1.Comparative analysis of inflammatory responses in lung tissue of K18-hACE2 transgenic mice infected with SARS-CoV-2 prototype and variant strains

Objective To compare the immune responses in lung tissue of K18-hACE2 transgenic mice infected with SARS-CoV-2 prototype strain Prototype and its variants Delta and Omicron BF.7, so as to provide experimental evidence for the research and development of related drugs. Methods K18-hACE2 transgenic mice were infected with Prototype, Delta or Omicron BF.7 by nasal drip. On the 7th day after infection, the lung tissues of mice were taken aseptically for subsequent analyses. Viral RNA copy numbers and cytokine gene expression levels in the lung tissues were quantified by qPCR. Histopathological examination of lung tissue damage was performed using HE staining. Results The copy number of viral nucleic acid in lung tissue of mice in Delta group was significantly higher than that in Prototype and Omicron BF.7 groups(t = 5. 419-6. 908, each P < 0. 01), the expression levels of interferon γ(IFN γ)and interleukin-13(IL-13)in lung tissue were significantly higher than those in Prototype and Omicron BF.7 groups(t = 3. 990-7. 282, each P < 0. 05), and the expression levels of tumor necrosis factor-α(TNF-α),and IL-5 were significantly higher than those in Prototype group(t = 7. 108 and 2. 908,each P < 0. 05). In addition, obvious pathological changes related to virus infection were observed in lung tissue sections of mice in three groups. Conclusion Delta variant strain induced the strongest inflammatory response in lung tissue of K18-hACE2 transgenic mice with the highest pathogenicity, followed by prototype strain and Omicron BF.7 variant strain.
2.Pharmacokinetic Study on the Triptolide in Normal Rats and Adjuvant Arthritis Model Rats in vivo

OBJECTIVE:To study the pharmacokinetics of triptolide in Tripterygium glycosides tablet in normal rats and adju-vant arthritis model rats in vivo,and provide reference for clinical rational drug use. METHODS:12 SD rats were randomly divid-ed into normal group and model group,6 in each group. Model group was subcutaneously injected complete Freund's adjuvant 0.1 mL to induce adjuvant arthritis model,normal group was subcutaneously injected the same volume of saline. After 14 d model-ing,2 groups were given Tripterygium glycosides tablet suspension 96 mg/kg intragastrically,the blood sample of eyes 0.4 mL were respectively taken before and 10,30,45,60,90,120,150,180,240,300,420 min after administration. The plasma con-centration of triptolide was determined by HPLC,the pharmacokinetic parameters were calculated by DAS 2.0 software,and the parameters were compared. RESULTS:The pharmacokinetic parameters of triptolide in normal group were cmax of(1.139±0.114)μg/mL,tmax of(2.167±0.606)h,t1/2α of(5.500±3.610)h,AUC0-7 h of(5.052±0.371)μg·h/mL,MRT0-7 h of(3.224±0.119)h, and CL of(11.616±2.986)mL/h;and those in model group were cmax of(0.916±0.103)μg/mL,tmax of(3.083±0.801)h,t1/2αof(5.593±1.795)h,AUC0-7 h of(4.707±0.347)μg·h/mL,MRT0-7 h of(3.429±0.139)h,and CL of(11.246±2.638) mL/h. Compared with normal group,cmax in model group was significantly decreased,tmax and MRT0-7 h were significantly prolonged(P<0.05). CONCLUSIONS:Adjuvant arthritis can affect the pharmacokinetics of triptolide in rats in vivo,and promote its absorption and removal.
3.Distribution of α-synuclein truncations in cells

Aim To study of the expression and distribution of four α-synuclein truncations in three cells.Method Four α-synuclein gene truncations were obtained by PCR method,followed by subcloning into the pEGFP-N1 eukaryotic expression vector.Four obtained recombination plasmids were transfected into MN9D cells,PC12 cells and SH-SY5Y cells using Lipofectamine 2000 respectively.The expression and distribution of four α-synuclein truncations were observed by Confocal.Results Distribution of four α-synuclein truncations was discrepant obviously,the truncations,with more C terminal remained,were prone to emerging in nuclei.Conclusion Localization of α-synuclein protein in cells may be related to the C terminal,and the whole C terminal plays an important role in distribution of α-synuclein into nuclei.
4.Predictive value of amino-terminal pro-B-type natriuretic peptide on short term prognosis in patients with acute coronary syndrome

Objective: To evaluate the predictive value of plasma amino-terminal pro-B-type natriuretic peptide (NT-proBNP) on short term prognosis in patients with acute coronary syndrome (ACS). Methods: Plasma NT-proBNP levels were measured in ACS patients within 24 h after admission with ELISA method. The 600 ACS patients were divided into two groups: event group (n=108)and no-event group (n=492)according to occurrence of cardiovascular events(including cardiac death, cardiac shock and heart failure), The NT-proBNP levels were compared between two groups. Single factor and multiple factor Logistic regression analyses were used to evaluate if NT-proBNP was an independent predictor for short term prognosis. Results: The plasma NT-proBNP levels of event group were significantly higher than those of no-event group [1361.2 (965.3, 2088.6) pmol/L vs.605.7 (274.5, 1177.8) pmol/L, p<0.01]. Multiple factor Logistic regression analyses demonstrated that plasma NT-proBNP was an independent predictor for short term prognosis (OR=1.000, p<0.01) in ACS patients. Conclusion: Plasma NT-proBNP was an independent predictor for short term prognosis in ACS patients.

