1.Study of the Effect of Dimethylsulfoxid(DMSO)on Function of Platelet in Vitro

OBJECTIVE:To explore the effect of dimethylsulfoxid(DMSO)on the activity and function of platelet in vit?ro.METHODS:Fresh random-donor platelet unit(n=60)were divided into3equal groups.A DMSO-free group was chosen as a control group.The other two groups included a washed platelet group and a DMSO group to which DMSO was added with a final concentration of3%.All the groups were stored on a horizontal shaker at room temperature.Changes of adherence rate,aggregating function and recovery rate from hypotonic shack of platelet were observed.RESULTS:In the DMSO group,platelet adherence rate,aggregating function,and recovery rate from hypotonic shack decreased during storage comparing with control and washed platelet groups.CONCLUSION:DMSO has a protective effect on platelet and its action is reversible.

3.Original meaning of mai and its academic evolution.

Mai is the important concept in TCM and this term has been found in the early classic work of TCM. With the development and perfection of TCM theory, the original meaning of mai is "blood vessels entirely distrusted in the body". In five dynasties, the meaning of it was supplemented as "distribution of qi and blood in five zang and six fu organs to the four extremities", in which, not only blood but also qi has been included. With the constant understanding on mai, qi and blood, the unique concept of meridian and collateral is gradually generated. In terms of the modern study on the original meaning of mai and the academic evolution of meridian and collateral, it is discovered that qi is predominated and distributed in meridian and collateral as compared with blood. It is very significant to study the original meaning and academic evolution of mai, as well as the origin of the concept of meridian and collateral and to provide the reference support for the terminology standardization and basic study.

4.Clinical study of gabapentin in the treatment of refractory uremic pruritus in hemodialysis patients

Objective To assess the safety and efficacy of gabapentin in treatment of refractory uremic pruritus in maintenance hemodialysis (MHD)patients. Methods A randomized controlled trial was performed.Forty-nine MHD patients with severe pruritus were randomly divided into treatment group(gabapentin 100-300 mg at dialysis day before sleep)and the control group(loratadine 10 mg daily).The efficacy was assessed by visual analogue score,pruritus score and improved Duo's VAG Score after 12-week treatment,and the side effect was also observed to assess the safety. Results After treatment,the frequency,degree and area of pruritus in 25 patients of treatment group were significantly reduced,and VAS score(1.46±1.38vs 8.71±1.17,P<0.01),VAG score(2.92±1.63 vs 8.29±0.68,P<0.01)and the improved Duo's pruritus score(11.33±3.99 vs 30.75±4.87,P<0.01)decreased significantly compared with that of prior treatment.The symptoms of 24 patients in control group were improved partly,which were not as good as those in treatment group(P<0.01).The side effects of gabapentin were drowsiness and dizziness,but most symptoms were relieved or disappeared within 1 week and no patient interrupted therapy.No serious adverse events were observed. Conclusion Gabapentin is safe and effective for refractory uremic pruritus,but long-term efficacy and safety requires larger sample and long-term observation.
5.Protein C genetic variation was associated with the susceptibility to acute respiratory distress syndrome in Chinese Han population

Objective To investigate the genetic variants in the protein C (PC) and endothelial protein C receptor (EPCR) genes associated with the risk and outcome of acute respiratory distress syndrome (ARDS) patients in Chinese Han race.Methods Five tagSNPs (single nucleotide polymorphism,SNP) in the PC and EPCR genes were genotyped in patients with ARDS (n =275) and non-ARDS (n =337) in order to find the association between them in this case-control study.The SNPs were genotyped by SNPstream Beckman platform.Then,the correlation between the associated SNPs and plasma levels of activated protein C (APC) in patients with ARDS was investigated.The APC levels were measured using enzyme linked immunosorbent assay (ELISA) method.Results Association analysis rcvealed that two PC SNPs in perfect linkage disequilibrium,rs1799809 and rs1158867,were significantly associated with susceptibility to ARDS.T allele frequency of rs1799809 in ARDS patients was significantly higher than that in non-ARDS patients (OR =1.569,95% CI:1.192-2.066).And the genotype frequencies of rs1799809 were also significantly different between these two groups (P =0.007).The association remained significant after adjustment for multiple comparisons.Haplotype consisting of three SNPs in the PC gene was also associated with susceptibility to ARDS.The frequency of haplotype CCC in the ARDS samples was significantly lower than that in the non-ARDS group (P < 0.01).Moreover,ARDS patients canrying rs1799809 TT genotype showed lower serum levels of APC than patients with TC and CC genotypes (Padj =0.02).However,genotype and allele analyses of EPCR did not show any significant difference between ARDS and non-ARDS patients.Conclusions These findings indicated that common genetic variation in the PC gene was significantly associated with susceptibility to ARDS in Chinese Han race.The PC genetic variation influenced plasma concentration of APC in patients with ARDS.

