1. Effect of polydatin on IL-1β in serum and protein expression of Bcl-2 in brain tissue of cerebral hemorrhage injured rats

Objective: To explore the effects and mechanisms of polydatin on rats with brain hemor-rhagic injury. Methods: The cerebral hemorrhage model in rats was induced by local injection of type VII collagenase and they were randomly divided into six groups. The treated groups were treated with polydatin and Naoxuekang; The normal and control groups were only treated with water. The changes of neuro logical defect were observed. The activity of SOD, the content of MDA, the protein expressions of Bcl-2 in brain tissue, and the contents of IL-1β in serum were measured. Results: Compared with the cerebral hemorrhage model group, polydatin (25, 50, and 100 mg/kg) could significantly reduce the symptoms of neurological deficits of cerebral hemorrhage rats, raise the activity of SOD, reduce the content of MDA, increase the protein expressions of Bcl-2, and decrease the contents of IL-1β in serum. Its effects were pos-itively correlated with the dose-effect. Conclusion: Polydatin has therapeutic effect on rats of cerebral hemorrhage, through against free radical damage and inhibiting neuronal apoptosis and inflammatory.
2.Study on Clinical and Immunophenotype of Acute Leukemia in Children

Objective To study the clinical significance and characteristic of acute leukemia(AL) immunophenotype expression in children.Methods Immunofluoscence labeled with three and two color monoclonal antibodies directly were used to analyze the surface antigen and cytoplastic antigen.Cytoplastic antigen was detected by flow cytometry additionally.Results Specimens from 85 patients with childhood acute leukemia were detected.1.The undifferentiated phenotype account for 1 case(1.2%);Mixed leukemia 7 cases(8.2%);Acute myeloid leukemia 18 cases(21.2%);Acute lymphoid leukemia 59 cases(69.4%).The subtypes of acute lymphoid leukemia:(B-ALL) 50 cases(84.7%),T-ALL 6 cases(10.2%),T/B 3 cases(5.1%).2.Among 59 cases of acute lymphoid leukemia,there were 31 cases ALL expression myeloid-associated antigen;There was no difference on the clinical and prognosis between the group of myeloid-associated antigen positive and the negative group.3.Of 18 patients with AML,5 patients(27.8%) expressed lymphoid antigen,CD7 was the most commonly expressed(60%);The frequency of CD_(14) expression in childhood M4,M5 was 75%.CD_(13),CD_(33) presented in 83.3% and 94.4% cases of AML,respectively.4.Among 7 cases acute mixed leukemia,B/M 6 cases,TB/M 1 case.Five cases were misdiagnosed as ALL by FAB classification.Conclusions With the multi-color flow cytometry,every acute leukemia patient was diagnosed accurately,especially on acute mixed leukemia and cross-lineage expression leukemia.Those help us choice the best treatment protocol for childhood AL.
3.Determination of Seven Antibiotics in Cosmetics by Ultra Performance Liquid Chromatography

Objective To establish an ultra performance liquid chromatography method for the determination of 7 kinds of antibiotics(minocycline hydrochloride,ox tetracycline,tetracycline hydrochloride,chlortetracycline hydrochloride,doxycycline hydrochloride,chloramphenicol,metronidazole) in anti-acne cosmetics.Methods Seven kinds of antibiotics were separated on a Waters UPLC BEH C18 column by gradient elution using A(methanol/acetonitrile = 1/2) and B(1% formic acid aqueous solution) as the mobile phase at a flow rate of 0.2 ml/min,with the detection wavelength of 268 nm.Results The detection limits were 0.3-0.5 ng,the calibration curves were linear in the range of 2-100 ng(1-50 mg/L),the precision was not more than 4.8% and the rate of recovery were 82.5%-105.6%.Conclusion The method established in the present paper is rapid,sensitive and suitable for the determination of 7 kinds of antibiotics in anti-acne cosmetics.
4.Application of Cationic Liposome and Direct Injection Method to Express hGH in Mice in vivo

An eukaryotic expression plasmid Rc/CMV GHcDNA containing CMV late promoter and hGH cDNA was constructed and introduced into mouse skeletal muscle by means of direct injection with or without the cationic liposome (Lipofectin and Lipofectamine reagent) intramuscularly. The expression of the gene was detected through both RT-PCR and IRMA (Immunoradiometric assay) at the level of transcription and protein respectively. The gene expression can be detected even after 90 days of single plasmid injection. Compared with the mice injected with the plasmid itself, the expression levels of mice injected with the plasmid-cationic liposome complex seem to be higher, and the expression period longer. Furthermore, the effect of Lipofectamine seems to be even better than that of Lipofectin. The results indicate that direct intramuscular injection with recombinant expression plasmid or plasmid-cationic liposome complex is a simple, efficient and economical way for foreign gene transfer and expression in experimental mice in vivo.
5.Study of the marker for a neuronavigation system.

