1.Mutation of SLC40A1 gene in newly diagnosed type 2 diabetic patients with iron overload

Objective To investigate the incidence of iron overload (liver biopsy tissue Prussian blue staining was positive)in newly diagnosed type 2 diabetic patients (T2DM) and to analyze the SLC40A1 gene (NG_009027.1) mutation in 12 newly diagnosed T2DM with iron overload.Methods 198 patients with newly diagnosed T2DM were collected in our hospital from December 2014 to December 2015.12 of them had elevated ferritin and liver biopsy tissue Prussian blue staining was positive.DNA was extracted from the paraffin section of liver tissue and the exons of SLC40A1 gene were amplified by polymerase chain reaction (PCR).The purified products were sequenced in two directions.Results The incidence of iron overload in these newly diagnosed T2DM patients was about 6%.One point mutation c.663T> C was found on the sixth exon region of SLC40A1 gene in all these 12 T2DM patients with iron overload.It's encoded amino acid did not change (221V>V),meaning a samesense mutation.Conclusion Iron overload has a certain incidence in patients with newly diagnosed T2DM.Iron overload may not due to the mutations in SLC40A1 gene,which need further investigation.
2.Cell proliferation and apoptosis in squamous carcinoma of cervix after neoadjuvant chemotherapy

Objective To determine the effect of neoadjuvant chemotherapy (NAC) for cervix cancer on cell proliferation and apoptosis. Methods A total of 24 patients with squamous carcinoma of the cervix (SCC) were treated with one cycle of cisplatin combined with hydroxycamptothecine, bleomycin regimen. The expression of proliferating cell nuclear antigen (PCNA) and apoptosis index (AI) were detected before and after NAC with flow cytometry and TUNEL. Results Clinical response was 58.3% (14/24). Before and after NAC, the expression of PCNA were (0.386?0.078) and (0.125?0.040) respectively and AI were (0.052?0.027) and (0.248?0.078). 3.Optimization of N2a cell transfection mediated by liposome

BACKGROUND:Cationic liposome-mediated celltransfection is reliable and repeatable. However the transfection efficiency is often low.
OBJECTIVE:To study the optimized methods for gene transfection mediated by liposome into N2a cells (mouse neuroblastma cells).
METHODS:Using traditional adherent method and improved suspension method, 500 ng recombinant plasmid pcDNA3-GFP carrying green fluorescence protein was transfected into N2a cells in 24-wel culture plate, which was mediated by 1.5μL Lipofectamine?LTX Reagent. The expression of green fluorescent protein was observed by inverted fluorescence microscope, and the transfection efficiencies at different transfection ways were calculated. By using improved suspension transfection method, 500 ng plasmid DNA was transfected with different doses of Lipofectamine?LTX Reagent (1.0, 1.5, 2.0, 2.5μL). The optimal ratio of liposome and DNA was explored.
RESULTS AND CONCLUSION:The transfection efficiency of suspension transfection method was significantly higher than that of the tranditional adherent method (P<0.01) when using 1.5μL liposome/500 ng DNA. The transfection efficiency of the 1.0, 1.5, 2.0, 2.5μL Lipofectamine?LTX on 500 ng plasmid DNA was respectively (76.60±3.85)%, (80.00±4.17)%, (88.00±5.89)%, (54.96±4.23)%. It showed the 500 ng DNA and 2.0μL liposome achieve the highest transfection efficiency.
4.Effect of TPh on cell HepG-2 cell cycle, apoptosis and related gene expression of hepatocellular carcinoma

