1.MOLECULAR IDENTIFICATION AND CLASSIFICATION OF DEEP SEA PSYCHROPHILIC BACTERIA FROM EAST-PACIFIC SEDIMENT

Many psychrophilic bacteria were isolated from east-pacific sediment which is 5,300m in depth. The genera of 13 isolations were identified and classified based on the comparison and phylogenetic study of 16S rRNA gene sequences. The results indicated that they belong to four genera including Parococcus, Pseudomonas, Halomonas and Pseudoalteromonas.
2. Process optimization of automatic dispensing machine in outpatient pharmacy

OBJECTIVE: To investigate the automatic dispensing process optimization program in outpatient pharmacy. Automatic dispensing machine integrated drug storage device, HIS and network communication as a whole to improve the drug distribution accuracy and reduce the management cost. The machine emancipated pharmacists from the traditional dispensing mode to provide much more time for pharmaceutical service. METHODS: According to the characteristics of drug use in the outpatient department of our hospital and the hospital information system, a lot of work to be, optimized the automatic dispensing process. RESULTS: After optimization, drug distribution efficiency increased significantly, and the time patient take medicine shorten greatly. CONCLUSION: Automatic dispensing machine could improve the quality of pharmaceutical care and the promotion of information technology in the field of drug dispensing and management. Automatic dispensing machine is the inevitable trend to hospital pharmacy.
3.Wnt/β-catenin signal pathway mediated Salidroside induced directional differentiation from mouse mesenchymal stem cells to nerve cells.

OBJECTIVETo explore the molecule mechanism of Salidroside inducing directional differentiation of mouse mesenchymal stem cells (MSCs) into neuronal cells.
METHODSThe mouse multipotent mesenchymal precursor cell line (D1) was taken as the objective. Cultured MSCs were divided into the negative control group (complete culture solution), the positive control group (containing 1 mmol/L β-mercaptoethanol), the Salidroside induced group (20 mg/L Salidroside), and the blocked group (20 ng/ ml DKK1, a special inhibitor of Wnt/β-catenin signal pathway). All cells were inoculated in a 6-well plate (1 x 10(4) cells/cm2) and grouped for 24 h. The expression of p-catenin was detected by fluorescence Immunochemistry in the negative control group, the positive control group, and the Salidroside induced group. The expression of neuron-specific enolase (NSE), beta 3 class III tubulin (β-tubulin III), nuclear receptor related factor 1 (Nurr1), glial fibrillary acidic protein (GFAP) mRNA, Wnt3a, β-catenin, low-density lipoprotein receptor-related protein6 (LRP6), Axin mRNA were detected using reverse transcrip- tion PCR (RT-PCR). The expression of β-catenin and NSE protein were analyzed by Western blot in the negative control group, the positive control group, and the Salidroside induced group. Ca2+ chelating agents (EGTA), L-type Ca2+ channel blocker (Nifedpine), and IP3Ks special inhibitor (LY294002) were used to block Ca2+ signal pathway respectively. The expression of Wnt3a, LRP-6, Axin, glycogen syn- thase kinase (GSK-3), and β-catenin mRNA were detected by RT-PCR. The β-catenin protein expression was analyzed using Western blot.
RESULTSCompared with the positive control group, β-catenin protein was strong positively expressed; the expression of Wnt3a, β-catenin, LRP6, Axin, NSE, β-tubulin III, Nurr1 mRNA, and NSE protein were obviously up-regulated in the Salidroside induced group (P < 0.01). Compared with the positive control group and the Salidroside induced group, β-catenin, NSE, Nurr1, and β-tubulin III mRNA expression decreased; β-catenin and NSE protein expression were also down-regulated in the blocked group (P < 0.01). Compared with the Salidroside induced group, the expression of Wnt3a, LRP-6, β-catenin, and Axin mRNA were down-regulated in the Ca2+ signal blocked group and the salidroside induced group (P < 0.01, P < 0.05).
CONCLUSIONSalidroside affected directional differentia- tion of MSCs into neuronal cells through Wnt/β-catenin and Ca2+ signal pathway.

