1.Expression of miR-19a in gastric cancer tissues and serum and its effect on proliferation and invasion ability of human gastric cancer AGS cells

Objective To investigate the expression of micro RNA-19a(miR-19a) in gastric cancer tissues and serum and its effect on the proliferation, invasion and regulation of thrombospondin 1(THBS1) expression in human gastric cancer AGS cells.Methods Real-time fluorescence quantitative PCR(RT-qPCR) was used to measure the expression of miR-19a in 22 gastric cancer tissues, corresponding adjacent noncancerous tissues, serum, and the serum of 22 healthy individuals. Then,the human gastric cancer AGS cells were cultured in vitro and divided into control, inhibitor NC, mimics NC, miR-19a inhibitor, miR-19a mimics, miR-19a inhibitor + si-NC and miR-19a inhibitor + si-THBS1 groups, three repeated wells for each group. The control group was without intervention, and the other groups were transfected with inhibitor NC, mimics NC, miR-19a inhibitor, miR-19a mimics, miR-19a inhibitor + si-NC and miR-19a inhibitor + si-THBS1 by using Lipofectamine~(TM)2000 liposomes respectively. After 24 h of transfection, each group was detected for the miR-19a expression, cell viability, proliferation rate, invasion number and the related proteins expression by RT-qPCR, cell counting kit-8(CCK-8), 5-ethynyl-2'-deoxyuridine(EdU), Transwell chamber and Western blot, separately.Results The expression level of miR-19a in gastric cancer tissues was significantly higher than that in corresponding adjacent tissues(t = 5. 061, P < 0. 001), and the expression level of miR-19a in serum of patients with gastric cancer was significantly higher than that of healthy people(t = 6. 299, P < 0.001). In vitro, the expression level of miR-19a in AGS cells in the miR-19a inhibitor group was significantly lower than that in the inhibitor NC group(t = 11. 120, P < 0. 001), the expression level of miR-19a in the miR-19a mimics group was significantly higher than that in the mimics NC group(t = 11. 637, P < 0. 001), and the mRNA and protein expression levels of THBS1 in the miR-19a inhibitor + si-THBS1 group were significantly lower than those in the miR-19a inhibitor + si-NC group(t = 10. 070 and 13. 164, P = 0. 001 and < 0. 001, respectively). Compared with the inhibitor NC group, the AGS cell viability, proliferation rate, invasion number, and the protein expression levels of Cyclin D1 and proliferating cell nuclear antigen(PCNA) in the miR-19a inhibitor group significantly decreased(t = 16. 679, 10. 072, 17. 737, 11. 173, and 11. 549,respectively, each P < 0. 001), and the THBS1 protein expression level increased significantly(t = 16. 774, P < 0. 001).Compared with the mimics NC group, the AGS cells in the miR-19a mimics group had significantly higher viability, proliferation rate, invasion number, and the expression levels of Cyclin D1 and PCNA proteins(t = 15. 004, 22. 746, 13. 349, 11. 484,and 19. 022, respectively, each P < 0. 001), while the expression level of THBS1 protein significantly decreased(t = 16. 384,P < 0. 001). Compared with miR-19a inhibitor + si-NC group, the viability, proliferation rate, invasion number, and the protein expression levels of Cyclin D1 and PCNA of AGS cells in miR-19a inhibitor + si-THBS1 group significantly increased(t = 13. 180, 8. 669, 14. 493, 6. 828, and 13. 587, respectively, each P < 0. 001), while THBS1 protein expression level significantly decreased(t = 14. 824, P < 0. 001).Conclusion The miR-19a is highly expressed in gastric cancer tissues and serum of patients with gastric cancer, and knockdown of miR-19a may inhibit the proliferation and invasion of human gastric cancer AGS cells by activating the expression of THBS1.

3.Distribution and risk factors of thyroid cancer in China

Incidence of thyroid cancer has continuously increased in the last three decades globally. A similar trend of thyroid cancer incidence appeared in China. New cases and deaths of thyroid cancer in China accounted for 15.6% and 13.8% of the global new cases and deaths in 2012. The incidence in East China was the highest while that in Middle China was the lowest. Thyroid cancer was more common in urban areas than in rural counterparts. The incidence was substantially higher in female patients than male. Thyroid cancer occurred most commonly in middle age. Radiation exposure, iodine suffciency or deifciency, sex hormone and genetic mutations were risk factors for thyroid cancer. To prevent it, potassium iodide should be taken to prevent radiation fallout in nuclear accidents, unnecessary exposure to radiation from medical imaging is reduced, iodine surveillance system is improved and appropriate standards for iodized salt are established.
4.Effect of extracts from Dendrobii ifficinalis flos on hyperthyroidism Yin deficiency mice.

