1.Differentiation of HL-60 cell induced by trichostatin

Objective To detect whether trichostatin(TSA) can induce HL-60 differentiation in vitro. Methods MTT method was used to test the effect of TSA on HL-60 cell growth. Cell cycle was tested by flow cytometry. CD11b expression was tested for detecting cell differentiation, RT-PCR was used to detect the mRNA expression of c-myc in the cells treated by TSA. Results Down-regulation of cell proliferation was observed and cells significantly accumulated at the G0 and G1 phase in HL-60 cells treated with TSA( P <0. 01). Dif-ferentiation rate was 15. 24% after being treated by TSA for 48 h. mRNA of c-myc was down regulated in time-dependent manner. Conclusion TSA can inhibit proliferation and induce differentiation in HL-60 cells.
2.The correlation between serum uric acid concentration and obesity in children

Objective To investigate the correlation between hyperuricemia and obesity in children.Methods 316 children who went to the hospital for treatment of obesity were enrolled in the study as obesity group.Age and sex-matched children with normal body mass index(BMI)were randomly recruited as control group.Fasting venous blood samples were collected for the detection of serum uric acid(UA),glucose(GLU),total cholesterol(CHOL),triacylglycerol (TG),high-density lipoprotein cholesterol(HDL-C) and low-density lipoprotein cholesterol(LDL-C).Results Compared with the control group,serum concentrations of UA,CHOL, TG and LDL-C were higher while HDL-C was lower in obese children,the difference was statistically significant(P <0.05 ).The detection rate of hyperuricemia in obese children was 24.68%,and in control group was 9.49%,the difference was statistically sig-nificant(P <0.05 ).Serum UA concentration was positively correlated with TG,CHOL and LDL-C,negatively correlated with HDL-C.Conclusion Obesity can lead to higher serum UA concentration and abnormal lipid metabolism.The level of serum UA was correlated with abnormal lipid metabolism.
3.Change of immune function in children patients with Henoch-Schonlein purpura and its clinical significance

Objective To discuss the change of cellular immune and humoral immune function in children patients with Henoch-Schonlein purpura(HSP)and to investigate its clinical significance.Methods The expression levels of T lymphocyte CD3 + ,CD3 +CD4 + ,CD3 + CD8 + ,CD3 + CD4 + CD8 + ,B lymphocyte CD3 - CD19 + and NK lymphocyte CD3 - CD16 + CD56 + in periphetal blood of 156 patients with HSP was detected by flow cytometry.The level of immunioglobins(IgG,IgA,IgM)and complement(C3,C4)of serum were evaluated by nephelometry immunoassay.The level of immunioglobin IgE was measured by immunochemiluminometric assays and compared with 25 healthy controls.Results Compared with control group,the ratio of CD4 +/CD8 + and the contents of CD3 + ,CD4 + cells in HSP patients were decreased significantly(P <0.01),while the content of CD19 + was significantly increased (P <0.01).The content of CD16 + CD56 + was decreased(P <0.05 ),while the content of CD3 + CD4 + CD8 + was increased(P <0.05),the content of CD8 + had no obvious change(P >0.01).The level of IgA and IgE were increased significantly(P <0.01).The level of IgG and C3 were increased (P <0.05),The level of IgM and C4 had no obvious change(P >0.05).Conclusion The cellular immune and humoral immune function in children with HSP are in disorder.To detect lymphocyte subsets,immunioglobin and com-plement will be of important significance to understand the immune function from the perspective of the cellular immune and humor-al immune to explore the pathogenic mechanisms and to assist diagnosis and patient′s prognosis.
