1.The correlation between serum uric acid concentration and obesity in children
International Journal of Laboratory Medicine 2015;(3):322-323
Objective To investigate the correlation between hyperuricemia and obesity in children. Methods 316 children who went to the hospital for treatment of obesity were enrolled in the study as obesity group.Age and sex-matched children with normal body mass index(BMI)were randomly recruited as control group.Fasting venous blood samples were collected for the detection of serum uric acid(UA),glucose(GLU),total cholesterol(CHOL),triacylglycerol (TG),high-density lipoprotein cholesterol(HDL-C) and low-density lipoprotein cholesterol(LDL-C).Results Compared with the control group, serum concentrations of UA,CHOL, TG and LDL-C were higher while HDL-C was lower in obese children, the difference was statistically significant(P <0.05 ).The detection rate of hyperuricemia in obese children was 24.68%,and in control group was 9.49%,the difference was statistically sig-nificant(P <0.05 ).Serum UA concentration was positively correlated with TG,CHOL and LDL-C,negatively correlated with HDL-C.Conclusion Obesity can lead to higher serum UA concentration and abnormal lipid metabolism. The level of serum UA was correlated with abnormal lipid metabolism.

Objective To discuss the change of cellular immune and humoral immune function in children patients with Henoch-Schonlein purpura(HSP)and to investigate its clinical significance. Methods The expression levels of T lymphocyte CD3 + ,CD3 +CD4 + ,CD3 + CD8 + ,CD3 + CD4 + CD8 + ,B lymphocyte CD3 - CD19 + and NK lymphocyte CD3 - CD16 + CD56 + in periphetal blood of 156 patients with HSP was detected by flow cytometry. The level of immunioglobins(IgG,IgA,IgM)and complement(C3,C4)of serum were evaluated by nephelometry immunoassay. The level of immunioglobin IgE was measured by immunochemiluminometric assays and compared with 25 healthy controls. Results Compared with control group, the ratio of CD4 +/CD8 + and the contents of CD3 + ,CD4 + cells in HSP patients were decreased significantly(P <0.01),while the content of CD19 + was significantly increased (P <0.01).The content of CD16 + CD56 + was decreased(P <0.05 ),while the content of CD3 + CD4 + CD8 + was increased(P <0.05),the content of CD8 + had no obvious change(P >0.01).The level of IgA and IgE were increased significantly(P <0.01).The level of IgG and C3 were increased (P <0.05),The level of IgM and C4 had no obvious change(P >0.05).Conclusion The cellular immune and humoral immune function in children with HSP are in disorder. To detect lymphocyte subsets, immunioglobin and com-plement will be of important significance to understand the immune function from the perspective of the cellular immune and humor-al immune to explore the pathogenic mechanisms and to assist diagnosis and patient′s prognosis.

Objective To evaluate the influence of SOCS3 conditional ablation in macrophage polarization and functional recovery after spinal cord injury in mice. Methods The Socs3fVfl Nes cre mice were used to obtain nervous-system-restricted SOCS3 deletion models as experimental group (n=40),and littermate Socs3fl as control group (n=40).Mice (n=35) in each group were underwent T10 spinal cord compressing injury (the left 5 were as sham-operated controls);on the third,7th and 14th d of injury, injured tissues from 15 mice of each group were harvested;immunofluorescence was performed to detect the changes of M2a and M2c phenotypic marker Arginase1 and M1 and M2b phenotypic marker CD86;on the first,third,7th and 14th d of injury, injured tissues from the left 20 mice from each group were harvested;fluorescent quantitative real time-PCR was used to detect the macrophage phenotypic markers (Arginasel,CD206,iNOS and CD32),SOCS3 and STAT3 gene expressions;the above data from the sham-operated controls were used as base data.BMS scale was used to evaluate the motor functions of posterior limbs and bodies on the first,third,7th and 14th d of injury. Results The behavioral assessments of the experimental group were superior on the third,7th and 14th d of injury as compared with those of control group (P<0.05).Immunofluorescence showed that as compared with the controls, the Socs3fVfl Nes cre mice had a predominance in distribution and population of Arginasel-positive macrophages in spinal cords on the third,7th and 14th d of injury;in contrast, the control group showed superiority of CD86 expression on the 7th and 14th d,with significant differences (P<0.05).Similarly,fluorescent quantitative real time-PCR analysis demonstrated that gene expression of A rginase I on the 7th d of injury, that of CD206 on the 7th and 14th d of injury, and that of STAT3 on the third,7th,and 14th d of injury were significantly increased, whereas that of CD32 on the 14th d of injury and SOCS3 on the first and third d of injury were significantly reduced in the experimental group,as compared with those in the control group (P<0.05).Conclusion SOCS3 conditional knockout can promote the formation of M2 macrophage,as well as good functional recovery after spinal cord injury in mice.