Some unhealthy life habits, such as long-term smoking, heavy drinking, sexual overstrain and frequent stay-up could induce the Yin deficiency symptoms of zygomatic red and dysphoria. Stems of Dendrobii officinalis flos (DOF) showed the efficacy of nourishing Yin. In this study, the hyperthyroidism Yin deficiency model was set up to study the yin nourishing effect and action mechanism of DOF, in order to provide the pharmacological basis for developing DOF resources and decreasing resource wastes. ICR mice were divided into five groups: the normal control group, the model control group, the positive control group and DOF extract groups (6.4 g · kg(-1)). Except for the normal group, the other groups were administrated with thyroxine for 30 d to set up the hyperthyroidism yin deficiency model. At the same time, the other groups were administrated with the corresponding drugs for 30 d. After administration for 4 weeks, the signs (facial temperature, pain domain, heart rate and autonomic activity) in mice were measured, and the facial and ear micro-circulation blood flow were detected by laser Doppler technology. After the last administration, all mice were fasted for 12 hours, blood were collected from their orbits, and serum were separated to detect AST, ALT, TG and TP by the automatic biochemistry analyzer and test T3, T4 and TSH levels by ELISA. (1) Compared with the normal control group, the model control group showed significant increases in facial and ear micro-circulation blood flow, facial temperature and heart rate (P < 0.05, P < 0.01), serum AST, ALT (P < 0.01), T3 level (P < 0.05), TSH level (P < 0.05) and notable deceases in pain domain (P < 0.01), TG level (P < 0.01). (2) Compared with the model control group, extracts from DOF (6 g · kg(-1)) could notably reduce facial and ear micro-circulation blood flow, facial temperature and heart rate (P < 0.05, P < 0.01) and AST (P < 0.05) and enhance pain domain (P < 0.01) and TG (P < 0.01). Extracts from DOF (4 g · kg(-1)) could remarkably reduce AST and ALT levels (P < 0.01, 0.05). Extracts from DOF (6 g · kg(-1) 4 g · kg(-1)) could significantly reduce T3 and increase serum TSH level (P < 0.05). DOF could improve Yin deficiency symptoms of zygomatic red and dysphoria in mice as well as liver function injury caused by overactive thyroid axis. According to its action mechanism, DOF may show yin nourishing and hepatic protective effects by impacting thyroxin substance metabolism, improving micro-circulation and reducing heart rate.

5.Analysis of HOXD13 gene in polydactyly in dispersion type.

OBJECTIVETo analyze HOXD13 gene in polydactyly in dispersion type of Fujian Han population in order to know whether there is mutation in HOXD13.
METHODSAll members were evaluated physically and radlologically. Genomic DNA was extracted from peripheral blood of the patients who were treated from Dec. 2012 to Apr. 2013, their parents, grandparents, and normal volunteers from our department. The polymerase chain reaction ( PCR) , agarose gel electrophoresis and DNA sequence analysis were adopted to analyze HOXD13 from six cases with polydactyly and forty normal volunteers.
RESULTSAll patients had no family history. A heterozygous synonymous mutation, c. 291 C > T( p. A60A), was detected in exon 1 of the HOXD13 Gene in five of the polydactyly patients. Similar mutation was not detected in one brachy dactyly patient and the forty normal volunteers.
CONCLUSIONA heterozygous synonymous mutation, c.291C > T (p. A60A), of the HOXD13 gene may be related with polydactyly in dispersion type of Chinese han population.

6.The application of medpor and split-thickness skin graft in formation of cranioauricular sulcus during auricular reconstruction with Nagata method.

OBJECTIVETo investigate the application of medpor and split-thickness skin graft in formation of cranioauricular sulcus during auricular reconstruction with Nagata method.
METHODSThe first stage operation was fulfilled according to the Nagata two-stage method which involves fabrication and grafting of the costal cartilage framework. The second-stage ear elevation operation was undertaken 6 months later to form the cranioauricular sulcus. Split-thickness skin was taken from temporal and accipital area. After releasing the auricular framework and transplanting C shaped medpor at the rear side of framework, the temporaparietal fascia flap was transferred to cover postauricular medpor and framework. Then the split-thickness skin graft was implant on the fascia surface.
RESULTSFrom July 2010 to August 2012, 20 cases (22 ears) were treated. Partial necrosis of temporaparietal fascia flap and framework exposure happened in 1 case. Successful ear reconstruction was achieved in other cases with satisfactory cranioauricular sulcus during the follow-up period of 6-18 months (average, 13 months).
CONCLUSIONSThe application of medpor and split-thickness skin graft in the ear elevation of Nagata method for auricular reconstruction for microtia can achieve satisfactory results. It not only avoids the obvious scar in the donor site due to harvesting full-thickness and intermediate-thickness skin, but also reduces chest trauma due to harvesting costal cartilage.