7.Clinical effect of bone melon extract combined with loronoxicam in elderly patients after total hip arthroplasty

Objective To investigate the clinical effect of bone melon extract(BME)combined with lornoxicam(LN)in elderly patients after total hip arthroplasty.Methods Elderly patients with femoral neck fracture attending the Department of Orthopedics of Xixi Hospital in Hangzhou City from January 2020 to June 2023 were retrospectively selected as study subjects.According to different postoperative treatment programs,they were divided into the BME group(LN combined with BME treatment)and the LN group(LN treatment).The visual analog scores(VAS)and Oswestry dysfunction index questionnaire(ODI)scores of patients in the two groups preoperatively and postoperatively,the changes of inflammatory factors[C-reactive protein(CRP),interleukin-6(IL-6),and tumor necrosis factor-α(TNF-α)]before surgery and 4 weeks after surgery,and the clinical efficacy of patients in the two groups preoperatively and at 6 months postoperatively,the hip joint function(Harris score),daily living ability[Barthel index(BI)]and quality of life[Mos36-Item Short Form Health Survey(SF-36)]were compared.Results A total of 100 patients were included in the study,with 56 in the BME group and 44 in the LN group.Before treatment,the differences in VAS scores,ODI scores,inflammatory factors,Harris scores,BI and SF-36 between the two groups were not statistically significant(P>0.05).After treatment,VAS scores,ODI scores,CRP,IL-6 and TNF-αdecreased significantly in both groups compared with those before treatment(P<0.05);and these indicators in the BME group were lower than those in the LN group(P<0.05).In terms of therapeutic efficacy,the therapeutic efficiency of the BME group was significantly higher than that of the LN group(P<0.05).Harris scores,BI and SF-36 significantly increased in both groups(P<0.05),and these in the BME group were greater than those in the LN group(P<0.05).Conclusion Compared with LN treatment alone,BME combined with LN can promote rapid recovery in elderly patients after total hip arthroplasty.
8.Care experience of family caregivers for patients with amyotrophic lateral sclerosis: a Meta-synthesis of qualitative research

Objective:To systematically evaluate the care experience of family caregivers for amyotrophic lateral sclerosis (ALS) patients, providing guidance for home care of ALS patients.Methods:Qualitative studies on the care experience of family caregivers for ALS patients were retrieved from PubMed, American Psychological Association Psychnet, CINAHL, Web of Science, Medline, Cochrane Library, Embase, China Biology Medicine disc, WanFang Data, VIP, and China National Knowledge Infrastructure. The search period was from database establishment to October 30, 2022. The quality of literature was evaluated using the quality evaluation criteria for qualitative research of the Australia Joanna Briggs Institute Evidence-Based Health Care Center (2016 edition). The aggregative integration method was used to integrate the results using NVivo 11 software.Results:A total of 11 studies were included. A total of 22 complete and clear results were extracted and combined to form eight new categories, which were integrated into three results, namely, family caregivers facing personal development limitations and various family difficulties, experiencing complex and chaotic emotional experiences, and craving for diversified care support.Conclusions:Nursing staff should pay attention to the impact of ALS on their patient's family caregivers, provide caregiver knowledge and skill guidance, promote positive emotional experiences, and offer diversified care support to improve the quality of life of patients and family caregivers.
9.Effect of tetrandrine on bax, bcl-2 and TGF-β2 mRNA expressions in cultured human Tenon's capsule fibroblasts.

OBJECTIVETo investigate the effect of tetrandrine (Tet) on the expression of bax, bcl-2, and transforming growth factor-β2 (TGF-β2) mRNA in cultured human fibroblasts of Tenon's capsules (TCFS) and explore its possible mechanism.
METHODSThe third passage of TCFS cultured in vitro were exposed to 1×10(-5) mol/L Tet for 24 h, and real-time fluorescence quantitative PCR was used to detect the changes in the expressions of bax, bcl-2, and TGF-β2 mRNA.
RESULTSThe expression level of bax mRNA was obviously higher, while bcl-2 and TGF-β2 mRNA levels were significantly lower in Tet-treated TCFS than those in the control cells (P<0.05).
CONCLUSIONTet can inhibit the proliferation of TCFS possibly by reducing the expressions of bcl-2 and TGF-β2 mRNA, enhancing the expression of bax mRNA and inducing cell apoptosis, suggesting its potential in preventing fibrous scar formation after glaucoma filtration surgery.

10.Establishment and application of the training system for laboratory animal employees

Laboratory animals are an important part of life sciences and medical researches, as well an important support for the science and technology innovation in our country. Laboratory animal science is of great significance to the protection of human health,food safety and biological safety. Laboratory animals are indispensable in the development of food safety,drugs,vaccines and biological products and the studies of human disease pathogenesis. In order to adapt to the requirements for overall development of the laboratory animal industry in China, our institute has independently developed the Network Training System for Laboratory Animal Managers. This system is an online education and training platform which integrates the practical operation and theoretical knowledge of laboratory animals,including seven knowledge modules such as animal welfare,animal breeding,animal surgery and so on. The training subjects of the system include managers, experiment operators, laboratory animal doctors and breeders, aimed at accelerating the personnel training and team building of laboratory animal sciences,and promoting the transformation and development of personnel training in laboratory animal industry.