5.Helping to early diagnosis of SARS by flow cytometric three color absolute count of T lymphocyte subsets in blood

Objective To investigate the changes of T lymphocyte subsets of severe acute respiratory syndrome(SARS) patients during acute phase and recovery stage and study its clinical significance in early diagnosis of SARS.Methods T lymphocyte subsets of 40 patients of SARS and 22 cases of SARS recovery stage were detected by flow cytometry.Results Compared with normal group, T lymphocyte, Th and Ts cell counts of SARS patients decreased obviously( P =0.013, P 0.05). Conclusion The cellular immunity of SARS patients were badly damaged during acute phase. Their T lymphocyte, Th and Ts cell counts decreased obviously. This could be a guideline of early diagnosis of SARS. The counts of T lymphocyte subsets of SARS patients can return to normal after they were cured.

7.Processing of Gait Data of Hindlimbs of Spinal Cord Injured Rhesus Monkeys

Objective To establish a new processing method for gait data on Matlab to evaluate the hindlimbs behavior of non-human primates. Methods Gait analysis was tested on three rhesus monkeys 6 weeks after spinal cord injury, and kinematics data of hindlimbs were obtained using the VICON system. The raw data of kinematics were filtered and extracted, which were achieved through VICON 3D motion capture system with the Excel link combining Matlab with Microsoft Excel, and calculated in the Matlab environment. Results The kinematic parameters such as step length, step height, knee joint angle, and malleolus joint angle were gained by calculating. The mean values of step length (F=2.869, P=0.088) and step height (F=1.148, P=0.344) showed no significant difference at three speeds, which implied a higher repeatability of the data model. Angle-time curve reflected the joint function and movement. This system initially described the foot gait trajectory which could be used in gait repetitive analysis, and also generated the gait 2D/3D trajectories of hindlimbs. Conclusion The implement of these functions makes the post-processing of data more flexible and open whitout VICON system, and the calculated parameters and space tracing of gait trajectory basically meet the need of hindlimb behavior evaluation for nonhuman primate.
8.The value of thyroglobulin measurement in fine-needle aspiration for diagnosis of suspicious lymph nodes in patients with thyroid carcinoma after thyroidectomy.

OBJECTIVETo investigate the value of thyroglobulin measurement in ultrasound guided fine-needle aspiration(FNA-Tg)for detecting neck node metastasis in patients with papillary thyroid carcinoma(PTC)after thyroidectomy.
METHODSA total of 128 suspicious metastatic lymph nodes in 112 patients were retrospectively analyzed. Postoperative pathologic results were taken as the gold standard. The values of FNA and FNA-Tg combined FNA in the diagnosis of metastatic lymph nodes were compared with different ultrasonic features.
RESULTSThe sensitivity, specificity and accuracy of single FNA for the diagnosis of neck node metastasis were 67.5%, 98.0% and 79.7% respectively, and those of FNA-Tg combined FNA were 87.0%, 100.0% and 92.2% respectively. Compared with single FNA, the sensitivity and accuracy of FNA-Tg combined FNA were increased significantly(χ(2)=8.319, P=0.004; χ(2)=8.275, P=0.004). When the ultrasonographic characteristics met any one of five indicators for neck node metastasis, the sensitivity, specificity and accuracy of single FNA for the diagnosis of neck node metastasis were 38.1%, 95.7% and 68.2% respectively, and those of FNA-Tg combined FNA were 71.0%, 100.0% and 86.4% respectively. Compared with single FNA, the sensitivity and accuracy of FNA-Tg combined FNA were increased significantly(χ(2)=4.709, P=0.030; χ(2)=4.141, P=0.042). When the ultrasonographic characteristics met any two of five indicators for neck node metastasis, the sensitivity, specificity and accuracy of single FNA for the diagnosis of neck node metastasis were 55.0%, 100.0% and 73.5% respectively, and those of FNA-Tg combined FNA were 90.0%, 100.0% and 94.1% respectively. Compared with single FNA, the sensitivity and accuracy of FNA-Tg combined FNA were increased significantly(χ(2)=6.140, P=0.013; χ(2)=5.314, P=0.021). The ultrasonographic characteristics met any three, four or five of five indicators or did not meet any of the indicators, there was no significant difference in the value of the diagnosis of metastatic lymph nodes between single FNA and FNA-Tg combined FNA(P>0.05).
CONCLUSIONSThe combination of FNA with FNA-Tg could be helpful in diagnosis of lymph node metastasis. When the suspicious lymph nodes had one or two ultrasound characteristics for neck node metastasis, FNA-Tg could raise the sensitivity and accuracy of FNA, and FNA-Tg could not significantly improve in the diagnosis of FNA when presenting with no or with more than 2 ultrasonographic features.