7.Safety and effectiveness of GreenLight 120-W laser photoselective vaporization of the prostate for benign prostatic hyperplasia: A meta-analysis.

OBJECTIVETo evaluate the safety and effectiveness of GreenLight 120-W laser photoselective vaporization of the prostate (PVP) versus transurethral resection of the prostate (TURP) for benign prostatic hyperplasia (BPH).
METHODSWe searched PubMed, Medline, Embase, Cochrane Library, Wanfang, CNKI, and VIP for randomized control trials and their references addressing 120-W PVP versus TURP in the treatment of BPH. Based on the inclusion and exclusion criteria, two reviewers independently accomplished the screening, quality assessment, and data extraction of the identified studies and performed meta-analyses using RevMan 5.2.
RESULTSTotally, 6 randomized control trials were included in this analysis, involving 703 cases, 351 treated by PVP and 352 by TURP. Compared with TURP, PVP showed significantly decreased time of catheterization (by 32. 55 hours, 95% CI 15.3 -49.8, P < 0.01), hospital stay (by 1.85 days, 95% CI 1.2-2.5, P < 0.01), and intraoperative blood loss (by 15.6 g/L, 95% CI 10.0-21.2, P < 0.01), but increased time of operation (by 9.37 minutes, 95% CI 5. 1-13.6, P < 0.01). There was also a significant reduction in blood transfusion, TUR syndrome, and capsular perforation in the PVP group. At 12 months after surgery, no statistically significant differences were found between the two groups in the improvement of maximum urinary flow rate, IPSS, postvoid residual, and sexual function.
CONCLUSIONGreenLight 120-W laser PVP is a safe and effective procedure for the treatment of BPH, with similar effectiveness to TURP but less blood loss, shorter time of catheterization and hospital stay, and lower incidences of blood transfusion, TUR syndrome and capsular perforation.

10.Clonal expansion and genetic diversity of nalidixic acid-resistant Salmonella enterica serotype Paratyphi A in Yuxi city, China

Objective To understand the elonal expansion and genetic diversity of Salmonella en-terica semtype Paratyphi A (SPA) and to construct a typing method to determine the epidemic clones of the isolates. Methods Antimicrobial susceptibility testing was performed with 3980 SPA isolates by the cen-trolled Kirby-Bauer disc diffusion technique on Muller-Hinton agar plates. A total of 15 SPA with nalidixie acid resistance for mutations in gyrA, gyrB, gyrC and gyrE genes within the quinolone-resistant determina-tion region (QRDR) were examined. Subtyping of 121 isolates of SPA from seven counties in Yuxi were studied using pulsed-field gel eleetrophoresis (PFGE) analysis following digestion of chromosomal DNA with restriction endanucleases Spe Ⅰ and Xba Ⅰ. PFGE patterns were analyzed by duster analysis. Results The nalidixic acid-susceptible isolates predominated in 1999 but was replaced by nalidixic acid -resistant (NAR) isolates after 2000. Amplification by PCR and sequencing of the genes with subsets of 15 NAR strains re-vealed that the resistance mechanisms had resulted from single point mutations in the gyrA gene. Spe Ⅰ and Xba Ⅰ digestion of 121 isolates gave five and four different PFGE patterns with the predominance of the Spe Ⅰ 01 and Spe Ⅰ 02 (or the Xba Ⅰ 01) epidemic patterns, respectively. Spe Ⅰ 01 and Spe Ⅰ 02 consisted of 37.2% and 57.9% of isolates, respectively, or Xba Ⅰ 01 consisted of 95.0% of isolates. Conclusion The incidence of resistance to nalidixic acid of the isolates increased during the study period. PFGE patterns Spe Ⅰ 01 and Spe Ⅰ 02 (or Xba Ⅰ 01), the main clones of the epidemics, are highly prevalent in Yuxi. PFGE with Spe Ⅰ and Xba Ⅰ is a useful technique to differentiate SPA.