Malignant neoplasm recurrence is the most important prognostic factor for patients.Looking for tar-get gene is an important part of cancer research.Annexin Ⅰ (ANXA1 )is to be discovered in the seventies century, composition by 13 calcium -phospholipid binding proteins annexin superfamily members of the first molecules. ANXA1 expressed in a variety of malignant tumors,ANXA1 expression levels were significantly reduced or absent in most tumor tissues.The study found that changed ANXA1 expression in tumor cells may have a causal relationship with the malignant phenotype of tumor cells.This paper aims to investigate the relationship between annexin Ⅰ and malignant neoplasm metastasis.
7.Construction of a multispecific internal control for competitive RT-PCR analysis of genes

Objective:To construct a mutispecific internal control for competitive RT-PCR analysis of Fas、FasL、GB、P、TIA-1 and ?-actin.Methods:Invitro synthesized fragments were amplified by PCR,there are two products,of which 145 and 147 bp.The 145 bp one contained 5′ primer sequences of Fas、FasL、GB、P、TIA-1 and ?-actin,another contained 3′ primer sequences of the same genes.The products with restriction sites were inserted into the vector PKF_3.Results:Restriction enzyme analysis and DNA sequencing were used to identify the recombinant plasmid,corresponding internal control was obtained by the amplification of the recombinant plasmid with each primer pair.The mutispecific internal control was then used for quantitative detection of TIA-1 in peripheral blood leukocytes from a patient with acutely rejecting allograft.Our study on Fas、FasL、GB、P、TIA-1 and ?-actin showed that the coamplified templates accumulated in a parallel manner throughout not only the exponential phase.Conclusion:The mutispecific internal control can be used for quantitative detection of the six genes. [
8.An Automated Dual-gradient Liquid Chromatography-MS/MS Method for Simultaneous Determination of Four Kinds of Plants Hormones in Different Parts of Soya Beans and Its Application to a Real Analysis

There are some common analysis challenges in the hormone detection in agriculture science, including matrix interference, complicated sample preparation, poor reproducibility, trace analyte content. An automated on-line SPE and innovative fast polarity switch analysis method employing dual-gradient liquid chromatography ( DGLC ) coupled with tandem mass spectrometry ( DGLC-MS/MS ) was established and validated for the simultaneous determination of gibberellic acid ( GA3 ) , indole acetic acid ( IAA ) , zeatin ( ZT) and abscisic acid ( ABA) in the soybean plant ( leaf, grain and pod) . The method was applied in the actual sample detection successfully. In order to acquire higher sensitivity, recovery, stability and precision, some conditions including SPE column, analytical column, mobile phase, additive etc were optimized according to the selection and retain of hormone. Beans were cryogenically grinded by liquid nitrogen, extracted by 80% methanol, certrifugatel and dilluted with water, and then injected directly. Samples were transported and gradient eluted on the analytical column Acclaim PA2 by 0 . 1% formic acid in water and methanol, after retaining and separation on the SPE column Hypersep Retain AX. All analytes were detected in selection reaction monitoring ( SRM) mode in both positive and negative channels. The quantification was based on linear regression. The linear ranges of GA3, IAA and ZT were 0. 1-50 μg/L with the LOQ of 0. 0002 μg/g, and the linear of ABA was 0. 5-50 μg/L with the LOQ of 0. 0010μg/g. The recoveries of four kinds of plants hormones were 76 . 1%-93 . 5%, and RSDs were 0 . 82%-6 . 02% at low ( 0 . 8 μg/L ) , medium (4. 0μg/L) and high (40μg/L). The results noted that the content of ABA in seeds was apparently higher than others. This method could be used for the rapid and accurate detection of hormone in different parts of soya beans.

Perioperative venous thromboembolism (VTE) of patients after thoracic cancer surgery has proved to be a serious or even fatal postoperative complication, which calls for great attention. China National Research Collaborative Group released the first edition of Chinese experts consensus on the perioperative VTE prophylaxis in 2018. This article is to interpret it on the aspect of epidemiological characteristics of perioperative venous thromboembolism and the current status of prophylaxis in China and other countries, in order to provide a better understanding of Chinese experts consensus for readers.
10.Optimal diagnosis and treatment for renal allograft fibrosis

Renal allograft fibrosis is one of the common and severe complications after kidney transplantation, which seriously affects the function and survival rate of renal allograft, and may even lead to organ failure and patient death. At present, the researches on renal allograft fibrosis are highly complicated, including immunity, ischemia-reperfusion injury, infection and drug toxicity, etc. The diagnosis and treatment of renal allograft fibrosis remain extremely challenging. In this article, the latest research progress was reviewed and the causes, novel diagnosis and treatment strategies for renal allograft fibrosis were investigated. By improving diagnostic accuracy and optimizing treatment regimen, it is expected to enhance clinical prognosis of kidney transplant recipients, aiming to provide reference for clinicians to deliver proper management for kidney transplant recipients.
