The results of study of glucocorticoid receptors (GCR) in mononuclear leukocytes (MNL) of peripheral blood in 25 patients with chronic glomerular disease are presented.lt is found that out of patients with primary nephrotic syndrome (PNS), those having a relatively higher content of GCR before taking GC and an obvious reduction of GCR content after taking GC will quickly respond to GC.
2.The relationship between the cyclin kinase inhibitor p27 and the hypertrophy of mesangial cell cultured in high glucose

Objective Our study aimed at the role of p27 on the hypertrophy of mesangial cell (MC) cultured in high glucose. Methods The p27 protein of MC lysate was detected with western blotting analysis. The degree of cultured MC hypertrophy was estimated through [3H]-thymidine incorporation and [3H]-leucine incorporation. The effect of reducing p27 expression on cell hypertrophy was analysed with p27 antisense oligodeoxynucleotide (ODN) phosphorothioate. Results in MC cultured in high glucose (450mg/dl) serum-free DMEM compared with MC cultured in normal glucose (100mg/dl) serum-free DMEM, p27 increased, [3H]-leucine incorporation increased and[3H]-thymidine incorporation decreased;p27 antisense ODN transfection reduced [3H]leucine incorporation, increased [3H] thymidine incorporation of MC cultured in high glucose senumfree DMEM. Increasing medium osmolarity with D-mannitol failed to induce p27 expression of MC. Conclusion p27 protein increased in MC cultured in high glucose. High level of p27 played an important role in MC hypertrophy induced by high glucose. Because the cell cycle is controlled by the interaction between the positive cell cycle regulatory proteins(CCRP) and negative CCRP, further research is needed to study the expression of the positive and negative CCRP in MC in order to better understand the role of CCRP in MC hypertrophy.
3.Changes of Urine Thromboxane A_2-Prostacyclin Balance in Patients with Lupus Nephritis and Their Clinical Significance

The levels of urine thromboxane B, (TXB2) and 6-keto-PGF1a were measured by radioimmunoassay in 24 patients with lupus nephritis and 12 normal subjects. The results showed that the level of TXB2 and the ratio of TXB2/6- keto- PGF1x in urine were significantly higher, while the level of urine 6-keto-PGF1x( was lower in patients with lupus nephritis than that in normal group. The ratio of TXB2/6-keto-PGF1a in urine was correlated positively to the content of urine protein. These results indicate that there is TXA2-PGI2 imbalance in the kidney in patients with lupus nephritis. TXA2 can play an important pathogenetic role in the progression of lupus nephritis in patients. The ratio of TXB2/6-keto-PGF1x in urine may serve as a parameter in evaluating patients with lupus nephritis.
4.The Clearance Differences of Different Molecule Weight Uremic Substances in Different Hemopurifications

In this study, authors observed and compared the clearances of different molecule weight substances between different dialysis membranes and different hemopurification methods. The results showed that the clearances of BUN by hemodialysis (HD) with cuprophan membrane, hemodiafiltration (HDF) with polysulfone and high flux dialysis (HFD) with polysulfone were all satisfactory. TACurea were all below 50 mg. Hemodialysis with cuprophan could clean out some middle molecule substances (MMS), but not ?2-microglobulin (?2m). Hemodiafiltration with polysulfone had the best clearances of MMS and ?2m. Hemofiltration (HF) with polysulfone had fair clearances of MMS and ?2m, but the clearances of BUN and Cr by HF were not so good. The clearances of MMS and ?2m by high flux dialysis were lower than those of HDF and HF. The clearances of MMS and ?2m by continuous ambulatory peritoneal dialysis (CAPD) at a single time were lower than those of HF, HDF and HFD, but the general clearances of MMS and ?2m by CAPD per week were two to four times higher than those by hemodialysis with cuprophan. CAPD is a choice method for those hospitals where HF or HDF can not be performed.
5.Changes of cyclin kinase inhibitor P27 in glomerulus of Thy1 glomerulonephritis in rats

Objective: To investigate the role of cyclin kinase inhibitor P27 on the proliferation of mesangial cell(MC) in Thy1 glomerulonephritis(GN) rats. Methods: The Thy1 model of experimental mesangial proliferative glomerulonephritis was induced by intravenous injection of rabbit anti-thymocyte plasma. The P27 protein of glomerular lysate was detected with Western blotting analysis. The P27 mRNA was determined with RT-PCR. The extracellular matrix (ECM) protein (fibronectin and type Ⅳ collagen) of glomerular lysate was examined with ELISA. Results: Glomerular P27 protein of Thy1 model rat decreased compared with that of normal rat. The level of P27 was associated with the degree of MC proliferation:P27 began to decrease on day 1 in Thy1 model, then reached the lowest on day 3 when MC proliferation was maximal, then returned to 60% of the baseline level on day 5 when MC proliferation began to resolute. 6.Experimental study of the nephrotoxicity of Guangfangji

AIM To study the nephrotoxicity of various doses of Guangfangji . METHODS Normal Wistar rats were given 1, 5 and 10 g?kg -1 of Guangfangji respectively. Renal pathology and function were observed. RESULTS Rats given 1 g?kg -1 of Guangfangji for 8 weeks showed normal renal function and histology Rats given 5 g?kg -1 of Guangfangji significantly increased 24 hour urinary protein excretion Tubular degeneration and interstitial edema was observed Blood urea nitrogen (BUN) and serum creactinine (Scr) remained in the normal range BUN and Scr increased significantly in the group given 10 g?kg -1 of Guangfangji for 4 weeks The tubulointerstitial abnormalities were more severe in the group given 5 g?kg -1 of Guangfangji CONCLUSION Longterm use of pharmacopoeial dose of Guangfangji shows no harm to the kidney.Renal injury may occur if relatively large dose of Guangfangji is given and the period of treatment using this drug is relatively longer

OBJECTIVE: To investigate the role of p27 in the inhibition of emodin on the mesangial cell (MC) proliferation induced by tumor necrosis factor-alpha(TNF-alpha). METHODS: p27 protein of MC was detected with western blotting analysis. The degree of MC proliferation was estimated through [(3)H] thymidine ([(3)H] TdR) incorporation. Different dosage of emodin (50 mg/L,100 mg/L) was added into MC stimulated by TNF-alpha. RESULTS: TNF-alpha (200 kU/L) decreased p27 level of MC cultured in serum-free DMEM for 24 hours and increased[(3)H] TdR incorporation. Emodin increased p27 level of MC stimulated by TNF-alpha and decreased [(3)H] TdR incorporation. The more the emodin was added, the greater the above-mentioned effect of emodin. CONCLUSION: The increment of p27 level maybe play an important role in the inhibition of emodin on MC proliferation induced by TNF-alpha.

10.Evaluating the adequacy of hemodialysis with neural calculating method

Objective: To study the feasibility of evaluating the adequacy of hemodialysis using neural calculating method. Methods: The adequacy of hemodialysis patients were evaluated using Daugirdas, TACurea and neural calculating method respectively, the results of the 3 method; were compared with the clinical assessment of the patients. Results: The coincidence rate among the 3 methods was 84.6%, coincidence rate between neural calculating method and the clinical outcome of the patients was 92.3%, which was significantly higher than that of Daugirdas method (76.9%) and of TACurea (80.8%). Conclusion: Neural calculating method has higher accuracy in assessing the adequacy of hemodialysis patients and is clinically practical.
