of multiple factors.This study found that the delay of the first pregnancy,the higher Body Mass Index,too much preserved food and seafood in the diet,higher level of education,and negative live event are apparently related to female thyroid tumor,and these factors may be major risk factors for thyroid tumor based on the current epidemiological study.
10.Perioperative psychological management of 11 male patients with breast cancer

Objective To summarize the experience of peri-operative emotion management of male patients with breast cancer. Methods The clinical data of 11 male patients with breast cancer were analyzed to summarize methods for the emotion management before, during and after chemo-radiotherapy. Result All the patients took a good attitude and complete the therapy after implementation of emotion management. Conclusion Peri-operative psychological management is of help for their alleviating the negative moods of helplessness, strengthening their confidence, stabilizing and improving the emotions, and finally making them cooperative in the treatment and nursing.