UNLABELLEDWith different ratios, Omnipague mixed with cod liver oil and Meglumine Diatrizoate mixed with cod liver oil are compared with the stardard marker under the scanning of CT and MR in order to find the better ratio and materials for the marker. The experimental results show that the performances of both Omnipague mixed with cod liver oil and Meglumine Diatrizoate mixed with cod liver oil are better than the stardard marker.
CONCLUSIONOmnipague mixed with cod liver oil and Meglumine Diatrizoate mixed with cod liver oil can be used in making marker for neuronavigation system.

8. Hyaluronic acid derivative-modified nano-structured lipid carrier for cancer targeting and therapy

To reduce the problems of poor solubility, high in vivo dosage requirement, and weak targeting ability of paclitaxel (PTX), a hyaluronic acid-octadecylamine (HA-ODA)-modified nano-structured lipid carrier (HA-NLC) was constructed. HA-ODA conjugates were synthesized by an amide reaction between HA and ODA. The hydrophobic chain of HA-ODA can be embedded in the lipid core of the NLC to obtain HA-NLC. The HA-NLC displayed strong internalization in cluster determinant 44 (CD44) highly expressed MCF-7 cells, and endocytosis mediated by the CD44 receptor was involved. The HA-NLC had an encapsulation efficiency of PTX of 72.0%. The cytotoxicity of the PTX-loaded nanoparticle HA-NLC/PTX in MCF-7 cells was much stronger than that of the commercial preparation Taxol®. In vivo, the HA-NLC exhibited strong tumor targeting ability. The distribution of the NLCs to the liver and spleen was reduced after HA modification, while more nanoparticles were aggregated to the tumor site. Our results suggest that HA-NLC has excellent properties as a nano drug carrier and potential for in vivo targeting.
9. Clinical investigation of CT-guided radiofrequency ablation for liver cancer in hepatic dome area

Objective: To investigate the technical procedure, clinical efficacy and safety of extrathoracic and transhepatic CT-guided radiofrequency ablation (CT-RFA) for liver cancer in hepatic dome area. Methods: A retrospective analysis was conducted in 50 patients who were diagnosed of primary liver cancer or metastatic liver cancer and received CT-RFA between September 2003 and October 2012. The total number of the malignant lesions in the hepatic dome area were 59. The detailed technical procedure, clinical efficacy and the safety as well as the factors related to local recurrence-free survival were anaylized and summarized. Results: The pathway for needle puncture of RT-RFA in treatment of the malignant lesions in the hepatic dome area was established by using a new "cube" model. The rates of technical success in RFA and complete necrosis were 94.92% (56/59) and 84.75% (50/59), respectively. The median local recurrence-free survival time of patients with completely necrotic lesions was 1 2.05 months, which for primary liver cancer and metastatic liver cancer were 14.23 and 8.07 months, respectively (P = 0.037). Three patients developed severe complications after RFA, and recovered after supportive treatment. The multivariable analysis revealed that the tumor size was an independent factor related to the local recurrence-free survival (P = 0.028). Conclusion: CT-RFA for the treatment of malignant lesions in the hepatic dome area is safe and effective. The tumor size is an independent risk factor related to the local recurrence-free survival. 10.Effect of Zhuyun Capsule on Expression of Ovarian Cytokine of Polycystic Ovary Syndrome Rats

Objective To observe the effect of Zhuyun Capsule(Pregnant-promoting Capsule)on serum sex hormone and expres- sion of ovarian cytokine of polycystic ovary syndrome(PCOS)rats.Methods Progestogen plus human chorionic gonadotrophin (HCG)was adopted to make PCOS rat models.Totally 40 SD rats were randomized into control group,PCOS model group,Zhuyun Capsule group,and clomiphene citrate group with 10 in each.Radioimmune assay(RIA)was used to measure the sex hormone and immunohistochemical test was done to measure the expression of tumor necrosis factor-?(TNF-?)of ovarian tissue,insulin-like growth factors-1(IGF-1),and leukemia inhibitory factor(LIF)to analyze the relations among these factors.Results Zhuyun Capsule could reduce greatly serum testosterone(T)and luteinizing hormone(LH),decrease expression of TNF-?and IGF-1,but increase expres- sion of LIF.Conclusion Zhuyun Capsule could regulate endocrine function and expression of ovarian cytokine to improve gonad func- tions of PCOS rats and to promote the maturation of follicle and ovulation.