Objective To investigate the effect of different concentrations of TPh on hepatoma cells , cell viability and and its possible mechanisms of anti-tumor.Methods The inhibitory rate of hepatoma cells and cell viability on different concentrations of TPh and time were measured by MTT assay;The morphological changes of apoptosis were observed by Hoechst 33258 straining; Cell cycle distribution was evaluated by flow cytometry (FCM).The protein expression of Bcl-2 and Bax were detected by Western blot analysis.Results MTT assay showed that TPh inhibited the proliferation of HepG-2 cells in a dose-dependent manner, and the inhibitory rate increased with the increase of concentration.The inhibitory rate was 50.9% (P<0.01).The results of Hoechst 33258 staining showed that the fluorescence intensity of hepatocellular carcinoma cells was light blue in fluorescence microscopy and bright blue fluorescence in apoptotic hepatocarcinoma cells, and the apoptosis of hepatocarcinoma cells increased with the increase of drug concentration.The percentage of cells in G0/G1 phase increased with the increase of cell cycle, and the ratio of cells in S phase was decreased in G2/M phase compared with blank control group (P<0.05);Western blot results showed that compared with the blank control group, TPh inhibited the proliferation of Bcl-2 cells in a concentration-dependent manner ( P <0.05 ) , and the number of apoptotic cells increased with the increase of drug concentration (P<0.05), and the ratio of Bcl-2/Bax in the TPh group decreased significantly (P<0.05), and the expression of Bax gene increased. Conclusion TPh inhibits cell proliferation, promotes apoptosis and induces HepG-2 to block G0/G1 phase.Its mechanism may increase the expression of Bax and decrease Bcl-2 protein expression.
5.Studies of Quality Control of Shiwei Shujin Huoluo Powder

Objective To set up the quality standard of Shiwei Shujin Huoluo Powder. Methods Rhizoma Polygoni cuspidati and Cortex Cinnamomi, the main ingredients of the powder, were identified by thin layer chromatography ( TLC) , and the content of emodin in the powder was detected by high performance liquid chromatography (HPLC) . Results The spots showed by TLC were clear without interference of the negative control. HPLC results showed that linear range of emodin was 9.30 ~ 46.52 μg·mL-1, the recovery of emodin was 96.22%, and RSD was 1.76% ( N=6) . Conclusion The established method is reliable and accurate, and can be used for quality control of Shiwei Shujin Huoluo Powder.
6.Expression of the plasma IL-11 and soluble glycoprotein 130 in the patients with acute leukemia

Objective To explore the levels and the significance of IL-11 and soluble glycoprotein 130(sgp130) in the treatment of new-diagnosed acute leukemia(AL) during the induced remission. Methods The levels of IL-11 and sgpl30 in 47 patients with acute leukemia were determined by ELISA respectively before treatment and after completed remission(CR), and the number of white blood cell (WBC) and platelet (Plt) were valued by hametometry. Results Plasma IL-11 level of AL patients was significantly lower than normal (P <0.05), and increased obviously to the normal level when complete remission was achieved. And it correlated with PLT counts. While sgp130 level was higher than normal (P<0.05),and declined after CR and correlated with WBC counts. Conclusion The plasma IL-11 and sgp130 levels can be helpful for confirming the diagnosis,evaluating the efficiency and predicting the prognosis of AL.
7.Evaluation on the efficacy of human umbilical cord derived-mesenchymal stem cell transplantation in liver cirrhosis patients with ascites in a prospective and control trial

ObjectiveTo evaluate the one-year follow-up of the therapeutic efficacy of human umbilical cord derived-mesenchymal stem cell (UC-MSC) transplantations in decompensated liver cirrhosis patients with ascites.MethodsFifty-four liver cirrhosis patients with ascites in Research Center for Biological Therapy in 302 Military Hospital were divided into treatment group (n=38) and control group (n=16) in a prospective controlled single-blinded trial.UC-MSC (0.5-1.0) × 106/kg and saline were intravenously transplanted into patients monthly for 3 times in treatment group and control group,respectively.The liver function,hepatitis B virus (HBV) DNA level,ascites and the model for end-stage liver disease(MELD)scores at different time points were compared between two groups.The comparison between groups was done by Mann-Whitney U test,and the data before and after transplantations were compared by Wilcoxon signed rank sum test.ResultsThere were no significant differences of alanine transaminase (ALT),total bilirubin (TBil),cholinesterase (CHE),HBV DNA positive rate and MELD scores at different time points between two groups (P>0.05).However, the albumin ( A1b)level was significantly increased after 36 weeks of UC-MSC transplantation in treatment group, which were (28.47±4.45)g/L at week 0 and ( 34.82±4.50)g/L at week 48 (P=0.046). Meanwhile, the ascites reduced markedly in treatment group with (46.6 ±30.6) mm at week 0 and (6.6±13.6) mm at week 48,which were significantly different from control group at the end of follow-up (P =0.037). Conclusion UC-MSC transplantations may help to increase A1b level and reduce ascites in patients with decompensated liver cirrhosis.
8.Analysis of drug procurement model of multiple areas based on joint inventory in public hospitals