9. Prediction of occult carcinoma in contralateral nodules based on the ultrasonic features of unilateral papillary thyroid carcinoma

Objective:
To investigate the occurrence of occult carcinoma in contralateral lobes based on the ultrasonic features of unilateral papillary thyroid carcinoma.
Methods:
The study included 202 consecutives cases of unilateral papillary thyroid carcinoma with benign nodules in the contralateral lobe identified by preoperative ultrasound or fine-needle aspiration from June 2014 to December 2015. All patients received total thyroidectomies, and with postoperative pathological examination they were divided into two groups, one including 60 cases with positive occult cancer and another one consisting of 142 cases with negative occult cancer. Univariate and multivariate analyses were performed to analyze the sonographic features of unilateral papillary thyroid carcinoma relevant to the occurrence of occult carcinoma in the contralateral nodules.
Results:

10.Clinical value of minimally invasive posterior lumbar interbody fusion assisted by X-Tube system in the treatment of low back disorders.

OBJECTIVETo evaluate the clinical outcomes of minimally invasive posterior lumbar interbody fusion (PLIF) assisted by X-Tube system for the management of degenerative lumbar diseases.
METHODSA total of 31 patients, 17 male and 14 female with ages from 38 to 75 (average 54.2 years), underwent minimally invasive PLIF assisted by the X-Tube system from May 2005 to March 2006. The index diagnosis was lumbar spondylolisthesis in 14 cases, spinal stenosis in 8 cases, separation of the posterior ring apophysis in 5 cases, intervertebral space stenosis with disk herniation in 4 cases. Before operation, conservative management for at least 6 months was proved to be failure in all these patients. The operative duration and blood loss were estimated . The changes of postoperative serum level of creatinine kinase was measured as well, and compared with the control group of 31 cases who were managed with traditional open PLIF operation during the same period at our department. The clinical functional outcomes were evaluated according to Oswestry disability questionnaire.
RESULTSThe operation lasted for 140-225 min, with a mean duration of (176 +/- 22) min. Blood loss during the operation was 270-750 ml, with a mean of (406 +/- 96) ml. Postoperative serum level of creatinine kinase was obviously lower in minimally invasive PLIF cases than in the open control cases. Although 2 pedicle screws in 2 cases were not in ideal position, there was no nerve root irritation or fixation failure and hence no revision was required. One case with spinal stenosis complained of numbness in the area dominated by left L5 nerve root after operation, but the symptom was relieved within 2 weeks through conservative management. All the 31 patients were followed up for 7-17 months, with a mean duration of 12.2 months. Lumbar radiography, and three-dimensional CT reconstruction in some cases, was performed and revealed solid fusion of the surgical segments half a year after the operation. The average Oswestry scores decreased from preoperative 40.6 +/- 5.1 to 17.4 +/- 6.5 at the first postoperative day and to 9.5 +/- 4.0 at the final follow-up. The outcome of this operation were rated as excellent.
CONCLUSIONSMinimally invasive PLIF assisted by X-Tube system has the characteristics of less blood loss, tissue trauma and operative time, quick recovery and bony fusion. The short-term outcomes are excellent.