Objective To evaluate the application effect of joint inventory management method in drug procurement and control management in multiple hospital areas.Methods Based on the joint inventory management model,four pilot drugs were selected from a certain group hospital by reasonable data processing methods for research.The effects of the model application were compared and analyzed from aspects such as inventory cost,turnover situation,and supply situation.Results After applying the joint inventory management model led by the central hospital,the inventory and amount of drugs in the three pilot hospitals were significantly reduced,with the inventory reduced by 31.93%and the average inventory amount decreased by 16.23%;The inventory turnover days had significantly decreased,with the turnover days of all three branches decreasing by more than one day;The drug shortage rate had significantly decreased,with the most significant change among the pilot drugs being the doxorubicin liposome injection,which had a 6.7%decrease in the shortage rate;The comparison results of each group of data showed statistical significance(P<0.05).Conclusion Adopting a central hospital led joint inventory management model in multiple hospital areas could significantly improve the effectiveness of drug procurement management and inventory management,which enhanced the efficiency of hospital fund utilization.
9.Reliability and validity of the Chinese version of the Type 2 Diabetes Stigma Assessment Scale

Objective To translate the English version of Type 2 Diabetes Stigma Assessment Scale(DSAS-2) into Chinese,and to test the reliability and validity of the Chinese version of DSAS-2. Methods The DSAS-2 was translated and adapted according to Chinese culture following the translation and back-retranslation procedure.The reliability and validity of the Chinese version of DSAS-2 was tested among 294 patients with type 2 diabetes. Results The Chinese version of DSAS-2 included three subscales: Treated Differently (6 items), Blame and Judgment (7 items), and Self-stigma (6 items) and contained a total of 19 items.The Cronbach α coefficient of the Chinese version of DSAS-2 was 0.879,the Cronbach α coefficient of three factors was 0.832,0.815 and 0.844,respectively;the test-retest reliability was 0.835. The content validity index was 0.916. The scores of DSAS-2 correlated with the scores of Rosenberg Self-esteem Scale, Patient Health Questionnaire 8- Item Scale and Generalized Anxiety Disorder 7-Item Scale (r =-0.452, 0.443, 0.412, P<0.01). Three factors were extracted by exploratory factor analysis and could explain 55.75% of the total variance. Conclusions The Chinese version of DSAS-2 has acceptable reliability and validity,which can be used to evaluate stigma among patients with type 2 diabetes in China.
10.Prospective association between peripubertal diet quality and puberty timing

Objective:
To examine the prospective association between peripubertal diet quality and pubertal timing.
Methods:
Multivariate regression analyses were performed using data obtained from 1 588 SCCNG (Southwest China Childhood Nutrition and Growth) study participants, which included dietary data, anthropometric parameters, and information about potential confounders during peri puberty, two and four years before the onset of menarche or voice break. Dietary intake in peri puberty was assessed using a validated food frequency questionnaire. Diet quality was determined using the Chinese Children Dietary Index (CCDI), which measures adherence to current dietary recommendations. Age at menarche or voice break (M/VB) were used as pubertal markers.
Results:
The CCDI score ranged from 58.0 to 132.5 for girls and from 48.3 to 129.9 for boys. The pubertal markers consistently indicated that girls and boys with a higher peripubertal diet quality entered puberty later than their counterparts with lower CCDI scores (higher vs. lower CCDI tertiles: age at M was 13.1 years (11.2-13.3) and 12.5 years (12.0-13.9)(P<0.01); VB was 13.8 years (12.1-14.8) and 13.4 years (12.5-15.1)(P=0.03), after adjusting for paternal education level, energy intake, and pre pubertal body fat.
Conclusion
Children with a higher diet quality during peri puberty entered puberty at a later age. Dietary guidance and intervention are needed for better dietary quality among children during pubertal transition.
